Nutritive value of protein of barley and rice Missitgaru, which are roasted and pounded powder and supplementary effect of soybean protein on the foods were studied in young rats. In evaluating the nutritive value of food protein in the animal experiment the turnover rate of plasma albumin-Ⅰ^131, as a possible single direct indicator of protein status, was estimated and compared with weight gain, hemoglobin and plasma protein. Barley Missitgaru diet supported significantly better growth (1.60g/day) than did rice Missitgaru diet (0.14g/day) at the same nitrogen content (protein 11.3%, N X 6.25), suggesting the more destruction of protein in rice through the roasting mothod. When the Missitgaru diet was partially replaced with soybean protein, rats grew at a significantly faster rate (2.30g/day) even though the supplementary effect was inferior than that of casein (3.91g/day). The turnover rate of plasma albumin-Ⅰ^131 of the rice Missitgaru fed rats, which showed extremely limited body weight gain, was significantly slower than that of rats fed the diet partially replaced with barley Missitgaru (weight gain; 0.88g/day). However, there was no noticeable difference in the rate among the paritially supplemented food groups, i.e., with barley Missitgaru, soybean protein and casein, where considerable different in growth were observed. The turnover rate of albumin, which was impressed as to be regulated within normal so far as possitive nitrogen balance is maintain, was hasten by excess protein intake. No significant difference was found in hemoglobin, hematocrit and plasma protein among these groups except a slight lower value of hemoglobin in rice Missitgaru fed group. Above result suggests that the estimation of turnover rate of plasma albumin would be useful in assessing the protein status.
HPLC-UVD를 이용한 수련(Nymphaea tetragona Georgi)의 Ellagic Acid 정량 분석

A high performance liquid chromatography (HPLC) method for the quantitation of ellagic acid in Nymphaea tetragona was developed for the quality control of functional cosmetic ingredient, the extract of N. tetragona. Separation and quantitation were successfully achieved with a Kromasil C18 column (5 mm, 250 mm ? 4.6 mm, i.d.) by isocratic elution of a mixture of acetonitrile containing 0.1% trifluoroacetic acid and water containing 0.03% phosphoric acid at a flow rate of 1.0 ml/min. The UV detector was used for the detection and the wavelength for quantitation was set at 254 nm. The presence of ellagic acid in the extract was determined by comparison of retention time and spiking with authentic standard. Analytical results showed good linearity (R2 = 0.99996) in relatively wide concentration ranges. The R.S.D. for precision test was less than 3.0%. Recovery of the compound was 98.55~101.72% with R.S.D values less than 4.0%. In conclusion, this method has been successfully applied to the determination of ellagic acid in N. tetragona.
