        축·수산·전통식품 중 황색포도상 구균의 정량적 분석을 위한 Petrifilm<sup>TM</sup> Staph Express Count Plate의 성능 평가

        본 연구는 식품으로부터 S. aureus를 분리하는 데 사용되는 선택배지들의 성능을 평가하였다. 먼저 BPA와 MSEY agar의 민감도 비교를 통해 BPA를 선택하였고, 이 배지의 회수율을 $3M^{TM}$ $Petrifilm^{TM}$ Staph Express Count Plate(STX petrifilm)의 회수율과 비교하였다. 축산식품 중 돼지고기 주물럭과 육회용 꾸릿살, 수산식품 중 쥐포와 반건조 오징어, 전통식품 중 떡과 잡채를 선정하여 S. aureus를 접종한 후 BPA와 STX Petrifilm 배지를 사용하여 회수율 분석 실험을 실시하였다. 6가지 식품에 대해 인위적으로 S. aureus를 저농도에서부터 고농도까지 접종한 후 0시간과 24시간 후의 검출률을 측정하였다. 두 배지 간의 유의 수준을 분석한 결과 식품의 종류, 접종 농도에 관계없이 두 선택배지 간에 통계적 유의차는 없는 것으로 확인되었다(P>0.05). 따라서 STX Petrifilm은 BPA를 대체할 수 있을 뿐만 아니라 배지의 준비 및 도말에 소요되는 분석시간을 절감하고 배양에도 최소한의 공간이 필요하게 되어 신속, 정확한 분석에 크게 기여할 수 있을 것으로 판단된다. To enumerate Staphylococcus aureus in food, Baird-Parker Agar (BPA) is usually used in the conventional method, However it requires time and space for the preparation and plating, and incubation. Thus, use of the $3M^{TM}$ $Petrifilm^{TM}$ Staph Express Count Plate (STX Petrifilm) might be appropriate to solve these challenging problems. The purpose of this study was to compare the efficiency of STX Petrifilm with BPA for enumeration of S. aureus in various foods. A mixture of S. aureus strains ATCC29213, ATCC25923, and ATCC13565 was inoculated on marinated pork chop, beef (chuck tender), dried filefish, semi-dried squid, rice cake, and Japchae (stir-fried glass noodles) at 2, 3, 5, and 7 Log CFU/g. S. aureus cell counts were enumerated by spread-plating on STX Petrifilm and BPA after 0 and 24 hours at $4^{\circ}C$ (marinated pork chop, beef, semi-dried squid, and stir-fried glass noodles) and $25^{\circ}C$ (dried filefish and rice cake). Recovery of STX Petrifilm for S. aureus from various food samples was compared with BPA, and the results showed that there were no significant differences between two selective media in all cases. The results indicated that STX Petrifilm had enough efficiency to recover S. aureus from various foods as well as saving time and space.

        Six cases of congenital coronary arteriovenous fistulae were diagnosed radiographically and confirmed after operation at Seoul National University Hospital in recent three and a quarter years since 1979. The clinical and radiological findings with the emphasis on the cinecardiographic findings were analysed. The summaries of the analysis are as follows: 1. Among 6 cases, 2 cases were male and 4 cases were female. Age distribution was from 3 months to 19 years. 2. In chest X-ray findings, 5 cases had cardiomegaly. Among these 5 cases, increase of pulmonary vascularity was in 3 cases. Chest X-ray finding of the rest 1 case was within normal limit. 3. The coronary arteriovenous fistula orginated from the right coronary artery in 5 cases and from the single left coronary artery in 2 case. In 5 cases, a coronary arteriovenous fistula drained into the right ventricle and in 1 case, a fistula from a SA node branch of the right coronary artery into the right atrium. 4. In 5 cases, a main right coronary artery was dilated. Among these 5 cases, aneurysmal dilatation of main right coronary artery was in 2 cases, saccular aneurysmal dilatation of distal portion of right coronary artery was in 2 cases and in 1 case, diffuse dilatation without aneurysm formation of a main right coronary artery. In 1 case, SA node branch of the right coronary artery was dilated with aneurysm formation. 5. For accurate diagnosis of congenital coronary arteriovenous fistula, biplane cinecardiography is an essential procedure.

