        미세다륜침(MTS)관리가 여성의 미세주름 및 탄력개선에 미치는 효과

        The aim of this study was prove that the effect of skin's light wrinkle and elasticity with treating by MTS and Vitamin E together and the validity of the efficiency in MTS, which is typical feature of skin aging and propose the criteria for effective treatment method. The study objects were 16 women with age of 30's and 40's, who were laxity of their skin elasticity condition. The first experimental group women was treated by MTS(Micro needle Therapy System), the second experimental group women was treated by MTS and Vitamin E together, studying each 8 persons respectively. The study objects were treated once a week for total 6 times and put in 30 min. per one treatment. The measurement was checked by total 4 times such as before the treatment, after one time treatment, after 6 times treatment and 2 weeks later after the final treatment, which was measured light wrinkle, elasticity, moisture, smoothness and pores on skin condition respectively. As for the result, this study turned out that the application of MTS and Vitamin E treatment together was more effective than MTS only, which brought more effective improvement for wrinkle, elasticity, moisture ratio and smoothness. As questioning about the condition of skin after treatment, we got a positive response in terms of applying for MTS and Vitamin E treatment together, which will be effectively considered for skin aging improvement program in the future. 본 연구는 미세다륜침(MTS)의 유효성에 대한 검증과 미세다륜침(MTS)과 비타민 E제품을 병행하였을 때 피부의 미세주름 및 탄력개선에 미치는 영향에 대해 알아보고 그 효과적인 관리방법의 기준을 제시하고자 하였다. 피부 탄력상태가 저하된 30, 40대 중년여성 16명을 선정하여 MTS를 실시한 8명은 MTS군, MTS와 비타민 E를 동시에 실시한 8명은 MTS+비타민 E군으로 명명하여, 실험을 진행하였다. 실험은 주 1회 총 6회 관리로, 1회 관리 시간은 총 30분 동안 시행하였다. 측정은 실험 전, 실험 1회 후, 실험 6회 후, 실험 종료 2주 후로 총 4회에 걸쳐 미세주름, 탄력도, 수분도, 거칠기, 모공 등의 피부상태를 측정하였다. 측정 결과 MTS만 단독으로 시행하였을 때 보다 MTS와 비타민 E 제품을 같이 시행하였을 때 미세주름, 탄력도, 수분도, 거칠기 개선에 효과가 큰 것으로 나타나 MTS+비타민 E관리가 미세주름 및 탄력개선 등 피부 노화개선에 효율적인 피부 관리 프로그램 방법으로 적용이 가능할 것으로 사료된다.

        미세 다륜침(MTS)과 AHA필링이 남성피부의 모공과 색소침착에 미치는 영향

        The aim of this study is to prove the effect of MTS on acne scars and large pores and propose the data on men's skin care which has been in the blind spot. MTS is the abbreviation of Microneedle Therapy Syetem and has a roller with 200 microneedle. It induces the natural regeneration of collagen by making the artificial wounds by microneedle roller. In aesthetic salons, AHA treatment has been mainly applied to large pores and acne scars. Recently, MTS arose a new treatment for them. Therefore, this study compared the effect of MTS and AHA. The study objects were 24 men with age of 20's with acne scars and large pores. The first experimental group was treatd by MTS. The second experimental group was treated by AHA. The control group was not treated. The study objects were treated once a week for 6 weeks. The experimental variables were the number of visible pores, the number of large pores among visible pores, the number of blackhead, the number of wrinkle, the area of hyperpigmentation and the brightness of complexion. They were measured before and after the experiment. As for the control group, there was no significant decrease in all of experimental variables. As for the experimental groups, all of experimental variables except the brightness of complexion decreased significantly. The experimental group treated by MTS was decreased in all of significant experimental variables more than the experimental group treated by AHA. Conclusively, this study proved that MTS treatment was effective to men's skin with acne scars and large pores. 본 연구는 미세 다륜침(MTS)의 효능을 검증하고, 피부 관리의 사각지대에 있는 남성피부를 대상으로 보다 과학적이고, 효율적인 자료를 제시하고자 하였다. 모공 관리와 여드름 자국 개선에 효과적인 관리법으로 AHA필링관리가 가장 많이 시행되고 있으며, 최근 MTS관리가 새로운 관리법으로 떠오르고 있다. 본 연구는 두 관리의 효과를 비교하기 위해 시행되었다. 대상자는 여드름 자국과 모공이 넓은 24명의 20대 남성으로 MTS관리와 AHA필링관리로 실험군을 설계했고, 무 처치 군을 대조군으로 두었다. 실험은 6주 동안 주 1회 총 6번 진행 하였고, 실험 전과 실험 후에 모공 수, 블랙헤드 수, 소 색소침착의 수와 면적, 피부밝기를 측정 하였다. 실험 결과 대조군은 모든 실험 변수가 증가하여 실험의 효과가 없는 것으로 나타났고, AHA군과 MTS군은 모든 실험 변수에 대해 효과가 나타났다. MTS군과 AHA군을 비교하면 MTS군이 AHA군에 비해 변화의 폭이 더 컸다. 결론적으로 MTS관리가 전형적인 남성피부의 특징인 모공이 넓고, 여드름 자국이 있는 색소침착 피부에 효과를 나타낸다는 사실을 검증하였다.

