        사묘용안탕(四妙勇安湯) 물추출물이 DNCB로 유발된 접촉성피부염에 미치는 영향

        Samyoyongantang (SMY), a herbal medicine, has been used as cure for gangrene. To evaluate anti-inflammatory effect of samyoyongantang, we treated samyoyongantang extract in animal model system induced contact hypersensitivity. Contact hypersensitivity, a local inflammatory response of the skin, was induced by 1% DNCB on the right ear of BALB/c mouse. Samyoyongantang was prepared as water extract and administrated everyday for 2 weeks per oral. A right ear of mouse was potently swelled by 1% of DNCB treatm ent, but a mouse ear thickness was significantly reduced by samyoyongantang after 2 we eks treatment. Samyoyongantang reduced IgG in serum obtained from blood of 1 % DNCB-treated mouse. IgE in serum was not changed by samyoyongantang treatment. From these results, anti-inflammatory effect of samyoyongantang, especially reduction of ear swell ing, might be partly due to reduction of IgG in serum.

        소풍산물추출물이 DNCB로 유발된 접촉성피부염에 미치는 영향

        Sopung-san(SPS; prescription No. DO27), a herbal medicine prescribed by oriental hospital of Daegu fanny University, has been used as cure for pruritus and psoriasis. To evaluate anti-inflammatory effect of Sopung-san, we treated Sopung-san extract in animal model system induced contact hypersensitivity. Contact hypersensitivity, a local inflammatory response of the skin, was induced by 1% DNCB on the right ear of BALB/C mouse. Sopung-san was prepared as water extract and administrated everyday for 2 weeks per oral. A right ear of mouse was potently swelled by 1% of DNCB treatment, but a mouse ear thickness was significantly reduced by Sopung-san after 2 weeks. treatment. Sopung-san reduced IgG in serum obtained from blood of 1 % DNCB-treated mouse. IgE in serum was not changed by DNCB and/or Sopung-san treatment. $IL-1{\beta}$ and IL-6 from ear tissue were significantly increased by DNCB, Dut were not reduced by Sopung-san. From these results, anti-inflammatory effect of Sopung-san, especially reduction of ear swelling, might be partly due to reduction of IgG in serum.

        DNCB로 유발된 알레르기성 접촉피부염에 황련해독탕이 미치는 영향

        Objectives: HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-inflammatory effect of HRHDT, we treated HRHDT-skin in Balb/c mice model induced contact hypersensitivity. Methods: Contact hypersensitivity, a local inflammatory respinse of skin, was induced by spreading the back skin of Balb/c mice with 1% DNCB. HRHDT-skin was prepared by dissolving 3% 1,3-butylene glycol extract of HRHDT in solution and treated 2 weeks on the back skin. Results: HRHDT-skin significantly reduced TEWL and erythema by 0.4-1% of DNCB treatment compared with control group. HRHDT-skin reduced IgE on serum obtained from blood of DNCB-treated Balb/c mice. Histopathological examination showed that thickening of the epidermis, hyperkeratosis and the infiltration of inflammatory cells were found in Balb/c mice under conventional circumstances. Conclusions: These results showed that HRHDT-skin could be used as a pharmaceutical material with antiinflammatory effects by reducing IgE in contact hypersensitivity dermatitis Balb/c mice by DNCB.

      • The Effect of Ultraviolet B Irradiation-Induced Epidermal Cytokines on the Generation of Delayed-Type Hypersensitivity Response in Mice

        Exposure of mice to UV radiation results in a systemic suppression of the immune response to contact allergens applied at a distant irradiated site. In addition, the immune response to alloantigens can be suppressed by injecting allogenic spleen cells into mice previously exposed to UV. In both cases, antigen-specific suppressor T cells are present in the spleens of the UV-irradiated animals. What is not entirely clear, however, is the mechanisms by which irradiation of the dorsal skin of the mice can lead to the induction of splenic suppressor cells. Recent studies have suggested that cytokines released from UV-irradiated epidermal cells may play a role in the systemic suppression of contact hypersensitivity. The purpose of the present study was to investigate the spectrum of immune responses that are suppressed by the cytokines released by UV-irradiated epidermal cells.