        Interleukin-10 이 $interleukin-1{\beta}$로 유도되는 골흡수에 미치는 효과

        The cytokines released by osteoblasts induce bone resorption via the differentiation of osteoclast precursors. In this process, $interleukin-1{\beta}$($IL-1{\beta}$)-induced bone resorption is mediated by granulocyte macrophage-colony stimulation factor(GM-CSF), interleukin-6 (IL-6), and tumor necrosis factor ${\alpha}$($TNF-{\alpha}$) released from osteoblasts. Since these cytokines (GM-CSF, IL-6, $TNF-{\alpha}$) are produced by not only osteoblasts but also monocytes, and interleukin-10(I1-10) inhibits the secretion of these cytokines from monocytes, it may be speculated that IL 10 could modulate the production of GM-CSF, IL-6, and $TNF-{\alpha}$ by osteoblasts, then control $IL-1{\beta}-induced$ bone resorption. Therefore, the aims of the present study were to examine the effects of IL-10 on bone resorption. The sixten or seventeen-day pregnant ICR mice were injected with $^{45}Ca$ and sacrificed one day after injection. Then fetal mouse calvaria prelabeled with $^{45}Ca$ were dissected out. In order to confirm the degree of bone resorption, mouse calvaria were treated with Lipopolysaccharide(LPS), $TNF-{\alpha}$, $IL-1{\alpha}$, IL-8, $IL-1{\beta}$, and $IL-1{\alpha}$, Then, IL-10 and $interferon-{\gamma}$ ($IFN-{\gamma}$) were added to calvarial medium, in an attempt to evaluate the effect of $IL-1{\beta}-induced$ bone resorption. In addition, osteoclasts formation in bone marrow cell cultures, and the concentration of IL-6, $TNF-{\alpha}$, and GM-CSF produced from mouse calvarial cells were investigated in response to $IL-1{\beta}$ alone and simultaneously adding f $IL-1{\beta}$ and IL-10. The degree of bone resorption was expressed as the ratio of $^{45}Ca$ release(the treated/the control). The osteoclasts in bone marrow cultures were indentified by tartrate resistant acid phosphatase(TRAP) stain and the concentration of the cytokines was quantified using enzyme linked immunosorbent method. As results of these studies, bone resorption was induced by LPS(1 ng/ml ; the ratio of $^{45}Ca$ release, $1.14{\pm}0.07$). Also $IL-1{\beta}$(1 ng/ml), $IL-1{\alpha}$(1 ng/ml), and $TNF-{\alpha}$(1 ng/ml) resulted in bone resorption(the rations of $^{45}Ca$ release, $1.61{\pm}0.26$, $1.77{\pm}0.03$, $1.20{\pm}0.15$ respectively), but IL-8 did not(the ratio of $^{45}Ca$ release, $0.93{\pm}0.21$). The ratios of $^{45}Ca$ release in response to IL-10(400 ng/ml) and $IFN-{\gamma}$(100 ng/ml) were $1.24{\pm}0.12$ and $1.08{\pm}0.04$ respectively, hence these cytokines inhibited $IL-1{\beta}$(1 ng/ml)-induced bone resorption(the ratio of $^{45}Ca$ release $1.65{\pm}0.24$). While $IL-1{\beta}$(1 ng/ml) increased the number of TRAP positive multinulcleated cells in bone marrow cultures($20{\pm}11$), simultaneously adding $IL-1{\beta}$(1 ng/ml) and IL-10(400 ng/ml) decreased the number of these cells($2{\pm}2$). Nevertheless, IL-10(400 ng/ml) did not affect the IL-6, GM-CSF, and $TNF-{\alpha}$ secretion from $IL-1{\beta}$(1 ng/ml)-activated mouse calvarial cells. From the above results, it may be suggested that IL-10 inhibites $IL-1{\beta}-induced$ osteoclast differntiation and bone resorption. However, the inhibitory effect of IL-10 on the osteoclast formation seems to be mediated not by the reduction of IL-6, GM-CSF, and $TNF-{\alpha}$ production, but by other mechanisms.

        보편적 학습 설계를 적용한 고등학교 통합사회 수업이 일반, 교과학습부진, 기초학력부진, 지적장애 학생의 학업성취도와 흥미도에 미치는 효과

        The purpose of this study is to investigate the effects of social studies classes based on universal learning design (including graphic organizers) and is based on the academic achievement and interest of students in high school inclusion classes. 77 boys and 111 girls from six classes in the first year of high school participated. The intervention period was from May 2019, right after the first paper-delivered examination, to June 2019; a total of 14 classes were held three times a week. The intervention was conducted by the lead author and the main findings are as follows. There were statistically significant differences in both the academic achievement and interests among all students in this study, showing that social classes based on universal learning design had a positive effect. Results were derived that showed a much higher improvement rating post-test, than what was observed pre-test. Based on these findings, the possibility of including learners with various academic needs for universal learning (design-based instruction) in inclusion classes was discussed. 본 연구는 보편적 학습 설계를 적용한 사회 수업이 고등학교 일반, 교과학습부진, 기초학력부진 학생과 지적장애 학생의 사회과 학업성취도와 흥미도에 미치는 효과를 알아보고자 하였다. 고등학교 1학년 총 6개 학급에서 남자 77명, 여자 111명의 학생들이 참여하였다. 중재 기간은 2019년 1학기 중간고사 직후인 5월부터 1학기 기말고사까지 주 3회, 총 14차시의 수업이 진행되었다. 중재는 연구자가 직접 실시하였으며, 주요 연구결과는 다음과 같다. 일반학생의 학업성취도는 실험-비교집단 간 통계적으로 유의미한 차이가 없었고, 흥미도는 실험집단이 비교집단에 비해 통계적으로 유의미하게 높게 나타냈다. 또한 교과학습부진 학생의 학업성취도와 흥미도는 모두 통계적으로 유의미한 차이가 나타나 보편적 학습 설계를 적용한 통합사회 수업이 긍정적 영향을 미치는 것으로 나타났고, 기초학력부진과 지적장애 학생의 경우도 학업성취도와 흥미도에서 사전에 비해 사후 결과가 훨씬 더 높은 향상을 나타내는 결과가 도출되었다. 이러한 연구결과를 바탕으로 통합학급에서의 보편적 학습 설계를 적용한 수업에 대한 다양한 학업적 요구를 지닌 학습자를 포괄할 수 있는 가능성에 대해 논의하였다.
