      • Isolation and characterization of the insect growth regulatory substances from actinomycetes

        <P><B>Abstract</B></P> <P>Insect growth regulators (IGRs) are attractive alternatives to chemical insecticides. Since it has been reported that secondary metabolites from actinomycetes show insecticidal activities against various insect pests, actinomycetes could be a potential source of novel IGR compounds. In the present study, insect juvenile hormone antagonists (JHANs) were identified from actinomycetes and their insect growth regulatory and insecticidal activities were investigated. A total of 363 actinomycetes were screened for their insect growth regulatory and insecticidal activities against <I>Aedes albopictus</I> and <I>Plutella xylostella</I>. Among them, <I>Streptomyces</I> sp. AN120537 showed the highest JHAN and insecticidal activities. Five antimycins were isolated as active compounds by assay-guided fractionation and showed high JHAN activities. These antimycins also exhibited significant insecticidal activities against <I>A</I>. <I>albopictus</I>, <I>P</I>. <I>xylostella</I>, <I>F</I>. <I>occidentalis</I>, and <I>T</I>. <I>urticae</I>. Moreover, dead larvae treated with these antimycins displayed morphological deformities that are similar to those of JH-based IGR-treated insects. This is the first report demonstrating that the insecticidal activities of antimycins resulted from their possible JHAN activity. Based on our results, it is expected that novel JHAN compounds potentially derived from actinomycetes could be efficiently applied as IGR insecticides with a broad insecticidal spectrum.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>Streptomyces</I> sp.       • Construction of A Recombinant Bacillus thuringiensis 19-22 Strain Exhibiting Antifungal and Dual Insecticidal Activities

        A new Bacillus thuringiensis isolate 19-22 (Bt 19-22) exhibited high anti-fungal activity against barley powdery mildew (Blumeria graminis f. sp. hordei). The cry gene content of Bt 19-22 comprised cry1Aa, cry1Ab, cry1Ac and cry1D which have high insecticidal activity against lepidopteran larvae. We tried to confer a dipteran insecticidal activity to Bt 19-22 for constructing a recombinant strain which has multiple functions, anti-fungal and dual insecticidal activity. The insecticidal cry11Aa gene of B. thuringiensis was constructed under cry1Ac promoter in an E. coli-B. thuringiensis shuttle vector (pPro11A). The plasmid, pPro11A was introduced into Bt 19-22 isolate by electroporation and four transformants which had different cry gene contents were identified by PCR with cry11Aa and cry1-type specific primers. Among them, a Bt 19-22 transformant (11A/19-22 No. 7) expressed Cry11A protein (approximately 70 kDa) successfully without change of its inherent characteristics such as Cry protein expression and antifungal activity. The insecticidal activity of 11A/19-22 No. 7 was checked against Plutella xylostella and Culex pipiens. These results suggests that the recombinant strain shows dual insecticidal activity against lepidopteran and dipteran larvae as well as antifungal activity.

      • Insecticidal activity of Streptomyces sp. with Juvenile hormone antagonist activity against Plutella xylostella

        Streptomyces has been reported to produce various secondary metabolites which have the potential to become environmentally safe insecticides. In this study, 1,274 streptomyces culture filtrates were screened for their JHAN activity in order to identify novel insecticidal compounds. 34 isolates with high levels of JHAN activity were selected, and their insecticidal activities were tested against Plutella xylostella larvae. Among them, IMBL-263 which was revealed to be Streptomyces anulatus by 16s rRNA sequencing showed the highest insecticidal activity. Also, systemic activities of secondary metabolites extracted from the S. anulatus on plant, Brassica napus, were investigated. These results suggested that secondary metabolites from the S. anulatus might be useful for development of novel environmentally benign insecticides.

        Insecticidal spectrum of fluralaner to agricultural and sanitary pests

        Fluralaner, as an isoxazoline insecticide, has been developed as antiparasitic (namely Bravecto®) with selective toxicity for insect versus mammals. Even though the toxic effect of fluralaner to several insects have been already examined, but also more agricultural and sanitary insects should be tested before it could be used widely. Four insects, including two agricultural insects, one sanitary insect and one stored product insect were tested in the present study. Compared to the broad-spectrum and high activity insecticide of fipronil, fluralaner showed mostly higher activity to tested pests, Chilo suppressalis, Tribolium castaneum, Spodoptera litura except Blattella germanica with topical application. Through summarizing and analyzing various published data, we concluded that, to date, the toxic activity of fluralaner to 19 insects from 11 orders were already examined, and fluralaner displayed excellent activity. Compared to fipronil, fluralaner mostly shows more excellent activity to tested insect with maximal 1.67 ×104-fold difference so much. In the present study, the toxicity activity of fluralaner was compared to the broad-spectrum phenylpyrazole insecticide fipronil to tested insect pests and the results exhibited that fluralaner would be a novel alternative insecticide with broad insecticidal spectrum and be widely used in more other areas not only as veterinary drug.

      • Insecticidal activities of methanol extracts and compounds from Asiasarum sieboldii against Plutella xylostella and Nilaparvata lugens

        To find an alternative for synthetic pesticides, methanol extract from plant samples were tested for their insecticidal activity against insect. The extract of Asiasarum sieboldii had strongly insecticidal activity against Plutella xylostella. Roots of A. sieboldii were extracted with methanol, and the concentrated extract was partitioned with n-hexane, ethylacetate, n-buthanol and H2O. The highest activity was shown in the hexane fraction. Activity-guided fractionation led to the isolation of two amides from hexane fration through the repeated silica gel column chromatographic separations. From the interpretation of spectropic data including NMR, MS, IR, the chemical structures of compounds were determined as dodeca-2E,4E,8Z,10Z-tetraenoic acid isobutylamide and dodeca-2E,4E,8Z, 10E-tetraenoic acid isobutylamide. These compounds showed insecticidal activity on P. xylostella by 96.7% at 100ppm. The liquid formulation controlled on cabbage effectively. The extract and compounds from A. sieboldii showed insecticidal activity against Nilaparvata lugens. As a naturally occurring pesticide, A. sieboldii could be useful as a new botanic insecticide.