        Multicellular tumor spheroid (MTS) 배양에 의한 EMT에서 HMGB1의 역할

        암조직의 내부에서 hypoxia와 glucose depletion 등의 microenvironmental stress를 받게 되면 necrosis가 유도되고, 실제로 암 조직 내부에서 necrotic core 형성이 관찰된다. Necrotic cells은 high mobility group box 1(HMGB1)를 extracellular space로 방출하는 것으로 알려져 있다. 방출된 HMGB1은 tumor-promoting cytokine으로 작용함으로써 tumor development 시 inflammation, metabolism 및 metastasis에 기여한다. 본 연구에서 non-invasive breast cancer cells MCF-7이 solid tumor의 in vitro model인 multicellular tumor spheroid (MTS) 배양을 통해 완전한 구형의 MTS를 형성하며 MTS가 성장함에 따라 inner region에 necrosis가 유도됨을 밝혔다. 또한 MCF-7 세포의 MTS 배양은 Snail 의존적으로 epithelial-mesenchymal transition (EMT)를 유도함을 관찰하였다. HMGB1의 cell surface receptors인 RAGE, TLR2, TLR4 발현이 MTS 배양에 의해 증가됨을 발견하였다. RAGE, TLR2, TLR4 를 knockdown한 결과 MTS 성장을 억제할 뿐만 아니라 MTS에 의해 증가되는 Snail 발현을 억제함을 밝혔다. 이는 MTS-induced Snail 발현이 RAGE/TLR2/TLR4의존적으로 조절되며 RAGE/TLR2/TLR4-Snail이 MTS 성장에 관여하는 것으로 보인다. 또한 Snail, RAGE, TLR2, TLR4 shRNA는 MTS 배양에 의해 유도되는 EMT를 억제함을 밝혔다. 실제 인간 암조직에서 정상조직에 비해 RAGE, TLR2, TLR4 유전자의 발현이 높음을 관찰하였다. 따라서 HMGB1이 RAGE/TLR2/4-Snail axis를 통해 MTS 배양에 따른 성장 및 EMT에 중요하게 작용할 것으로 예상된다. As tumors develop, they encounter microenvironmental stress, such as hypoxia and glucose depletion, due to poor vascular function, thereby leading to necrosis, which is observed in solid tumors. Necrotic cells are known to release cellular cytoplasmic contents, such as high mobility group box 1 (HMGB1), into the extracellular space. The release of HMGB1, a proinflammatory and tumor-promoting cytokine, plays an important role in promoting inflammation and metabolism during tumor development. Recently, HMGB1 was shown to induce the epithelial-mesenchymal transition (EMT) and metastasis. However, the underlying mechanism of the HMGB1-induced EMT, invasion, and metastasis is unclear. In this study, we showed that noninvasive breast cancer cells MCF-7 formed tightly packed, rounded spheroids and that the cells in the inner regions of a multicellular tumor spheroid (MTS), an in vitro model of a solid tumor, led to necrosis due to an insufficient supply of O2 and glucose. In addition, after 7 d of MTS culture, the EMT was induced via the transcription factor Snail. We also showed that HMGB1 receptors, including RAGE, TLR2, and TLR4, were induced by MTS culture. RAGE, TLR2, and TLR4 shRNA inhibited MTS growth, supporting the idea that RAGE/TLR2/TLR4 play critical roles in MTS growth. They also prevented MTS culture-induced Snail expression, pointing to RAGE/TLR2/TLR4-dependent Snail expression. RAGE, TLR2, and TLR4 shRNA suppressed the MTS-induced EMT. In human cancer tissues, high levels of RAGE, TLR2, and TLR4 were detected. These findings demonstrated that the HMGB-RAGE/TLR2/TLR4-Snail axis played a crucial role in the growth of the MTS and MTS culture-induced EMT.

