      • 화학공학 전공 학생의 취업을 위한 복합 STC 운영

        우리 영동대학교에서 1998년 이후 지금까지 진행하여 온 특별 주제 학습과정인 STC(Special Topics COurse)프로그램을 기존의 운영틀에서 탈피하여 한층 더 발전시킨 복합STC로 활용하고자 하였으며, 특히 이를 졸업생의 취업 및 진학의 방법으로 최대한 활용하고자 계획·운영하였다. 그 결과 취업 및 진학과 같은 진로 확립은 물론 전공지식의 확대, 자격시험 대비능력의 향상, 학생과 교수간의 유대강화 등과 같은 괄목할 만한 성과를 얻을 수 있었으며, 교수들의 산업체 방문을 통한 산학협동의 계기 마련에도 커다란 성과를 얻었다. We have upgraded the STC(Special Topics Course) program, which has been carried out in our university since 1995, to more structured program so that we could train our chemical engineering students better. This program was geared to help students find better jobs and enter well-recognized graduate schools. Consequently, we had most students successful in job searches as well as entrance exams for graduate school; besides, those who actively participated in our STC program were able to attain better professional knowledge and acquire various professional engineers certificates. On top of that, this program allowed us to build up closer relationships with our students. We also made university-industry cooperation possible through our visits to industries nearby.

        대장균군 검사용 간이 시험지 개발

        대장균군 검사용 간이 시험지는 본 실험실에서 국내 최초로 고안, 개발하였으며 이 간이 시험지법은 현장 검사법의 하나로 대장균군이 내는 succinic acid dehydrogenase 때문에 tetrazolium salt 가 환원되어 적색 반점을 형성하는것을 이용한 방법으로서 이 간이 시험지의 제조는 대체로 종래의 표준 평판법과 거의 동일한 조성의 배지와 시약을 사용하여 여지에 흡착시킨 후, 건조시켜 (60℃) 멸균한 것으로 표준 평판법과 어떤 상관관계가 있는가를 검토하였다. 이 간이 시험지의 제조에서는 bile salt No.3를 deoxycholate로 대체하여 제조 원가를 절감하였고, 또한 일본에서 현재 시판되고 있는 제품과 품질 비교시험을 하여 더 좋은 결과를 얻었으며 종래의 표준 평판법과 비교하였을 때도 오히려 표준 평판법(24-48시간 배양)보다 빠른 시간(16-20시간 배양)내에 판정할 수 있는 이점이 있으며, 표준 평판법에서는 없어서는 안될 배지나 배양 접시, pipette등의 자료및 기구가 일체 필요없고 언제 어디서나 현장에서 직접 시험할 수 있어 매우 간편하며 또한 저렴한 가격으로 제조 할 수 있는 경제성이 높은 이점을 갖고 있다. The objective of this study was to develop a paper strip which could determine E. coli qualitatively and quantitatively in water, wastewater, drinks, or food. This paper strip method was a simple and rapid test method that determine E. coli by visual identification. In this study, nutrient culture media were formulated and characterized for optimum conditions. Paper strips were then prepared by impregnating into the media and dried at 60℃. The test procedure is quite simple to use. The paper strip was dipped into a sample, and excess sample was removed. The strip was then incubated at 37℃ for 16 to 20 hours and the number of colonies on the strip was counted. The color of the colony spots produced by microorganisms varied depending on the media formulation. Violet-red spots were produced by E. coli. The test method was simple, rapid and no special laboratory equipment was necessary for visual identification. Therefore, this test method is applicable to on-site tests such as field tests or home tests. The paper strip method was compared with the standard agar plate method and Japanese commercial product. The method of the economical preparation of test strips was studied for production on industrial scale.

