        오레가노 잎 추출물에 대한 항염증 및 미백 활성

        In this study, the anti-inflammatory and whitening effects of hot water and 70% ethanol extracts of Origanum vulgare leaf were examined. First, the results from measurement of the total polyphenol content confirmed that OLD was 649.16 ±0.41 mg/100 g and OLE was 646.62 ±0.85 mg/100 g. Analysis of electron donating activity and ABTS radical scavenging activity was performed for measurement of the antioxidant activity, which confirmed that both extracts exerted antioxidant activity in a concentration-dependent manner. The results of measurement of tyrosinase inhibition activity showed an inhibitory effect of 41.52% for OLE at a concentration of 1,000 μg/mL. Assessment of cytotoxicity in RAW 264.7 cells and B16-F10 cells was then performed for measurement of anti-inflammatory and whitening effects at the cellular level. RAW 264.7 cells showed cell viability of more than 95% at a concentration of 500 μg/mL, and B16-F10 cells showed cell viability of more than 95% at a concentration of 50 μg/mL. Measurement of the inhibitory activity on production of NO was performed in order to verify anti-inflammatory activity. The results confirmed that production of NO was decreased by both OLD and OLE in a concentration-dependent manner compared to the group treated with LPS alone. The inhibitory activity on melanin synthesis was determined for evaluation of the whitening effect. The results indicated that melanin synthesis was inhibited by both OLD and OLE in a concentration-dependent manner compared to the group treated with α-MSH alone. In conclusion, the results confirmed the antioxidant, anti-inflammatory, and whitening effects of OLD and OLE, indicating the potential for their use as new natural materials.

        LPS로 자극된 대식세포에 대한 딸기 줄기의 항염증 효능 검증

        In strawberry farming, most parts of strawberry stems but the fruit have been dumped. Therefore, this study attempted to investigate the antioxidant and anti-inflammatory effects of strawberry stems which are thrown away after farming. For this, strawberry stem extracts were obtained, using hot water and 70% ethanol. First, total polyphenol contents of the hot water and ethanol extract were checked (265.4 ± 0.12 mg TAE/100 g, 503.88 ± 0.2 mg TAE/100 g). For analysis of antioxidant activities, electron donating ability (EDA) and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity were measured. Both extracts increased in a dose-dependent fashion, and similar effects with vitamin C (control group) were confirmed. In terms of cell viability of the hot water and ethanol extracts of strawberry stems, ‘RAW 264.7’ was 99% or higher at 500 µg/ml. In addition, cell experiments were conducted at 50, 100 and 500 µg/ml where cell viability is above 99%. In terms of inhibition of the inflammatory mediator ‘nitric oxide (NO)’, the hot water and ethanol extracts of strawberry stems were 37.9% and 38.8% respectively, confirming the inhibition of NO production. To check anti-inflammatory activities, protein and mRNA expressions of ‘iNOS’ and ‘COX-2’ were measured, using RAW 264.7. Compared to the LPS group, the protein expression of the inflammatory mediators was inhibited in the hot water and ethanol extract-treated groups. The above results confirmed that the hot water and ethanol extracts of strawberry stems are valuable as natural substances with antioxidant and anti-inflammatory activities.

        LPS로 유도한 대식세포에서 레드향 가지 (Citrus reticulata 'Kanpei') 추출물의 항염증 활성

        Consumers’ interest in natural materials has shown a recent increase; this study was conducted for evaluation of antioxidant activity and anti-inflammatory efficacy in order to determine the potential for application of natural materials using hot water (CBD) and 70% EtOH extract (CBE) of Citrus reticulata 'Kanpei' branches. The results of measurement showed that the polyphenol content of CBD and CBE was 372.97 ± 0.79 mg TAE/100 g and 371.85 ± 0.63 mg TAE/100 g, respectively. The antioxidant activity of DPPH radical scavenging activity and ABTS radical scavenging activity of CBD and CBE was greater than 90% at a concentration of 1,000 μg/mL . The result of evaluation showed that the cell viability of CBD and CBE was greater than 90% at a concentration of 100 μg/mL. The nitric oxide production inhibitory activity of CBD and CBE was 25.41% and 29.26% at a concentration of 100 μg/ mL, respectively, compared with the control group. According to the results from measurement of the level of protein expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), inhibition of iNOS by CBD and CBE was 7.66% and 28.09% at a concentration of 100 μg/mL, respectively, and 11.69% and 35.43% for COX- 2, respectively. Therefore, the findings of this study verified the antioxidant and anti-inflammatory effects of Citrus reticulata 'Kanpei' branches, confirming their potential for use as natural materials.