        Mobile Trading System 앱 UI 디자인의 사용성 연구 - 국내 MTS앱 4종을 중심으로 -

        코로나19 바이러스의 영향으로 국내 주식 시장의 20 대 개인투자자들이 증가하여, 이들의 지속적인 Mobile Trading System(MTS)앱 사용과 서비스 만족도 제고 를 위해 UI 디자인의 사용성에 관한 연구가 필요하다. 본 연구에서는 20대 개인투자자들이 가장 많이 찾은 국 내 MTS 앱 4종을 대상으로 사용성 평가를 진행하였다. 먼저 선행연구를 통해 MTS 앱의 UI 디자인 구성요소 와 UI 사용성 평가원칙을 도출하였고, 국내 MTS 앱 4 종의 UI 디자인 구성요소를 비교 분석하였다. 사용성 평가는 20대 신규투자자를 실험 대상자로 선정하여, 각 MTS 앱 별로 태스크를 수행한 후 설문과 심층 인터뷰 를 진행하였다. 그 결과를 종합하여 발견한 문제점은 첫 째, 일관성에 관한 문제점으로 버튼의 위치가 화면별로 달라지는 경우 사용자에게 혼란을 주고, 메뉴와 버튼이 과다하게 제공되면 사용에 방해요소가 된다는 것이다. 둘째, 심미성과 관련된 문제점으로는 UI 색상의 종류가 많으면 사용자의 시선을 분산시킨다는 점이다. 셋째, 학 습 용이성과 관련된 문제점으로 앱의 기능과 용어에 대한 설명이 부족하면 사용자가 이용하지 못한다는 것과 유용한 정보를 제공하는 첫 화면이 없는 경우 원하는 정보를 찾는 데 어려움을 겪는다는 점이다. 넷째, 효율 성과 관련된 문제점은 개인화 작업 시에 작은 버튼과 아이콘은 사용자가 발견하기 어려워 기능을 이용하지 못한다는 점이다. 마지막으로 가시성과 관련된 문제점으 로는 앱의 작동 방식과 기능에 대한 설명 및 안내가 눈 에 띄지 않아 사용자가 알기 어렵다는 점이다. 본 연구 결과를 통해 발견된 문제점을 개선함으로써 20대 신규 투자자를 위한 MTS 앱의 UI 디자인 사용성과 서비스 만족도가 제고되기를 기대한다. Due to the influence of COVID-19, the number of individual investors in their 20s in the domestic stock market has increased, and research on the usability of UI design is needed to continuously use Mobile Trading System (MTS) apps and improve service satisfaction. In this study, usability evaluation was conducted on four domestic MTS apps that individual investors in their 20s visited the most. First, through previous studies, the UI design components of the MTS app and the UI usability evaluation principle were derived, and the UI design components of four domestic MTS apps were compared and analyzed. For usability evaluation, new investors in their 20s were selected as subjects, and each MTS app was used to perform tasks, and then surveys and in-depth interviews were conducted. The problem found by synthesizing the results is that first, the problem of consistency is that if the position of the button varies by screen, it confuses the user, and if the menu and buttons are provided excessively, it interferes with use. Second, the problem related to aesthetics is that if there are many types of UI colors, the user's gaze is distributed. Third, problems related to ease of learning are that users cannot use the app if there is a lack of explanation of its functions and terms, and that it is difficult to find the information they want without the first screen that provides useful information. Fourth, the problem related to efficiency is that small buttons and icons are difficult for users to find during personalization work, so they cannot use the function. Finally, the problem related to visibility is that it is difficult for users to know because there is no noticeable explanation and guidance on how the app works and functions. By improving the problems found through the results of this study, it is expected that the UI design usability and service satisfaction of MTS apps for new investors in their 20s will be improved.

