Investigation of meridian line and acupoints as anatomical entities should be very important, as acupuncture and meridian massage for medical purpose in the hospital or for skin care in beauty shop are each based on the traditional meridian theory. We isolated tubular structures under the skin of rabbits where the traditional meridian lines are located. The characteristics of these tubular structures isolated from the abdominal wall matches those of Bonghan tubes and Bonghan tubules found by Bong Han Kim. The Bonghan theory established by Bong Han Kim suggests that meridian lines consist of Bonghan tubes and Bonghan tubules. We could observe the presence of large cells inside the Bonghan tubule and the presence of small granules referred to as Sanal in Bonghan theory inside the striae consisting of the Bonghan tubules. Present research suggests that there exist a third unique tubular structure where Sanals flow other than Bonghan tube and Bonghan tubules.
Purification and Characterization of $\beta$-Glucuronidase from Rat Uterus.

쥐의 자궁에서 베타-글룩유로니다아제를 $(NH_4)_2S0_4$ 분획분리, DEAE-셀루로오즈 크로마토그라피법, 젤여과법 및 Concanavalin-A Sepharose 흡착 크로마토그라피법등을 사용하여 완전분리 정제 하였다. 정제된 효소는 pH 8.3, 7.5% 아크릴아마이드 젤에서 전기영동한 결과 단일 단백질로 나타났다. 젤여과법에 의해 얻어진 분자량은 288,000 Dalton 이었고 SDS 폴리아크릴아마이드 젤 전기 영동법에 의해 이 효소는 75,000의 분자량을 갖는 단위 조각으로 구성 됐음을 알아냈다. 이 효소의 에 따른 활동도는 기질의 농도나 이온 강도의 변화에 따라 변화됨을 볼 수 있었다. 또한 이 효소는 중성보다 산성 완충용액일때 비교적 높은 온도에서 더욱 안정하였다. P-Nitrophenyl-$\beta$-D-glucuronide를 기질로 사용하였을 Km 치는 0.25mM 이고 $Hg^{++}$, $Cu^{++}$, $Ag^{+}$ 같은 중금속 이온에 의해 효소의 활동도가 방해 되었다. citrate, heparin 이나 식초산도 역시 효소의 활동도를 방해 하였다. 이온강도가 0.4에서 여러 염들도 효소의 활동도를 20%정도감소시켰다. $\beta$-Glucuronidase was purified from rat uter us by $(NH_4)_2S0_4$ fractionation, DEAE-cellulose chromatography, gel filtration on Sephadex G-200, CM-cellulose chromatography, and affinity chromatography on Concanavalin-A Sepharose. The pH profile of the enyzme varied when changing substrate concentration and ionic strength. The enzyme was more stable at high temperature in acidic buffers than in neutral ones. The Km value for p-nitrophenyl-$\beta$-D-glucuronide as substrate was 0.25 mM, and the enzyme was inhibited by heavy metal ions, such as $Hg^{2+}$, $Cu^{2+}$ and $Ag^+$. Citrate, heparin, and acetate also inhibited the enzeme. Various salts decrease the enzyme activity by 20% at ionic strength of 0.4.

Alkaline reduced water produced by mineQ showed anti-cancer and anti-diabetic effect

Special mineral combination(mineQ) was developed to produce alkaline reduced water(ARW) with high pH and low oxidation-reduction potential (ORP) and with the torsion information. ARW produced by mineQ showed anti-diabetic effectand anti-cancer effect. ARW spurs the growth of Otsuka Long-Evans Tokushima Fatty (OLETF) rats with spontaneous development of diabetic conditions, and long-term ingestion of ARW resulted in a reduction in the levels of glucose, triglycerides, and total cholesterol in the blood. When B16 melanoma cells were inoculated subcutaneously and intra-peritoneally, C56BL/6 mice fed with ARW showed tumor growth delay and thesurvival span was significantly lengthened. ARW also showed the inhibition of metastasis by reducing the numbers of B16 melanoma colonies when injected through tail vein. The amount of reactive oxygen species (ROS) was reduced when fed with ARW except for spleen, whichis a major organ for immunity. Even for normal mice, ARW intake invoked systemic cytokines, such as, Th1. (IFN-g, IL-12) and Th2 (IL-4, IL-5), suggesting strong immuno-modulation effect. Both ROS scavenging effect and immuno-modulation effect might be responsible for anti-cancer and anti-diabetic effect of alkaline reduced water.

Radioactive technetium-99m pertechnetate (<sup>99m</sup>Tc) injected to a specific acupoint flowed upward along the meridian line as was suggested by traditional meridian theory. Tracing of injected <sup>99m</sup>Tc to an acupoint revealed that the meridian is a unique ductal system through which the materials flow.
Cordyceps scarabaeicola KEFC-C252의 균사체 배양에 의한 수용성 색소의 생산과 색소의 항돌연변이 효과

Cordyceps scarabaeicola KEFC-C252 균사체의 배양으로 생산되는 천연색소의 생산조건 및 색소의 정제방법을 조사하였으며, 분리된 색소의 항돌연변이 효과를 조사하였다. 색소의 생산에 적합한 배지는 1.5% sucrose, 2.5% yeast extract, 초기 pH5.5였다. C. scarabaeicola KEFC-C252를 26℃에서 108시간 배양한 후에 색소생산이 최대에 달하였으며, 최적조건에서 C. scarabaeicola KEFC-C252는 1.2 g/liter의 색소를 생산하였다. 색소는 ethylacetate 추출, 산침전 및 결정화과정을 거치면서 정제하여 황색의 육각기둥 모양의 결정을 얻었다. 분리된 색소는 용액의 pH변화에 따라 색상이 변하는 특성을 나타내었는데 알칼리성 용액에서 청색, 산성 용액에서 붉은색을 나타내었다. 분리된 색소는 4-NQO로 유도된 돌연변이를 억제하는 항돌연변이원성 물질이었다. 또 분리된 색소는 Salmonella typhimurium TA98과 TA100에서 자발적 돌연변이를 감소시켰다. Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at 26℃ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TA100.
