        홍고추 중 matrine의 가공계수

        This study was carried out to investigate the residual characteristics and calculate processing factor of the environment friendly material matrine in fresh chilli pepper by drying. Spray solution of matrine was prepared by dilution of the commercial product (2% active ingredient) with water at 1 : 1000 (v/v) ratio and sprayed onto chilli pepper plants at seven day intervals. Samples were collected at 0, 1, 3, 5 and 7 days after last application and then dried using a hot air dry oven at 60℃ for 36 hours until the water content was reduced to 14%. Recoveries and storage period stabilities of matrine in the samples ranged from 106.6 to 119.1% and 106.6 to 113.1%, respectively. The residual concentrations of matrine in fresh chilli pepper and dried chilli peppers treated only once were found to be from less than 0.01 to 0.11 and from 0.03 to 0.25 mg/kg, respectively. In case of plants sprayed twice with matrine, the residual concentrations ranged from 0.02 to 0.12 and from 0.04 to 0.4 mg/kg, respectively. Processing factor of matrine in the fresh chilli pepper by drying was found to be from 1.5 to 3.3, indicating that the residual concentration of matrine in dried chilli pepper increased about two or three times by drying.

        Optimization of Submerged Fermentation Medium for Matrine Production by Aspergillus terreus, an Endophytic Fungus Harboring Seeds of Sophora flavescens, Using Response Surface Methodology

        Different endophytes isolated from the seeds of Sophora flavescens were tested for their ability to produce matrine production. Response surface methodology (RSM) was applied to optimize the medium components for the endophytic fungus. Results indicated that endophyte Aspergillus terreus had the ability to produce matrine. The single factor tests demonstrated that potato starch was the best carbon source and the combination of peptone and NH<sub>4</sub>NO<sub>3</sub> was the optimal nitrogen source for A. terreus. The model of RSM predicted to gain the maximal matrine production at 20.67 μg/L, when the potato starch was 160.68 g/L, peptone was 24.96 g/L and NH<sub>4</sub>NO<sub>3</sub> was 2.11 g/L. When cultured in the optimal medium, the matrine yield was an average of 20.63 ± 0.11 μg/L, which was consistent with the model prediction. This study offered an alternative source for the matrine production by endophytic fungus fermentation and may have far-reaching prospect and value.

        상추와 오이 중 친환경 농자재 matrine의 잔류특성을 구명하기 위하여 고삼의 뿌리 추출액인 matrine을 1,000배 희석한 후 비닐하우스에서 생육중인 상추와 오이에 7일 간격으로 1회 및 2회 살포하고 경시적으로 시료를 채취하여 잔류농약을 분석하였다. 상추와 오이 중 matrine의 검출한 계는 각각 0.02와 0.001 mg/kg이었으며, 시험 약제의 회수율은 두 농약 모두 80.49-96.92%, 변이 계수는 모두 5% 미만으로 양호하였다. 상추 중 matrine의 잔류량은 0.05-0.34 mg/kg 의 범위이었 으며, 경시적으로 잔류량은 감소하는 경향이었다. 오이의 경우 0일차의 잔류량은 0.003 mg/kg이었 으나 1, 3, 5, 7일차의 경우 1회 및 2회 처리구 모두 검출한계 미만으로 검출되었다.

        Matrine promotes trophoblast invasion and reduces inflammation via miR-19a-3p to prevent preeclampsia

        Background During preeclampsia (PE), reactive oxygen species (ROS) production and infl ammation are increased. Matrine exerts diverse pharmacological eff ects, including anti-infl ammatory and anti-apoptotic eff ects. Objective The present study aimed to study the therapeutic eff ects of matrine on PE. Human trophoblasts (HTR-8/SVneo cells) were treated with H 2 O 2 to simulate oxidative stress, followed by treatment with various concentrations of matrine. Next, fl ow cytometry was used to examine apoptosis. Cell invasion and migration were examined by Transwell and scratch wound assays, respectively. The concentrations of IL-6, IL-8, and TNF-α were quantifi ed by enzyme-linked immunosorbent assays. The expression levels of miR-19a-3p and ZEB2 were quantifi ed by qRT-PCR. The interaction between miR-19a-3p and ZEB2 was examined by a dual-luciferase reporter assay. Result Matrine attenuated H 2 O 2 -induced increases in ROS levels, apoptosis, and cytokine release in HTR/SVneo cells. Matrine also increased cell migration and invasion during oxidative stress by reducing miR-19a-3p expression. Moreover, miR-19a-3p can target ZEB2 directly. Conclusion During oxidative stress, matrine can reduce ROS levels, apoptosis, and infl ammation in human trophoblasts and promote migration and invasion via miR-19a-3p, off ering a potential new approach for the treatment of PE.

