      • 실크 세리신의 가수분해 특성과 응용 II. 수용성 세리신의 특성과 피부에의 응용

        수용성 세리신의 전자공여능을 측정하고, FT-IR, DSC 등의 분석을 통하여 물리 화학적 특성을 조사하고 부직포에 세리신을 처리하여 pack test 실시하여 다음과 같은 결론을 얻었다. 1. 전자공여 작용의 항산화 실험 결과 high>low>PK>PP>PA 순서로 전자공여능이 높았다. 2. FT-IR 분석 결과 난용성 세리신에 나타난 3285 $cm^{-1}$ /에서의 피크가 수용성 세리신에서는 단파장쪽으로 이동되었고, 가수분해가 되어 분자량이 작을수록 아미드 II밴드의 크기는 작아지고 단파장쪽으로 이동하는 현상이 나타났다. 3. DSC 분석 결과 모든 세리신에서 20$0^{\circ}C$와 30$0^{\circ}C$부근에서 흡열피크가 나타났으며, 난용성 세리신의 경우 25$0^{\circ}C$부근에서 흡열 피크가 하나 더 나타나고 30$0^{\circ}C$부근의 흡열피크는 수용성 세리신보다 높은 온도에서 나타났다. 4. 백도는 low>high>PP>PA>PK>IN 순서로 높았고, 황도는 PK>PA>PP>low>high 순서로 높았다. 5. Pack test 결과 세리신 처리된 부직포로 얼굴에 pack 처리한 경우는 처리 전에 비하여 번들거림이 줄어들고 피부가 편평한 상태가 되었다. This study was carried out to investigate the characteristics of the soluble sericins after degumming and after hydrolysis of insoluble sericin with various enzymes. Physical and chemical characteristics of the soluble sericins were also studied and pack test was conducted. Electro donating ability of the various sericins became higher as high>low>KP>PP>PA. In FT-IR analysis, the band of insoluble sericin at 3,285cm$\^$-1/ was shifted to the lower wavelengths in soluble sericins. The intensity of the amide II band was reduced and shifted to lower wavelengths as a result of hydrolysis. In DSC analysis, all sericins showed endothermic peaks around 200$^{\circ}C$ and insoluble sericin showed another peak at 250$^{\circ}C$. The endothermic peak of the insoluble sericin was found at higher temperature compared with those of the soluble sericins which showed at 300$^{\circ}C$. Whiteness of the various sericins became higher as low>high>PP>PA>PK>IN and yellowness were PK>PA>PP>low>high. In pack test, the skin packed with the non-woven fabric treated with the various sericins was more smooth and less oily.

        Enhanced Mechanical Property of Silk Sericin Beads Prepared from Ethanol-precipitated Sericin

        Sericin beads were prepared from ethanol-precipitated sericin. The addition of ethanol into hot-water extracted sericin solution induced precipitation of sericin and thereby some sericin could be fractionated. The ethanol-precipitated sericin (EpSS) had narrower molecular weight distribution than original sericin. The EpSS had mainly random coil structure with small portion of β-sheet structure. With the EpSS, spherical beads could be prepared at lower concentration than with original sericin due to higher viscosity. The EpSS beads had better compressive strength than the original sericin beads and had rubber-like property. Our results suggest that EpSS is more compatible in the polymeric field, since it has better mechanical strength than original sericin.

        키토산/세리신 블렌드 필름의 구조 및 특성

        The uncrosslinked and crosslinked by ethylene glycol diglycidyl(EGDE) chitosan/sericin blend films were prepared by solvent blending method. Physicochemical properties of the prepared blend films according to various blend ratios were investigated. This was carried out by using universal testing machine, thermogravimeter(TGA), differential scanning calorimeter(DSC), wide angle X-ray diffractometer(WAXD), Fourier transform infrared spectrum(FTIR) and scanning electron microscopy(SEM). With an increase in the sericin mixing ratio, tensile stress increased whereas the tensile strain decreased for uncrosslinking blend films. For a cross linked chitosan/sericin blend film by EGDE, both the tensile stress and strain decreased. Chitosan/sericin blend films exhibit an excellent thermal stability when sericin mixing ratio is 25%. In the infrared spectra, chitosan/sericin blend films showed an absorption band of amide I peak at 1630 $cm^{-1}$ and it is concerned with C=O stretching vibration by strong hydrogen-bond between inter-molecular. These also showed an absorption band of amide II peak at 1521 $cm^{-1}$ concerned with N-H bending vibration. The absorption band of amide I peak decreased with an increase in the chitosan mixing ratio. We identified that there was good miscibility between chitosan and sericin. The water-retention values(WRV) of chitosan/sericin blend films increase as the amount of sericin is raised and this owes to the hydrophilic nature of sericin. Weight loss of the citosan/sericin blend films by cleaning with an aqueous sodium hydroxide increased with an increase in the sericin mixing ratio. This loss is the loss of acetic acid and sericin in the chitosan/sericin blend films and this is because of the molecular weight of the sericin is 1/5 of the chitosan and sericin is dissolved into NaOH solution. Weight loss increased when sericin content was more than 50% for the cross linked chitosan/sericin blend films. From the structure analysis, we identified that there was good miscibility and strong intermolecular interaction between the chitosan molecules that resulted from intermolecular hydrogen bonds.

