        Emerging roles of polyunsaturated fatty acid synthesis pathway in colorectal cancer

        The development of colorectal cancer typically involves the accumulated influences of geneticalterations, medical issues, lifestyle, and diet. Dietary fatty acids appear to affect thetumorigenesis and progression of colorectal cancer. Despite conflicting results, the currentconsensus on the effects of very long-chain polyunsaturated fatty acids on colorectal cancer isthat low levels of eicosapentaenoic acid and docosahexaenoic acid, and high levels ofarachidonic acid are associated with an increased risk of colorectal cancer. Altered levels ofarachidonic acid in membrane phospholipids can change the levels of prostaglandin E2, whichaffect the biological activities of cancer cells in multiple stages. Arachidonic acid and other verylong-chain polyunsaturated fatty acids can affect tumorigenesis in prostaglandin E2-independent manners as well, including stabilization of β-catenine, ferroptosis, ROS generation,regulation of transcription factors, and de novo lipogenesis. Recent studies have revealed anassociation between the activities of enzymes synthesizing very long-chain polyunsaturatedfatty acids and tumorigenesis and cancer progression, although the mechanisms are stillunknown. In this study, PUFA effects on tumorigenesis, the endogenous very long-chainpolyunsaturated fatty acid synthesis pathway, metabolites of arachidonic acid and their effectson tumorigenesis and progression of CRC, and current knowledge that supports the associationof the enzymes involved in the polyunsaturated fatty acid synthesis pathway with colorectalcancer tumorigenesis and progression are reviewed.

        Simple Purification of Adeno-Associated Virus-DJ for Liver-Specific Gene Expression

        Recombinant gene expression using adeno-associated viruses (AAVs) has become a valuable tool in animal studies, as they mediatesafe expression of transduced genes for several months. The liver is a major organ of metabolism, and liver-specific expressionof a gene can be an invaluable tool for metabolic studies. AAV-DJ is a recombinant AAV generated by the gene shuffling of various AAV serotypes and shares characteristics of AAV2 and AAV8. AAV-DJ contains a heparin-binding domain in its capsid, which suggests that a heparin column could be used for the purification of the AAV. Given that AAV-DJ has been only recently available, relatively little is known about the optimal preparation/purification and application of AAV-DJ. Here, we present a simplelarge-scale preparation method that can generate 3×1013 viral particles for in vivo experiments and demonstrate liver-specific gene expression via systemic injection in mice.

        The SCAP/SREBP Pathway: A Mediator of Hepatic Steatosis

        Nonalcoholic fatty liver disease (NAFLD) is strongly associated with insulin resistance, obesity, and dyslipidemia. NAFLD encompassesa wide range of states from the simple accumulation of triglycerides in the hepatocytes to serious states accompanied by inflammationand fibrosis in the liver. De novo lipogenesis has been shown to be a significant factor in the development of hepatic steatosisin insulin-resistant states. Sterol regulatory element binding protein-1c (SREBP-1c) is the main transcription factor that mediatesthe activation of lipogenesis, and SREBP cleavage activating protein (SCAP) is required for the activation of SREBPs. Here, recentanimal studies that suggest SCAP as a therapeutic target for hepatic steatosis and hypertriglyceridemia are discussed.

