        Experimental Study of Ginkgo-Chunghyul-dan on Anti-oxidant, Anti-platelet Aggregation, and Anti-hyperlipidemic Activity

        Objectives:Ginkgo-Chunghyul-dan (GCHD) is newly developed herbal medicine to prevent and treat stroke. In this study, we investigated whether the GCHD had antioxidant activity and anti-platelet aggregation effect in vitro and hypolipidemic activities in vivo. Methods:Anti-oxidant activity of GCHD was measured using the Blois method, anti-platelet effect of GCHD was assessed by the Born method, and hypolipidemic activities of GCHD were evaluated in corn oil- or Triton WR-1339-induced and cholesterol-fed rats. Results:GCHD showed anti-oxidant activity in the study inhibiting the formation of 1-diphenyl-2-picrylhydrazyl radicals and xanthine oxidase activity. GCHD had anti-platelet aggregation activity. GCHD significantly lowered total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) in high cholesterol diet and Triton WR-1339 induced model TG in corn oil-induced model. GCHD had no acute toxicity at a single dosage. Conclusion:These results suggest that GCHD has the potential to treat hyperlipidemia and stroke.         Hypolipidemic and antioxidant effects on hypercholesterolemic rats of polysaccharide from Salicornia bigelovii seed

        The hypolipidemic and antioxidant effects of dietary supplementation with polysaccharide isolated from Salicornia bigelovii seed (SBS) were evaluated in hypercholesterolemic rats. Following administration of SBS polysaccharide at 200mg/kg BW/day (HPS group), total cholesterol (TC), and low density lipoprotein cholesterol (LDLC) levels were 32.45% and 58.09% lower than those in negative controls (NC) group, respectively. However, in the case of high density lipoprotein cholesterol (HDLC), it was 36.37% higher than that of NC group. The arteriosclerosis and cardiac risk indices were 64.23% and 50.53% lower than in the NC group. However, the HTR (HDLC/TC) value was higher 2.05-fold than that of the NC group. Hydroxy-3-methylglutaryl CoA reductase activity in HPS group was 55.1% lower than that of the NC group. Activities of alanine transferase, aspartate aminotransferase, and alkaline phosphatase in HPS group were 29.71%, 26.49%, and 32.24%, lower than that of the NC group. SOD, CAT, and GSH-Px activities in HPS group were 31.05%, 49.29%, and 54.62% higher than that of the NC group. These results indicate that SBS polysaccharide has both hypolipidemic and antioxidant effects and shows potential for ameliorating hypercholesterolemia.

        Hypolipidemic Effect of Proteoglycans Isolated from Sweet Potato (Ipomoea batatas LAM) in Hyperlipidemia Rats

        Effects of sweet potato proteoglycans (SPG) inrats fed high lipid diets were investigated. Serum lipidindices for SPG were evaluated. SPG had hypolipidemiceffects that were correlated with dosage. There were nosignificant (p>0.05) differences in hypolipidemic effectsbetween SPG varieties of different molecular weights. SPG-1 (Mw value: 508.3 kDa) significantly (p<0.05)reduced serum total cholesterol (TC), triacylglyceride(TAG), low density lipoprotein cholesterol (LDL-C), andapolipoprotein (Apo) B levels, and the arteriosclerosisindex (AI), and induced serum high density lipoproteincholesterol (HDL-C), ApoA-I, lecithin cholesterol acyltransferase(LCAT), and liver lipoprotein lipase (LPL)activities, compared with controls. The whole blood viscosityand red blood cell (RBC) membrane microviscosity werealso decreased. Hypolipidemic effects were mediated viaelevation of the serum HDL-C concentration by inductionof LACT activity, an increased ApoA-I concentration, anda reduction in the serum lipid concentration due toregulation of LPL activity.

      • Hypolipidemic effect and antioxidant activity of glycoprotein isolated from Ulmus davidiana Nakai in Triton WR-1339-treated mouse

        <P>The glycoprotein isolated from Ulmus davidiana Nakai (UDN) (UDN glycoprotein) has a molecular weight of 116 kDa and consists of 78.65% carbohydrate content and 21.35% protein content. In the present study, we investigated the hypolipidemic effect of UDN glycoprotein on Triton WR-1339-induced mice. With pretreatment with UDN glycoprotein, the triacylglycerol (TAG), total cholesterol and low density lipoprotein-cholesterol (LDL-C) concentrations were significantly reduced, whereas high density lipoprotein-cholesterol (HDL-C) concentration was increased in the plasma of Triton WR-1339-induced mice. With respect to antioxidative activity, UDN glycoprotein significantly decreased the level of thiobarbituric acid reactive substances (TBARS) and improved activities of catalase and glutathione peroxidase (GPx), without an apparent change of superoxide dismutase (SOD) activity. Also UDN glycoprotein significantly increased nitric oxide (NO) production in Triton WR-1339-induced mice. These results indicate that UDN glycoprotein has a hypolipidemic effect, possesses antioxidant activity and has an ability to stimulate NO production. Thus, we speculate that UDN glycoprotein is an example of natural compound that lowers plasma lipid level together with having an antioxidant function in Triton WR-1339-induced mice.         Hypolipidemic and Antioxidative Effects of Dika Nut (Irvingia gabonensis) Seeds and Nkui (Trimphetta cordifolia) Stem Bark Mucilages in Triton WR-1339 Induced Hyperlipidemic Rats

        Two different mucilages were extracted from dika nut (Irvingia gabonensis) kernels and nkui (Trimphetta cordifolia) stem barks and the hypolipidemic and antioxidant effects were studied on Triton WR-1339induced hyperlipidemic rats. The GC analysis revealed that, dika nut mucilage is an arabinogalactan type while nkui is a rhamnogalacturonane type. The mucilage of nkui (200 mg/kg) showed the greatest hypolipidemic effect (p<0.05). An oxidative stress marker analysis suggested that the level of malonyl dialdehyde was significantly decreased in serum by the 2 mucilages (p<0.05). There was also a significant decrease in hydroperoxide level for the tested fibers. Moreover, nkui mucilage gave the highest levels in liver homogenate and in the hemolysate and tissues (p<0.05). The results suggest that the hypolidemic effect of mucilages varies with their chemical nature. They would be effective in the prevention of increase of serum lipid levels, in the reduction of lipid peroxidation and in the improvement of antioxidant status.

