        Pulmonary inflammation caused by silica dioxide nanoparticles in mice via TXNIP/NLRP3 signaling pathway

        Background Silica dioxide nanoparticles (SiONPs) have been used for various medical applications, including therapeutics and imaging, and the use of SiONPs has increased gradually over the years. However, despite an increase in the use of SiONPs, not much is known about mechanism of action of SiONPs and their pulmonary toxicity. Objective The present study investigated the pulmonary toxicity of SiONPs and explored the underlying mechanism of action, primarily focusing on thioredoxin-interacting protein (TXNIP)/NOD-like receptor pyrin domain-containing 3 (NLRP3) in SiONPs-treated mice. We investigated the toxic effects of SiONPs in the lung of BALB/c mice administered 5, 10, and 20 mg/kg SiONPs for 3 days. Results Exposure to SiONPs markedly increased inflammatory cell counts, including those of neutrophils and macrophages, and levels of inflammatory mediators, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α in a dose-dependent manner in the bronchoalveolar lavage fluid. Moreover, the inflammation was verified upon histopathological analysis. In addition, exposure to SiONPs increased the expression of TXNIP in a dose-dependent manner and, in turn, upregulated NLRP3 inflammasome proteins, which subsequently induced IL-1β production. Conclusion Collectively, exposure to SiONPs induced inflammation in the lungs of mice, which resulted in the activation of IL-1β production via the TXNIP-NLRP3 axis. Our results provide useful information on the pulmonary toxicity induced by SiONPs and provide insights into the underlying mechanism of action.

        척추분리증이 동반된 요추 추간판 탈출증

        연구계획 : 척추분리증이 동반되어 있는 추간판탈출증의 수술적 치료에 관한 후 후향적 연구이다. 연구목적 : 본 연구의 목적은 추간판탈출증 환자 중 척추분리증이 동반되어 있는 빈도를 알아보고, 추간판탈출증을 단순히 수핵제거술 만으로 치료한 경우에 단순한 추간판탈출증의 치료에 비하여 예후에 차이가 있는지를 비교하고자 하였다. 대상 및 방법 : 1989년 3월부터 1999년 2월까지 수핵제거술을 시행받은 환자 273명 중 척추분리증이 있으나, 척추불안정이 없고, 1년 이상 추시 관찰이 가능하였던 9명을 대상으로 하였다. 남자가 5명, 여자가 4명이었고, 평균 연령은 42.7세(30세 64세)였고, 평균 추시 기간은 54.6개월(15개월 114개월)이었다. 추간판탈출의 형태, 척추분리증이 있는 부위 등을 조사하였고 환자들의 이환 기간, 증상의 정도 및 양상, 신경학적 소견 등을 술 전 및 추시 시의 결과를 Kim등의 평가기준에 따라 평가하였다. 결과 : 척추분리증이 동반된 추간판탈출증의 비율은 3.7%로 나타났다. 임상적 증상을 보면 동통은 7례(78%)에서 양성이었는데, visual analogue scale에 의한 통증평가의 평균은 술전 2.1(0 8), 최종 2.1(0 5)이었고, 방사등 9례(100%)모두에서 나타났으며 통증평가의 평균은 술전 7.6(4 10)에서 최종 0.8(0 3)로 호전되었다. 하지 직거상 검사상 7례(78%)에서 양성 소견을 보였고 7명 모두에서 증상의 호전이 있었다. 근력의 약화는 6례(67%)에서 관찰되었는데 6례모두 증상의 호전이 있었다. 감각의 저하는 8례(89%)에서 나타났으며 ?M 우 7례는 증상의 호전을 보였다. 심부 건반사의 이상은 5례(56%)에서나타났으며 2례에서 증상의 호전을 보였다. 임상적 결과는 탁월 2명, 우수 6명, 양호가 1명이었다 결론 : 추간판탈출증에서 척추분리증이 동반된 비율은 3.7%로 일반적인 척추분리증의 빈도와 큰 차이는 없었다. 술후 임상증상의 회복율이나 동통지수의 변화를 볼 때 척추분리증이 동반된 추간판탈출증환자들의 수술결과도 일반적으로 추간판탈출증만이 있는 환자를 치료한 결과와 큰 차이가 없는 것으로 보아 척추분리증이 동반되었다고 하여 척추고정술을 고려하는 등 추간판탈출증 자체의 수술방법을 변경할 필요는 없다고 판단된다. Study Design : This is a retrospective study determining the surgical result of lumbar HlVD associated with spondylolysis. Objectives : To analyze the incidence of lumbar HlVD associated with spondylolysis and to compare the results of open discectomy for lumbar HlVD associated with spondylolysis to simple lumbar HlVD. Summary of Literature Review : Lumbar HlVD associated with spondylolysis need be treated by spinal fusion. Materials and Methods : Nine patients(5 males and 4 females) who had lumbar HlVD with spondylolysis, no instability. follow-up period of 1yr were identified out of 273 patients with lumbar HlVD, treated by open discectomy from March 1989 to Feb. 1999. The type of HlVD and level of spondylolysis were evaluated, the clinical symptoms and signs including SLR. motor deficit, sensory deficit, change of DTR and severity of radiating pain were periodically followed up on the predesigned protocoL Results : The incidence of lumbar HlVD associated with spondylolysis is 3.7%. The recovery of back pain was 2.1 to 2.1 by visual analogue scale, radiating pain was 7.6 to 0.8. The recovery rate of SLR was 100%, motor deficit; 100%. sensory deficit: 85%, change of DTR 4O%. The clinical evaluation was excellent(2), good(6). fair(1). Conclusions : According to the recovery rate of the clinical symptoms, the results of open discectomy for lumbar HlVD associated with spondylolysis without spinal instability and simple HIVD was not different. Therefore, we conclude that lumbar HlVD associated with spondylolysis need not be treated by spinal fusion.