        SD-rat에 KIOM-MA128을 경구 투여 한 후 혈장 중 Matrine의 약물 동태

        KIOM-MA128 is a novel Korean herbal medicine with anti-atopic, anti-inflammatory and anti-asthmatic effects. This article presents the first pharmacokinetic study on KIOM-MA128. The purpose of this study was to characterize a pharmacokinetic characteristic of matrine, a potential marker of KIOM-MA128, in rats using population pharmacokinetic model. 1, 2 and 8 g/kg of KIOM-MA128 were administered to rats orally and plasma concentrations of matrine was determined by HPLC-MS/MS. Non-compartmental analysis (NCA) was performed using Phoenix?? and pharmacokinetic model was built using NONMEM??. This model was validated with internal validation which is visual predictive check (VPC) and bootstrap. The NCA result of matrine showed that Cmax was 294.24, 552.22 and 868.65 ng/ml, AUCinf was 1273.05, 2724.76 and 9743.25 ng · hr/ml and Tmax was 1, 1.3 and 2.3 hr for the doses of 1, 2, and 8 g/kg, respectively. The rat plasma concentrations were described very well with one-compartment model. Pharmacokinetic model for matrine was successfully developed and evaluated. Finally, our model is helpful to understand pharmacokinetic characteristic of KIOM-MA128.

      • LC-MS/MS를 이용한 농산물 중 생물 유래 유기농자재(nicotine, matrine) 시험법 검증

        최근 우리나라에서도 유기농업 혹은 무농약 재배 농가가 증가함에 따라 친환경유기농자재의 안전사용 등 안전관리를 위해서는 유기농자재 주요성분의 다양한 작물에 대한 잔류실태조사가 필요하다. 본 연구는 유기농자재의 작물 잔류성 자료를 확보하기 위해 제조, 유통되는 병해충 관리용 유기농자재의 식물 추출물 원료 중 matrine과 nicotine의 실태조사에 앞서 시험법을 검증하고자 수행하였다. 대상 작물은 5종의 대표 농산물(감귤, 감자, 고추, 대두, 현미)을 대상으로 진행하였다. 시험법은 아세토니트릴 10mL을 첨가하고 pH를 8.5로 조절한 뒤 d-SPE(dispersive-solid phase extraction)로 추출하여 LC-MS/MS로 분석하였다. 시험법의 유효성 검증을 위하여 PLS 수준 0.01 mg/kg을 포함하는 0.00025∼0.1 mg/kg 범위에서 검량선을 작성하였다. 모든 검체에서 상관계수(R<sup>2</sup>)는 0.99 이상 확보하였고, 정량한계(LOQ)는 0.01 mg/kg 이하로 나타났다. 각 작물별로 정량한계, 정량한계의 10배, 정량한계의 50배 수준으로 각각 처리하여 회수율을 검증한 결과, 평균 회수율은 72.1∼110.6%이었고 반복 간의 상대표준편차(CV) 값이 18.4% 이내로 나타났다. nicotine과 matrine 분석법의 검증 결과 유효성을 확인하였으며 이를 통해 국내에서 유통되고 있는 친환경 농산물들에 대한 nicotine과 matrine의 잔류실태를 조사하여 유기농자재 안전관리의 기초자료로 활용하고자 한다.

