The Effect of Hematin and Hemoglobin on Bovine Platelet Aggregation and Thromboxane Synthesis

Washed bovine platelets were stimulated with hematin or hemoglobin in the presence or absence of ADP and the extent of aggregation and thromboxane formation were estimated. When hematin was added to the bovine platelet suspension, the aggregation increased in proportion to the concentration of hematin added. Hemoglobin, even in higher concentration than hematin, did not gave the comparable result. Addition of ADP to the washed platelet suspension in the presence of hematin was found to induce considerable aggregation, depicting synergistic effect of ADP and hematin on platelet aggregation. Synthesis of thromboxane was also measured by incorporating [1-^14C] arachidonic acid in to the platelets and analyzing the radio-labeled products fromed during the reaction by TLC and liquid scintillation counting. It was found that the stimulatory effect of hematin and hemoglobin on platelet aggregation was accompanied with the increased synthesis of TXB_2. In addition aggregation of platelets and formation of thromboxane induced by hematin could be blocked by pretreatment of platelets with indomethacin. The amount of cAMP and cGMP were reduced (63% and 45%, respectively) in platelets treated with 3μM of hematin. These findings suggest that hematin stimulates platelet aggregation by activating thromboxane synthesis in the platelets.

Ion-exchange polymeric stationary phases presenting amino acid and amino groups were prepared by the surface grafting of glycidyl methacrylate onto a silica gel surface and subsequent amination. Three kinds of amino acids-L-arginine (Arg), D-lysine (Lys), and D-histine (His)-were used in this study. An ion-exchange polymeric stationary phase presenting ethylene diamine (EDA) was also prepared by surface graft polymerization. Separation of the model proteins bovine serum albumin (BSA), chick egg albumin (CEA), and hemoglobin (Hb) was performed using the amino acid- and amine-derived columns. In separating the CEA/BSA mixture, the resolution time of BSA was longer than that of CEA when using the EDA column, whereas the resolution time of BSA was shorter than that of CEA when using the Arg, Lys, and His columns. In the separation of the Hb/BSA mixture, the resolution time of BSA was longer than that of Hb in the EDA column, whereas the resolution time of BSA was shorter than that of Hb in the amino acid columns (D-Lys, L-Arg, and D-His).
Hb Heathrow [β103(G5)Phe→Leu], a First Report in an Asian Patient with Erythrocytosis

Congenital erythrocytosis (CE) is a rare and heterogeneous disease. The high oxygen affinity hemoglobin (Hb) variants are the most common cause of CE. Herein, we report a Korean patient with isolated erythrocytosis. A 25-year-old man was referred to our hospital for evaluation of high Hb level (Hb 20.4 g/dL, hematocrit 58%, reticulocyte count 2.90%, white blood cell count 6.83×109/L, and platelet count 195×109/L). Bone marrow biopsy revealed normocellular marrow without myeloproliferative features. JAK2 (V617F, exon 12), CALR (exon 9), and MPL W515K/L mutations were not detected. P50 (partial pressure at which Hb is half saturated with oxygen), which is an indicator of left-shift of oxygen dissociation curve (high oxygen affinity state), was 14.3 mm Hg (reference value 22.6–29.4 mm Hg). He was suspected to have CE. Mutation analysis of the HBB gene revealed the known Hb variant, Hb Heathrow [β103(G5)Phe→Leu]. This is the first report of Hb Heathrow in Asian.