        패션프루트 껍질의 생리활성에 대한 연구

        In this study, the effects of the physiological activity of the Passiflora edulis f. peel (PEP) were verified and the applicability as a cosmetic material was reviewed. As a result of the total polyphenol content, the content was expressed as 95.88±0.45mg/100g dry in the hot water extract and 119.99±1.17mg/100g dry in the 70% ethanol extract. The antioxidant effects were measured through an experiment based on evaluating the electron-donating ability and ABTS radical scavenging activities, and the results showed that the effects were better in the 70% ethanol extract than in the hot water extract. In order to verify the whitening effect, the measurement of tyrosinase inhibitory activity showed an effect of 30.73±0.77% at 1,000 ㎍/㎖ of PEP in the 70% ethanol extract, which showed a greater whitening effect than of the hot water extract. Active suppression of nitric oxide (NO), an inflammatory inducer, was measured to verify the anti-inflammatory efficacy. Prior to that, an MTT assay was conducted in RAW 264.7 cells to assess the cell survival rate; it was found that no toxicity was apparent at any concentration. In the NO assay, experiments were conducted using a concentration of 500 ㎍/㎖ with a 100% survival rate. The NO assay was measured using Griess reagent; compared with the lipopolysaccharide treatment group and non-treatment group, it showed better activity in the ethanol extract than in the hot water extract. With the above result, it has been confirmed that PEP extract is an effective antioxidant, has whitening effects, and is an anti-inflammatory agent. In particular, the use of ethanol extracts as natural materials for cosmetics was confirmed to have excellent effects.

        거봉포도 가지 추출물의 항산화 및 항염증 효능

        The antioxidant and anti-inflammatory activity of 70% ethanol extracts of 'Kyoho' grape branches (Vitis labruscana L.) was examined in order to develop a new natural material. First, an evaluation of total polyphenol content and antioxidant activity of 70% ethanol extracts of 'Kyoho' grape branches was performed. The result of measuring the total polyphenol content of 70% ethanol extracts of 'Kyoho' grape branches was 862.41±1.09 mg/100 g. Electron donating ability and 2’-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging ability were confirmed for evaluation of the antioxidant activity of 70% ethanol extracts of 'Kyoho' grape branches. The results of measurement of antioxidant activity at a concentration of 1,000 µg/mL confirmed that the electron donating ability was 93.09% and the ABTS radical scavenging ability was 99.71%. The MTT assay was performed using RAW 264.7 cells for evaluation of cell viability; the result showed more than 99% at a concentration of 500 µg/mL. Nitric oxide (NO) production inhibitory activity was measured, and Western blot was performed in lipopolysaccharide-induced RAW 264.7 cells for confirmation of the anti-inflammatory activity. The NO production inhibitory activity of 70% ethanol extracts of 'Kyoho' grape branches showed an inhibitory effect of 28.74% at a concentration of 500 µg/mL. Western blot was performed for confirmation of inflammation-related factors iNOS and expression of COX-2 protein. The result confirmed that expression of iNOS and COX-2 protein was inhibited in a concentration-dependent manner. Based on the results described above, the antioxidant and anti-inflammatory activity of 'Kyoho' grape branches, as well as its use as a natural material was confirmed.

        Study on Anti-Inflammatory and Moisturizing Effect of Extracts from Abelmoschus esculentus