        신규 개인제독키트, SDK시제품(가칭)의 면역독성평가에 관한 고찰

        To evaluate immunotoxicity of skin decontamination kit(SDK) newly-developed in Agency for Defense Development(ADD), delayed contact hypersensitivity (maximization) test and passive cutaneous anaphylaxis(PCA) test of SDK were performed and the results were compared with those of M 291. In maximization test, sensitization reaction was induced by id injection (2.5 mg / 0.1 $\textrm{m}{\ell}$/ guinea pig or 2.5 mg+CFA/0.1 $\textrm{m}{\ell}$/guinea pig) and topical application (2.5 mg/$\textrm{m}{\ell}$/guinea pig) with SDK or M291 at an interval of 1 week, and 2 weeks later, challenged by topical application with 25 mg/$\textrm{m}{\ell}$/guinea pig. SDK and M291 did not induce any reactions, showing 0 point of sensitization score and 0% of sensitization rate. In conclusion, it is suggested that SDK and M291 do not induce delayed contact hypersensitivity. In PCA test, rats were administered id with mouse anti-SDK serum and challenged iv with a mixture of antigen SDK and Evan's blue. SDK did not induce blue spots at the injection sites of both high (2.5 mg/mouse) and low (1.25 mg/mouse) dose-induced antisera. In contrast, BSA, positive control produced spots larger than 5 mm in diameter at the injection sites of BSA-induced antiserum up to $2^2$ ~ $2^4$dilution. In conclusion, it is suggested that SDK do not induce IgE production and is not a PCA-reaction inducer.

      • Selenium이 마우스의 일차 체액성 면역반응과 접촉성 과민반응에 미치는 영향

        Effects of Selenium (Se) on the primary humoral immune response and contact-delayed type hypersensitivity (CDTH) to 2,4-dinitro-l-fluorobenzene (DNFB) in normal and cyclophosphamide (CY)-treated mice. Se was orally administered via drinking water for 18 days (1ppm or 3ppm) or via intraperitoneal (i.p.) injection with a single dose of 0.05, 0.25, 0.5 mg/kg b.w. to normal and CY-treated mice. CY was i.p, injected to mice with a single dose of 10 or 50 mg/kg b.w. on the last day of Se-administration. When only Se was administered simultaneously with or after immunization, splenic IgM plaque forming cells (PFC) and hemmaglutinin-titer (HA-titer) in mice were not changed, however, decreased in the mice which were administered with high dose of Se before immunization. In addition, they were significantly increased in the concomitant treatment of CY and Se, compared to them in the only CY-treatment. Se significantly decreased DNFB-induced CDTH and inhibited the CY-enhanced CDTH. These results indicate that Se may inhibit the immunotoxicity induced by CY.

      • The effect of HIF-2α inhibitor in DNFB-induced contact hypersensitivity

        Background: Allergic contact dermatitis (ACD) is caused by exposure of foreign material and various cytokines are induced by direct stimulation of hapten on keratinocytes or by sensitized T lymphocytes. T cells have an important role in the pathogenesis of ACD providing rapid response to previously encountered pathogens. Hypoxia-inducible factors (HIFs) are the main regulator of cellular response to reactive oxygen species and T cells are expected to be suppressed by HIF-2α antagonist. Objectives: The purpose of this study is to evaluate cytokine profile and the effect of HIF-2α antagonist in contact hypersensitivity mouse model induced by DNFB. Methods: Contact hypersensitivity was induced in C56BL/6 mouse by DNFB. HIF-2α antagonist was treated by oral gavage in contact hypersensitivity group. We measured level of ear thickness, epidermal thickness and CD4+ T cells by immunohistochemical staining. RNA level of various cytokines was evaluated using real-time PCR. Results: Ear thickness, epidermal thickness, and CD4+ T cell percentage were increased in contact hypersensitivity group. Proinflammatory cytokines (IL-1α, IL-1β, TNF), alarmin (IL-33), Th1 (IFN-γ, CXCL-10) and Th2 (IL-4, CCL-11, CCL-24) cytokines, and EPAS1 were elevated, but no significant difference was identified when treated by HIF-2α antagonist. Conclusion: Various cytokines were elevated in contact hypersensitivity mouse model induced by DNFB, but no significant differences were found when treated by HIF-2 α antagonist.

