        넓패(Ishige foliacea)의 최종당화산물 생성저해 활성 및 신장세포 보호 효과

        본 연구에서 국내 자생 갈조류 중 하나인 넓패를 이용하여 추출방법에 따른 생리활성 물질의 함량을 정량적으로 분석하였다. 또한 당뇨병성신장증의 합병증을 유발하는 물질로 알려진 최종당화산물의 생성 억제 효과, 최종당화산물과 콜라겐 사이의 교차결합 형성 억제 효과 및 교차결합 절단 효과를 평가하였다. 넓패 70% 에탄올 추출물은 총 페놀, 총 플라보노이드 및 탄닌과 같은 생리활성 성분을 가장 많이 함유하는 추출 조건임을 확인하였으며, 최종당화산물의 생성 억제, 최종당화산물과 콜라겐 사이의 가교결합 형성 저해 및 생성된 가교결합을 절단하는 효과 또한 가장 높았다. 특히, 70% EtOH IFE는 양성대조군으로 처리한 AG보다 더 높은 교차 결합 억제 및 교차결합 절단 효과를 나타내는 것을 확인할 수 있었다. 또한 70% EtOH IFE는 MGO에 의해 유도되는 산화적 스트레스로부터 신장 세포인 mesangial 세포의 세포 내 산화적 스트레스 수준을 유의적으로 감소시킬 뿐만 아니라 apoptosis로 인한 세포 사멸을 감소시켜 신장 손상을 보호하는 효과를 보였다. 따라서 넓패는 부작용이 적은 당뇨합병증 예방 및 치료제 개발 후보물질로서 가능성이 충분한 것으로 판단되며, 향후 국내 자생 해양자원의 가치 향상을 위해 넓패의 생리활성 성분의 동정 및 당뇨합병증 예방과 치료에 작용하는 메커니즘 구명 연구가 추가적으로 진행되어야 할 것으로 사료된다. The present study was conducted to investigate the anti-glycation activities and renoprotective properties of Ishige foliacea. The yield of extract ranged from 3.20 to 35.59%. Among the extracts, 70% aqueous ethanol extract of Ishige foliacea (70% EtOH IFE) showed the highest total phenolic, condensed tannin, and total flavonoid contents (1.570 ± 0.165 mg GAE/g DW, 0.407 ± 0.008 mg CE/g DE, 0.353 ± 0.050 mg CE/g DE, respectively). 70% EtOH IFE showed the highest breaking activity against preformed AGEs-BSA cross-link than ALT-711, the well-known cross-link breaker. In addition, 70% EtOH IFE observedthe highest inhibitory effect on the AGEs-BSA cross-link and exhibited more potent than aminoguanidine, a well-known AGEs-BSA cross-link inhibitor. Treatment with the 70% EtOH IFE, at 2, 10, and 50 μg/mL, reduced oxidative stress to the level of approximately 186.3-95.9%. The intracellular antioxidative effect of the 70% EtOH IFE was stronger to that with amino-guanidine at 500 μM. Pre-treatments of cells with the 70% EtOH IFE at concentrations of 2, 10, and 50 μg/mL led to 60.3%, 38.0%, and 20.8% decrease in apoptotic cells, respectively. These result suggest that 70% aqueous ethanol extract of Ishige foliacea can serve as a good source of natural anti-diabetic complications agents via breaking and inhibition of AGEs-BSA cross-link and renoprotective effects against MGO-induced oxidative stress.

        넓패(Ishige foliacea) 추출물의 생리활성 탐색

        Ishige foliacea belongs to class Phaeophyceae and family Ishigeaceae. This study investigated the physiological activities of the Korean marine algae I. foliacea. Its solvent extracts were prepared with 70% ethanol, 80% methanol, and distilled water. The ethanol and methanol extracts had higher α-glucosidase (half-maximal inhibitory concentration, IC50: 0.67–0.73 mg/mL), xanthine oxidase (IC50: 0.25–0.28 mg/mL), and angiotensin-converting enzyme (IC50: 25.29–38.28 μg/mL) inhibitory activities than those of the water extract. The ethanol and methanol extracts possessed high acetylcholinesterase inhibitory activity (IC50: 0.78–0.97 mg/mL). Conversely, the water extract exhibited the highest β-secretase inhibitory activity (IC50: 0.48 mg/mL). These results indicate that I. foliacea may be useful as a functional substance in food and pharmaceuticals with anti-diabetic, anti-gout, anti-hypertension, and anti-dementia properties.