        In this study, we studied the anti-inflammatory and physiological activities Abelmoschus esculentus extracts by hot water (AEW) and 70% ethanol (AEE) and confirm the possibility to use it as a natural ingredient. For this study measure the antioxidative activity, total polyphenol content was measured, and DPPH and ABTS scavenging activity assays were conducted. Total polyphenol content of AEW and AEE was measured, and the results showed that they were 126.76 mg TAE/100 g and 144.21 mg TAE/100 g, respectively. DPPH and ABTS radical scavenging activities were measured to determine the antioxidative activity, and the results indicated that DPPH and ABTS radical scavenging activities increased in both extracts concentration-dependently. The moisturizing effect was measured by measuring the amount of hyaluronic acid (HA) produced within HaCaT cell using the ELISA kit. AEW and AEE increased the amount of HA production in a concentration-dependent manner. In order to determine the anti-inflammatory activity of AEW and AEE, the NO assay was conducted, and the inhibitory effects were found to be 11.46% and 25.28%, respectively in 100 µg/ml. In order to measure the anti-inflammatory activity, nitric oxide (NO) inhibitory activity was measured, and the inhibition of expression of iNOS, COX-2 proteins was measured and shown. Furthermore, inhibition of expression was found in inflammatory inducing factors iNOS and COX-2 proteins showing concentration-dependent inhibition. This study found the excellent effects of antioxidative, moisturizing effect, anti-inflammatory activity in Abelmoschus esculentus extracts, which indicates that can be used as functional materials for aesthetics, food and functional cosmetics.

        대추나무 새순 추출물의 보습 및 항염증 효능

        . In this study, Ziziphus jujuba Miller bud were extracted with hot water (ZBD) and 70% ethanol (ZBE) to examine the potential for antioxidant, anti-inflammatory, and moisturizing effects. The results of measuring the total polyphenol content showed that the contents of ZBD and ZBE were 525.39 ±0.94 mg TAE/100 g and 581.63 ±0.94 mg TAE/100 g, respectively. Regarding electronic donating ability, at a concentration of 1,000 μg/mL, ZBD and ZBE were 76.06% and 80.92%, respectively, and the results of 2,2’-azino-bis (3-ethylbenzo-thiazoline-6-sulfonic acid) radical scavenging showed that both extracts exerted excellent antioxidant activity of greater than 99% at a concentration of 500 μg/mL. Cell experiments were then performed according to the results of RAW 264.7 and HaCaT cytotoxicity measurements. At a concentration of 500 μg/mL, the results of measuring nitric oxide inhibitory activity showed that both extracts exerted an inhibitory activity of greater than 11% compared to the lipopolysaccharide (LPS) treatment group alone, and the amount of hyaluronic acid production by ZBE was greater than 126% compared to the groups treated with P. acnes alone. By confirming the protein expression of iNOS and COX-2 compared to the LPS treatment group alone, at a concentration of 500 μg/mL, ZBD inhibited 24.21% and 29.78% and the inhibitory efficacy of ZBE was 68.83% and 47.71%, respectively. Therefore, ZBD and ZBE can be regarded as being effective in exerting anti- inflammatory and moisturizing effects and can be used as a material for natural cosmetic.

        천혜향 잎 추출물의 항산화 활성 및 RAW 264.7 세포에서의 iNOS와 COX-2의 발현 저해 효과

        In this study, evaluation of antioxidant and anti-inflammatory activity was performed to confirm the potential for use of Cheonhyehyang leaves as a natural ingredient in cosmetics. Extraction of Cheonhyehyang leaves was performed using hot water extracts (CLD) and 70% ethanol extracts (CLE). The total polyphenol content of CLD and CLE was 210.21 ± 0.19 mg TAE/100 g and 204.14 ± 0.50 mg TAE/100 g, respectively. Electron donating ability was confirmed after measurement, verifying the effectiveness of CLD (79.00%) and CLE (78.91%) at a concentration of 1,000 μg/ mL. The ABTS radical scavenging capacity for CLD and CLE was 99.91% and 99.69% at a concentration of 1,000 μg/mL, respectively. Assessment of cell viability in RAW 264.7 cells performed using an MTT assay showed greater than 90% at a concentration of 100 μg/mL. For assessment of anti-inflammatory activity, production of nitric oxide was inhibited using RAW 264.7 cells induced using lipopolysaccharide; the results showed a decrease according to concentration. Measurement of the inhibitory effects of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) was performed using western blot. The result of treatment with CLD and CLE confirmed that expression of iNOS and COX-2 protein was suppressed in a concentration-dependent manner. In conclusion, the findings of this study confirmed the antioxidant and anti-inflammatory effects of CLD and CLE, as well as their potential for use as natural materials.

