        정향 및 목향과 백지를 혼합한 한약재 에탄올 추출물의 항산화 효능

        Objectives : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix. Each of herbal medicines has studied on anti-oxidant effect. So this study was conducted to investigate efficacy and potency of JMB on anti-oxidation. Methods : The JMB was extracted at room temperature by 80% ethanol. And total polyphenol contents, total flavonoid contents in JMB ethanol extract were determined by colorimetric method. Also, DPPH, ABTS free radical scavenging capacity and reducing power of JMB ethanol extract were measured at 100, 500, 1000, 5000 ㎍/㎖ concentrations by spectrometric assay. Positive control was used BHA (butylated hydroxyanisole). Results : The total polyphenol contents and total flavonoid contents of the extract were 55.38 ㎎/TAEg, 513.72 ㎎ /RUEg, respectively. Also, DPPH free radical scavenging capacity and reducing power of JMB ethanol extract in treated concentrations (100, 500, 1000, 5000 ㎍/㎖) increased dose dependently. In particular, DPPH free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ㎍/㎖ was similar to positive control (BHA) at high concentration (50, 100 ㎍/㎖). ABTS free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ug/ml was similar to BHA at high concentration (50, 100 ㎍/㎖). Also, reducing power was showed that JMB ethanol extract at 5000 ㎍/㎖ was similar to BHA at high concentration (50, 100 ㎍/㎖). Conclusions : These results suggest that JMB ethanol extract has effects to scavenge free radicals. Therefore, JMB has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

        차전초 에탄올 추출물의 항산화 효능

        Objectives : Butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA) were well known as anti-oxidant, but they were limited to use because of toxicity. So, many studies are being done to develope natural anti-oxidant. Total phenolic and flavonoid contents along with total antioxidant capacity of the ethanolic extract of Plantaginis Herba (PH) were evaluated to explore the reliable and potential sources of novel natural antioxidants. Methods : Total polyphenol contents and total flavonoid contents in PH ethanol extract were determined by colorimetric method. And DPPH(1.1-diphenyl-2-picrylhydrazyl), ABTS(2'-azino-bis (3-ethylbenzothiazoline-6-Surfonicacid)) free radical scavenging capacity and reducing power inhibition activities of PH ethanol extract were measured at 100, 500, 1000, 5000 ㎍/㎖ concentrations by spectrometric assay. Results : The total polyphenol contents and total flavonoid contents of the extract were 161.99 ㎎/g, 144.05 ㎎/g, respectively. Also, DPPH, ABTS free radical scavenging capacity and reducing power of PH ethanol extract in treated concentrations (100, 500, 1000, 5000 ㎍/㎖) increased dose dependently. In particular, DPPH free radical scavenging capacity of PH ethanol extract from 500 ㎍/㎖ was significantly increased compared to positive control (BHA). ABTS free radical scavenging capacity of PH ethanol extract from 1000 ㎍/㎖ was significantly higher than BHA. Also, reducing power showed that PH ethanol extract from 500 ㎍/㎖ was significantly increased compared to BHA. Conclusions : These results suggest that PH ethanol extract has effects to scavenge free radicals, thus PH has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

        Much attention has been focused on the activity of the natural antioxidants present in fruits and vegetables, because potentially these components may reduce the level of oxidative stress. Especially, mulberry leaves containing many natural components are considerable resource for natural antioxidants. The antioxidant capacity of mulberry leaves was investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The antioxidant capacity of 16 varieties was 3303.4 nmol at opening stage of five leaves in spring. The highest stage of antioxidant capacity (3708.0 nmol) and yield rate was just before the coloration stage with anthocyanin in fruits, whereas the lowest stage was middle of June (2231.6 nmol) and about two months growing stage after summer pruning (2064.6 nmol). But after summer pruning, the antioxidant capacity of mulberry leaves increased gradually until just before fallen leaves stage. Even if samples were same variety, antioxidant effect of those showed different results according to collected regions. Also, antioxidant effect of mulberry leaves were higher than that of branches. The antioxidant capacity of yield-type mulberry leaves and fruits (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from In-je, Won-ju and Yang-yang regions, Kang-won province, Korea, was investigated. The results indicated that total antioxidant capacity of yield-type mulberry leaves was 2711.2 nmol. In the antioxidant capacity analysis of Jeollabuk-Do genetic resources, autumn’s mulberry leaves showed higher antioxidant capacity than that of spring’s it. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their’s antioxidative capacity were 2,531.01 nmol, 1,867.42 nmol, 1,053.72 nmol, 292.71 nmol and 188.91 nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96 nmol.

        Long-Term Intake of High Doses of Vitamin C Down-regulates Anti-oxidant Enzymes in Human Erythrocytes

        We located a group of healthy young males (aged 20~30) who had been taking a high dose (more than 5 g) of vitamin C daily for more than one year. We observed that this vitamin C group had plasma levels of vitamin C that were more than three times that of the control group. The control group had not taken any additional vitamin C except for that included in their diets. But the vitamin C group showed significantly lower amounts of Cu/ZnSOD, catalase and glutathione-s-transferase and lower activities of glutathione peroxidase and glutathione reductase in erythrocyte lysates than the control group. However, there was no difference in the plasma levels of lipid peroxides between the two groups. These results suggest that vitamin C offsets its own contribution to anti-oxidant activity by repressing the expression of anti-oxidant enzymes and also excludes the possibility that vitamin C acts as a pro-oxidant in vivo.

