        생육온도에 따른 느타리류의 갓색, 수량 및 형태적 특성 변화

        To elucidate how cultivation temperature affected various traits including pileus color, yield and morphology of Pleurotusspp. Main results were as follows. Pileus lightness of all cultivars of Pleurotustested became higher as cultivation temperature increased, while those of Santari, Hwang-geumsantari and Sunjung at 21oC were lower than at 18oC. Redness and yellowness of pileus decreased as cultivation temperature increased; those of chromatic pileus cultivars showed noticeable difference. Yellowness of cultivar with chromatic pileus was higher than that of cultivar with achromatic pileus. Yield was increased as cultivation temperature increased, Wonhyeung 1ho; low temperature favored cultivar showed high yield when it was cultivated at low temperature andno fruiting body at 21oC. Valid number of stipes were generally higher at 18oC, and its correlation coefficient with yield was low. Length and stipe thickness changed consistently (larger and thicker) upon cultivation temperature; the coefficient of determination(R2) 0.514 for lengthof Heuktari and 0.963for stipe thickness of Santari were high. Correlation coefficient of one trait was highly related with multiple traits. In the future, we will conduct research on the changes of expressed genes involved in the pigments for pileus color by RNA expression analysis.

      • 산느타리버섯의 품종별 재배특성

        국내에서 육성된 산느타리버섯 8품종을 병재배 방식으로 재배특성을 조사하였다. 8품종의 자실체 형태는 전체적으로 갓직경이 크고 대는 짧은 형태였다. 대는 편심생으로 갓에 결합되어 있다, 자실체의 길이가 가장 긴 품종은 여름2호(72.8mm)이고 가장 짧은 품종은 여름1호(33.8mm)이다. 대의 두께는 여름2호, 호산의 순서로 두겁고 15.5mm, 12.8mm 였다. 갓의 직경은 45.8-62,3mm로 약산이 가장 작고 여름2호가 가장 큰 갓을 지닌 것으로 나타났다. 전체 무게(수확량)은 여름2호가 80.7g으로 가장 많은 생산량을 보였고 여름1호가 35.5g으로 가장 낮은 수치를 보였다. 가장 갓색이 밝은 품종은 여름2호로 55.6이었고 호산도 54.1이었다. 특징적으로 갓색이 가운데 부분은 엷은 편이었으며 가장자리가 더 진하게 착색되었다. 평균적 발이소요일과 생육소요일은 각각 4.6일과 8.0일이어서 발이 후 3.4일 만에 수확을 완료하였다. Pleurotus pulmonarius 8 cultivas, domestically bred, were cultivated and evaluated for cultural properties. The morphological properties of 8 cultivars were big diameter of pileus and short stipe. Stipe was eccentrically combined with pileus. The longest length of stipe was Yeoleumneutali 2ho(72.8mm) while shortest cultivar was Yeoleumneutali 1ho(33.8mm). The diameter of pileus ranged 45.8-62,3mm, Yaksan was the smallest one and Yeoleumneutali 2ho was largest cultivar. Yield was largest in Yeoleumneutali 2ho(80.7g) whereas smallest was in Yeoleumneutali 1ho(35.5g). The highest L value was in Yeoleumneutali 2ho and Hosan. Lightness was not even around pileus but thicker in rim area and thinner in center of pileus. Average period of pinheading and harvest were 4.6 and 8.0 days thus only 3-4 days after pinheadng were required for harvest.

