      • Streptozotocin 투여 용량에 따른 당뇨병성 마우스의 신경전도도 변화에 대한 연구

        본 연구는 streptozotocin 유발 type 1 당뇨병 mouse 동물 모델에서 당뇨병성 신경병증과 streptozotocin 용량과의 상관관계를 규명하고자 하였다. 구체적으로 streptozotocin의 용량을 50mg/kg, 100mg/kg, 150mg/kg 으로 투여하여 mouse에서 당뇨병을 유발하였고, 31일 경과 후 mouse 좌골신경에서 운동신 경 및 감각신경의 신경전달 속도를 측정 하여 당뇨병성 말초신경병증의 정도를 평가하였다. Streptozotocin 투여용량별 체중변화는 streptozotocin을 투여하지 않은 그룹과 비교하여 streptozotocin을 투여한 그룹들의 체중이 감소했고 streptozotocin의 용량증가에 따라 체중 감소율은 유의적으로 증가하였다. Streptozotocin 투여용량에 따른 평사 (non fasting)와 공복시(fasting) 의 혈당은 streptozotocin의 투여용량이 증가할수록 혈당은 증가하는 것으로 나타났다. Streptozotocin 투여용량에 따른 신경전달속도 변화를 평가 사streptozotocin 투여 31일 경과시 감각신경의 신경 전달속도는 의미 있는 차이가 나타나지 않았으나 운동신경의 신경전달속도는 streptozotocin올 투여한 그룹에서 유의미하게 감소를 나타냈고 streptozotocin 용량의존적으로 감소하는 경향을 보여주었다. 따라서 운동신경 및 감각신경 양자 모두에서 신경전달속도의 뚜렷한 감소를 확인하기 위해서 는 streptozotocin 투여 후 고혈당증의 지속기간이 좀 더 필요한 것으로 사료된다.

      • 톳【Hijikia fusiforme (Harvey) Okamura】이 Streptozotocin 유발 당뇨성 흰쥐의 혈당 및 지질성분에 미치는 영향

        Streptozotocin (55㎎/㎏ of body weight, I.P. injection)으로 인한 당뇨 유발 Sprague Dawley계 숫흰쥐에 있어서 톳녹즙액의 급여가 혈당 및 혈청의 지질개선효과를 규명하기 위해 4주간 실험 사육한 결과, 혈당 농도는 대조군인 1군(물)에 비해 Streptozotocin 투여군(2군, 3군, 4군, 5군)에서 유의적으로 높았으나, 2군(Streptozotocin+물)에 비해서 3군(Streptozotocin+톳녹즙액)이, 4군(Streptozotocin+콜레스테롤+물)보다 5군(Streptozotocin+콜레스테롤+톳녹즙액)이 낮았다. 혈청 총콜레스테롤 및 LDL-콜레스테롤 농도는 1군이 가장 낮았고 2군에 비해 3군이 유의적으로 낮았고, 4군보다 5군이 약간 낮게 나타났다. 혈청 HDL-콜레스테롤 농도는 1군이 가장 높았으나, 2군과 4군에 비해 톳녹즙액 급여군인 3군과 5군에서 각각 높게 나타났다. 총콜레스테롤에 비해 HDL-톨레스테롤 농도비는 톳녹즙액 급여군이 높게 나타났다. 동맥경화지수는 2군에 비해 톳녹즙액 급여군인 3군이 낮았다. 혈청 유리콜레스테롤 및 콜레스테롤 에스테르 농도는 톳녹즙액 급여군)3군, 5군)에서 약간 감소하였으며, 혈청 중성지질농도는 2군에 비해 3군이, 4군보다 5군이 유의성있게 감소하였다. 인지질의 농도 또한 2군보다 3군이 낮은 경향을 보였다. 이상의 결과에서 톳 녹즙액이 당뇨병 흰쥐에 대한 혈당 및 혈청지질 저하 효과가 있는 것으로 나타났다. This study was designed to observe the effects of the feeding Hijikia fusiforme(Harvey) Okamura juice extract on the improvement of the blood glucose, lipids in the serum of sterptozotocin (55㎎/㎏ of body weight, I.P. injection) induced hyperglycemic rats (S.D. strain, ♂)fed the experimental diets for 4 weeks. Concentrations of blood glucose were significantly higher in the streptozotocin administration(I.P.) groups(groups 2, 3, 4 and 5) than those in the control group(group 1, basal diet+water). Blood glucose concentrations were lower in the group 3(streptozotocin+Hijikia fusiforme). than in the group 2(streptozotocin+water), and then those of the group 5(streptozotocin+cholesterol+Hijikia fusiforme) were lower than in the group 4(streptozotocin+cholesterol+water). Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the group 3 than those in the group 2, and the group 5 were rather lower than in the group 4. Concentrations of HDL-cholesterol in serum were the highest in the group 1. Those in the groups 3, 5 were higher than those in the groups 2, 4. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, Hijikia fusiforme juice extract administration groups were higher percentage than the other groups. Atherosclerotic index were lower in the group 3 than those in the group 2. Concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3, 5 than the other groups. Triglyceride concentrations in serum were significantly lower in the groups 3, 5 than those in the groups 2, 4. Concentrations of phospholipid in serum were decreased in the group 3 than in the group 2. From the above research, the Hijikia fusiforme juice extracts were effective on the improvement of the blood glucose and lipid compositions in serum of streptozotocin induced hyperglycemic rats.

