        AhR 발현을 통한 콤부차 분획물의 안티폴루션 효과 연구

        본 연구에서는 설탕을 넣은 녹차나 홍차에 유익균을 첨가해 발효시킨 음료인 콤부차를 컬럼 크로마토그래피를 이용해 분획한 후, TLC를 통해 플라보노이드류인 퀘르세틴 글리코사이드(quercetin glycoside)의 유효성분 유무를 확인하고 그 분획물을 이용하여 공해로부터 피부를 보호 및 개선하는 안티폴루션 효과를 확인하였다. 인간 각질형성세포에 Kombucha-quercetin glycoside (K-QG)를 처리하여 세포 생존율을 확인한 결과, 100 ㎍/mL 농도까지 90% 이상의 생존율을 보였으며, 안티폴루션 효과를 보기 위해 benzo[e]pyrene과 미세먼지 자극에 의한 세포 생존율을 측정한 결과, 100 ㎍/mL에서 각각 68.79%, 50.68%의 개선율을 보였다. 또한, benzo[a]pyrene에 의해 활성화되는 aryl hydrocarbon receptor (AhR) 발현을 웨스턴 블롯을 통해 확인한 결과, 대조군에 대비하여 100 ㎍/mL에서 31.08%의 억제율을 보였다. 본 연구의 결과를 통해 K-QG는 벤조피렌과 미세먼지로부터 자극받은 피부를 보호, 개선해주며 안티폴루션 기능성 소재로써 가치가 있는 것으로 사료된다. In this study, Kombucha, a fermented drink by adding beneficial bacteria to green tea or black tea with sugar, was classified using column chromatography, and the presence or absence of quercetin glycoside, a type of flavonoids, was confirmed through TLC. In addition, the anti-pollution effect of protecting and improving the skin from pollution was confirmed using the fraction. Keratinocytes were treated with K-QG to confirm the cell viability, showing a survival rate of 90% or more up to a concentration of 100 ㎍/mL, and the cell viability by benzo[e]pyrene and fine dust stimulation to see the anti-pollution effect was measured. At 100 ㎍/mL, the improvement rates were about 68.79% and 50.68%, respectively. In addition, as a result of confirming the expression of AhR activated by benzo[a]pyrene through Western blot, it showed an inhibition rate of about 31.08% at 100 ㎍/mL compared to the control group. Through the results of this study, K-QG is thought to be valuable as an anti-pollution functional material, protecting and improving skin irritated from benzopyrene and fine dust.

        최근 IT기술발달로 스마트헬스 산업이 성장하면서, 의약품을 복용하는 사람들에게 편리함을 가져다 줄 수 있는 서비스들이 쏟아져 나오고 있다. 하지만 사용자의 유형을 체계적으로 분류하여 알맞은 서비스를 제안한 연구는 미흡한 실정이다. 이러한 문제점을 해결하기 위해 의약품 복용 경험이 있는 참가자를 대상으로 심층인터뷰를 진행하였다. 이를 통해 본 논문에서는 사용자를 4가지 유형으로 분류하고, 각각의 니즈(needs)를 도출하여 2가지 의약품 서비스 콘셉트를 제안하였다.

        찔레꽃 추출물의 RAW 264.7 세포에 대한 항염증 및 항산화 활성

        Studies on antioxidant activity, antibacterial activity and antiseptic effect have been reported on Rosa muliflora flower, but research on anti-inflammatory activity is insufficient. The purpose of this study was to confirm the possibility of using an extract of Rosa multiflora flower as an antioxidant and anti-inflammatory functional material. R. multiflora flower was extracted with hot water, 70%v/v ethanol, and 70%v/v acetone solvents. Polyphenol and flavonoid content, DPPH and ABTS radical scavenging ability, SOD-like activity, cytotoxicity (MTT assay with RAW 264.7 macrophages), and NO inhibitory activity were analyzed. Anti- inflammatory was analyzed by, Western blotting. The polyphenol content of R. multiflora flower water extract (RMW) was 236.58 mg/g; that of 70% ethanol extract (RME) was 198.15 mg/g; that of 70% acetone extract (RMA) was 216.97 mg/g. The flavonoid content was 16.91 mg/g for RMW, 7.80 mg/g for RME, and 20.24 mg/g for RMA. The DPPH radical-scavenging ability was highest in RME (59.88%); the ABTS radical–scavenging ability was highest in RMA (99.1%); and the SOD-like activity was highest in RMW (62.04%). Cell viability was determined by setting the concentration range for a survival rate of 90% or more. RME showed the highest inhibitory activity (70.86%). Further, iNOS protein activity was inhibited by 53.94% by RME, and that of COX-2 protein by 61.48% by RMA. Collectively, the R. multiflora flower extract has excellent potential for use as a functional anti-inflammatory and antioxidant food material.