      • CLASSIFICATION OF THE GENUS <i>ISHIGE</i> (ISHIGEALES, PHAEOPHYCEAE) IN THE NORTH PACIFIC OCEAN WITH RECOGNITION OF <i>ISHIGE FOLIACEA</i> BASED ON PLASTID <i>rbc</i>l AND MITOCHONDRIAL <i>cox</i>3 GENE SEQUENCES<sup>1</sup>

        <P>The taxonomy and biogeography of a genus with species that occur in geographically isolated regions is interesting. The brown algal genus <I>Ishige</I> Yendo is a good example, with species that apparently inhabit warm regions of both the northwestern and northeastern Pacific Ocean. We determined the sequences of mitochondrial <I>cox</I>3 and plastid <I>rbc</I>L genes from specimens of the genus collected over its distributional range. Analyses of the 86 <I>cox</I>3 and 97 <I>rbc</I>L sequences resulted in congruent trees in which <I>Ishige sinicola</I> (Setch. et N. L. Gardner) Chihara consisted of two distinct clades: one comprising samples from Korea and Japan, and the other comprising samples from the Gulf of California. Additional observations of the morphology and anatomy of the specimens agree with the molecular data. On the basis of results, we reinstated <I>Ishige foliacea</I> S. Okamura (considered a synonym of <I>I. sinicola</I> from the Gulf of California) for plants from the northwest Pacific region and designated a specimen in the Yendo Herbarium (SAP) as the lectotype. <I>I. foliacea</I> is distinguished by large (up to 20 cm) and wide (up to 20 mm) thalli, with a cortex of 4–7 cells, and a medulla composed of long, tangled hyphal cells. Both <I>cox</I>3 and <I>rbc</I>L sequence data strongly support the sister-area relationship between the northwest Pacific region and the Gulf of California. A likely explanation for this pattern would be the presence of a species ancestral to contemporary species of <I>Ishige</I> in both regions during the paleogeological period, with descendants later isolated by distance.</P>

      • Efficient approach to purification of octaphlorethol A from brown seaweed, <i>Ishige foliacea</i> by centrifugal partition chromatography

        <P><B>Abstract</B></P> <P>The brown seaweed, <I>Ishige foliacea</I> is distributed along the coast of Jeju Island. The main compound that this seaweed produces, octaphlorethol A (OPA), has been recently reported to exhibit strong antidiabetic and whitening activities. However, obtaining pure OPA requires repetitive complex processes and its antioxidant effect <I>in vivo</I> has not been demonstrated. Therefore, we devised a method to efficiently detect and isolate OPA from <I>I. foliacea</I> using 2.2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS<SUP>+</SUP>) on-line HPLC and centrifu-gal partition chromatography (CPC), respectively. And we evaluated its antioxidative properties using zebrafish as an <I>in vivo</I> model system. OPA among compounds from <I>I. foliacea</I> ethyl acetate (EtOAc) fraction had the lowest absorbance in ABTS<SUP>+</SUP> on-line HPLC. A total of 11.2mg OPA was rapidly isolated from the EtOAc fraction (500mg) of <I>I. foliacea</I> extract by using CPC with a two-phase solvent system. In particular, OPA strongly inhibited reactive oxygen species and lipid peroxidation in 2.2′-azobis (2-amidinopropane) dihydro-chloride-induced zebrafish embryos.</P>

      • Phlorotannin-rich fraction from <i>Ishige foliacea</i> brown seaweed prevents the scopolamine-induced memory impairment via regulation of ERK-CREB-BDNF pathway