        부위별 흰색 느티만가닥버섯 추출물의 항산화 활성

        본 연구에서는 천연물 유래 항산화 소재로서 흰색 느티만가닥버섯(Hypsizygus marmoreus)의 부위별 이용 가능성을 알아보기 위하여 흰색 느티만가닥버섯을 갓(pileus)과 대(stipe)로 분리하여 열수와 80% 에탄올에 침지한 다음 부위별 항산화 활성을 조사하였다. 흰색 느티만가닥버섯은 에탄올 추출물(갓 부위 923.47±0.18 mg GAE/100g, 대 부위 324.05±0.03 mg GAE/100 g)에 비해 열수 추출물(갓 부위 1137.39±0.38 mg GAE/100 g, 대 부위 700.86±0.06 mg GAE/100 g)의 총 폴리페놀 함량이 높게 나타났고 대 부위보다는 갓 부위의 총 폴리페놀 함량이 높게 나타났다. DPPH 라디칼 소거 활성은 10 mg/ml의 농도에서 에탄올 추출물 갓 부위의 활성이 47.32±0.23%로 가장 높았으며 ABTS 라디칼 소거 활성도 10 mg/ml의 농도에서 에탄올 추출물 갓 부위의 활성이 57.33±0.10%로 가장 높았다. 환원력과 ORAC 지수는 에탄올 추출물에 비해 열수 추출물이 높게 나타났고 환원력은 갓 부위와 대 부위에 큰 차이가 없었으나 ORAC 지수는 대 부위보다는 갓 부위가 높게 나타났다. 흰색 느티만가닥버섯 추출물의 세포 독성은 WST-1 assay를 이용하여 추출물이 RAW 264.7 세포에 미치는 영향으로 평가하였다. 이상의 결과를 종합해보면, 흰색 느티만가닥버섯의 총 폴리페놀 함량과 항산화 활성은 추출물의 농도가 증가함에 따라 증가하는 경향을 나타내었으며 대 부위보다는 갓 부위의 항산화 활성이 우수하였다. Hypsizygus marmoreus (white cultivar), also called white beech mushrooms, are edible mushrooms commercially cultivated in Korea and Japan. This study was carried out to evaluate the antioxidant properties of H. marmoreus. H. marmoreus fruit bodies were divided into pileus and stipe. The pileus and stipe were extracted into water and 80% ethanol and their antioxidant activities were analyzed. The total polyphenol content was highest in the water extract (pileus 1137.39±0.38 mg of GAE (gallic acid equivalents)/100 g, stipe 700.86±0.06 mg of GAE/100 g) compared to the ethanol extract (pileus 923.47±0.18 mg of GAE/ 100 g, stipe 324.05±0.03 mg of GAE/100 g). Ethanol extracts from pileus showed better scavenging ability on DPPH (47.32±0.23% at 10 mg/ml) and ABTS (57.33±0.10% at 10 mg/ml) than the stipe and water extract groups. Water extract from pileus were more effective in reducing power and ORAC (oxygen radical absorbance capacity) value than stipe and ethanol extract. The toxicity of water and ethanol extracts was investigated using WST-1 (Water Soluble Tetrazolium salt) assay on the mouse macrophage cell line RAW 264.7. Overall, total polyphenol content and antioxidant activities of extracts from H. marmoreus increased in a dose dependent manner while pileus was showed better total polyphenol content and antioxidant activities than stipe.

        갈색 느티만가닥버섯 추출물의 부위별 항산화 활성

        본 연구에서는 기능성 식의약품 소재로써 갈색 느티만가닥버섯(Hypsizygus marmoreus)의 이용 가능성을 조사하기 위해 갈색 느티만가닥버섯의 부위별, 추출 용매별 항산화 활성을 조사하였다. 열수 추출물 갓과 대의 총 폴리페놀 함량은 17.15±0.19 mg GAE/g과 7.37±0.16 mg GAE/g이었으며 에탄올 추출물 갓과 대의 총 폴리페놀 함량은 각각 10.23±0.14 mg GAE/g과 3.76.±0.19 mg GAE/g으로 에탄올 추출물에 비해 열수 추출물의 폴리페놀 함량이 높게 나타났고 대에 비해 갓의 폴리페놀 함량이 높게 나타났다. 또한 갈색 느티만가닥버섯 추출물의 DPPH, ABTS, ORAC 지수, 환원력도 10 mg/ml의 농도에서 에탄올 추출물에 비해 열수 추출물에서 높게 나타났고 대에 비해 갓에서 높게 나타났으며 흰색 느티만가닥버섯에 비해 갈색 느티만가닥버섯의 총 폴리페놀 함량, DPPH, ORAC 지수, 환원력이 높은 것으로 나타났다. 추출물의 세포독성은 WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate)를 이용하여 열수 추출물의 처리 농도에 따른 대식세포주 RAW 264.7의 세포 생존율로 확인하였으며 갈색 느티만가닥버섯 열수 추출물 처리구에서 대식세포주 RAW 264.7의 세포 생존율이 증가하였기 때문에 세포독성은 없는 것으로 판단된다. 이상의 결과를 종합하면, 느티만가닥버섯은 품종에 따라 항산화 활성에 차이가 있고, 갈색 느티만가닥버섯은 다른 버섯보다 항산화 활성이 높기 때문에 기능성 식의약품 소재로서 이용 가능성이 있다고 판단된다. This study was carried out to evaluate potential of Hypsizygus marmoreus (brown cultivar) as a functional food and drug materials. H. marmoreus were divided into pileus and stipe and extracted in hot water and 80% ethanol. The total polyphenol content was highest in the hot water extracts (pileus 17.15±0.19 mg of GAE g, stipe 7.37±0.16 mg of GAE/g) and pileus compared to the ethanol extracts (pileus 10.23±0.14 mg of GAE/g, stipe 3.76.±0.19 mg of GAE/g) and stipe. Also, hot water extracts of pileus from H. marmoreus (brown cultivar) was more effective DPPH, ABTS, ORAC value, reducing power than ethanol extracts and stipe extracts. The pileus and stipe extracts were confirmed to be non-cytotoxic in the mouse macrophage cell line RAW 264.7 determined by WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) assay. Overall, extracts of H. marmoreus (brown cultivar) was higher antioxidant activity than other mushrooms, and no cytotoxicity. Therefore, H. marmoreus (brown cultivar) showed potential as a functional food and drug materials. The brown cultivar of H. marmoreus have higher antioxidant activity than white cultivar, H. marmoreus seem to have different antioxidant activity depending on the cultivar.