        저강도 treadmill 운동이 streptozotocin 유도 당뇨쥐의 혈당, 인슐린 및 지질 농도에 미치는 영향

        본 연구는 streptozotocin으로 당뇨병을 유도하는 조건들을 농도에 따른 변화를 관찰하고, 적절한 조건에서 당뇨쥐에서의 저강도의 treadmill 운동이 체중과 혈중의 당, 인슐린 및 지질의 변화에 미치는 영향을 알아보고자 실시하였으며, 연구결과를 요약하면 다음과 같다. 실험 I에서 STZ 주사량이 40 mg/mg으로 높아질수록 정상군에 비해 혈당이 유의하게 증가하고, 반대로 체중과 인슐린 농도는 상대적으로 감소하였다. 실험II에서 STZ 주사를 한 모든 군에서 혈당이 유의하게 증가하지만 BE군은 C군에 비해 그 증가율이 유의하게 억제되었으며, 운동이 인슐린 농도와 혈중지질 농도를 정상적으로 유도하는 것으로 나타났다. 특히, 당뇨 유발 전부터 운동을 실시한 경우 그 효과가 더욱 유의하게 나타났으며, 앞으로 당뇨 유발의 정도와 운동 빈도 및 강도와 관련된 더욱 많은 연구가 이루어져야 할 것으로 사료된다. Exercise is beneficial to the diabetic patients and streptozotocin-induced diabetic rat has been used for the study of exercise effect. The purpose of this study was to establish the optimal condition of induction of hyperglycemic diabetic rat using streptozotocin and to examine the preventive effect of treadmill exercise on the diabetic rat before and after streptozotocin injection. Intraperitoneal injection of increasing amount of streptozotocin up to 40 mg/kg dose-responsively induce hyperglycemic diabetic rat and inversely reduced the blood insulin level. Body weight was also gradually reduced with the increasing amount of streptozotocin. Control and diabetic rats exercised for 4 weeks before streptozotocin injection. The exercise was performed in the treadmill for 25 minutes a day and 5 times a week with low intensity (0 degree tilt, 15 m/min velocity). Following streptozotocin injection, the blood glucose level was measured every week and the rat was sacrificed after 4 weeks to measure the concentration of insulin and blood lipids. The blood levels of glucose and insulin was significantly reduced with exercise before streptozotocin injection, while those were not changed after streptozotocin injection. The levels of blood lipids such as total cholesterol, HDL-cholesterol, LDL-cholesterol and triglyceride were close to normal control rats. From this study, researchers found the optimal condition of preparation of streptozotocin-induced hyperglycemic diabetic rat, and the mild treadmill exercise has beneficial effect on preventing hyperglycemia and hyperlipidemia. Thus, even low intensive running prevent not only diabetes but also diabetic vascular complications.