      • 세리신잠(Nd-s)의 발육특성과 혈액아미노산 조성

        세리신 잠의의 발육, 실샘 성장, 세리신 량, 혈액 아미노산 조성 등 세리신 잠의 특성을 알아보기 위하여 잠120과 같은 조건으로 사육하여 얻은 결과는 다음과 같다. 1. 부화율에 있어서 세리신 잠은 85%이며 잠120은 95%이었다. 특히 세리신 잠은 부화가 고르지 않았다. 2. 유충 기간에 있어서, 세리신은 20일 23시간이었고, 잠120은 21일 22시간이었다. 3. 실샘에 있어서 세리신 잠은 중부실샘이 대부분이며 후부 실샘의 길이는 약2∼3cm 이었고 굴곡은 없었다. 4. 세리신 잠에 있어서 상족후 나방까지의 기간은 약12일 이었으며, 화용후 화아까지는 7일로 대단히 짧은 특성을 보였다. 5. 고치에 있어서 세리신 잠은 아주 얇고 가벼웠으며 견층중은 2.7cg 이며 견층 비율은 3.65% 이였다. 6. 세리신 (sericin protein)양에 있어서, 세리신잠의 견층이 sericin만으로 구성되어 있고 잠120은 28%가 sericin 이었다. 세리신 잠의 견층당 sericin 율은 잠120의 34.6% 정도였다. 7. 혈액 아미노산의 양에 있어서 세리신 잠은 histidine, lysine, glutamic acid 가 많았으며 5령 발육에 따른 조성의 변화에 있어서 대부분의 혈액 아미노산들은 증가하였다. This experiment was to know properites of Sericin Jam that development, growth of silkgland, content of sericin and composition of amino acid in hemolymph. The characters of Sericin Jam can be seen form this experiment. Hatching ratio was 85% in Sericin Jam and 95% in Jam 120. Especially hatching period of Sercin Jam was longer than Jam was 20 days and 23 hours, and Jam 120 was 21 days and 22 hours. In Sericin Jam, middle silkgland contain a great p art s in silkgalnd and posterior silkgand is short and no curves. The period of mounting to emergence was 12 days in Sericin Jam. The period of pupa was 7day. It is property of Sercin Jam that the period is short. Cocoon was very thin and light in Sericin Jam. Weight of cocoon shell of Sericin Jam is 2.7cg. The sericin protein quantity was 100% in cocoon shell of Sericin Jam, about 28% in cocoon shell of jam 120, however the sericin ratio per Sericin Jam cocoon was 34.6% compare to cocoon shell of jam 120 in sericin protein quantity. The amino acids in hemolymph of Sericin Jam was much hidtidine, lysine, glut amic acid. And the amount of almost amino acids were increased depends upon development at t he 5th instar.

        Characteristics of Carboxymethyl Cellulose/Sericin Hydrogels and the Influence of Molecular Weight of Carboxymethyl Cellulose

        Carboxymethyl cellulose (CMC),which is a hydrophilic polysaccharide obtained from the carboxymethylation of natural cellulose, has been formulated as hydrogel for various biomedical applications. However, the functional crosslinking agents were required to form the stable CMC hydrogels. In this study, sericin which is a glue-like protein that envelops the fibroin fiber in the silkworm cocoons was introduced to blend with CMC to form a mechanically stable hydrogel without further crosslinking. Furthermore, the effect of molecular weight of CMC on characteristics of the CMC/sericin hydrogels obtained was investigated. We found that the incorporation of sericin in CMC hydrogels enhanced the stability and the hydrogel’s resistance to hydrolysis and enzymatic degradation while the porous structure and swelling abilities of the CMC hydrogels were preserved. The in vitro biodegradation of the CMC/sericin hydrogels and the release of sericin from the hydrogels were evaluated. The biodegradability of the CMC/sericin hydrogels depended on the molecular weight of CMC. The high molecular weight CMC/sericin hydrogels showed the slowest rate of degradation. The slow-degrading high molecular weight CMC/sericin hydrogels resulted in the lowest amount of sericin release while the fast-degrading low and medium molecular weight CMC/sericin hydrogels released high amount of sericin. The high amount of sericin released from the fast-degrading low and medium molecular weight CMC/sericin hydrogels could consequently promote the collagen production of L929 cells. These CMC/sericin hydrogels could be further investigating to be applied as a wound dressing material.

