Micropropagation of Hypericum erectum Thunbergby using Thidiazuron

The effect of plant growth regulators was investigated on in vitro shoot proliferation from axillary bud explantsof Hypericum erectum. To determine the optimal cytokinin for proliferation of axillay buds, we carried out screening four cytokinins(BA, kinetin, 2iP, TDZ). When nodal segments were cultured on MS medium supplemented with 4.5 μM TDZ (thidiazuron),a number of shoots were induced. Our results indicated that the addition of TDZ to culture medium resulted in theinduction of significantly more axillary buds than in the addition of other cytokinins. The optimal concentration of TDZ forproliferation of axillary buds was 10 μM. 92% of shoots spontaneously rooted without any plant growth regulator (PGR) andformed whole plantlets within one month. More than 95% of these regenerants survived and they did not show any detectablevariation in morphology or growth characteristics compared to their donor plants.

cDNA for oxidosqualene cyclase was cloned by a homology-based PCR method and sequenced from Centella asiatica. In a sequences analysis, the putative polypeptide of C. asiatica cycloartenol synthase (CaCYS) deduced from the 2,274 bp nucleotide sequence, consisted of 758 amino acids and had a molecular mass of 86.3 kD. The predicted amino acid sequence exhibited high homology to that of PNX (cycloartenol synthase) from Panax ginseng (89%). Southern blot analysis suggests that CaCYS may be present in one copy of the C. asiatica genome. If methyl jasmonate (MJ) is applied exogenously to plants, not only triterpene saponins are accumulated in tissues, but also it produces effects such as growth inhibition and the promotion of ethylene production. In order to investigate the effect of MJ and thidiazuron (TDZ), a cytokinin that plays a role as an antisenescence agent in several plants, on the level of CaCYS mRNA, we performed northern blot analysis. When MJ is alone treated by adding to culture medium, CaCYS transcripts were inhibited. However, sustained levels of the expression of CaCYS, by adding TDZ to the medium despite MJ treatments, were demonstrated in C. asiatica leaves.

The aspect of character building through physical education is an essential issues and has been rarely studied till recently. In this paper, I investigated physical education and character building, especially concentrating on the attitudes and ethics of physical education. Attitude in physical education is as important as the elements of understand and ability when evaluating students, but in fact it has not been given much value. The development and guidance of a student's attitude is very important. Moreover, it is an important duty in the instruction of physical education to transfer the socialization learned or attitudes experienced in the area of physical education and sports to general life. The development of social skills or attitudes through the area of physical education and sports is obvious, owing to the particular nature of the field. But in fact, we don't want to admit that transfer of such stills or attitudes is yet more difficult because of the particular nature of the field. The positive aspect of sportsmanship developed through modern sports is often limited to one's personal ethics or groop ethics. Moreover this fact not only leaves an individual helpless against new issues but also possibility invites other important evils. Consequently nowadays, the new principle of character building through physical education and sports, and at the same time, it needs to be constructed with a symbiotic point of view towards the environment, nature, and life. We must pay attention to the idea that we must concede the right to life of nature, to recognize the ethical responsibility of the present generation towards future generations, and to do so through a global perspective rather than personal one.

The purpose of this study was to investigate relationship between parents' child rearing attitude and physical activity of middle-school student. In order to meet purpose, the following three subsequent research questions were analyzed and these were: 1) relationship between fathers' rearing attitude and middle-school physical activity, 2) relationship between mothers' rearing attitude and middle-school physical activity. The subject of this study were 254 (127 male; 127 female) middle-school student. The data collected from questionnare designed for this study were consisted of responses to items constructed to repression analysis. The finding of this study were as follows; First, variable such as sex among sociodemographic characteristics have influenced fathers' rearing attitudes. Variables such as grade sociodemographic characteristics has influenced on technique test, theory test, and attitudes test. Second, the more fathers' rearing attitudes are affection- oriented, the more fathers' rearing attitudes has influenced on technique test, and the more fathers' rearing attitudes are rationality-oriented, the more fathers' rearing attitudes has influenceed on attitudes test. Third, the more mothers' rearing attitudes are rationality-oriented, the more mothers' rearing attitudes are rationality-oriented, the more mothers' rearing attitudes has influenceed on attitudes are rationality-oriented, the more fathers' rearing attitudes has influenceed on attitudes test. Third, the more mothers' rearing attitudes are rationality-oriented, the more mothers' rearing attitudes has influenceed on theory test.
병풀 (Centella asiatica L. Urban) 식물체 배양을 이용한 Triterpene Glycoside 생산

기내에서 병풀의 식물체 배양을 확립하였고 기본배지 설정 및 대량원소들, sucrose 농도가 triterpene glycoside (Made-cassoside와 Asiaticoside: M&A) 생산에 대한 영향을 조사하였다. 병풀 식물체가 각각의 배지에서 배양되었을 때, MS와 RCM배지는 각각 식물체 생장과 M&A 생산에 대해 가장 좋은 결과를 나타냈다. 하지만, M&A에 대해 가장 높은 생산성은 B5 배지로 나타내었다. B5 배지의 대량영양소인 KNO$_3$는 25 mM, NaH$_2$PO$_4$는 1 mM, CaC1$_2$는 1 mM 그리고 MgSO$_4$는 1~10 mM에서 M&A 생산이 최적을 이루거나 증가시켰다. Sucrose를 농도별로 처리했을 때, 6% sucrose 농도에서 가장 높은 M&A 생산을 얻을 수 있었다. Whole plant cultures of Centella asiatica (L.) Urban in vitro were established and the effects of basal media, some macro elements and sucrose concentration on productivity of triterpene glycosides (madecassoside and asiaticoside: M$\varepsilon$A) were investigated. Among the media (MS, B5, RCM) tested, MS and 0.5 RCM medium were the best for plant growth and M$\varepsilon$A production, respectively. However, taking into account the M$\varepsilon$A productivity, B5 medium was superior (M$\varepsilon$A: 14.28 mg/g dry wt.). Major macronutrients of B5 medium adjusted with the concentration of 25 mM KNO$_3$,1 mM NaH$_2$PO$_4$, 1 mM CaCl$_2$ and 1~10 mM MgSO$_4$, caused elevated or optimized levels of M$\varepsilon$A. On sucrose concentration, the highest yields of M$\varepsilon$A were obtained from 6% sucrose.