      • Methyl Iinoleate가 mouse 혈청 및 간의 단백질 효소 활성에 미치는 영향

        순수한 methyl linoleate를 mouse에 장기간 경구투여 하였을 때의 생체에 미치는 영향을 구명하기 위하여 methyl linoleate의 일정량을 1일 1회 20일 동안 경구투여하여 혈청 및 간장에서의 단백질 및 효소활성에 미치는 영향을 검토한 결과는 다음과 같다. 실험군의 혈청에서는 정상군에 비하여 TBA값이 증가하였으며, TBA값의 증가에 따라 실험군의 혈청내 acid phosphatase, alkaline phosphatase, glutamic-oxaloacetic transaminase(GOT) 및 glutamic-pyruvic transaminase(GPT)의 활성은 모두 증가하고 있었다. 실험군의 간에서의 acid phosphatase와 alkaline phosphatase의 활성은 증가하였으나, GOT와 GPT의 활성은 감소하는 경향을 나타내었다. 한편, 혈청단백질의 전기영동상은 실험군에서는 정상군에 비하여 albumin과 α₂-globulin은 증가하였고, β-globulin과 r-globulin은 감소하였다. 전기영동 결과 lactic dehydrogenase(LDH) isozyme들의 백분율은 LDH₁은 증가한 반면, LDH₄, ??의 활성은 감소하였다. 이상의 결과로부터 methyl linoleate를 mouse에 장기간 투여하였을 때 혈청 및 간의 단백질 및 효소활성에 크게 영향을 미치고 있음을 알 수 있었다. In order to investigate the effect of pure methyl linoleate fed to the mouse on the protein and enzyme activity in the mouse serum and liver, we adminstered the fixed dosage of methyl linoleate to mice once per day for 20 days by using stomach tube. And the following results were obstaind. TBA value in the serum of test group was increased than those of normal group. The activities of acid phosphatase, alkaline phosphatase, glutamic oxaloacetic transaminase(GOT), and glutamic pyruvic transaminase(GPT) in the serum of test group were increased than those of normal group. The activities of acid phosphatase and alkaline phosphatase in the liver of test group were increased than those of normal group but the activities of GOT and GPT were slightly decreased in the test group. In the serum protein of test group, the amounts of albumin and α₂-globulin were increased, on the other hand the amounts of β-globulin and r-globulin were decreased. According to the electrophoresis, the activity of LDH₁were increased while these of LDH₄and ?? were decreased. From these results we conclude that the pure methyl linoleate fed to mouse influences on the protein and enzyme activity in the mouse serum and liver.

        Risk of Subsequent Events in Patients With Minor Ischemic Stroke or High-Risk Transient Ischemic Attack

        This study aimed to present the prognosis after minor acute ischemic stroke (AIS) or transient ischemic attack (TIA), using a definition of subsequent stroke in accordance with recent clinical trials. In total, 9,506 patients with minor AIS (National Institutes of Health Stroke Scale ≤ 5) or high-risk TIA (acute lesions or ≥ 50% cerebral artery steno-occlusion) admitted between November 2010 and October 2013 were included. The primary outcome was the composite of stroke (progression of initial event or a subsequent event) and all-cause mortality. The cumulative incidence of stroke or death was 11.2% at 1 month, 13.3% at 3 months and 16.7% at 1 year. Incidence rate of stroke or death in the first month was 12.5 per 100 person-months: highest in patients with large artery atherosclerosis (17.0). The risk of subsequent events shortly after a minor AIS or high-risk TIA was substantial, particularly in patients with large artery atherosclerosis.

      • Cdk5 regulates N-cadherin-dependent neuronal migration during cortical development