        Changes of Methyl trans-cinnamate Levels During Fruit-body Development in Tricholoma matsutake

        The relationship between the contents of methyl trans-cinnamate and the ratio of DP/DS (diameter of pileus / diameter of stalk) in the fruit-body of Tricholoma matsutake during its development was investigated. The stages of development were divided as follows: stage A is less than 1, stage B is from 1 to less than 2, stage C is from 2 to less than 3, and stage D is more than 3 of the values of DP/DS. The contents of methyl trans-cinnamate in pileus and stalk of pine mushroom ranged from 77 mg/g to 824 mg/g and from 7.6 mg/g to 22.4 mg/g, respectively during its development. In the part of pileus, there is no relevance of the methyl trans-cinnamate content of pine mushroom between the stage A and B, but there was significantly different among the stage of B, C and D. In the case of stalk, the relevance of the methyl trans-cinnamate content of pine mushroom between stage D and other stages showed a low difference. In addition, as pileus of pine mushroom developed the level of the aroma compound increased as well and showed higher correlation relationship (r2=0.877) between the contents of methyl trans-cinnamate in the pileus and the ratio of DP/DS. From the results of this study, we can conclude that the aromatic component of pine mushroom can be deduced from the value of DP/DS, which indicates the stage of the development appearance.

        HPLC를 이용한 한국산 송이의 methyl trans-cinnamate 함량 분석

        The contents of methyl trans-cinnamate, a significant and major aromatic compound in Tricholoma matsutake (pine mushroom), were determined by HPLC (High Performance Liquid Chromatography.). Mushroom sample collected from five sites in Korea were extracted with methanol by ultrasonication. The contents of methyl trans-cinnamate in the pileus and stalk of pine mushroom ranged from 84.8 to 995.7 μg/g and from 4.5 to 45.2 μg/g, respectively. The average contents of methyl trans-cinnamate in the pileus and stalk of pine mushroom from the five sites were 559.1 μg/g and 35.7 μg/g, respectively. From the results of this study, we could conclude that the contents of methly trans-cinnamate varied by different parts (pileus and stalk) and collecting sites. The amounts of methyl trans-cinnamate from tresh T. matsutake were higher than those of dried T. matsutake, Pileus of pine mushroom appeared to contain more methyl trans-cinnamate than did stalk. Additionally, as pileus of pine mushroom developed, the aroma content increased as well.

        병재배용 백색느타리 신품종『백선』의 균사배양 및 생육특성

        Oyster mushrooms are an economically important crop, accounting for 35% of the total mushroom production in Korea. In this study, we developed a new cultivar of Pleurotus ostreatus, known as ‘Baekseon,’ which is characterized by a white pileus with a white stipe. It was bred by mating monokaryons isolated from white mutant oyster mushrooms that were naturally generated from ‘Gonji-7ho’ and ‘Wonhyeong-1ho’ at the Mushroom Research Institute, GARES, Korea in 2018. The optimum temperature for mycelial growth on potato dextrose agar medium was approximately 28-31 °C, and the optimum temperatures for primordia formation and growth of fruit bodies on sawdust media were 22 °C and 20 °C, respectively. The time required for the bottle-cultured mushrooms to complete spawn running, primordia formation, and growth of fruit bodies was 30 days, 4 days, and 4 days, respectively. The fruit bodies were bundle-shaped, the pilei were round type and white, and the stipes were white. The stipes were slender and longer than those of the control (‘Miso’). In the productivity test, the yield per bottle was 185 g/1100 mL, which was 45% greater than that of the control (‘Miso’). In the farm test, the yield per bottle for Farm A (Pyeongtaek) and Farm B (Yeoju) was 184 g/1100 mL and 178 g/850 mL, respectively. With regard to the physical properties of fruit bodies, the springiness, cohesiveness, gumminess, and brittleness of stipe tissue were 80%, 57%, 720 g, and 57 kg, respectively. These values were lower than those of the control (‘Miso’). To test the shelf life, the fruit bodies were wrapped with antifogging film and stored at 4 °C for 28 days and then at room temperature for 4 days; such conditions were sufficient for maintaining edibility.