        아티초크 추출물의 항산화, 항염증, 미백 효능 활성에 관한 연구

        The objective of this study was to investigate the biological activities of 70% ethanol extracts from Cynara scolymus L. (CSEE). The total polyphenol content of CSEE were found to be 158.5 mg tannic acid/100 g dry weight of CSEE. To confirm the antioxidant effect, electron donating ability and ABTS radical scavenging effect were measured. As a result, the extent of electron donating and ABTS radical scavenging effect on CSEE increased in a dose-dependent manner. The tyrosinase inhibitory effect of CSEE, which is related to skin-whitening, was observed to be 36.25% at a concentration of 1,000 μg/mL. Furthermore, MTT assay was performed to confirm the cell viability in RAW264.7 and B16F10 cell lines. In both RAW 264.7 cell and B16F10 cells, the cell viability was observed more than 95% at 100 μg/mL in all extracts. The anti-inflammatory effect of CSEE on lipopolysaccharide (LPS)-induced RAW264.7 cells was also investigated. The CSEE significantly suppressed NO production in a concentration-dependent manner. In order to measure the whitening effect on CSEE, the expressions of proteins related to whitening, such as microphthalmia-associated transcription factors, tyrosinase-related proteins-1 were observed using western blot. In the section treated with the extracts, it was confirmed that the protein expression decreased in a concentration-dependent manner. In conclusion, CSEE was verified to have antioxidant, anti-inflammatory and whitening effects, and it was confirmed that it is a potential natural cosmetic material.

        애플망고 잎의 추출물의 항산화 및 대식세포(RAW 264.7)에서 iNOS, COX-2 발현 저해 효과

        본 연구는 애플망고 잎 추출물의 항산화 및 항염증에 활성을 검증하여 연구하였다. 총 폴리페놀 함량은 Folin-Denis의 방법에 의해 측정하였다. 그 결과 애플망고 잎의 열수와 70% 에탄올 추출물은 각각 440.83±1.02, 475.63±1.3 mg/100 g TAE의 함량을 나타내었다. 항산화 활성을 측정하기위해 전자공여능 측정과 2,2"-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) 라디칼 소거능 활성을 측정하였으며, 모든 추출물이 우수한 효과를 나타내었다. 대식세포내에서 애플망고 잎 추출물의 세포 생존율을 MTT assay의 방법으로 측정하였다. 세포의 독성이 나타나지 않은 농도구간에서 이하의 세포실험을 진행하였다. 항염증 활성을 효과적으로 측정하기 위해 LPS로 염증반응을 유도시키고 Griess assay를 이용해 nitric oxide를 측정하였다. 그 결과 애플망고 잎 추출물은 농도의존적으로 NO의 생산을 억제함을 나타내었다. LPS로 유도된 RAW 264.7세포에서 iNOS와 COX-2의 염증인자 생성억제를 mRNA발현 억제를 RT-PCR을 통해 확인하였다. 따라서 본 연구에서는 애플망고 잎 추출물의 항산화 및 항염증 활성의 우수한 효과를 확인하였고, 그 결과 천연소재로서의 유용성과 함께 화장품의 기능성소재로서의 활용가능성을 확인하였다. The present study investigates the antioxidant and anti-inflammatory activities of Mangifera indica L. leaf extract. The total polyphenol content was measured using the Folin-Denis method. Results showed that the M. indica L. leaf extract of water and 70% ethanol showed a content of 440.83±1.02, 475.63±1.3 mg/100 g tannic acid equivalent. To assess antioxidant activity and electron-donating ability, 2,2"-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity were measured, and all extracts were found to be highly efficacious. To assess cell viability of the extract from M. indica L. leaf on macrophage cells (RAW 264.7), a 3-[4,5-dimethyl-thiazol-2- yl]-2,5-diphenyl-tetrazoliumbromide assay was performed. The following experiments were conducted in section where cells was not shown of toxicity. In order to effectively determine anti-inflammatory activity, inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells was examined using a Griess assay. The result showed that M. indica L. leaf extract concentration-dependently inhibited NO production. M. indica L. leaf extract was measured using Western blot, reverse transcription- polymerase chain reaction (RT-PCR) that to find the production of pro-inflammatory factor on stimulated RAW 264.7 cells of LPS. According to the results of this study, the M. indica L. leaf extract showed excellent effectiveness in antioxidant and anti-inflammatory activity, thus confirming its usability as a natural material and a functional raw material for cosmetics.