        <P><B>Abstract</B></P> <P>Cyclin-dependent kinase 5 (Cdk5) controls neuronal migration in the developing cortex when multipolar newborn neurons transform to become bipolar. However, by which mechanisms Cdk5 controls cell adhesion in migrating neurons are not fully understood. In this study, we examined the functional interaction between Cdk5 and N-cadherin (Ncad) in newborn neurons when they undergo the multipolar to bipolar transition in the intermediate zone (IZ). Detailed expression analysis revealed that both Cdk5 and Ncad were present in GFP-electroporated migrating neurons in the IZ. Misexpression of dominant negative Cdk5 into the embryonic brains stalled neuronal locomotion in the lower IZ in which arrested cells were round or multipolar. When Ncad was co-introduced with Cdk5DN, however, cells continue to migrate into the cortical plate (CP) and migrating neurons acquired typical bipolar morphology with a pia-directed leading process. Similarly, downregulation of CDK5 resulted in lesser aggregation ability, reversed by the expression of Ncad <I>in vitro</I>. Down-regulation of activity or protein level of CDK5 did not alter the total amount of NCAD proteins but lowered its surface expression in cells. Lastly, expression of CDK5 and NCAD overlapped in the IZ of the human fetal cortex, indicating that the role of Cdk5 and Ncad in neuronal migration is evolutionarily conserved.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Reduced Cdk5 activity inhibits the multipolar-bipolar transition of migrating neurons. </LI> <LI> Ncad expression rescues migration defects caused by the reduction of Cdk5 activity. </LI> <LI> Downregulation of Cdk5 impairs the membrane localization of Ncad. </LI> <LI> Cdk5 and Ncad are present in developing mouse and human brains. </LI> </UL> </P>

        Properties of Low - Molecular Alginate by Ultrasound

        Response Surface Methodology was applied for optimizing the processing parameters of ultrasound treatment in order to produce low-molecular alginate. The use of ultrasound significantly reduced viscosity of alginate solutions. Suggested parameters of ultrasound treatment for maximum reduction of alginate molecular weight were: specific intensity, 115.81 W/㎠at 20㎑ frequency; treatment time, 35.55 min; temperature, 20.08℃; alginate concentration, 2.5%. Low-molecular alginate obtained by ultrasound had two peaks on Sepharose CL-6B gel filtration. The viscosities of control, fraction I, and fraction II at 0.1% concentration and 25℃ were 3.07, 1.23, and 0.82cps, respectively. Molecular weights of control, fraction I, and fraction II alginates were 336,500, 70,400, and 52,800 daltons, and their solubilities were 3, 6, and 14%, respectively. The lower molecular weight of alginate, the lower the alcohol precipitation and the higher Ca^(2+) ion binding capacities. Heavy metal ion binding capacities of alginates were high in the following order of Pb, Cd, Zn, and Co.

        Optimizing Boiling Condition for the Preparation of Fish Extracts

        The optimum boiling condition was determined for fish extracts by response surface model. Model equations were designed with effect of time (T) and the amount of added water (W) on the level of released free amino acid Based on the high ($gt;0.9) coefficient of determination and low ($lt;0.01) level of significant, those model was approved to be significant. The added water amount of higher regression coefficient (β₂), showed a greater influence on releasing free amino acids than boiling time. The optimum boiling times are 6 hours for crucian carp, 5 hours for bastard halibut, 7 hours for loach and 5 hours for jacopever. The ratio of added water to sample 1 (v/w) could be applied to all fish samples at 100 ± 2℃.

        High Temperature Cooking of Fish Protein Extracts for Plastein Reaction

        High temperature-cooking conditions of cultured fishes(loach, crucian carp, bastard halibut, and jacopever) were optimized by response surface methodology(RSM), and plastein products were prepared using enzymatic hydrolysis. Four models were proposed with regard to effects of time(t), temperature(T), and water/fish meat (w/f) ratio on the amount of 0.3M TCA soluble fractions. The model coefficients were ranged from p<0.0001 for jacopever to p<0.0433 for bastard halibut. Cooking conditions for 60% hydrolysis were optimized at 1) 140℃ except for crucian carp(l36.7℃); 2) 10.08 hours(loach), 7.25 hours(crucian carp), 9.85 hours(ba-stard halibut), and 9.38 hours(iacopever); 3) l:l(w/f) ratio except for the crucian carp(l.l:l). When protein hydrolyzates were employed for the plastein synthesis, optimum plastein-reaction conditions were determined to be pH 9.0 with chymotrypsin for the loach and crucian carp hydrolyzates, pH 9.0 with papain for the bastard halibut hydrolyzate, and pH 11.0 with trypsin for the jacopever hydrolyzate. Plastein reaction could be performed in water at concentration up to 20% (w/f).
