Novel Germline Mutations of BRCA1 and BRCA2 in Korean Familial Breast Cancer Patients

Breast cancer is the second most common cancer in Korean women. Germline mutations in the BRCA1 and BRCA2 genes cause hereditary breast cancer and are detected in 15-20% of hereditary breast cancer. We investigated the BRCA1 and BRCA2 mutations in 114 familial breast cancer patients using next-generation sequencing. We confirmed 20 different mutations of BRCA1 and BRCA2 in 25 subjects (21.9%). Two such mutations in eight patients were novel (not reported in any variant database or previous study). Six mutations have been reported as disease-causing mutations in public databases. Seven mutations were found only in a single nucleotide polymorphism database and one mutation has been reported in Korea. The BRCA1/2 mutation frequency was similar to that of other studies on familial breast cancer patients in the Korean population. Further studies should examine more cases and mutations of whole exons.

The surface rolling method which is one of the plastic deformation processes increases the surface roughness with reduction of diameter and hardness. In this study, three NACHI 6000 ZZ bearing were used for surface rolling tool on a mild steel. The following results have been obtained with the mild steel. 1) The load is major factor in getting fine surface roughness of roller fininshing after grinding. The optimal surface roughness of SS40 steel can be obtained at the contact pressure of 210 kgf/㎠ But, Better surface roughness can not be expected, Due to flaking phenomena at more than 300 kgf/㎝ of contact pressure. 2) At the contact pressure range of 200kgf/㎠∼210kgf/㎠ for optimal surface roughness, The surface hardness increased to Hv 200 ∼ Hv 240 from Hv 125 before surface rolling. 3) Within the diameter variation of 713㎛ the surface roughness and the surface hardness were increased, but out of variation of 14㎛. The surface roughness become worse and the surface hardness was increased.

The magnetic polishing is the usefiil method to finish using magnetic power of magnet. This method is one of precision polishing techniques and has an aim of the clean technology using for the pure of gas and inside of the clean pipe for transportation. The magnetic abrasive polishing method is not so common for machine that it is not spreaded widely. There are rarely researcher in this field because of non-effectiveness of magnetic abrasive. Therefore, in this paper deals with development of the magnetic abrasive using sludge. In this development, abrasive grain WA and GC used to resin bond fabricated low temperature. And magnetic material was fabricated from the sludge which were crused into 200 mesh and average diameter 01.2mm ball type. The XRD analysis result show that only WA and GC abrasive and sludge crystal peaks detected which explains resin bond was not any more chemical reaction. From SEM analysis it is found that WA and GC abrasive and sludge were strong bonding with each other by bond.