        <P><B>Abstract</B></P> <P>In this study, the effect of the MW on the structure and properties of sericin film, sponge, and gel was examined. As the MW of sericin increased, the gelation of the sericin aqueous solution was found to be accelerated, and the gel strength, and the gel-sol transition temperature increased. Irrespective of the casting solvent (water and formic acid) and form of sericin (gel, film, or sponge), the crystallization of the sericins was accelerated. The mechanical properties of the sericin sponge were remarkably improved upon increasing the MW of sericin. The MW of sericin almost did not have an effect on the cell toxicity. As the MW of sericin is increased, the sericin sponge becomes denser and its porosity is reduced, leading to a decrease in the swelling ratio. These results indicate that various characteristics of the sericin forms can be modulated by controlling the MW of sericin, with enhanced potential for biomedical and cosmetic applications.</P>

        A Novel Approach for Sericin-Conjugated Silver Nanoparticle Synthesis and Their Potential as Microbicide Candidates

        Silver nanoparticles have been widely applied for biomedical areas owing to their potent antiviral and antibacterial activities. Synthesis of silver nanoparticles using biomacromolecules is more efficient, environment-friendly, and cost-saving compared with the traditional approach. In this paper, a novel approach was developed to establish a reaction system with Ag<sup>+</sup>-BH4--sericin to synthesize silver nanoparticles conjugated to sericin (AgNPs-Sericin). Sericin could be as a good dispersant and stabilizing agent, which is able to modify nanoscaled AgNPs, the average diameter of which was only 3.78 ± 1.14nm prepared in a 0.3 mg/ml sericin solution. The characterizations of the AgNPs-Sericin were determined by FTIR, thermogravimetry, and XRD analyses. The results showed that the synthesized AgNPs conjugated with sericin as organic phase. Via SAED and XRD analysis, we showed that these AgNPs formed polycrystalline powder with a face-centered cubic structure of bulk metals. Moreover, we investigated the antiviral and antibacterial activities of AgNPs-Sericin, and the results showed that AgNPs-Sericin exhibited potent anti-HIV-1 activity against CCR5-tropic and CXCR4-tropic strains, but no significant cytotoxicity was found toward human genital epithelial cells compared with free silver ions, which are accepted as a commonly used potent antimicrobial agent. Moreover, its antibacterial activity was determined via flow cytometry. The results showed that AgNPs-Sericin could suppress gram-negative (E. coli) and grampositive (S. aureus) bacteria, but more was potent for the gram-negative one. We concluded that our AgNPs-Sericin could be a potential candidate as a microbicide or antimicrobial agent to prevent sexually transmitted infections.

        Effect of Extraction Time on the Rheological Properties of Sericin Solutions and Gels

        Recently, silk sericin has attracted the attention of researchers owing to its useful properties as a biomaterial including 1) good wound healing and cell activities, 2) fast gelation character, and 3) high water retention property. In the present study, silk sericin was prepared using different extraction times in hot water and the effect of extraction time on the rheological properties of sericin solutions and gels was examined. It was found that the production yield of sericin increased with extraction time. The shear viscosity of sericin solutions and gels decreased with increasing extraction time due to a decrease in sericin molecular weight. When the sericin solution transformed to a gel, the viscosity increased and the shear thinning behavior was more evident. In addition, the shear stress measurements indicated that the slip between the sericin samples and the measuring plate of the rheometer was increased by the gelation of sericin. The compression strength of sericin gel could be increased remarkably (by more than 100 fold) by preparation using the freezing and thawing method.

        Recovery of Silk Sericin from Soap-Alkaline Degumming Solution

        Sericin is usually abandoned after the degumming process. However, it could be a valuable bioresource if an economically efficient recovery process could be set up. In this study, sericin was recovered directly from the degummed waste solution by adding calcium chloride, which induced the precipitation of the surfactant, sodium oleate, by charge interaction. The recovery yield was maximum when 10% of calcium chloride was added. Further increase in the calcium chloride concentration induced the precipitation of sericin. The recovered sericin had a molecular weight distribution similar to that of the hot-water-extracted sericin; but some highmolecular- weight sericin could not be recovered. The secondary structure and amino acid composition of the recovered sericin were similar to those of conventional hot-water-extracted sericin. We expect that sericin recovered from the degummed waste solution could be an alternative to the hot-water-extracted sericin, which is widely used in various applications.