        Streptozotocin 유도 당뇨 Rat에 한약제제의 급여가 혈당 및 지질 농도에 미치는 영향

        Objectives: To examine the effect of Oriental herb medicines on serum glucose and lipid concentration in streptozotocin-induced diabetic rats. For this study, the materials concerned with the useful Oriental herb medicines for Diabetes MellitudDM) were collected at the clinic of Daegu Haany University. Methods: To verify the effects of Oriental herb medicines on serum glucose through streptozotocin-induced diabetic rats, some rats were injected 1 time with 50 mghg streptombcin intraperitoneally(1.P.) to induce diabetes, and other rats were injected with buffer I.P. as control group. Results: The concentration of serum glucose in SZ(streptozotocin) plus Oriental herb medicines extract group was significant decrease than SZ(streptozotocin) control group(p<0.05). The concentration of cholesterol in SZ(streptozotocin) plus Oriental herb medicines extract group did not shown significant decrease than SZ(streptozotocin) control group(p<0.05). Also the concentration of triglycende in SZ(streptozotocin) plus Oriental herb medicines extract group did not shown significant decrease than SZ(streptozotocin) control group(p<0.05). Conclusion: These results suggest that Oriental herb medicines extract used in this experimental could be applied to the treatment of diabetes mellitus.

        흰쥐에서 streptozotocin에 의해 유도된 당뇨병에 대한 운동과 melatonin의 효과

        The effects of melatonin and exercise on diabetes mellitus induced by streptozotocin in male Sprague-Dawley rats were studied. Diabetes was induced by the intraperitoneal injection of 65 mg of streptozotocin per kg of body weight. To know the effect of melatonin, a dose of 10 mg of melatonin per kg of body weight was administered intraperitoneally. And to know the effect of exercise, rats were performed exercise by a treadmill running at the speed of 10 m/minute, 8% of slope, for 60 minutes in a day for 1 week. The parameters including the levels of blood glucose, hemoglobin A1C, and malondialdehyde analysed to evaluate diabetic state and the degree of lipid peroxidation by oxygen free radicals. The injection of streptozotocin caused significant increases in the levels of blood glucose, hemoglobin A1C and malondialdehyde. Melatonin and exercise reduced significantly the levels of blood glucose, hemoglobin A1C and malondialdehyde which were increased by streptozotocin injection. When the effect of melatonin and exercise in the diabetic rats was compared the exercise was more effective for normalizing the parameters which were changed by streptozotocin injection than melatonin. These results may suggest that melatonin and exercise are effective the oxidative damage in the diabetes induced by streptozotocin. Exercise is more protective for diabetes mellitus than melatonin.

        Streptozotocin 유발 당뇨 백서에 상엽 지각(桑葉 枳殼) 혼합물의 항당뇨 및 혈관탄성개선 효과

        Objectives : This study was designed to evaluate the anti-diabetes and vasoelasticity effects of Mori Folium and Aurantii Fructus in streptozotocin-induced type II diabetes mellitus model. Methods : The anti-diabetic effect of Mori Folium and Aurantii Fructus on rats induced with diabetes by streptozotocin was investigated through analyses of changes in body weight, blood glucose, urine volume of rats, viability of human umbilical vein endothelial cells(HUVECs), and elasticity of descending thoracic aorta in rats. The subjects in this study were divided into four groups(n=15): a normal group without any treatment (Con), a normal group with Mori Folium and Aurantii Fructus treatment(Con+P), a diabetes group induced by streptozotocin(STZ), and a Mori Folium and Aurantii Fructus treatment group under diabetes induced by streptozotocin(STZ+P). Rats were administered streptozotocin to induce diabetes. Results : The study showed that Mori Folium and Aurantii Fructus significantly reduced highly increased blood glucose levels(p<0.01) and prevented the diabetic rats from weight loss(p<0.01) and polyurea(p<0.05), Mori Folium and Aurantii Fructus also recovered decreased viability of HUVECs(p<0.01) and damaged elasticity of aorta induced by the streptozotocin (p<0.01). Conclusions: It was concluded from the results that Mori Folium and Aurantii Fructus have a distinct anti-diabetes effect and they also prevent damage of blood vessel induced by diabetes. resulting in prevention of cardiovascular diseases ascribed to diabetes.