        님트리(Azadirachta indica)잎 에탄올추출물의 약리활성 및 안티폴루션 효능 검증

        In this study, we investigated the anti-oxidant, anti-inflammatory, whitening, anti-wrinkle, and anti-pollution effects of Neem tree (i.e., Azadirachta indica leaf) 70% ethanol extract. More specifically, the polyphenol content was 51.37 mg/g, and the DPPH and ABTS radical scavenging activities were 34.09 and 72.18%, respectively, at a final concentration of 1,000 μg/mL. SOD experiments showed an effect of 24.13% at a final concentration of 1,000 μg/mL. In addition, to investigate the whitening effect, the tyrosinase inhibition effect was measured, giving a value of 47.21% at a final concentration of 1,000 μg/mL. Furthermore, MTT assay experiments were carried out on macrophages (RAW 264.7) to determine the cytotoxicity using the neem tree leaf extract, it was confirmed that the cell viability was unaffected up to a concentration of 100 μg/mL. Moreover, anti-inflammatory (NO) analysis of the cells using macrophages stimulated with LPS induced an inflammatory response of 68.86%, in addition to an anti-inflammatory activity of 66.45% at a final concentration of 100 μg/mL. The iNOS protein showed an inhibition rate of 97.49% at a concentration of 100 μg/mL, and in the cytotoxicity assay (MTT assay) using keratinocytes (HaCaT), the cell viability was similar to that of the untreated control at 10 μg/mL. Western blotting analysis confirmed the inhibition rate of MMP-1. Additionally, HaCaT and CCD-986sk human skin cells were stimulated with benzo[e]pyrene, which contains fine dust and heavy metals, and then treated with neem tree extract to measure the effect of the extract on the cell viability. Indeed, the anti-pollution efficacy of the extract was confirmed. Overall, the benzo[e]pyrene-stimulated cells presented improvements of >92 and 81% for the HaCaT and CCD cells at a concentration of 25 μg/mL, while improvements were observed at a concentration of 100 μg/mL HaCaT for fine dust-stimulated cells. Moreover, HaCaT cell and CCD cells showed improvements of more than 85 and 76%, respectively. The obtained results therefore indicate that Neem tree 70% ethanol extract is expected to have value for application in cosmetics due to its pharmacological activity and its fine dust-blocking properties.

