The safety of fake anti-impotence drugs (fake Viagra : 26 samples, fake Cialis : 25 samples) distributedin Gyeonggi province was studied by monitoring the concentrations of anti-impotence pharmaceutical ingredientand their analogues. The concentrations of anti-impotence pharmaceutical ingredient 4 specis and theiranalogues 17 specis were estimated using by HPLC/PDA, LC-MS/MS. The range of concentration of sildenafil infake viagra was 40~199 mg/tablet, among them the portion of the concentrations of sildenafil over 150 mg/tabletexceeded 65%. 3 cases in tested samples contained sildenafil and tadalafil. The range of concentration of sildenafil infake cialis was 102~249 mg/tablet, among them the portion of the concentrations of sildenafil over 150 mg/tabletexceeded 88%. One case in tested samples contained demethylhongdenafil (90 mg/tablet). These results indicate thatthere were many fake anti-impotence drug contained high level of anti-impotence pharmaceutical ingredients, a sustainablemonitoring and the blocked distribution of fake anti-impotence drugs recommended.

본 연구는 국내/수입 유통되는 식품과 건강기능식품의 발기부전치료제 및 그 유사물질 혼입 농도를 모니터링하고, 그 결과를 바탕으로 식품의 안전성을 확보하고자 하였다. 전국에서 유통중인 식품 및 건강기능식품 총 226건을 구입하고 24종의 발기 부전치료제 및 그 유사물질을 HPLC/PDA 및 LC-ESI-MS-MS를 이용하여 분석하였다. 고체 및 액체 시료의 전처리는 메탄올을 이용하여 추출하였다. 식품 및 건강기능식품 시료에서 확립된 시험법을 이용하여 식품과 건강기능식품 226건을 수거하여 모니터링한 결과, 5건의 성기능 표방 제품에서 acethylvardenafil, sildenafil, tadalafil이 검출되었다. 분석 결과, 검출량은 acethylvardenafil이 1종에서 21,476 ㎎/㎏(15 ㎎/capsule)이었으며, sildenafil이 2종에서 각각 52,778 ㎎/㎏(29 ㎎/capsule), 71,535 ㎎/㎏ (48 ㎎/capsule)이었고, tadalafil이 4종에서 9,772 ㎎/㎏(6 ㎎/capsule) -55,545 ㎎/㎏ (33 ㎎/capsule)이었다. 따라서 식품 및 건강기능식품 중의 발기부전치료제 및 그 유사물질에 대한 지속적인 실태조사가 요구된다. This study was conducted to monitor the concentrations of 24 anti-impotence drugs and their analogues in various foods and dietary supplements, with the aim of ensuring the safety of the foods and supplements. The measurements were done in 226 samples using high performance liquid chromatography/photodiode array detector (HPLC/PDA) and liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). Five male sexual function enhancing products have been detected as follows: acethylvardenafil (21,476 ㎎/㎏; 15 ㎎/capsule from one sample), sildenafil (52,778 ㎎/㎏, 29 ㎎/capsule in one sample; 71,535 ㎎/㎏, 48 ㎎/capsule in one sample), and tadalafil (9,772-55,545 ㎎/㎏, 6-33 ㎎/capsule in four samples). A sustainable monitoring of anti-impotence drugs and their analogues in various foods and dietary supplement is recommended.

식품중 발기부전 치료제 및 유사물질 함량에 관한 조사연구

To det ermine 8 anti-impotence drug/drug-like compounds such as sildenafil, vardenafil, tadalafil, homosildenafil, hydroxyhomosildenafil, aminotadalafil, pseudovardenafil and hongdenafil in foods, simultaneously, high performance liquid chromatograrhy(HPLC) and liquid chromatograthy-mass spectrometry (LC/MS) were used. The HPLC/UV analysis was performed on a column of capcellpak C₁? with 0,1 % sodium-1-hexansulfonate in 0.2M ammonium formate/acetonitrile as a mobile phase. Mass spectra of the compounds by LC/MS were investigated with SCAN mode(Mass range and Fragment voltage) and SIM (Selected Ion Monitoring) mode (Ion target and Fragment voltage). The results follow as; 1. The HPLC/UV analysis was detected from 5 out of 63 samples. The content of sildenafil was in the range of 32.80 ppm~60,13 ppm from 4 out of 5 samples, The contents of sildenafil, vardenafil, homosildenafil were in the range 47,14 ppm from lout of 5 samples. 2. The conformed result of LC/MS was equal of detected from 5 out of 63 samples in HPLC/UV analysis. An easily available, simultaneous determination of 8 standards in adulterated health related foods was established by using a combination of LC/MS methods.

HPLC 및 ESI-tandom Mass Spectrometry를 이용한 인터넷에서 판매되는 기능성표방식품 중 부정유해물질 모니터링

인터넷에서 판매되는 건강기능성표방식품 51개를 구매하여 발기부전치료제 5종과 그 유사물질 29종, 이카린, 요힘빈, 시부트라민 그리고 시부트라민 유사물질인 데스메틸시부트라민, 디데스메틸시부트라민 총 39종의 부정유해물질을 3개의 혼합표준용액으로 만든 후 HPLC를 이용하여 총 50분 동안 이동상 용액 A(5mM sodium hexanesulfonate와 0.1% phosphoric acid을 함유한 물), B(95% acetonitrile) 두 가지를 기울기 용매 조건으로 분석하였다. 검출된 화합물의 확인을 위하여 LC-ESI-tandom MS의 MRM 조건을 최적화하고 정성분석 한 결과 1개의 홍삼제품과 영양보충용제품 2건에서 실데나필과 슈도바데나필, 디메틸티오실데나필 성분이 1회 복용량당 1.3-82.1 mg 검출되었으며, 2개의 기타가공품에서 부작용이 우려되어 사용이 금지된 시부트라민의 유사물질인 디데스메틸시부트라민이 각각 1회 복용량 당 2.2, 1.7 mg이 검출되었다. 시중에 판매되는 비아그라의 실데나필 1회 최소복용량은 25 mg으로 시료에서의 검출량은 이보다 높은 수준으로 함유되었으며, 의사의 처방 없이 식품 중에 불법적으로 함유된 이들 물질 혹은 이들의 유사물질은 소비자들의 건강상에 문제를 일으킬 수 있으므로, 시중에 유통되는, 더욱이 온라인 상에서 음성적으로 판매되고 있는 건강관련식품에 대한 모니터링이 지속적으로 필요한 실정이다. It was carried out to detect anti-impotence drug-like compounds, sibutramine and their analogues in dietary supplements, which are doubtful whether they include illegal compounds. A total of 51 food products were bought online and have been investigated. The separation was achieved on a C18 column, with the mobile phase made up of water (5 mM sodium hexanesulfonate and 0.1% phosphoric acid) and 95% acetonitrile, at a flow rate of 1.2 mL/min with gradient elution using by HPLC-DAD. The UV signals were monitored at 220 nm and 291 nm. LC-ESI-tandom MS was utilized to confirm that detected compounds in samples are the same as the reference materials. Two nutrient supplement foods and ginseng products were found to contain 1.3-82.1 mg of sildenafil, dimethylthiolsildenafil and pseudovardenafil per serving size. In addition, two other processed products were detected to contain 1.7 and 2.2 mg of didesmethylsibutramine, derived from sibutramine per serving size.
