To find active components in Ganoderma Iucidum. their free amino acid and free amino acid derivatives were analyzed. After extracting with hot water, the extracts were filtrated by three steps. So, supernatants below 10,000 dalton were obtained. Filtrates were derivativated with PITC (phenylthiocarbamyl) derivative reagent and PITC amino acid was obtained. Then, they were analyzed by RP-HPLC. 13 amino acids were analyzed in cultured Korean Nok-kak ji (one of Ganoderma Iucidum), 15 amino acids in cultured Korean Ganoderma Iucidum, 12 amino acids wild Ganoderma Iucidum, 16 amino acids in cultured Taiwan Ganodenna Iucidum.

N/A The activity of taurine transporter is affected by various extracellular stimuli such as ion, hormone and stress. To assess effects of steroid hormones and cyclosporin A (CsA) on the taurine transporter activity, murine monocytic RAW264.7 cell line was stimulated with dexamethasone (DM), triamcinolone (TA), cortisone (CS), hydrocortisone (HCS), prednisone (PSN), prednisolone (PSL) and methylprednisolone (MPSL) in the presence of 12-O-tetradecanoylphorbol-l3-acetate (TPA). Treatment of TPA on the cell line led to significant reduction of taurine transporter activity. However, in case of stimulation of the cells with steroid hormones in the presence of TPA, all of them recovered TPA-induced reduction of the taurine transporter activity. Treatment of the cells with CsA led to significant reduction of the taurine transporter activity. Ionomycin (IM) recovered the reduced taurine transporter activity by CsA, but failed in the presence of EDTA, a calcium chelating agent. These results showed that glucocorticoid hormone recovered TPA-induced reduction of taurine transporter activity and that IM recovered CsA-induced reduction of the transporter activity by increasing intracellular free Ca^(++) concentration.

Antitumor effect of LC43, a protein-bound ploysaccharide (M.W. 43 kDa) that was purified from intergeneric protoplast fusant of Lentinus edodes and Coriolus versicolor, was elucidated against mouse sarcoma 180 cell in vitro and in vivo. By injecting LC43 into ICR mice bearing solid or ascitic sarcoma 180, tumor regression and survival rates were investigated. To examine the effects of LC43 on immunopotentiation activity, immunoorgan weight, B cell differentiation, T cell activity and macrophage activation were determined. LC43 showed antitumor effects against both solid tumor and ascitic tumor of sarcoma 180. It did not change significantly the immunoorgan weight but potentiated immune responses such as B cell differentiation and the release of superoxide anion from macrophages. These results suggest that the protein-bound polysaccharide of LC43 exhibited antitumor activities through the activation of immune-related cells and acted as an immun-modulator.

It has been known that Ganoderma lucidum and Lentinus edodes have anticancer activity and immune enhancing activity. These two mushrooms were grown in liquid culture and harvested. From these<br/> mycelia, DNA was isolated and EtBr-CsCl density gradient ultracentrifugation was performed to purify it further. Then mitochondrial DNA was isolated by bisbenzimide-CsCl density ultracentrifugaton. Mitochondrial DNA of Ganoderma lucidum was digested by restriction enzymes, EcoR I, Hind III, and Pst I, then electrophoresed. It showed 12, 22, 4 fragments. Mitochondrial DNA of Lentinus edodes was digested by EcoR I. Electric pattern showed 6 fragments. 4 fragments had appeared by Pst 1 digested mitochondrial DNA. Hind III couldn't digest mitochondrial DNA of Lentinus edodes. Mitochondrial DNA of fusants was isolated to compare to those of parents. The results showed that fusant P₂S₂ has new, recombined mitochondrial DNA. But P₂S₂ had the same<br/> DNA that Ganoderma lucidum had.
영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향

영지버섯 균사체의 ethanol 용성분획을 극성에 따라 6개의 fraction으로 나누었다. 사염화탄소와 galactosamine으로 독성을 유도한 일차배양 간세포를 이용하여 검색한 결과 전반적으로 열수출물분획과 n-butanol 분획의 극성분획이 GPT, GOP 값을 가장 크게 저하시켜 간 보호작용을 나타냈으며, 부분 간 절제수술 후의 일차배양 간세포에 대해 열수추출물, 고분자분획, n-butanol 분획이 유의성있게 DNA, RNA, Protein 합성을 증가시켜 손상된 간 조직에 대해 회복 효과를 보였다. 간암세포에 대해서는 ethyl acetate 분획이 가장 강한 세포독성을 나타내었다. The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl_4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rat primary cultured hepatocytes with [^3H]-thymidine, [^3H]-uridine and [^3H]-leucine, the high-polarity fractions such as aqueous, high-molecular weight and n-butanol fractions increased synthetic activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma (SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 ㎍/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.