        Hypolipidemic Effects of Hickory Nut Oil using Cold Pressure Extraction

        Optimal conditions of hickory nut oil cold press technology were studied. L9(34) orthogonal experiment results showed that optimal conditions were a pressing pressure of 15 MPa, pressing temperature of 50oC, pressing cycle of 4 s, and stop cycle of 9 s. Fatty acid compositions were determined using GC-MS and hypolipidemic effects in mice were investigated. Compared to a high fat diet group, hickory nut oil administration decreased serum and visceral total cholesterol, triacylglycerol, and serum low density lipoprotein cholesterol levels. Serum high density lipoprotein cholesterol values were increased. Hickory nut oil can be used as a valuable bioactive source of natural hypolipidemic compounds.

      • Hypolipidemic and Antioxidant Effects of Dandelion ( <i>Taraxacum officinale</i> ) Root and Leaf on Cholesterol-Fed Rabbits

        <P>Dandelion (<I>Taraxacum officinale</I>), an oriental herbal medicine, has been shown to favorably affect choleretic, antirheumatic and diuretin properties. Recent reports have indicated that excessive oxidative stress contributes to the development of atherosclerosis-linked metabolic syndrome. The objective of this current study was to investigate the possible hypolipidemic and antioxidative effects of dandelion root and leaf in rabbits fed with a high-cholesterol diet. A group of twenty eight male rabbits was divided into four subgroups; a normal diet group, a high-cholesterol diet group, a high-cholesterol diet with 1% (w/w) dandelion leaf group, and a high-cholesterol diet with 1% (w/w) dandelion root group. After the treatment period, the plasma antioxidant enzymes and lipid profiles were determined. Our results show that treatment with dandelion root and leaf positively changed plasma antioxidant enzyme activities and lipid profiles in cholesterol-fed rabbits, and thus may have potential hypolipidemic and antioxidant effects. Dandelion root and leaf could protect against oxidative stress linked atherosclerosis and decrease the atherogenic index.</P>

        고지질이 유발된 흰쥐에서 버섯된장 추출물의 Hypolipidemic 효과

        버섯된장 추출물이 고지혈증에 미치는 영향을 알아보기 위하여 고지혈증이 유도된 흰쥐에 세 종류의 버섯된장 추출물을 경구투여한 후 혈청 및 간장에서의 지질 성분을 측정하였다. 그 결과 체중 증가량에 있어서 영지버섯 된장군(ESG)이 대조군에 비해 10%가 유의하게 낮았으나, 다른 군들은 차이가 없었다. 식이 섭취량, 식이 효율 및 간과 신장의 무게에 있어서는 버섯된장 추출물 투여군들이 대조군과 별다른 차이가 없었으나, 비장 무게는 ESG군이 다른 군에 비해 유의하게 낮았다. 혈청 총 cholesterol, LDL-cholesterol 및 간장 총 cholesterol 농도는 버섯된장 추출물 투여군 모두가 대조군에 비해 유의하게 낮았으며, 특히 ESG군이 대조군보다 혈청 총 cholesterol 20%, LDL-cholesterol 33%, 및 간장 총 cholesterol 17%가 낮게 나타났다. 버섯된장 추출물 투여군들이 대조군에 비해 유의적인 차이는 없었지만, 혈청 HDL-cholesterol은 높았고, triglyceride와 동맥경화지수는 낮게 나타났다. 혈청 총 cholesterol에 대한 HDL-cholesterol의 농도비는 버섯된장 추출물 투여균 모두에서 대조군에 비해 유의적으로 높았으며, 특히 ESG군이 대조군에 비해 35%가 높게 나타났다. 이상의 결과로 버섯된장 추출물들, 특히 ESG가 고지혈증 억제작용뿐만 아니라 관상동맥경화증을 비롯한 각종 동맥 경화증을 예방할 수 있는 가능성과 간장에서 지질 농도의 억제효과가 있음을 시사해주고 있다. The hypolipidemic effects of soybean paste containing mycelia of Phellinus linteus, Cordyceps militaris, Ganoderma lucidum were evaluated in hyperlipidemic rats. Body weight of the rats fed with soybean paste containing mycelia of Ganoderma lucidum (ESG) were significantly lower than control group whereas, there was no significant difference in body weight in other diet groups. Food intake, food efficiency ratio, and liver and kidney weights were not significantly different among the diet groups examined. However, the spleen weight of ESG group was markedly lower than other groups. Significant decreases in serum total cholesterol, LDL-cholesterol and liver total cholesterol were also observed in all diet groups tested. All groups fed with soybean paste containing mycelia of mushroom were almost equally effective in increasing plasma HDL level and decreasing the atherogenic index, whereas the ratio of HDL to total cholesterol was significantly increased in all experimental diet groups.
