국내 자생하는 야생버섯 추출물의 생리활성 성분을 평가하기 위하여 각지에서 수집된 야생버섯 70% 에탄올추출물에 대한 DPPH 라디컬 소거능, 총 폴리페놀 및 총 플라보노이드 함량, 철 환원 항산화능, 환원력의 항산화 활성, 아질산염 소거능 및 야생버섯의 건조시료를 이용하여 베타글루칸 함량을 분석하였다. 본 연구에서 수집된 10종의 야생버섯류 중에서 영지(OK1362) 에탄올추출물의 DPPH 라디컬 소거능(73.2%), 총 폴리페놀 함량(28.9 mg GAE/g) 및 총 플라보노이드 함량(10.0 mg QE/g)철 환원 항산화능(0.134), 환원력(0.155), 아질산염 소거능(56.3%)이 다른 버섯류에 비하여 유의적으로 가장 높았다. 그 이외에도 아까시흰구멍버섯(OK1360), 광대버섯속의 A. lanigera (OK1398), 졸각무당버섯(OK1406)이 높은 항산화능 및 아질산염 소거능을 가지고 있음을 확인하였다. 또한 베타글루칸 함량은 영지(OK1362)가 25.2%를 나타내며 가장 높았으며, 그 외에 구름버섯속의 T. lactinea (OK1457)가 24.5%로 높은 것으로 나타났다. 본 연구의 결과는 국내 자생하는 야생버섯류 중 새로운 천연물 유래 생리활성 물질을 탐색하기 위한 기초자료로서 활용 가능성을 기대한다. The aim of the study was to obtain the extracts of various native wild mushrooms and select the useful resources though biological activity evaluation. The anti-oxidant potential, nitrite scavenging activity, and -glucan content of wild mushrooms collected from Eumseong and Bonghwa in Korea were investigated. Based on the results of this study, Ganoderma lingzhi (OK1362) showed the highest DPPH radical scavenging activity (73.2%), ferric reducing anti-oxidant power (0.134), reducing power (0.155), nitrite scavenging activity (53.6%), total polyphenol content (28.9 mg GAE/g), flavonoid content (10.0 mg QE/g), and -glucan content (25.2%) when compared to other wild mushrooms sampled in this study. In addition, it was confirmed that Perenniporia fraxinea (OK1360), Amanita sp. (OK1398), and Russula sp. (OK1406) had relatively high anti-oxidant and nitrite scavenging potentials. In conclusion, our results can provide fundamental data for extracting beneficial compounds from wild mushrooms.

Ureteral polypoid endometriosis causing hydroureteronephrosis

A 42-year-old nulliparous woman visited our hospital due to the left hydronephrosis which had been found during her regular check-up. Abdominal ultrasonography revealed the left hydroureteronephrosis and about 6-cm intramural uterine fibroid locating at the left lateral wall. She had mild dysmenorrhea during her periods but did not need to take a pill. Her pelvic examination revealed no evidence of the pelvic endometriosis. She underwent the laparoscopic surgery to remove the fibroid and find the reason of the hydronephrosis. The uterine fibroid had no relationship with the left hydronephrosis, but there was an adhesive peritoneal endometriotic lesion at the left pelvic brim. After dissection of the left retroperitoneal space, we found the swollen portion of the ureter beneath the adhesive peritoneum, which length was about 3cm. We performed the segmental resection of the swollen ureter and the ureteroureterostomy, After procedure, the resected specimen showed a 3-cm polypoid mass inside the lumen. Histologically, the mass was ureteral endometriosis. The postoperative course was uneventful. Ureteral polypoid endometriosis causing hydroureteronephrosis is rare. Hence, we report this case.