        모바일 증권거래시스템(Mobile Trading System)의 지속적 이용의도에 미치는 영향요인에 관한 연구

        스마트폰 등 다양한 스마트 디바이스의 확산으로 모바일 증권거래시스템을 활용한 주식매매와 주식정보를 습득하는 스마트 증권투자방식이 새로운 이슈로 떠오르고 있다. 이러한 변화는 금융회사에게 경쟁력 확보를 위한 기회이자 새로운 경쟁시장을 선보이고 있다. 따라서 본 연구에서는 MTS 이용만족에 영향을 미치는 MTS 특성에 대해 알아보고, MTS의 지속이용에 영향을 미치는 고객 특성을 밝히고자 하였다. 이를 위해 MTS 특성에 대한 연구를 통해 반응성, 접근성, 정보성 등의 요인을 도출하고, MTS 이용만족에 미치는 영향관계를 살펴보았다. 또한 MTS 지속이용 고객의 특성을 파악하여 증권사에 시사점을 제공하고자 MTS를 이용하는 고객의 개인적 특성을 발굴 및 그 영향관계를 실증하였다. 그 결과 MTS 특성에서 반응성, 접근성, 정보성이 MTS 이용만족에 유의한 영향을 미치는 요인으로 나타났고, 개인의 투자성향이 MTS의 지속적 이용의도에 유의한 조절효과가 있는 것을 확인하였다. 이 연구의 결과들은 향후 MTS의 활성화 연구 및 증권사의 전략수립에 유용한 시사점을 제공할 것으로 기대한다. As smart phones are widely used, smart stock investment method enabling people to trade stock and access related information through mobile stock trading system is getting to catch public`s attention. This transition of paradigm is providing a chance to enhance competitiveness and also envisioning new market to financial companies. Thus this study is to distinguish user satisfaction factors and establish correlation between Mobile Trading System user satisfaction and intention of continuous use. It is intended to verify empirically effects of independent variables; reactivity, accessibility, informativity, design and earning cost which are set from the characteristics of MTS on user satisfaction of MTS as a parameter and ultimately effects of personal investment propensity and occupational profession as moderating variables on intention of continuous use of MTS as a dependent variable. It was shown that reactivity, accessibility and informativity from characteristics of MTS have valid effect on MTS user satisfaction and personal investment propensity has valid moderating effect on intention of continuous use of MTS. Having above conclusion, it is expected to provide useful implications for study on revitalization of MTS and establishing stock firm`s business strategy.

        MTS 서비스품질이 증권회사의 경영성과에 미치는 영향

        본 연구는 최근 온라인 증권거래에서 이용 비중이 증가하고 있는 MTS 이용자를 대상으로 MTS의 어떤 서비스 품질요인이 증권회사의 경영성과에 유의한 영향을 미치는 지를 분석하였다. 본 연구결과를 요약하면 다음과 같다. 첫째, MTS 서비스 품질요인과 재무적 성과의 회귀분석 결과 유형성, 신뢰성, 반응성, 공감성 및 확신성의 다섯 개 요인이 매출액증가율, 매출액이익률, 총자산회전율, 유동성비율 및 부가가치율의 재무적 성과에 유의한 정(+)의 영향을 미치는 것으로 나타났다. 하지만 신속성은 유의한 영향을 미치지 않았다. 둘째, MTS 서비스 품질요인과 비재무적 성과의 회귀분석 결과 유형성, 반응성, 공감성 및 확신성의 4개 변수가 1% 수준에서 통계적으로 유의한정(+)의 부호를 나타내었다. 신속성과 신뢰성은 유의한 영향을 미치지 않았다. 이와 같은 연구결과는 MTS를 이용하는 고객의 요구사항을 인식하고 이를 개선하기 위한 구체적 활동에 관한 정보를 제공함으로써, 고객과 증권회사 모두가 만족하는 윈윈전략의 수립에 많은 정보를 제공할 것으로 기대된다. The objectives of this study are identify the factors that affect the quality of MTS services, to determine the effects of these service quality factors on securities firms. A total of 251 questionnaires were used for the relationship between MTS services quality and business performance in securities firms. Frequency analysis, reliability analysis, validity analysis, correlations analysis, multiple regression analysis were used. After analyzing the effects of the factors affecting the quality of MTS services on business performance in securities firms. The results are as follows: First, The Five factors, i.e., tangibles, reliability, responsiveness, empathy and assurance represent statistically marginal significance in explaining the relationship between MTS service quality factors and business performance in securities firms. Second, The Four factors, i.e., tangibles, responsiveness, empathy and assurance represent statistically marginal significance in explaining the relationship between MTS service quality factors and business performance in securities firms. Securities firms should consider this result in order to increase business performance.

