        Toxic Effects on the Nonspecific Immune System of the Rock Bream Oplegnathus fasciatus upon Exposure to Di-2-ethylhexyl Phthalate

        The aim of this study was to investigate the in vivo toxicity of di-2-ethylhexyl phthalate (DEHP), on the immune system of the rock bream, Oplegnathus fasciatus. Fish were injected twice intraperitoneally with DEHP (200, 400, and 800 mg/kg BW), and the cellularity and functional activity of phagocytes in the spleen and head kidney were measured. The number of head kidney leukocyte cells was significantly greater in fish treated with 800-mg DEHP/kg BW. Nonspecific immunity, as determined by the phagocytic index, was significantly decreased at 800-mg DEHP/kg BW in the head kidney. A significant reduction in phagocytic capacity was observed in the head kidney at ${\geq}$400-mg DEHP/kg BW. Furthermore, significantly increased levels of serum glutamic oxaloacetate transaminase and glutamic pyruvate transaminase indicated a marked hepatic dysfunction in immunosuppressed fish. Total serum protein was significantly reduced at ${\geq}$400-mg DEHP/kg BW; however, there were no significant changes in lysozyme activity. These results demonstrate that immune responses in the rock bream, Oplegnathus fasciatus can predict immunotoxicity at doses ${\geq}$400-mg DEHP/kg BW.

        TBT 노출에 따른 돌돔, Oplegnathus fasciatus의 성장 및 혈액성분의 변화

        Experiments were carried out to investigate the growth and hematological parameters of rock bream Oplegnathus fasciatus exposed to several different concentrations (0, 1, 2, 4, 8 ㎍/L) of tributyltin (TBT) for 4 weeks. Growth rate of the fish exposed to 8 ㎍/L of TBT concentration was significantly lower than that of control group fish. The major hematological findings were significant decreases in the red blood cell count, hematocrit value and hemogobin concentration in the fish exposed to 8 ㎍/L. Although serum glucose concentration was significantly reduced compared to the control group, total protein concentration was found to be significantly increased over the control group. Exposure to 8 ㎍/L of TBT concentration resulted in significant increase in the enzyme activities, such as glutamic oxalate transminase and glutamic pyruvate transminase in the fish. The present findings suggest that exposure to 8 ㎍/L concentration of TBT can cause significant changes in growth and hematological parameters of rock bream Oplegnathus fasciatuso.

        사료 내 해조류(톳, 감태) 첨가가 돌돔(Oplegnathus fasciatus) 치어의 비특이적 면역반응에 미치는 영향

        Two feeding trials were conducted to determine the effects of dietary supplementation with Hizikia fusiformis or Ecklonia cava on the non-specific immune responses of parrot fish Oplegnathus fasciatus. Fish were fed experimental diets to which H. fusiformis or E. cava powder were added to final concentrations of 0, 2, 4 and 6%, respectively. After feeding for two weeks, phagocytic activity was significantly higher in fish fed diets containing H. fusiformis, but not E. cava, than in fish fed the basal diet. Lysozyme activity was significantly increased in the fish fed diets containing 6% H. fusiformis and E. cava. Myeloperoxidase activity was also significantly higher in fish fed diets containing 2 and 4% H. fusiformis, as compared to the basal diet, but not in those fish fed E. cava. These two studies indicate that dietary supplementation with H. fusiformis or E. cava could enhance the innate immune responses of parrot fish during their growth stage.

        옥시벤존(Benzophenone-3)이 돌돔(Oplegnathus fasciatus)의 배 발생 과정에 미치는 영향

        Oxybenzone (Benzophenone-3; BP-3) is a compound used in sunscreens and enters the oceans due to improper disposal. It is known to disrupt the endocrine signaling in marine organisms, leading to immune and reproductive abnormalities in corals, shellfish and fishes. In this study, we investigated the effects of different concentrations of BP-3 (0, 10, 100 and 1,000 μg/L) on the fertilized eggs and hatched larvae of Oplegnathus fasciatus. The morphological changes and hatching and survival rates during embryo development were assessed. In addition, the levels of triiodothyronine (T3) were also measured. The hatched larvae exposed to BP-3 at concentrations of 10 and 100 μg/L for 24 h displayed malformation of the tail. After 36 h of exposure to BP-3, spinal deformity was observed at all tested concentrations. The hatching rate was significantly low when exposed to 100 μg/L of BP-3. A high levels of T3 was observed when the larvae were exposed to BP-3 at a concentration of 1,000 μg/L for 96 h (the end of the experiment). This may be related to increased size of larvae at 1,000 μg/L BP-3. In conclusion, our results suggested that BP-3 may interfere with embryo development, resulting in a reduction in hatching rate and malformation of larvae.

