        새로운 퀴놀론 항균제 DW-116의 살균 작용

        The bactericidal activities of DW-116, a new fluoroquinolone was estimated by comparing the minimal bactericidal concentrations (MBCs) of it against some Gram-positive and -negative bacterial concentration with the minimal inhibitory concentrations (MICs). The MBCs against the test organisms were equal to or two times higher than MICs. The results support that the antibacterial activity of DW-116 is bactericidal.

        은 나노 용액의 식물병원성 세균에 대한 살균활성

        We have conducted in vitro experiments with nano-silver liquid for their effect against various plant pathogenic bacteria. Different types of nano-silver liquid WA-CV-WA13B, WA-AT-WB13R and WA-PR-WB13R were used. These are classified based on different manufacturing processes. The tested bacteria were provided by KACC. We experimented ten bacterial isolates in Clavibacter, Erwinia, Pseudomonas, Ralstonia, and Xanthomonas genera. In order to determine the level of concentrations of control effects, different concentrations (10, 25, 50, and 100 ppm) of each different nano-silver liquid were added in the culture media. As a result, WA-CV-WA13B showed high inhibition effect against C-1 at 10 ppm, and showed minor inhibition effects against P-6, X-1, and X-2. WA-AT-WB13R showed bactericidal effect against P-6 at 10 ppm. At 10 ppm, WA-AT-WB13R showed relatively high inhibition effects against C-1, X-1, and X-2. WA-PR-WB13R showed bactericidal effects against P-5, P-6 and X-2 at 10 ppm or higher concentrations. All the tested three nano-silver liquid showed bactericidal effects against all the tested plant pathogenic bacteria at concentrations of 25 ppm or higher. These results indicated the possible use of nano-silver liquid for the control of plant pathogenic bacteria.         Bactericidal Activity of Sawa-wasabi (Wasabia japonica) Against the Fish Pathogenic Bacteria

        In this study, the bactericidal activity of each extract from Sawa-wasabi (Wasabia japonica) root, stem and leaf against the fish pathogenic bacteria were examined. The main component related to bactericidal activity in Sawa-wasabi was well known to AlT. The Sawa-wasabi roots showed the highest AIT amount with 1.18 mg/g. Stems was 0.41 and leaves was 0.38 mg/g. All of them showed bactericidal activity against 2 strains of Vibrio hollisae, V. anguillarum, and 2 strains of Edwardsiella tarda, but weak effect against Staphylococcus capitis. The Sawa­wasabi leaves showed the strongest bactericidal activity with minimal bactericidal concentrations (MBCs) of 156.3mg of dry weight/mL against 2 strains of V. hollisae, V. anguillarum and 2 strains of E. tarda. The roots and stems showed a little weak bactericidal activities with 312-1,250mg of dry weight/mL against them. These results suggest that certain components besides AIT in Sawa-wasabi are affective in killing fish pathogenic bacteria.

        Bactericidal Effect of Combination of Atmospheric Pressure Plasma and Nisin on Meat Products Inoculated with Escherichia coli O157:H7

        This study was conducted to investigate the bactericidal effect of nisin (Nisin) only, atmospheric pressure plasma (APP) only, and a combination of APP and nisin (APP+Nisin) on beef jerky and sliced ham inoculated with Escherichia coli O157:H7, gram-negative bacteria. The bactericidal effect against E. coli O157:H7 and Listeria monocytogenes was confirmed using a nisin solution at a concentration of 0–100 ppm, and APP+Nisin was tested on beef jerky and sliced ham using 100 ppm nisin. Beef jerky and sliced ham were treated with APP for 5 min and 9 min, respectively. In the bacterial solution, 100 ppm nisin out of 0–100 ppm nisin exhibited the highest bactericidal activity against L. monocytogenes (gram-positive bacteria; p<0.05); however, it did not exhibit bactericidal effects against E. coli O157:H7 (gram-negative bacteria). The APP+Nisin exhibited a 100% reduction rate in both E. coli O157:H7 and L. monocytogenes compared to the control group, and was more effective than the Nisin. The APP+Nisin decreased the number of colonies formed by 0.80 and 1.96 Log CFU/g for beef jerky and sliced ham, respectively, compared to the control, and exhibited a higher bactericidal effect compared to the Nisin (p<0.05). These results demonstrate the synergistic bactericidal effect of APP and nisin, providing a possible method to improve the limitations of nisin against gram-negative bacteria. In addition, this technology has the potential to be applied to various meats and meat products to control surface microorganisms.