        The present study investigated the sedative effects of Sophora flavescens (SF) and its bioactive compound, matrine through performing locomotor activity test and the electroencephalography (EEG) analysis in the rat. The underlying neural mechanism of their beneficial effects was determined by assessing c-Fos immunoreactivity and serotonin (5-HT) in the brain utilizing immunohistochemical method and enzyme-linked immunosorbent assay. The results showed that SF and matrine administration had an effect on normalization of caffeine-induced hyperactivity and promoting a shift toward non-rapid eye movement (NREM) sleep. c-Fos-immunoreactivity and 5-HT level in the ventrolateral preoptic nucleus (VLPO), a sleep promoting region, were increased in the both SF and matrine-injected groups. In conclusion, SF and its bioactive compound, matrine alleviated caffeine-induced hyperactivity and promoted NREM sleep by activating VLPO neurons and modulating serotonergic transmission. It is suggested that SF might be a useful natural alternatives for hypnotic medicine.

        복숭아혹진딧물, Myzus persicae, 방제를 위한 제충국, 데리스, 고삼 추출물의 살충농도와 살충시간 결정

        제충국(Tanacetum cineariaiaefolium), 데리스(Derris elliptica), 고삼(Sophora flavescens) 추출물은 다양한 해충을 방제하는데 사용되고 있다. 하지만, 국내에서 판매되고 있는 식물추춞물 자재는 유효성분의 표기가 없고, 살충농도와 살충시간에 대한 자료가 전무한 상황이다. 본 연구에서는 상용화된 주요 식물추출물의 살충유효성분의 농도를 결정하고 복숭아혹진딧물에 대해 살충농도와 살충시간을 측정하였다. 식물추출물의 살충활성성분인 pyrethrins, rotenone, matrine과 oxymatrine의 농도는 액체 크로마토그래피에서 표준물질을 활용하여 질량분석을 통해 측정하였다. 식물추출 물을 농도별로 희석하여 복숭아혹진딧물에 살포하여 살충력을 측정하였다. 표준화합물과 비교한 후 질량분석 및 결정했습니다. Myzus persicae에 대한 lethal concentation과 lethal time을 조사했다. 살포 후 48시간 후 치사 농도(LC50)는 pyrethrins (20.4 ppm), roteone (34.1 ppm), matrine (29.6 ppm)였고, 100 ppm 살포한 LT50은 pyrethrins (13.4시간), rotenone (15.1시간), matrine (14.4시간)로 측정되었다. Kaplan-Meier 생존분석 결과, 100 ppm에서 세 가지 식물 추출물의 LT50은 대조구인 화학 살충제인 Sulfoxaflor를 살포 처리구보다 유의하게 빨랐습니다. 본 결과는 복숭 아혹진딧물 방제를 위해 식물추출물의 제형화에 단일 또는 혼합 제제를 개발하는데 기준 살충농도와 살충시간을 제고하는데 의미가 있다. Botanical extracts are employed in management of aphids. Extracts from Tanacetum cineariaiaefolium, Derris elliptica, and Sophora flavescens are widely used to control various insects. In this study, we determined concentrations of insecticidal active ingredients in commercial botanical extracts of these plants, and we investigated the time and concentration for lethal results with the green peach aphid, Myzus persicae. The concentrations of active ingredients, pyrethrins from T. cineariaiaefolium, rotenone from D. elliptica, and matrine and oxymatrine from S. flavescens, were determined after their fractionation by liquid chromatography followed by mass analysis and comparison with standard compounds. The extracts were tested for lethality in a bioassay with green peach aphids. Sprays at defined doses were applied to tobacco leaves infested with aphid nymphs. The lethal concentrations (LC50) were 20.4 ppm for pyrethrins, 34.1 ppm for rotenone, and 29.6 ppm for matrine at 48 h after treatments. At 100 ppm application levels, the lethal time LT50 was 13.4 h for pyrethrin, 15.1 h for rotenone, and 14.4 h for matrine. Kaplan-Meier analysis indicated the lethal times for the three botanical extracts at 100 ppm were significantly faster than application of a chemical insecticide, Sulfoxaflor, applied at the recommended level. These results provide baselines to develop and formulate single or mixed preparations containing botanical extracts to control green peach aphids on commercial crops.