        S-MTS를 이용한 강판의 표면 결함 진단

        Steel plate faults is one of important factors to affect the quality and price of the steel plates. So far many steelmakers generally have used visual inspection method that could be based on an inspectors intuition or experience. Specifically, the inspector checks the steel plate faults by looking the surface of the steel plates. However, the accuracy of this method is critically low that it can cause errors above 30% in judgment. Therefore, accurate steel plate faults diagnosis system has been continuously required in the industry. In order to meet the needs, this study proposed a new steel plate faults diagnosis system using Simultaneous MTS (S-MTS), which is an advanced Mahalanobis Taguchi System (MTS) algorithm, to classify various surface defects of the steel plates. MTS has generally been used to solve binary classification problems in various fields, but MTS was not used for multiclass classification due to its low accuracy. The reason is that only one mahalanobis space is established in the MTS. In contrast, S-MTS is suitable for multi-class classification. That is, S-MTS establishes individual mahalanobis space for each class. Simultaneous implies comparing mahalanobis distances at the same time. The proposed steel plate faults diagnosis system was developed in four main stages. In the first stage, after various reference groups and related variables are defined, data of the steel plate faults is collected and used to establish the individual mahalanobis space per the reference groups and construct the full measurement scale. In the second stage, the mahalanobis distances of test groups is calculated based on the established mahalanobis spaces of the reference groups. Then, appropriateness of the spaces is verified by examining the separability of the mahalanobis diatances. In the third stage, orthogonal arrays and Signal-to-Noise (SN) ratio of dynamic type are applied for variable optimization. Also, Overall SN ratio gain is derived from the SN ratio and SN ratio gain. If the derived overall SN ratio gain is negative, it means that the variable should be removed. However, the variable with the positive gain may be considered as worth keeping. Finally, in the fourth stage, the measurement scale that is composed of selected useful variables is reconstructed. Next, an experimental test should be implemented to verify the ability of multi-class classification and thus the accuracy of the classification is acquired. If the accuracy is acceptable, this diagnosis system can be used for future applications. Also, this study compared the accuracy of the proposed steel plate faults diagnosis system with that of other popular classification algorithms including Decision Tree, Multi Perception Neural Network (MLPNN), Logistic Regression (LR), Support Vector Machine (SVM), Tree Bagger Random Forest, Grid Search (GS), Genetic Algorithm (GA) and Particle Swarm Optimization (PSO). The steel plates faults dataset used in the study is taken from the University of California at Irvine (UCI) machine learning repository. As a result, the proposed steel plate faults diagnosis system based on S-MTS shows 90.79% of classification accuracy. The accuracy of the proposed diagnosis system is 6-27% higher than MLPNN, LR, GS, GA and PSO. Based on the fact that the accuracy of commercial systems is only about 75-80%, it means that the proposed system has enough classification performance to be applied in the industry. In addition, the proposed system can reduce the number of measurement sensors that are installed in the fields because of variable optimization process. These results show that the proposed system not only can have a good ability on the steel plate faults diagnosis but also reduce operation and maintenance cost. For our future work, it will be applied in the fields to validate actual effectiveness of the proposed system and plan to improve the accuracy based on the results.

        突厥 阿史那氏의 原住地 再檢討 ― 阿史那氏의 發生과 移住, 그리고 勢力化 過程 ―

        The short, unclear record that says after Ju-Qu-Shi(沮渠氏) power was defeated by Northern Wei(北魏) in the mid-fifth century, he escaped to southern Altai(金山) Mts. and became an ironworker of Rou-Ran(柔然), makes it uncertain where Ashina clan(阿史那氏) occurred and when and why he moved to southern Altai Mts. In order to fill the temporal gap of that short record and figure out the birthplace and original residence of Ashina clan, this study newly examines another myth that says Ashina clan occurred when ten sons born by a female wolf in the enormous basin in the hole of the cave in the mountain located in the north of Turfan (Gao-chang-Guo, 高昌國) married the local women. And connecting the two records temporally, this study examines their feasibility as data, and also it re-examines and connects the extracts found as facts with the result of the field trip and previous research results. As a result, unlike the previous view that the birthplace of Ashina clan was in the base of Tian-shan(天山) Mts. in southern Jimsar(吉木薩爾), this study found the description of a large basin inside the hole of the cave in the mountain located in northern Turfan and the result that the Barköl(巴里坤) basin in the eastern mountain range of Tian-shan Mts. with similar topography could be the birthplace. Moreover, this could be more concertized through the process of arranging the fierce political competition around Tian-shan Mts. before and after the sixth century when Ashina clan living in the Barköl basin could not but move to the southern Altai Mts.. Therefore, by re-examining the birthplace of Ashina clan, this study could systematize the early history of Türks(突厥) more clearly which had been left as the time of mythology with no proper understanding. In other words, Ashina clan occurred in the Barköl basin in the late sixth century and had to move to southern Altai Mts. around 508 during the war between Gao-Che(高車) and Rou-Ran. After he grew up, he subjugated Gao-Che in Jimasar at the northern Tian-shan Mts. and secured power and also defeated even Rou-Ran in the steppe of Mongolia to found the nation in 552. All this process is closely connected to the competition between Chinese dynasties, Rou-Ran, Ephtalite, Gao-Che, Tu-Yu-Hun(吐谷渾), and many other Oasis powers divided into the north and the south in the Tian-shan Mts. region.