        Isolation of Novel Hepcidin Isoforms from the Rockbream Oplegnathus fasciatus (Perciformes)

        Three novel hepcidin isoforms were isolated and characterized from the perciform fish species Oplegnathus fasciatus. These hepcidin isoforms (designated rbhepc5, rbhepc6 and rbhepc7) were found to share a conserved, tripartite gene structure and a considerable sequence homology one another. A comparison of their mature peptide sequences with those of other perciform hepcidin orthologs indicated that these three hepcidin isoforms as well as four other isoforms previously identified in this species, appear to belong to the HAMP2 group of hepcidin genes. Analysis of the 5'-upstream sequences showed that the proximal non-coding regions of rbhepc5~7 do not possess canonical TATA signals; instead, they harbor several binding motifs for transcription factors involved in immune modulation. Reverse transcriptase-PCR analysis demonstrated that the rbhepc5~7 are expressed predominantly in the liver, and that the transcription of rbhepc5~7 is rapidly induced in the liver, but not in other tissues, by experimental challenge with any of three different bacterial species. However, transcription of rbhepc6 appeared to be negligible under both basal and stimulated conditions, as judged by the redundancy count of randomly chosen reverse transcriptase-PCR clones.

        Population Genetic Structure of Rock Bream (Oplegnathus fasciatus Temminck & Schlegel, 1884) Revealed by mtDNA COI Sequence in Korea and China

        The rock bream, Oplegnathus fasciatus, is a common rocky reef game fish in East Asia and recently has become an aquaculture species. Despite its commercial importance, the population genetic structure of this fish species remains poorly understood. In this study, 163 specimens were collected from 6 localities along the coastal waters of Korea and China and their genetic variation was analyzed with mtDNA COI sequences. A total of 34 polymorphic sites were detected which determined 30 haplotypes. The genetic pattern reveals a low level of nucleotide diversity (0.04 ± 0.003) but a high level of haplotype diversity (0.83 ± 0.02). The 30 haplotypes are divided into two major genealogical clades: one that consists of only Zhoushan (ZS, East China Sea) specific haplotypes from the southern East China Sea and the other that consists of the remaining haplotypes from the northern East China Sea, Yellow Sea, Korea Strait, and East Sea/Sea of Japan. The two clades are separated by approximately 330~435 kyBP. Analyses of AMOVA and F st show a significant population differentiation between the ZS sample and the other ones, corroborating separation of the two genealogical clades. Larval dispersal and the fresh Yangtze River plume are invoked as the main determining factors for this population genetic structure of O. fasciatus. Neutrality tests and mismatch distribution analyses indicate late Pleistocene population expansion along the coastal waters of Korea and China approximately 133–183 kyBP during which there were periodic cycles of glaciations and deglaciations. Such population information needs to be taken into account when stock enhancement and conservation measures are implemented for this fisheries species.

        The Molecular Profiling of a Teleostan Counterpart of Follistatin, Identified from Rock Bream Oplegnathus fasciatus which Reveals its Transcriptional Responses against Pathogenic Stress

        The follistatin (FST) gene encodes a monomeric glycoprotein that plays a role in binding and inhibiting the functions of members of the transforming growth factor (TGF)-${\beta}$ superfamily. Thus, FST facilitates a wide variety of functions, ranging from muscle growth, to inflammation and immunity. In this study, we sought to characterize an FST counterpart, RbFST, which was identified from rock bream Oplegnathus fasciatus. The RbFST cDNA sequence (2,419 bp) contains a 933-bp open reading frame (ORF) that encodes a putative amino acid sequence for RbFST (35 kDa). The putative amino acid sequence contains a Kazal-type serine protease inhibitor domain (51-98 residues) and an EF-hand, calcium-binding domain (191-226 residues). Additionally, this sequence shares a high identity (98.7%) with the Siniperca chuatsi FST sequence, with which it also has the closest evolutionary relationship according to a phylogenetic study. Omnipresent distribution of RbFST transcripts were detected in the gill, liver, spleen, head kidney, kidney, skin, muscle, heart, brain, and intestine of healthy animals, with significantly higher expression levels in the heart, followed by the liver tissue. Under pathogenic stress caused by two bacterial pathogens, Streptococcus iniae and Edwardsiella tarda, RbFST transcription was found to be significantly up-regulated. Altogether, our findings suggest the putative role of RbFST in immune related responses against pathogenic infections, further prefiguring its significance in rock bream physiology.

        Expression Analysis of Lily Type Lectin Isotypes in the Rock Bream, Oplegnathus fasciatus: in the Tissue, Developmental Stage and Viral Infection

        Lectins belong to the pattern-recognition receptors (PRRs) class and play important roles in the recognition and elimination of pathogens via the innate immune system. Recently, it was reported that lily-type lectin-1 is involved when a pathogen attacks in the early immune response of fish. However, this study is limited to information that the lectin is involved in the innate immune response against viral infection. In the present study, the lily-type lectin-2 and -3 of Oplegnathus fasciatus (OfLTL-2 and 3) have been presented to be included B-lectin domain and two D-mannose binding sites in the amino acid sequence that an important feature for the fundamental structure. To investigate the functional properties of OfLTLs, the tissue distribution in the healthy rock bream and temporal expression during early developmental stage analysis are performed using quantitative real-time PCR. OfLTL-2 and 3 are predominantly expressed in the liver and skin, but rarely expressed in other organ. Also, the transcripts of OfLTLs are not expressed during the early developmental stage but its transcripts are increased after immune-related organs which are fully formed. In the challenge experiment with RBIV (rock bream iridovirus), the expression of OfLTLs was increased much more strongly in the late response than the early, unlike previously known. These results suggest that OfLTLs are specifically expressed in the immune-related tissues when those organs are fully formed and it can be inferred that the more intensively involved in the second half to the virus infection.