        운동그릿 형성 과정에 대한 근거이론 접근

        목적: 이 연구의 목적은 실생활에서의 수많은 방해 요소에도 불구하고 오랜 기간 동안 운동을 지속할 수 있는 이유를 밝히는 것이다. 방법: 먼저 연구를 위해 오랜 기간 동안 꾸준하게 운동을 지속하는 능력을 운동그릿(exercise grit)이라고 조작적 정의한 후 그 형성 과정을 살펴보고 이론화하였다. 연구참여자는 중단 없이 3년이상 운동을 지속하고 있는 청장년층으로 여자 5명, 남자 6명, 총 11명이며 유목적 표집법에 따라 선정하였다. 연구참여자를 대상으로 심층면담을 통해 자료를 수집하여 Strauss와 Corbin이 제시한 개방코딩, 축코딩, 선택코딩을 순서대로 적용하여 분석하였다. 결과: 개방코딩 결과, 상위범주 20개, 하위 범주 55개, 개념 368개가 도출되었다. 이후 실시한 축코딩에서 패러다임 모형을 도출하였다. 패러다임 모형의 내용은 다음과 같다. 인과적 조건은 운동을 만나고 경험하게 된 계기로 자각한 운동 필요성, 환경적 영향이었고, 맥락적 조건은 운동이 주는 보상으로 심리적보상, 극강의 희열과 재미, 멋진 외모와 건강, 사교성 향상, 성실하고 규칙적인 생활 태도, 기술 습득 및 실력 향상 등으로 나타났다. 중심현상은 그릿 강화, 습관 형성으로 목표를 향한 의지, 가족, 지인의 지지, 운동을 향한 깊은 애정과 열정 등이었고, 중재조건은 운동 걸림돌로 주변의 부정적인 시선, 일은 운동 방해꾼 등으로 나타났다. 작용/ 상호작용은 운동을 위한 나만의 고유 전략으로 심리전략, 운동 일정 철통 사수, 유리한 환경 구축, 운동 시간 우선 확보 등이었고, 결과는 습관 유지로 삶이 된 운동, 평생의 동반자, 정신 수양 차원의 운동 등으로 나타났다. 선택 코딩에서는 모든 범주를 추상적으로 아우르는 핵심범주로 ‘삶에 유익한 변화를 주는 반려 운동’을 추출하였고 이에 따른 이야기 윤곽을 서술하였다. 마지막으로 이론화 모형을 제시하여 운동그릿 형성 과정을 입체적으로 설명하였다. 결론: 이 연구를 통해 패러다임 모형의 맥락적 조건인 ‘운동이 주는 보상’으로 일어나는 신체, 심리,사회적 변화가 운동 그릿을 형성하고 습관으로 정착되는 것에 대한 구체적인 논의가 이루어졌다. Purpose: The purpose of this study is to examine the reason that people can sustain exercise over a long period of time in spite of the numerous obstacles that may exist in other areas of their life. Methods: First of all, this study operationally defined the ability to sustain exercise activity over a long period of time as ‘exercise grit’, and then it examined its formation process and built a theory around its development. The participants in this study consisted of young adults who had been consistently exercising for more than three years, with a total of 11 participants, consisting of 5 females and 6 males, being selected according to purposeful sampling. Data were then collected through in-depth interviews with the research participants, and these data were applied and analyzed via open coding, axial coding, and selective coding, in that order, as suggested by Strauss and Corbin. Results: The open coding findings indicated 20 upper categories, 55 lower categories, and 368 concepts. The next step involved a paradigm model derived through axial coding to extract the story outline and core categories, and then the presentation of a theoretical model. The results of the paradigm model derived through axial coding are as follows. Causality conditions refer to an opportunity to encounter exercise, and they indicate the perceived need for exercise and environmental influences. Contextual conditions refer to the rewards provided by exercise, displayed as psychological rewards, extreme joy and enjoyment, good physical looks, and health, the improvement of sociability, a faithful and stable life attitude, skill acquisition, and skill improvement. Central phenomena indicated the strengthening of grit for habit formation, which included achieving goals, receiving support from family and acquaintances, and having a deep affection and passion for sports. Mediation conditions refer to the obstacles preventing exercise, which can be viewed as a negative gaze perception that acts as an exercise disruptor. Action/interaction refer to one’s own unique exercise strategy, and it is indicated through the display of a psychological strategy, tightly keeping to an exercise schedule, the establishment of a favorable environment, the prioritization of exercise time, etc. The results refer to habit maintenance that adapted exercise into one’s regular life, lifelong partners, and mental training. Finally, selective coding refers to a core category that abstractly encompasses all categories, and this study suggested ‘companion exercise that encourages beneficial changes to one’s life’. Conclusion: In this study, a specific discussion was conducted on how the physical, psychological, and social changes that occur within a contextual condition of the paradigm model, ‘reward from exercise’, form exercise grit and create long-lasting habits.

        주거용 단일유닛 모듈러의 활성화를 위한 국내외 사례연구

        The purpose of this study is to analyze architectural characteristics of domestic and foreign unit modular housing products for development of affordable housing as case study. A document research and field survey were used to analyze and classify the architectural characteristics of four unit modular housing products, Staco, Muto as domestic, Spacebox and Caledonian of Europe. Based on the results of the critical review, The framework of this study was composed of four dimensions such as ubiquity, flexibility, sustainability and affordability. The results of this study suggest that unit modular housing not only has typical benefits of prefabricated building such as shorter build time and budget reduction, but also can get flexibility, sustainability, affordability and proved quality via mass production. This research can be a basis for the application of the unit modular housing to the affordable housings for college students, further, officer and workers.
