        안전한 화장품 개발을 위한 동물실험대체법 연구 동향

        The AMOREPACIFIC Corp., a leading cosmetic company in South Korea, has been making an effort to use variety of alternative methods for many years. We have been making use of alternative methods on assessing phototoxicity of fragrances, corrosivity of hair care products, and ocular irritancy of eye creams and eye makeup products. In addition to existing protocols, there are also several trials on improving and applying current alternative methods. We have intended to apply the HET-CAM (Hen`s egg test on the chorioallantoic membrane) test to various studies on safety assessment of cosmetic products prior to ophthalmology test or anti-irritant material screening. A battery test was introduced for the purpose of improving the accuracy of in vitro phototoxicity method. We also make effort to get suitable alternative methods for predicting sensitizing potency of cosmetic ingredients and products. The one of the strong candidates is h-CLAT (human cell line activation study) and which is now under examination of its accuracy and utility.

        The Study of Age-related Skin Pigmentation Changes in Korean Female

        Since facial skin pigmentation is one of the visual characteristics of skin aging, it is important to evaluate skin pigmentation in the cosmetics and aesthetic fields. Several groups have investigated and developed the image analysis methods for skin pigmentation and some of the groups reported the age-related changes of the number and size of facial pigmented spots. However, they didn’t show the changes of the number and size of pigment spots by defined size, and there is no report for Korean female regarding pigmentation. A total of 194 Korean females aged 20 ~ 79 (48.97 ± 17.11 years) were analyzed for the number, size, and intensity of pigmented spots using various filters such as large high-pass filter and median filter in their digital facial images. There were significant correlations between age and total pigmented spot number (No.), size, and intensity (I) (pearson’s correlation coefficient r = 0.688, r = 0.645, r = -0.563), and significant correlations were also observed between the number and intensity of pigmented spots of different sizes. According to the ANOVA results, there were significant differences in the percentage of spot size of 2 ~ 4 mm2 and > 20 mm2 between 20’s and 70’s. In other words, with aging, pigmentation increases in the facial skin, and the large increase in pigmentation is particularly noticeable in Korean women.

        한국 한방화장품 발달 배경 및 연구 현황

        Traditional Asian medicine has an extensive evidence base built upon thousands of years of experience within Asia, of curing various diseases. Only recently, within the past two centuries, have modern medical scientists developed interest in traditional Asian medicine. Asian Medicine seems to be regarded only as an adjunctive medicine and viewed as alargely un-proven alternative medicine to complement western medicine, used in some cases to establish a new paradigm of “integrative medicine”. This article reviews how Korean herbal cosmetics emerged by applying traditional Asian medicine to the science of cosmetics. The characteristics of Korean herbal cosmetics are examined through examples of history, concepts and traditions. With the advancements in biotechnology, studies are now being conducted on the dermatological effects and processing methods of herbal ingredients, including ginseng. The authors explain the current research on the identification on the active ingredients of herbs, extraction methods, and bio-processing of ingredients to improve the biological efficacies of herbs on the skin. A summary of studies focused on modern reinterpretations of ageing theories, such as ‘Seven year aging cycle’, are provided. In conclusion, the development of Korean cosmetics products are based on the accumulated knowledge of thousands of years of experience including; 1) practical heritage of traditional Asian medicines such as Donguibogam; 2) excellent medicinal plants, such as ginseng, which are native to Korea; and 3) innovative attempts to modernize materials, processes, and principles. 동아시아의학은 오랜 역사를 가지고, 아시아인들의 다양한 질병 치료 경험들을 쌓아왔다. 최근의 현대 의과학자들은 의학의 새로운 패러다임을 정립하기 위해, 혹은 서양의학의 한계를 극복할 수 있는 역할로 동아시아 의학에 많은 관심을 갖고 있다. 한국의 한방화장품은 이러한 동아시아의 의학적 자산과 한방 원료들을 화장품과학에 적용하여 탄생하였으며, 최근 세계 화장품 시장에서 성장하고 있는 추세이다. 본 연구에서는 한방화장품의 의사학적 배경과 최근의 관련 영역에서의 연구 성과 등을 고찰하고자 한다. 구체적으로는 한약재의 피부 효능을 개선하기 위한 활성 성분 연구, 추출 방법이나 가공 기술 개발 사례, 한의학적 피부 노화 이론의 발굴 사례 등이다. 결론적으로, 한국의 한방 화장품 발달 배경을 의사학적인 관점으로 분석하면 다음과 같다. 첫째. 동의보감과 같은 실용적인 동아시아 의학 자산의 활용, 둘째는 한반도에 자생하는 인삼과 같은 뛰어난 약용식물들의 신규한 가치 발굴, 셋째는 각종 원료와 이론들을 현대화하려는 혁신적인 시도들이 바로 그 점이다

        안점막자극시험의 대체시험법으로서의 CAM-TBS assay 소개

        The hen`s egg test-chorioallantoic membrane (HET-CAM) assay, which employs the vascular membrane of a fertile hen`s egg, has been actively studied by several countries as an alternative method to the Draize rabbit eye irritation test. However, the scoring system of the original HET-CAM test is not satisfactory because of its lack of objectivity and quantitativeness. Moreover, the test required skill when the effects of test substance are to be regarded. To overcome these disadvantages, Hagino et al. developed and objective evaluation technique using the amount of trypan blue absorbed at the site of treatment as an indicator of injury to the CAM (Chorioallantoic membrane-trypan blue staining, CAM-TBS). In this study, we evaluate the CAM-TBS methods and used three materials (1%, 2%, 5% SLS, tween 20 and PBS) to evaluate the correlation of CAMTBS with HET-CAM test. The amounts of trypan blue adsorbed with the CAM showed a good correlation with the HET-CAM test score(r=0.958). According to these results, we suppose that the trypan blue staining assay using CAM-TBS may be a good method to replace the HET-CAM test as an alternative to Draize rabbit eye irritation test. Despite our results, additional studies with more materials are necessary to validate this method.

There are two methods to evaluate the eye irritation potential, reaction time method(RTM) and end point assessment(EPA), between which can be chosen as the apt method depending on the characteristics of test material. In this study, we employed the EPA method to compare with the Draize rabbit eye irritation test method and then calculated the Pearson correlation coefficient value. The Result of Pearson correlation was high(0.986, p=0.002) with 5 test materials: 3 raw materials(IPMS 03, VCP-IS-2Na, Seletinoid: Amorepacific R&D center, Korea), 1 commercial cleansing form(Amorepacific R&D center, Korea) and 1 surfactant (Amisoft LS-11, Ajinomoto, Japan). According to these results, we suppose that the HET-CAM assay is a good method to replace the Draize eye irritation test as the tests of 5 materials have proven. Despite our results, additional studies with more materials are necessary to validate this method as an alternative to the Draize rabbit eye irritation test method.