        Quartz 자극에 의한 iNOS 발현의 자가조절

        목적 : Rat2 섬유모세포에서 α-quartz에 의해 유도된 NOS의 조절기전을 알아보기 위하여 본 연구가시도 되었다. 방법 : Rat2 세포에 있어서 α-quartz 자극에 의한 NO와 H₂O₂의 분비 및 iNOS의 발현을 관찰하였고, iNOS 억제제인 L-NIL과 H₂O₂ 억제제인Catalase를 전처리하고 마찬가지로 NO와 H₂O₂분비 그리고 iNOS의 단백발현 정도를 western blot-ting을 이용하여 관찰하였다. 결과 : o-quartz는 Rat2 세포에서 iNOS에 의한NO와 H₂O₂의 분비를 유도하였다. L-NIL은 R3t2세포에서 D-quartz에 의한 H₂O₂의 분비와 iNOS발현을 완전히 억제하였다. Catalase로 전처리 하였을 때 o-quartz로 유도된 iNOS에 의하여 생산증가된 H2Oa가 iNOS를 억제하는 autoinhibitorypathway가 차단되어 결과적으로 H₂O₂의 분비와 iNOS의 발현이 증가하였다. 결론 : Rat2 세포에서 α-quartz에 의하여 유도된iNOS는 H₂O₂의 분비를 촉진하고 이 H₂O₂는 iNOS기능을 조절하는 자가조절기전에 중요한 역할을 하는 것으로 생각된다. Objectives : This study was performed in order to investigate the molecular mechanism regulating nitric oxide synthase (NOS) induced by α-quartz in Rat2 fibroblast. Methods : α-quartz-induced nitric Oxide (NO) anti H₂O₂ formation anti α-quarts-induced iNOS protein expression in Rat2 fibroblast were monitored. With iNOS inhibitor (L-N6- (1-iminoethyl) lysine hydrochloride, L-NIL) or antioxidant (catalase), we observed NO and H₂O₂ formation and iNOS protein expression in Rat2 fibroblast stimulated with α-quartz. Results : α-quartz stimulated iNOS-induced NO and H₂O₂ formation in Rat2 fibroblast. L-NIL inhibited H₂O₂ formation and iNOS protein expression by α-quartz in Rat2 fibroblast. Pretreatment with catalase blocked the autoinhibitory pathway of iNOS by iNOS-induced H₂O₂ therefore H₂O₂ and NO production and iNOS protein expression were increased in Rat2 fibrobalst stimulated with α-quartz Conclusion : α-quartz-induced iNOS Stimulated H₂O₂ formation in Rat2 fibroblast. INOS-induced H₂O₂ by α-quartz plays an important role in the autoinhibition pathway for regulating the iNOS function in Rat2 fibroblast

      • 연약지반 성토시 사면의 안정에 미치는 Geotextile의 효과

        Geotextiles have been extensively used in road constructions as effective and economical materials performing several functions such as separation, filtration, drainage, protection against frost break, and as reinforcement to improve the load bearing capacity and stability of road. In this study, a method for the analysis of embankment reinforced with geotextile is studied. Computer program based on the simplified Bishop method is modified in order to investigate quantitively the effect of gotextile on the slope stability of embankment on weak soils. Evaluation of the tensile strength required for the design safety factor is also possible utilizing the computer program in this study.

        Cardiac tumors, especially the primary tumors involving any part of the heart are extremely rare and its relative incidence has been reported to be approximately 0.02%. We report a patient with huge cardiac hemangioma who complained of shortness of breath, general weakness, and dizziness, Imaging study by echocardiography and computed tomography of thorax revealed a huge lobulating mass like a bunch of grapes in the right chamber of heart. The surgical exploration of thorax was performed and a histological diagnosis of spindle cell hemangioma was obtained by microscopy. The patient was treated by surgical resection of the tumor and doing well after surgery. Our experience indicated that prompt diagnosis and treatment of cardiac hemangioma is imperative for patients' prognosis.