        Streptozotocin을 이용한 제1형 당뇨 유발 흰쥐에서 KIOM-79의 효과

        We investigated the effect of KIOM-79, 80% ethanolic extract of herbal prescription isolated from Magnolia officinalis, Pueraria lobate, Glycyrrhiza uralensis, and Euphorbia pekinensis, on streptozotocin-induced diabetes in Sprague-Dawley rats. The rats were treated orally with KIOM-79 (500 mg/kg/day) 1) for 3 days prior to streptozotocin (60 mg/kg, intraperitoneal) injection or 2) for 9 weeks after establishing diabetes model to examine acute and chronic effects on hyperglycemia and biochemical variables, respectively. As a result, KIOM-79 had little effects on hyperglycemic changes in acute model. Sexual comparison, however, showed reduced hyperglycemia in female rats, especially 24 hours after streptozotocin injection with or without KIOM-79 pretreatment. In chronic model, streptozotocin-induced hyperglycemia was well established, but KIOM-79 treatment showed no statistically significant effects on all variables. Thus, based on our findings KIOM-79 might have little effects on streptozotocin-induced insulin-dependent diabetes although it has been known to have hypoglycemic and antidiabetic effects on non-insulin-dependent diabetes models.

        트레드밀 운동과 타우린 섭취가 streptozotocin 유도 당뇨쥐의 체중, 혈당, 인슐린 및 콜레스테롤 농도에 미치는 영향

        Exercise and taurine has been known to be benficial to the diabetic patients. But it combinatory effect has not been studied. This study aims to understand whether taurine intake has a repressive effect on blood levels of glucose, insulin and lipids of the streptozotocin-induced hyperglycemic diabetic rat, and also whether combinational treatment of taurine intake and low-intensity exercise has a synergistic repressive action. Taurine was freely supplied as 1 % solution. The blood glucose level was not suppressed in the taurine-intake diabetic rat, but the blood levels of insulin and lipid such as total colesterol, HDL-cholesterol, LDL-cholesterol and triglyceride was reverted. Exercise (25 mill/day, 5 time/week,15 m/min) alone has a repressive effect on the hyperglycemic index. The combinatory effect of taurine and exercise before and after streptozotocin injection was examined. When taurine was supplemented with exercise before streptozotocin injection, the blood glucose level was surprisingly unchanged. but the blood levels of insulin and lipids were mildly recovered. When taurine was supplemented with exercise after streptozotocin injection, the blood glucose level was also unchanged, but the blood levels of insulin and lipids were mostly recovered. These results suggest that taurine itself has no beneficial effect on the blood glucose level and no combinatory effect with exercise, although it has a positive effect on the blood insulin levels and also serum lipids.

      • Therapeutic potential of cyanobacteria against streptozotocin-induced diabetic rats

        <P>Hypoglycemic effect of cyanobacteria has evaluated in the normal and streptozotocin-induced diabetic male albino rats as a mammalian model. Normal and streptozotocin-diabetic rats were orally administered cyanobacteria for 60 consecutive days, and their blood levels of glucose, insulin, C-peptide, lipid peroxidation, body weight and enzyme changes were determined using standard methods. Cyanobacteria administration reduced blood glucose level, and increased plasma insulin, C-peptide levels and restored the body weight. Cyanobacteria administration significantly reduced lipid peroxidation in the diabetic rats. Hexokinase enzyme activity was increased, whereas glucose-6-phosphatase enzyme activity decreased in streptozotocin-diabetic rats compared to their respective controls. Cyanobacteria administration caused significant renormalization of serum hepatic enzymes in streptozotocin-induced diabetic rats. In conclusion, cyanobacteria have a protective effect on anti-oxidant and anti-diabetic in streptozotocin-induced diabetic rats.</P>