        Objectives : The purpose of this study is to prove the effect and mechanism of Gamikyejakjimogawusul-tang(GKHA) herbal acupuncture on induced rheumatoid arthritis model of DBA/1 mice. Methods : We check effect of GKHA extract on the AST, ALT, Creatinine, BUN of serum and cell viability of GK extract in RAW 264.7 cells to test the stability of this study. In vitro, we measure total phenol contents, total flavonoid contents, DPPH free radical scavenging activity, ABTS cation radical scavenging activity of Gamikyejakjimogawusul-tang, effect of GK extract on ROS(Reactive Ooxygen Species) production to estimate a anti-oxidant capacity, and we also measure effect of GK extract on NO (Nitric Oxid), IL-1β, IL-6, IL-17, IL-21, TNF-α, MCP-1, GM-CSF production in RAW 264.7 cells to estimate a anti-inflammatory efficacy. In vivo, we compare a rheumatoid arthritis manifestation between control and experimental group and estimate a AI. Then we check effect of GKHA on the level of WBC, neutrophil, lympocyte, monocyte in the blood to see the effect of immune cells in blood. In addition we measure effect of GKHA on the level of hs-CRP, IgM, IgG, IL-1β, IL-6, IL-17, IL-21, TNF-α, MCP-1, GM-CSF in serum. We observe effects of GKHA on imaging of cartilage degeneration using micro CT-arthrography in paw hind. And we calculate effects of GKHA that reduced BV ratio, BS/BV ratio using 3D Micro-CT. Lastly we observe effects of GKHA histopathologic examination analysis. Results : 1. The toxicity on liver and kidney was disregardable and the cytotoxicity against RAW 264.7 cells was also disregardable. < In vitro > 1. Total phenol contents and total flavonoid contents in GK extract were in high level. 2. DPPH free radical scavenging activity and ABTS cation radical scavenging activity were increased according to concentration of GK extract 3. ROS production was significantly decreased in GK extract (at 10, 100 ㎍/ml). 4. NO, IL-6, TNF-α, MCP-1 production were significantly decreased in GK extract(at 10, 100 ㎍/ml). IL-17, GM-CSF production were significantly decreased in GK extract(at 1, 10, 100 ㎍/ml). IL-1β, IL-21 production were also decreased but there was no statistical significance. 5. 25x observation after H&E and M-T staining, infiltration of immune cells and subsidence of the cartilage and damage to the synovial cells were decreased. Conclusions : This study showed that GKHA extract had anti-oxidant capacity, anti-inflammatory efficacy. GKHA extract also had inhibiting effect on the process of rheumatoid arthritis and can protect joint and cartilage. So we expect that GKHA extract can be a meaningful treatment to rheumatoid arthritis patients.

        Evaluation of the Antioxidant Activity of the Fruiting Body of Phellinus linteus Using the On-line HPLC-DPPH Method

        Phellinus linteus has traditionally been used as a medicinal mushroom in Oriental countries. Bioactive phytochemicals from the fruiting body of P. linteus were investigated using the on-line HPLC-DPPH system. The system was used to separate 10 antioxidants: protocatechuic acid (1) protocatechualdehyde, (2) caffeic acid (3), ellagic acid (4), hispidin (5), davalliallactone (6), interfungins A (7), hypholomine B (8), inoscavin A (9), and methyldavallialactone (10). Of these, compounds 6 and 7 showed the strongest inhibitory effect against the DPPH radical and superoxide anion radical-scavenging capacity. On comparison with quercetin (IC50, 44.0 µM), the IC50 value of compounds 6 and 7 for DPPH radical-scavenging capacity were 19.6 µM and 18.5 µM. Moreover, the antioxidative capacity of compound 6 at 40mM was 2.95 nmol Trolox equivalents per mg dry wt. These results indicated that hispidin dimers such as hispidin with hispidin or hispolon had a stronger effect on radical-scavenging capacity than hispidin itself.

        재배방법을 달리한 복령 에탄올추출물의 항산화 효과

        Objectives : Poria cocos Wolf has been widely used in Korean medicine as a medicinal fungus. In this study, we investigated that comparative anti-oxidant effects of ethanol extract from wild Poria cocos (WP) and plastic bag-cultivated Poria cocos (PBP). Methods : Total polyphenol and flavonoid contents in WP and PBP extract were monitored. And DPPH, ABTS, hydroxyl (·OH) free radical scavenging capacity, reducing power and xanthine oxidase inhibition activities of them were determined at 5, 1, 0.5 mg/ml concentrations. Results : Higher total polyphenol contents were found in the PBP extract (52.07±0.6 mg/TAEg) than in the WP extract (28.44±0.26 mg/TAEg). The flavonoid contents in WP and PBP extract were 17.29±0.30 mg/ RUEg, 21.36±0.40 mg/ RUEg, respectively. Also, DPPH, ABTS and ·OH free radical scavenging capacity of PBP extract in treated concentrations (5, 1, 0.5 mg/ml) significantly increased compared to those of WP-treated group. In particular, DPPH, ABTS free radical scavenging capacity and reducing power of PBP extract at 5 mg/ml concentration were similar to positive control (BHA or vit. C). Xanthine oxidase (XO) inhibition rate in both extract increased dose dependently. But it was significantly increased in PBP-treated group, only at 5 mg/ml, compared to those of WP-treated group. Then, their inhibition rate by PBP was similar to positive control (BHA). Conclusions : These results suggest that PBP extract is superior to WP extract in anti-oxidant capacity thus PBP can be applied in variable antioxidative products as a substitute for WP.