      • 줄눈콘크리트포장의 하중응력분포 해석

        This study focused on the development of an alternative stress estimation procedure to instantly calculate the critical stresses bonded concrete pavement. Closed form analysis is commonly used to analyze pavement structures. This type of analysis assumes linearelastic material properties and static loading conditions. The well-known ILLI-SLAB finite element program was used for the analysis. Bonded concrete overlay analyzed the stress distribution, behavior and load carrying capacity under track load is made evaluation standard of bonded concrete overlay. In the study, the following results were derived; The properties of strength is that compress and 3-point bending strength of existing pavement is deteriorated with 184 kg/㎠, 59kg/㎠ but compress and splitting tensile strength of overlay is satisfied with 464 kg/㎠, 45kg/㎠. Load transfers is happen at adjacent slab by interlocking under track load. The stress distribution under interior, corner and edge load is described high loading position surrounding then loading position.

        Teaching materials available for the effective performance of laboratory exp-erimental work in middle school were developed: Physics part ; A measuring apparatus of uniformly accelerated motion and normal force of inclined plane was developed. Chemistry part ; An imploved Eudiometer applicable in measuring reaction mole ratio and usful for a gas generating apparatus was developed. Biology part ; convenient teaching materials observing law of dominance, law of segregation, law of independence, incomplete dominance and dihybrid cross were developed. Earth science part ; A laboratory teaching materials observing formation of wave, current and graded bedding, and identification of sedimentary rocks were developed. Putting the above results of study together, the integrated suggestions were made to the effective development of science education in middle school : 1. The contents of science textbook should be simplified to the direction of the understanding of essential principles and concept. 2. The development and dirtrihbution of various supplementary teaching materials (film, T.P., cassettes film, supplementary textbook, program teaching materials individual learning materials) are required. 3. The experimental laboratory should be equipped with common instruments and equipements. 4. Efforts and time are needed to science teachers, and so remission of sundry duties and arrangement of assistant are hopefully considered. 5. Teacher's research activities should be encouraged by administrative route or relevant institute.

      • 폐주불사를 혼입한 콘크리트의 내구성에 관한 연구

        콘크리트 구조물은 자연적인 혹은 인위적인 온도의 상승, 강하로 인하여 동결-융해 작용을 받게 되어 구조물의 성능저하를 야기시킨다. 현대에 들어서면서 콘크리트의 내구특성을 파악하기 위한 연구적 요구가 증가되고 있다. 따라서 폐주물사 콘크리트를 실제 구조물에 적용하기 위해서는 내구특성에 대한 연구가 반드시 이루어져야 한다. 폐주물사를 혼입한 콘크리트의 내구특성을 파악하기 위한 동결-융해 실험은 폐주물사의 잔골재 치환율, 물-시멘트비, AE제 사용 여부를 주요 변수로 하여 실시하였다. 원형공시체를 제작하여, 동결-융해 시험기에 넣어 -18∼4℃로 급속 동결-융해를 진행시키면서 매 23싸이클마다 동탄성계수를 측정하였다. 실험결과 AE제를 첨가했을 때 물-시멘트비가 적을수록, 폐주물사의 치환율이 클수록 전반적으로 강도가 증가하는 것으로 나타났으며, 물-시멘트비가 증가함에 따라, AE제를 사용한 콘크리트가 동결-융해 저항성이 증가하는 것으로 나타났다. 특히 폐주물사 치환율이 50%일 때가 동결-융해 저항성이 가장 좋은 것으로 나타났으며 그 다음으로 25, 0% 순으로 저항성이 좋은 것으로 나타났다. 따라서 폐주물사 콘크리트가 내구성이 우수한 것으로 나타나 폐주물사를 콘크리트에 재활용 할 수 있는 것으로 나타났다. Concrete structures has been deteriorated by the freezing and thawing due to temperature gap. This study was conducted to evaluate durability of concrete which are increasingly demanded recently. Therefore the research of durability must be executed for application of waste foundry sand concrete real structures. Concrete durability properties incorporating waste foundry sand was performed with the variable of W/C ratio, Sand/Waste foundry sand ratio and Air entrainment-Non air entrainment. Cylinder specimens were made and subjected to freezing and thawing cycle at -18℃ and 4℃. Dynamic modulus of elasticity were evaluated as F/T cycle increase. The results show that strength of concrete is increased the W/C ratio decrease, the Sand/Waste foundry sand ratio increase when the concrete contains AE agent and decreasing W/C ratio and AE concrete makes improved resistance of freezing and thawing improved. Especially, resistance of freezing and thawing is improved by Fine aggregate/Waste foundry sand ratio which is 50%, 25%, 0% in a row. Therefore it is turn out the waste foundry sand could be applied to concrete from the experiment.

      • 양생온도변화에 따른 콘크리트의 강도 예측

        The maturity concept was adopted to predict the strength of concrete, which was subjected to several temperature levels and variable curing conditions. Penetration test and compressive test were conducted to measure the initial and final setting time and the compressible strength of concrete specimen, respectively. Also, the temperature and time were measured at some time intervals for calculating the maturity. The initial and final setting were delayed as the w/c ration increased and curing temperature decreased. The relationships at the relative strength and the equivalent age were proposed at different w/c ratio for the several temperature curing conditions, and these were applied for the variable curing conditions.