        MTS 도입 전·후 KOSPI200 현·선물시장에서의 정보효율성에 관한 연구

        동 연구는 우리나라 KOSPI200 현, 선물시장 간의 가격과 거래량 정보를 이용하여 모바일트레이딩시스템(MTS) 도입 전과 후의 정보 효율성 변화정도를 실증 분석하고자 하였다. 이를 위해 2007년 1월 2일부터 2012년 12월 28일까지의 한 시간대별 일 중 데이터를 이용하여 그랜저(Granger)인과관계를 분석하였으며 주요 실증분석결과는 다음과 같다. 첫째, KOSPI200 현물과 선물시장의 수준변수들은 단위 근이 존재하는 불안정한 자료이었으나 차분변수인 거래변화량과 수익률은 단위 근이 존재하지 않는 안정적인 시계열로 나타났다. 둘째, 현, 선물시장의 변수에 대한 각 시계열의 수준변수들 사이 장기적인 균형 관계를 분석하기 위해 공적분 관계를 분석한 한 결과, 통계적 유의한 수준에서 기각되어 장기적인 균형 관계가 존재함을 확인되었다. 셋째, 그랜저(Granger) 인과 관계 분석결과, MTS 도입 이전에 비해 도입 이후 선물수익률에 대한 현물수익률 및 거래변화량간의 영향이 다소 개선 된 것으로 나타났다. 넷째, KOSPI200 선물시장의 현물시장에 대한 영향력은 선물수익률에 대한 현물수익률의 영향이 MTS 도입 이후 다소 감소한 것으로 나타났다. 이러한 분석결과로 국내 KOSPI200 현, 선물시장 간의 정보효율성은 MTS 도입 이전에 비해 도입 이후 다소 개선 된 것을 확인할 수 있었다. This study empirically analyzes the degree of information efficiency change before and after the introduction of mobile trading system (MTS) using KOSPI200 futures and futures market price and volume information. To do this, we analyze Granger causality using daily data from January 2, 2007 to December 28, 2012, and the results of the empirical analysis are as follows. First, the level variables of KOSPI200 spot and futures market are unstable data with unit root, but the transaction variance and yield as differential variable are stable time series with no unit root. Second, the long-term equilibrium relationship between the time series variables of the current and futures markets is analyzed and the long-term equilibrium relation is rejected at the statistically significant level. Third, the Granger causality analysis showed that the effect of spot returns and trading changes on futures returns after introduction was somewhat improved compared to before MTS was introduced. Fourth, the influence of the KOSPI200 futures market on the spot market decreased slightly after the introduction of MTS. As a result of this analysis, the information efficiency of KOSPI200 and futures markets in Korea has been somewhat improved after introduction compared to before MTS introduction.

        Cationic Oligopeptide-Functionalized Mitochondria Targeting Sequence Show Mitochondria Targeting and Anticancer Activity

        Mitochondrial drug delivery systems require development of highly selective mitochondria-targeting carriers. In this study, we report that mitochondria targeting sequence (MTS)-hybrid cationic oligopeptide, MTS-H3R9, shows the dual role of a mitochondria targeting vector along with anticancer effect for cancer therapy. In cytotoxicity assays, MTS-H3R9 was shown to be more effective than MTS. MTS-H3R9 showed significant cell penetration and internalization activity compared to that of MTS along with more efficient escape from lysosome to the cytosol. We showed efficient targeting of MTS-H3R9 to mitochondria in HeLa cell line. Furthermore, we exhibited anticancer agent properties that mitochondrial-accumulated MTS-H3R9 caused cell death by reactive oxygen species generation and loss of mitochondrial membrane potential. MTS-H3R9 exhibited dramatically increased anticancer activity in 3D spheroids as well as in a 2D culture model. We demonstrated that MTS-H3R9 provides dual potentials both as a vehicle for targeted delivery and as a cancer treatment agent for therapeutic applications.