      • 뽕나무 뿌리껍질 추출물의 살균력과 안정성

        In order to develop a natural food preservative, root bark of Morus alba was extracted with ethanol and then bactericidal activity of the extract against microorganism was investigated. The stability for temperature, pH, storage conditions and metal ions of the extracted antimicrobial substance was investigated. The crude fat content of the extract from root bark of Morus alba was 62.9% and palmitic acid and stearic acid were pretty high in fatty acid composition. The extract from root bark of Morus alba had bactericidal activity against Gram positive bacteria such as Bacillus subtilis, and Staphylococcus aureus. While the extract had bacteriostatic activity against Gram negative bacteria such as Escherichia coli. The minimum bactericidal concentration(MBC) of the extract against B. subtilis and S. aureus was 32㎍/㎎ and 64㎍/㎎, respectively. The antimicrobial activity of the extract was stable by the change of pH, freezing and heat treatment. KCl, NaCl and MgCl_(2) didnot affect bactericidal activity of the extract from root bark of Morus alba but CaCl_(2) did negative affect bactericidal activity.

        Bactericidal Antibody Responses to Meningococcal Recombinant Outer Membrane Proteins

        Secretin PilQ is an antigenically conserved outer membrane protein that is present in most meningococci and PorA is a major protein that elicits bactericidal immune response in humans following natural disease and immunization. In the present study, BALB/c mice were immunized subcutaneously with rPilQ406-770 or rPorA together with Freund’s adjuvant (FA). Serum antibody responses to serogroup A and B Neisseria meningitides whole cells or purified proteins and functional activity of antibodies were determined by ELISA and serum bactericidal assay (SBA), respectively. Serum IgG responses were significantly increased in the immunized group with rPilQ406-770 or rPorA together with FA compared to control groups. IgG antibody response of mice immunized with rPilQ406-770 was significantly more than mice immunized with rPorA (OD at 450 nm was 1.6 versus 0.83). The booster injections were effective in increasing the responses of antirPilQ406- 770 or anti-rPorA IgG significantly. Antisera produced against rPilQ406-770 or rPorA demonstrated strong surface reactivity to serogroup B N. meningitides in comparison with control groups. Antisera raised against rPorA or rPilQ406-770 and FA demonstrated SBA titers from 1/1024 to 1/2048 against serogroup B. The strongest bactericidal activity was detected in sera from mice immunized with rPilQ406-770 mixed with FA. These results suggest that rPilQ406-770 is a potential vaccine candidate for serogroup B N. meningitidis.

        면역유충 체액으로 부터 항균물질의 유도

        It was known that normal-haemolymph from the 3rd instar larvae of Lucillia illustris contain a lysozyme (or lysozyme-like substance) with bactericidal activity to fram positive bacteria, and the bactericidal activity of injured-haemolymph was increased significantly after injuring the body wall. To elucidate the defence mechanism of insect against the nonpathogenic bacteria, the immune-haemolymph against Escherichia coli K-12 was prepared after immunization. The bactericidal activity between injured and immune-haemolymph was compared, and it was revealed that the immune-haemolymph showed higher titer of bactericidal activity to fram positive bacteria as well as to Escherichia coli. The bactericidal substance from the immune-haemolymph was purified through a successive chromatographies on Sephacryl S-300 and CM-Sepharose CL-6B, and it was characterized as a basic protein in nature with heat stable property at acidic conditions. 정상 상태의 연두금 파리 (Lucilia illustris) 제 3령충의 체액 (normal-haemolymph)으로 부터 그람양성 세균에 대하여 항균력이 있는 lysozymec (or lysozyme-like substance)을 확인하였으며 체벽을 손상시킨 체액 (injured-haemolymph)의 경우 그 항균력이 증가되었다. 특히 비병원성 세균인 Escherichia coli K-12로 면역하여 얻은 체액 (immune-haemolymph)의 경우 injured-haemolymph보다 그람양성 세율에 대하여 더 높은 항균력을 보였을 뿐만 아니라 그람음성 세균인 E. coli에 대하여 도 항균력을 나타내었다. Immune-haemolymph로 부터 유효 분획을 얻고 이에 대한 성상을 분석하기 위하여 Sephacryl S-300 및 CM-Sepharose CL-6B로 분리시킨 결과 상기 항균울질은 엽기성 만백질로서 산성하에서 내열성의 성질을 나타냈다.