The wound-inducible lipoxygenase obtained from maize is one of the nontraditional lipoxygenases that possess dual positional specificity. In this paper, we provide our results on the determination and comparison of the kinetic constants of the maize lipoxygenase, with or without detergents in the steady state, and characterization of the dependence of the kinetic lag phase or initial burst, on pH, substrate, and detergent in the pre-steady state of the lipoxygenase reaction. The oxidation of linoleic acid showed a typical lag phase in the pre-steady state of the lipoxygenase reaction at pH 7.5 in the presence of 0.25% Tween-20 detergent. The reciprocal correlation between the induction period and the enzyme level indicated that this lag phenomenon was attributable to the slow oxidative activation of Fe (II) to Fe (III) at the active site of the enzyme as observed in other lipoxygenase reactions. Contrary to the lagging phenomenon observed at pH 7.5 in the presence of Tween-20, a unique initial burst was observed at pH 6.2 in the absence of detergents. To our knowledge, the initial burst in the oxidation of linoleic acid at pH 6.2 is the first observation in the lipoxygenase reaction. Kinetic constants (Km and kcat values) were largely dependent on the presence of detergent. An inverse correlation of the initial burst period with enzyme levels and interpretations on kinetic constants suggested that the observed initial burst in the oxidation of linoleic acid could be due to the availability of free fatty acids as substrates for binding with the lipoxygenase enzyme.
Generation of Soluble Human Tumor Necrosis Factor-α Receptor 1-Fc Transgenic Pig

BACKGROUND.: Acute humoral xenograft rejection (AHXR) is an important barrier to xenograft survival. Human tumor necrosis factor-α (hTNF-α) is one of the essential mediators of AHXR and induces activation of porcine endothelial cells (PECs), resulting in upregulation of major histocompatibility complex molecules, adhesion molecules, and proinflammatory chemokines. We investigated whether introduction of a soluble human tumor necrosis factor receptor I-Fc (shTNFRI-Fc) fusion gene can suppress activation of PECs and, more importantly, produced shTNFRI-Fc transgenic pigs. METHODS.: The shTNFRI-Fc gene expression vector was constructed and inserted into PECs. The inhibitory effects of shTNFRI-Fc were tested by luciferase assay, reverse-transcriptase polymerase chain reaction, and flow cytometry. A shTNFRI-Fc transgenic pig was generated by somatic cell nuclear transfer. The expression of shTNFRI-Fc in the transgenic pig was evaluated by PCR, western blot, enzyme-linked immunosorbent assay, and immunohistochemistry. The inhibitory effects of shTNFRI-Fc in the serum obtained from the transgenic pig were also tested. RESULTS.: In comparison with control green fluorescent protein, shTNFRI-Fc protein showed much stronger inhibitory effects on NF-&kgr;B activation in the HEK293-NF-&kgr;B-luciferase reporting cell line, expression of chemokines and adhesion molecules in PECs, and TNF-α-mediated cytotoxicity. We successfully generated shTNFRI-Fc transgenic pig. Sera obtained from the transgenic pig inhibited induction of chemokines, and E-selectin in PECs stimulated with Human TNF-α. CONCLUSIONS.: We have generated transgenic pigs producing shTNFRI-Fc protein that can inhibit TNF-α-mediated activation of PECs. Because TNF-α is an important mediator of xenograft rejection, the use of xenografts that can produce shTNFRI-Fc proteins de novo could be an effective approach in overcoming a considerable component of the xenograft rejection process, especially AHXR.

<P>An Arabidopsis 관-glucosidase, AtBG1 is known to hydrolyze glucose-conjugated, biologically inactive abscisic acid (ABA) to produce active ABA, which increases the level of ABA in plants. Since an increase of ABA in plants confers tolerance against abiotic stress such as drought, we introduced the pCAMBIA3301 vector harboring the AtBG1 gene into creeping bentgrass through Agrobacterium-mediated transformation. After transformation, putative transgenic plants were selected using the BASTA resistance assay at a concentration of 0.8%. Genomic integration of the AtBG1 gene was confirmed by genomic PCR and Southern blot analysis, and gene expression was validated by Northern blot and Western blot analyses. Interestingly, the transgenic bentgrass plants overexpressing AtBG1 had a dwarf phenotype with reduced growth rates when compared to wild-type creeping bentgrass. In addition, the transgenic plants accumulated higher ABA levels and displayed enhanced drought tolerance. These results suggest that the expression of AtBG1 in plants induces the accumulation of higher ABA levels, which results in the formation of dwarf creeping bentgrass and enhances the survival in water-limiting environments.</P>