        Insecticidal and Acaricidal Activities of Domestic Plant Extracts against Five Major Arthropod Pests

        Methanol extracts from 420 samples of 173 plant species in 58 families were tested at 5000 ppm for their insecticidal and acaricidal activities against five economically important arthropod pests by spray method. The responses varied with arthropod pest species, plant species and plant tissue sampled. In a test with Nilaparvata lugens Stal, extracts from Zanthoxylum piperitum barks, Chamaecyparis obtusa leaf and Quercus salicina leaf showed potent insecticidal activity. With Plutella xylostella L., potent larvicidal activity was observed from extracts of Platycarya strobilacea wood, Meliosma myriantha barks, Sophora japonica leaf, Zanthoxylum piperitum barks, and Pinus thunbergii wood. Methanol extracts of Sophora japonica leaf and Zanthoxylum piperitum barks showed high insecticidal activity against Spodoptera litura. In a test with Tetranychus urticae Koch, extract from Carpinus coreana leaf, Firmiana simplex barks, Elaeagnus macrophylla leaf, Aralia elata leaf, Comus controversa barks and Chamaecyparis obtusa leaf exhibited strong acaricidal activity. As a naturally occurring pest control agent, Zanthoxylum piperitum barks could be useful as new insecticidal and acaricidal products against various arthropod pests.

        Insecticidal activity of five Chinese medicinal plants against Plutella xylostella L. larvae

        Twenty-five extracts of five Chinese medicinal plant species were screened for insecticidal activity against the diamondback moth, Plutella xylostella L., larvae by a leaf dipping bioassay method. The ethyl acetate,ethanol, and acetone extracts of Veratrum nigrum L. root and rhizome, the ethyl acetate extract of Phytolacca americana L. root, and the petroleum ether extract of Pseudolarix kaempferi Gord. root bark were effective against P. xylostella. Among them, the ethyl acetate extract of V. nigrum showed the strongest insecticidal activity against the second and third instar larvae of P. xylostella, with LC50 values of 225 and 335 ppm,respectively. The other extracts gave little or no insecticidal activity against P. xylostella. These results indicated that V. nigrum may be a promising naturally occurring agent for P. xylostella larval control, and that the organic solvent used for plant extraction can affect its insecticidal activity.

        This study evaluated the activity of biocidal compounds isolated from natural products and confirmed applicable possibility as conservative agents for orgainic cultural heritages damaged by biological degradation. we used methanol extracts of 11 Korean trees and 17 medicinal plants as natural resources. Among these extracts, Phellodendron amurense was investigated on antifungal activity against Coniophora puteana, Lentinus lepideus, Tyromyces palustris and Aspergillus niger. Asarum sieboldii was investigated on insecticidal activity against adult of Lasioderma serricorne. First, the extract of Phellodendron amurense was partitioned into hexane, dichloromethan and ethylacetate fraction. Among them, the ethylacetate fraction had the most significant antifungal activity. In addition, Asarum sieboldii essential oil showed an effective antifungal and insecticidal activity. As a result, the extract of Phellodendron amurense and Asarum sieboldii essential oil could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and Lasioderma serricorne.

        Structural Characteristics that Influence on the Insecticidal Activity of 2-(n-Octyl)pseudothiourea Analogues against the Diamondback Moth (Plutella xylostella, L.)

        Structural characteristics that influence on the insecticidal activity ($pI_{50}$</TEx>) of 2-(n-octyl)isothiourea analogues (1-45) against the diamondback moth (Plutella xylostella, L.) based on three dimensional quantitative structure activity relationships (3D-QSARs) were discussed quantitatively using a comparative molecular field analysis (CoMFA) and a comparative molecular similarity indeces analysis (CoMSIA) methods. The statistical values of the CoMFA 2 model were better than those of the CoMSIA 1 model. The CoMFA 2 model was the optimized model with the correlativity (the training set: Ave. = 0.104 & PRESS = 0.613) and the predictability (the test set: Ave. = 0.086 & PRESS = 0.096). Insecticidal activities with the optimized CoMFA 2 model were dependent upon steric factors (79.4%) of $R_1-R_3$ substituents. From the analytical results of CoMFA contour maps, it is predicted that the R1 substituent of 1-45 which has a steric favor in a broad space, $R_2\;and\;R_3$ groups with a steric favor in a narrow space and a H-bond donor favor would have better the insecticidal activity.

      • Insecticidal activity of secondary metabolites from Streptomyces celluloflavus against Spodoptera exigua

        The beet armyworm, Spodoptera exigua, is one of the worldwide distributed agricultural pest insects and has been known to show high resistance to conventional chemical insecticides. Since it has been reported that secondary metabolites from actinomycetes show insecticidal activities against various insect pests, actinomycetes could be a potential source of insecticidal compounds. In this study, culture extracts of Streptomyces strains were tested for their insecticidal activity against Spodoptera exigua. Among them, culture extracts of IMBL-0003 strain which was identified as Streptomyces celluloflavus showed a high insecticidal activity (more than 90% mortality). These results suggested that secondary metabolites of this isolate could have potentials to be a efficient eco-friendly pesticide for controling Spodoptera exigua.