        Streptococcus mutans 에 대한 키토산의 항균효과

        S. mutans에 대한 키토산의 항균효과는 그 분자량에 따라 뚜렷한 차이를 보였으며, 약 30,000의 분자량을 갖는 키토산이 가장 높은 항균활성을 보였다. 키토산 처리에 의해 세포로부터 단백질, 핵산, 2가 양이온 등이 상당량 유출되는 것이 관찰되었으며, 세포내 물질의 유출정도는 키토산의 항균효과와 밀접한 상관성을 보였다. 이러한 세포내 물질의 유출은 양이온성을 띠는 키토산의 아민기와 세포벽내의 음이온 성분과의 강한 결합에 의해 세포벽의 결합력이 약화되면서 생기는 결과로서 추측되었다. 키토산처리에 의해 S. mutans의 세포벽이 원형질로부터 분리되고 세포내 물질이 세포벽 내에서 밖으로 유출되는 현상은 투과전자현미경(TEM)에 의해 확인되었다. Bactericidal effects of chitosans with varying molecular weight $(10,000{\sim}170,000)$ were investigated for Streptococcus mutans, a primary causative bacterium of dental caries. The molecular weight of chitosan was a significant contributor to the bactericidal effect, and a chitosan having approximately 30,000 of molecular weight exhibited the highest bactericidal effects on S. mutans. Treatment of chitosan resulted in leaking intracellular protein and nucleic acid out of S. mutans cells. In addition, the divalent cations such as $Ca^{2+}\;and\;Mg^{2+}$ were also significantly released out of the cell. Visible damage of chitosan treated cells was observed by transmission electron microscopy (TEM), in which the cell wall was notably distorted and cytoplasmic membrane was separated from the cell wall. The results suggested that the bactericidal effect of chitosan on S. mutans was attributable to both leakage of intracellular materials and structural disintegration of cell wall.

        Bactericidal Activity of Grapefruit (Citrus paradisl) Seed Extract-Based Disinfectant

        Bactericidal activity of grapefruit seed extract (GSE)-based disinfectant, as a safe disinfectant, was measured against five bacteria by Korean Food & Drug Administration (KFDA) dilution-neutralization method. GSE-based disinfectant showed a 99.9999% bactericidal activity against Escherichia coli ATCC 10536, Salmonella typhi ATCC 29629, Staphylococcus aureus ATCC 6538, Bacillus cereus ATCC 11778, and Listeria monocytogenes ATCC 1911 at the concentration of 2.15% GSE. It showed better bactericidal activity against Gram-negative bacteria of E. coli ATCC 10536 and S. typhi ATCC 29629 at lower concentration of GSE (0.43%). Based on the results, it was suggested that a possible bactericidal mechanism of GSE active ingredients was due to the abrupt osmotic shift during the bactericidal activity test by KFDA method.

        가두리 양식장의 Vibrio vulnificus 검출 및 제어 방법

        2000년 1 월부터 2000년 10월까지 가두리 양식장에서 Vibrio vulnificus를 검출하였고 이들의 억제 방법을 연구하였다. 이 세균의 검출은 선택적 분리법과 vvhA 유전자를 확인하는 방법을 이용하였다. V. vulnificus는 수온이 $17^{\circ}C$이상인 5월부터 검출되었고 $19^{\circ}C$ 이상인 6월부터 9월까지는 대부분의 시료에서 검출되었다. V. vulnificus를 제어하기 위한 방법 중 냉동 및 냉장 처리는 살균효과를 나타내지 못했다. Citric acid도 균의 생장을 억제하지 못했으나, 500 mg/1이상의 EDTA가 첨가될 경우 균이 완전히 사멸되었다. 분말 광촉매인 산화티타니움은 자외선을 조사할 경우 15분~1 시간 이내에 이 세균을 완전히 사멸시키는 효과를 나타내었다. 산화티타니움을 유리 구슬에 코팅한 광촉매 장치를 이용하여 0.2/min의 turnover rate로 사멸효과를 얻었다. Detection of Vibrio vulnificus in fish farm and searching for the bactericidal methods on this bacteria were studied. To detect this microorganism in sea water, mud, fish and mussels, selective isolation methods and detection of vvhA gene were used from January to October,2000. V. vulnificus was detected from May when the water temperature was over $17^{\circ}C$. From June to September, higher than $19^{\circ}C$, this bacteria could be isolated from most of the samples. Freezing and refrigerating did not inhibit the growth of V. vulnificus. Citric acid did not show the bactericidal effect, but more than 500 mg/l of EDTA did. With the aid of UV and photocatalyst, $TiO_{2}$ showed bactericidal effect after 15 minute treatment. Photocatalytic system consisted of glass bead coated with $TiO_{2}$ and UV illumination showed bactericidal effect on V. vulnificus at the turnover rate of 0.2/min.