        <P><B>Abstract</B></P> <P> <I>Ishige foliacea</I>, a species of edible brown algae, contains specific polyphenols called phlorotannins, which have antioxidant, anti-inflammatory, and anticancer properties. We hypothesized that the phlorotannin-rich fraction of <I>I. foliacea</I> (PRFI) is neuroprotective. We investigated the effects of PRFI on scopolamine-induced memory impairment in mice. PRFI (50 or 100 mg/kg) supplementation for 6 weeks improved scopolamine-induced memory impairment symptoms in the Morris water maze and passive avoidance tests. PRFI reduced acetylcholinesterase activity in the brain and significantly decreased lipid peroxidation levels, but increased glutathione levels and superoxide dismutase activity. Additionally, PRFI up-regulated the expression levels of brain-derived neurotrophic factor (BDNF) and tropomyosin receptor kinase B, and phosphorylated extracellular signal-regulated kinase (ERK) and cyclic AMP-response element-binding protein (CREB), which are involved in neuroplasticity in the cerebral cortex and hippocampus. Our results suggest that PRFI prevents scopolamine-induced memory impairment, possibly through oxidative scavenging and ERK-CREB-BDNF pathway regulation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PRFI improved scopolamine-induced memory impairment in mice. </LI> <LI> PRFI decreased AChE activity and restored scopolamine-induced oxidative stress. </LI> <LI> PRFI activated the ERK-BDNF-CREB signalling pathway. </LI> <LI> PRFI is a potent agent for the treatment of memory impairment-related diseases. </LI> </UL> </P>

        넓패추출물에 의한 RAW 264.7 세포에서의 항염효과

        본 연구는 RAW 264.7 세포를 이용하여 넓패추출물(Ishige foliacea, I. foliacea)의 피부 항염 효과를확인하고자 하였다. RAW 264.7 세포에 대한 넓패추출물의 항염증 효과는 세포 생존력 분석, mRNA 발현 및산화질소(NO)/프로스타글란딘 E2 (PGE2) 생성을 통해 확인하였다. 넓패추출물의 항염증 효과는 IL-1α/IL-1β /IL-6/TNFα mRNA 발현 및 NO/PGE2 생성을 분석하여 규명하였고, 정량적 실시간 중합효소 연쇄반응을 통해넓패추출물은 iNOS/COX2/IL-1α/IL-1β 및 IL-6의 mRNA 발현을 감소시키는 것을 확인하였다. 또한 PGE2와NO 생성의 비교를 통해 넓패추출물이 항염증 효과가 있음을 확인하였다. 넓패추출물의 항염 효과를 바탕으로코스메슈티컬 소재로 활용될 수 있는 가능성을 확인하였다. 넓패추출물의 항염증 메커니즘에 대한 후속 연구를바탕으로 코스메슈티컬뿐만 아니라 염증과 관련된 약리학적인 메커니즘 발굴에도 도움이 될 것으로 보인다. This study was carried out to identify the anti-inflammatory effects of Ishige foliacea (I. foliacea) extract on skin using RAW 264.7 cells. The anti-inflammatory effects of I. foliacea extract on RAW 264.7 cells were assessed by cell viability assay, mRNA expressions, and nitric oxide (NO)/prostaglandin E2 (PGE2) productions. The anti-inflammatory effects of I. foliacea extract were elucidated by analysis of IL-1α/IL-1β/IL-6/TNFα gene expressions and PGE2/NO production. Quantitative real-time polymerase chain reaction showed that I. foliacea extract decreased the gene expression levels of iNOS/COX2/IL-1α/IL-1β and IL-6. Furthermore, PGE2/NO production also revealed that I. foliacea extract exhibited anti-inflammatory properties. These results suggest that I. foliacea extract is an anti-inflammatory compound. It could be a potent cosmeceutical material for anti-inflammatory effects. Further studies on the anti-inflammatory mechanisms of broadleaf extracts are expected to help identify pharmacological mechanisms related to inflammation in addition to cosmeceuticals.

        넓패(Ishige foliacea) 추출물의 피부노화 억제활성

        In this study, the antioxidant and anti-skin aging properties of the Korean marine algae Ishige foliacea were investigated. Solvent extracts from I. foliacea were prepared with 70% ethanol, 80% methanol, and water. The extraction yields of various solvent extracts ranged from 9.55% to 35.12%. In terms of antioxidant activity, the ethanol extract showed the highest ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging activity, nitrite oxide scavenging activity, reducing power, and FRAP (ferric reducing antioxidant power). Regarding anti-skin aging activity, evaluation of the skin whitening and anti-wrinkle activities revealed that the methanol, water, and ethanol extracts possessed the highest tyrosinase (IC50=0.98 mg/mL), elastase (IC50=0.15 mg/ mL), and collagenase (IC50=0.06 mg/mL) inhibitory activities, respectively. These results suggest that I. foliacea holds potential as an antioxidant and anti-skin aging substance in food and cosmetic materials.