        알루미늄 박 및 플레이트 표면 미세 패터닝을 위한 상온 임프린팅 기술

        Nanoimprint lithography (NIL) has attracted much attention due to its process simplicity, excellent patternability, process scalability, high productivity, and low processing cost for pattern formation. However, the pattern size that can be implemented on metal materials through conventional NIL technologies is generally limited to the micro level. Here, we introduce a novel hard imprint lithography method, extreme-pressure imprint lithography (EPIL), for the direct nano-tomicroscale pattern formation on the surfaces of metal substrates with various thicknesses. The EPIL process allows reliable nanoscopic patterning on diverse surfaces, such as polymers, metals, and ceramics, without the use of ultraviolet (UV) light, laser, imprint resist, or electrical pulse. Micro/nano molds fabricated by laser micromachining and conventional photolithography are utilized for the nanopatterning of Al substrates through precise plastic deformation by applying high load or pressure at room temperature. We demonstrate micro/nanoscale pattern formation on the Al substrates with various thicknesses from 20 μm to 100 mm. Moreover, we also show how to obtain controllable pattern structures on the surface of metallic materials via the versatile EPIL technique. We expect that this imprint lithography-based new approach will be applied to other emerging nanofabrication methods for various device applications with complex geometries on the surface of metallic materials. 나노임프린트 리소그래피(Nanoimprint lithography, NIL) 공정은 패턴 형성을 위한 공정 단순성, 우수한 패턴 형성, 공정의 확장성, 높은 생산성 및 저렴한 공정 비용이라는 이유들로 인해 많은 관심을 받고 있다. 그러나, 기존의 NIL 기술들을 통해 금속 소재 상 구현할 수 있는 패턴의 크기는 일반적으로 마이크로 수준으로 제한적이다. 본 연구에서는, 다양한 두께의 금속 기판 표면에 마이크로/나노 스케일 패턴을 직접적으로 형성하기 위한 극압 임프린트 리소그래피(extremepressure imprint lithography, EPIL) 방법을 소개하고자 한다. EPIL 공정은 자외선, 레이저, 임프린트 레지스트 또는 전기적펄스 등의 외부 요인을 사용하지 않고 고분자, 금속, 세라믹과 같은 다양한 재료의 표면에 신뢰성 있는 나노 수준의 패터닝을 가능하게 한다. 레이저 미세가공 및 포토리소그래피로 제작된 마이크로/나노 몰드는 상온에서 높은 하중 혹은 압력을가해 정밀한 소성변형 기반 Al 기판의 나노 패터닝에 활용된다. 20 μm 부터 100 μm까지 다양한 두께를 갖는 Al 기판 상마이크로/나노 스케일의 패턴 형성을 보여주고자 한다. 또한, 다목적 EPIL 기술을 통해 금속 재료 표면에서 그 형상을 제어하는 방법 역시 실험적으로 증명된다. 임프린트 리소그래피 기반 본 접근법은 복잡한 형상이 포함된 금속 재료의 표면을요구하는 다양한 소자 응용을 위한 나노 제조 방법에 적용될 수 있을 것으로 기대한다.

      • 모바일 뉴스 제보 애플리케이션의 UI 개선안 제안

        본 연구는 우리나라 케이블방송 TV 뉴스를 대표하는 YTN 의 모바일제보시스템 UI 개선방향을 제시하고 있다. 온라인 설문과 추가 인터뷰 데이터 등을 통해 문제점을 분석하고, 사용자들이 더욱 효과적으로 제보와 제보상태에 대해 빠르게 인지하여 효율적인 제보를 할 수 있도록 YTN 애플리케이션 모바일 제보서비스를 개선하였다. This paper proposes the way to improve user interface of YTN, the key cable broadcasters, mobile report service. Based on analysis of problems through data collected from the online questionnaire and follow-up interviews, we improved UI of YTN mobile report services more effective and easy with highly cognitive state.

        금화규(Abelmoschus manihot) 꽃 추출물의 항산화 활성과 총페놀 및 플라보노이드 함량 분석

        In this study, we analyzed the antioxidant activity, total phenol, and flavonoid content of Abelmoschus manihot flowers by extracting, concentrating, and freezing the flower site with water and a 70% ethanol solution. The hydrothermal extract (AMW) and the 70% ethanol extract (AME) showed activities of ≥ 77% and ≥ 95% at 100 μg/mL, respectively, in ABTS+ radical scavenging assay. Electron donating availability experiments showed activities of AMW and AME of > 86% and ≥ 91% at 1,000 μg/mL, respectively. In the ferric reducing antioxidant power (FRAP) assay, AME had a reduction force of approximately twice that of AMW at 1,000 μg/mL; moreover AME exhibited reduction force higher than that of AMW. In the SOD-like availability asset, both AMW and AME showed low activities. Total phenol and flavonoid contents were higher in AME than in AMW, although both have similar contents. As a result, AME exhibited higher antioxidant activity than AMW, while their phenol and flavonoid contents were similar. Therefore, A.manihot flower extracts can be considered good natural biological resources for use as raw materials for food and cosmetics.