Hereditary hemolytic anemia comprises a group of disorders in which red blood cells are destroyed faster than they are produced in the bone marrow; various hereditary factors can cause this condition, including production of defective Hb and erythrocyte membrane. Recently, we identified Hb Koriyama, a rare Hb variant that was undetectable in Hb electrophoresis and stability tests, in a patient with severe hemolytic anemia. This is the first study to show the nucleotide-level sequence variations in Hb Koriyama. On the basis of our results, we conclude that unstable Hb may not be detectable by conventional Hb electrophoresis or stability tests. Thus, we suggest further genetic workup in cases of unexplained hereditary hemolytic anemia.
Effect of Carboxy-hemoglobin on Color Stability of Cooked Pork Sausage

To investigate the effect of carboxy-hemoglobin (CO-Hb) on the color of cooked pork sausage (CPS), 6treatments with 5 levels of CO-Hb (0, 0.2, 0.4, 0.6, and 0.8%) and 0.015% NaNO2 were manufactured. Increased CO-Hb decreased a* values, but increased L* and b*values significantly (p<0.05). Color of the 0.4% CO-Hb samples had no significant change (p>0.05) during the 29-day storage. Both CO-Hb and carboxy-myoglobin (COMb)were found in the pigments extracted from the 0.4%CO-Hb non-cooked sausages. Therefore, CO-Hb showed a potential color agent in the manufacture of sausages.
ARKRAY ADAMS A1c HA-8180의 분석능 평가

The hemoglobin A1c (Hb A1c) test is widely used to diagnose diabetes mellitus and monitor glycemic control in patients with diabetes. We evaluated the performance of the ARKRAY ADAMS A1c HA-8180 (ARKRAY KDK, Japan), an automated, HPLC-based Hb A1c analyzer. The ARKRAY ADAMS A1c HA-8180 was evaluated for its linearity and precision and compared to the HLC-723 G7 (Tosoh Corporation, Japan), according to the Clinical and Laboratory Standards Institute’s guidelines. The coefficients of variation (CVs) for within-run precision at low and high levels were 0.6% and 0.3%, respectively, and the total CVs at low and high levels were 0.8% and 0.6%, respectively. The coefficient of determination (R2) was 0.9975, with linearity in the range of 3.0-18.5%. A comparison between the ARKRAY ADAMS A1c HA-8180 and HLC-723 G7 revealed a good correlation (r=0.9955) in the range of 4.8-14.6%. The runtime was 57 s per sample. The ARKRAY ADAMS A1c HA-8180 showed good analytical performance and high throughput. Therefore, it is suitable for routine use for clinical measurements of Hb A1c. 당화혈색소 Hb A1c는 당뇨병의 진단기준과 혈당 조절의 모니터링 지표로 널리 이용되고 있다. 본 연구에서는 Hb A1c 측정 자동화 장비인 ARKRAY ADAMS A1c HA-8180 (ARKRAY KDK, Japan)의 성능을 평가하였다. ARKRAY ADAMS A1c HA-8180의 직선성, 정밀도 및 기존 검사실 장비인 HLC-723 G7 (Tosoh Corporation, Japan)과의 상관성을 CLSI 지침에 따라 평가하였고, 분석속도를 측정하였다. 저농도와 고농도의 Hb A1c 정도관리물질의 검사 내 변이계수는 각각 0.6%와 0.3%였으며, 총 변이계수는 0.8%와 0.6%였다. 직선성은 3.0-18.5% 범위에서 R2=0.9975였다. Tosoh HLC-723 G7과의 상관성 평가에서도 4.8-14.6% 범위에서 r=0.9955의 상관성을 보였고, 분석 속도는 검체 당 57초였다. ARKRAY ADAMS A1c HA-8180는 임상검사실에서 우수한 분석성능으로 대량 검체를 신속하게 검사할 수 있을 것으로 생각된다.

Objectives: We examined if their hematological status could be changed by blood loss in patients treated with bleeding pressure therapy. Methods: The patients were divided into two groups as follows: The Subject group(B) were 42 cases treated with bleeding pressure therapy. The Control group(N) were 43 cases treated with negative bleeding pressure therapy, We checked CBC & differ cell count of two groups. Results: The means of RBC, Hb count and Hct were slightly decreased after 1 week. but they were recovered after 2 weeks. Conclusions: These results indicate that Bleeding Pressure Therapy don't reduced RBC, Hb count and Hct in Patients.