Studies of AD and contact hypersensitivity in nummular eczema are limited and their associations remain unclear. Objectives: To investigate the association of AD and contact hypersensitivity with nummular eczema Methods: 97 patients with nummular eczema were retrospectively reviewed. The results of the patch tests and the history of AD were analyzed. Results: Out of 97 patients, 44 patients were males and 53 were females. The mean age of the patients was 40.21. Patch test was postive in 59.79% of the patients and nickel (44.83%) was most commonly sensitized, followed by cobalt and thiomersal (17.24%). 67.01% of the patients had a history of AD, and those with atopic sensitization had a significantly higher total IgE levels than those without (452.52 and 71.34 kU/L). Patients without atopic sensitization had shown a higher positivity in patch tests (66.15%) compared to those with a history of AD(46.88%), although the difference was statistically insignificant. Conclusion: Patients with nummular eczema accompanied by atopic dermatitis had a less tendency to have allergic contact dermatitis. However, because contact hypersensitivity is commonly found in these patients, numerous adult patients with nummular eczema may be already sensitized to multiple allergens.

      • Effects of Cyclophosphamide and Indomethacin on UVB-suppressed Contact Hypersensitivity in Guinea Pigs

        This experiments pursued the effect of cyclophosphamide and indomethacin on UVB-suppressed contact hypersensitivity in guinea pigs. Before sensitization with DNCB, animals were exposed to UVB at induction sites 15 mJ/㎝^2 and were given intraperitoneal injection of cyclophosphamide and indomethacin for 4 consecutive days (days-3,-2,-1,0). Fourteen days after sensitization, animals were challenged with application of DNCB, and skin reactions were evaluated 24 hours after challenge. UVB irradiated group B (64.1%) showed moderate suppression of contact hypersensitivity compared with positive control, group A (77.8%). Group C treated with indomethacin and group E treated with cyclophosphamide significantly augmented contact hypersensitivity (81.2%, 93.1%) compared with positive control (77.8%). Suppression of contact hypersensitivity elicited by UVB were reversed by indomethacin in the group D (69.1%) and cyclophosphamide in the group F (76.4%) compared with group B (64.1%).

        해독사물탕(解毒四物湯)이 피부 접촉성 과민반응에 미치는 영향(影響)

        Objectives : Haedoksamul-tang (HSTE), a water extract from a mixture of Phellodendri Cortex, Coptidis, Scutellariae Radix, Gardeniae Fructus, Angelica acutiloba Radix, Cnidii Rhizoma, Paeoniae Radix, Rehmanniae Radix, has been traditionally used for allergic skin diseases such as atopic dermatitis and contact dermatitis in oriental countries. However, little is known about the effects of aqueous extract of HSTE on trimellitic anhydride (TMA)-induced contact hypersensitivity (CHS) in a mouse model. Methods : In this study, we investigate the pharmacological effects of HSTE on TMA-induced CHS in Balb/c mice. Contact hypersensitivity was induced in mice by topically sensitizing and challenging with TMA in flank skin and ears during oral administration (for 17 days) and topical treatment (30 min before challenge) with HSTE. We examined the effects of HSTE on IgE and IgG1 levels, inflammatory parameters in ear tissues, CD4<sup>+</sup>/CD8<sup>+</sup> ratio, cytokine and chemokine production in sera, tissues, and immune cells from TMA-sensitized mice.Results : Oral and topical administration with HSTE reduced, in a dose dependent manner, thickness and leukocyte infiltration of ear tissues and IgE levels in serum from mice sensitized with TMA. In addition, auricula lymph node cells isolated from TMA-sensitized mice significantly elevated the expression ratio of CD4<sup>+</sup>/CD8<sup>+</sup> as well as increased the production of IL-4, IL-5, IL-13 and IFN-γ by ex vivo stimulation with antibodies against CD3 and CD28, and these inflammatory indexes, except for IFN-γ, were significantly suppressed by orally and topically administration of HSTE. Furthermore, stimulation of auricula lymph node cells from TMA-sensitized mice with antibodies against CD3 and CD28 increased the production of MCP-1/CCL2 and MIP-1α/CCL3, and these effects were inhibited in a dose-dependent manner in cells from mice treated with HSTE. Conclusions : These results suggest that HSTE can be used for treating contact hypersensitivity by inhibiting leukocyte infiltration as well as production of serum IgE and chemokine/Th2 cytokine in an animal model.