        35종 해조류 추출물의 병원성 세균 및 Candida sp. 진균에 대한 항균 활성 평가

        In the course of this study aimed at the development of functional food ingredients from seaweeds, the in vitro antimicrobial activities of methanol extracts prepared from 35 different seaweeds (17 phaeophyta, 11 rhodophyta and 7 chlorophyta) were determined against food-borne diseases and pathogenic microorganisms including multi-drug resistant (MDR) Pseudomonas sp. and Candida sp. Based on disc-diffusion assays at 500 g/disc concentration of the methanol extracts, Ishige okamurai, I. foliacea, Sargassum confusum, and S. yamade exhibited strong antibacterial activities in a broad-spectrum, except against Pseudomonas aeruginosa. In addition to the latter four seaweeds, Ecklonia stolonifera, E. cava and Eisenia bicyclis also demonstrated antifungal activity against C. albicans. Among these 8 selected seaweeds, I. okamurai, I. foliacea, and S. yamade exhibited strong hemolytic activity (55-93%) at 500 g/ml against human RBC. Organic solvent sequential fractions using hexane, ethylacetate and butanol, and water residues were prepared from the 8 selected seaweeds and their anti-Candida sp. activities were further determined. The ethylacetate and butanol fraction of I. okamurai, and the hexane fraction of I. foliacea demonstrated antifungal activity against MDR-pathogenic Candida sp. Although the solvent fractions had no activity against MDR-Pseudomonas sp., our results suggest that seaweeds, especially Ishige okamurai, I. foliacea, S. confusum, and S. yamade could be developed as broad-spectrum antimicrobial ingredients.

        Ishige foliacea ameliorates depressive‑like behaviors in stress hormone treated mice

        Chronic stress is an important factor to several physical and mental diseases. Ishige foliacea, an edible brown seaweed, has various biological properties; however, its role in alleviating stress-induced depression remains unclear. Thus, the antidepressant-like effects of I. foliacea ethanolic extract (IE) were investigated using corticosterone (CORT)-treated HT22 cells and CORT-induced depressive mice. IE significantly reduced the production of reactive oxygen species (ROS) and inhibited monoamine oxidase (MAO) activity and protein expression; furthermore, it up-regulated the extracellular signal-regulated kinase (ERK)-cAMP response element-binding protein (CREB)-brain derived neurotrophic factor (BDNF) signaling pathway. In the CORT-induced depressive mice, IE administration (500 mg/kg, bodyweight) exerted antidepressant effects according to behavioral tests. In addition, IE activated the ERK-CREB-BDNF signaling pathway owing to CORT exposure in the hippocampus. Therefore, IE inhibited ROS production and MAO activity and improved depressive behaviors caused by stress hormones, which demonstrates the therapeutic potential of IE in the treatment of depression.

        국내 자생 갈조류의 생리활성 물질 분석 및 총 항산화능력 비교 연구

        The inconsistent methods of harvest, identification, and preparation of brown algae samples make it difficult to compare the results of certain measurements across different studies, such as total phenolic contents (TPC), total flavonoid contents (TFC), and total antioxidant capacities (TAC). Therefore, this study aims to systematically measure and compare TPC, TFC, and TAC in 10 types of standardized brown algae extracts provided by the Bio-Resource Information System of Korea. Among the ten brown algae extracts, Ishige foliacea exhibited the highest levels of TPC and TAC, followed by Sargassum nigrifolium. TAC was measured by the following four assays: ABTS, DPPH, superoxide radical scavenging activities, and ferric reducing antioxidant power. TAC was found to be positively correlated with TPC (p<0.05) but not TFC, indicating that Ishige foliacea and Sargassum nigrifolium have potential use as marine antioxidative food material.
