        황기의 첨가.급여가 육계의 생산성 및 혈액의 성상과 육질에 미치는 영향

        This experiment was conducted to investigate the effect of dietary supplemental Astragalus membranaceus on performance and meat quality of broiler chicks for 5 weeks. Basal diets based on corn and soybean meal contained 21% crude protein for the first three weeks and 19% for the rest of two weeks. Four levels of dietary Astragalus membranaceus(0, 0.25, 0.50, 1.00%) were fed in a one way design. There were four replicates of 50 chicks per treatment. Weight gain, feed intake, feed efficiency, blood component, breast meat lipid and protein were measured at the end of experiment. No increased growth rate was observed in chicks fed the basal diet supplemented with Astragalus membranaceus. Chicks fed the diet containing 0.5 and 1.00% Astragalus membranaceus tended to depress the growth rate. Feed intake of 0.25% Astragalus membranaceus treatment was significantly lower than control group(P<0.05), Dietary supplemental Astragalus membranaceus improved the feed efficiency compared to the control group, but was not significantly different between them. However, Astragalus membranaceus treatment exhibited decreased serum cholesterol and AST compared to those of control group, but was not significantly different between supplemental groups. Serum hematocrit, triglyceride, phosphate and BUN decreased at 0.25% Asparagus membranaceous supplementation. Growth hormone and IGF-1 (ng/ml) of chicks fed more than 0.50% Astragalus membranaceus significantly decreased compared to those of control(P<0.05). The total lipid and protein content of chicks fed 0.25 and 0.50% Astragalus membranaceus tended to increase, but did not show consistency at more than 0.50% supplement. The results of this experiment indicated that optimum dietary Astragalus membranaceus level to improve the performance and meat quality of broiler chicks may be less than 0.25%. 본 연구는 황기의 사료내 첨가·급여가 육계의 항병성, 생산성 및 계육의 품질에 미치는 영향을 구명하고자 시행하였다. 기초사료는 전기 3주와 후기 2주에 각각 21과 19% 수준의 조단백질이 함유되었다. 시험설계는 황기의 첨가수준을 0, 0.25, 0.50, 1.00%로 4개의 처리구로 하였고, 처리구당 4반복으로 하였으며, 반복당 50수씩 800수를 이용하였다. 증체량, 사료섭취량, 사료요구율은 5주동안 매주 측정하였고, 시험 종료시에 혈액의 성상 및 계육의 지방과 단백질함량을 조사하였다. 황기 첨가구는 대조구에 비하여 성장에 영향을 미치지 못하였으나, 사료섭취량은 대조구에 비하여 현저하게 감소하였고(P<0.05), 사료효율은 0.25%첨가구에서 개선되는 경향을 보였다. 육계용 사료에 황기의 첨가수준이 증가함에 따라서 혈청 콜레스테롤과 AST는 감소하였으며, 헤마토크릿, 지방, 인과 BUN의 수준은 0.25%첨가구에서 감소하였다. 혈청 GH와 IGF-1은 0.50과 1.00% 황기 급여구에서 대조구에 비하여 낮았다. 0.25와 0.50%황기 첨가구에서 가슴육의 지방과 단백질함량은 대조구에 비하여 증가하는 경향을 보였으나, 0.50%이상의 수준에서는 일관성이 없었다. 본 시험의 결과 육계용 사료에 황기의 적정 첨가수준은 0.25%이하에서 생산성과 항병성 및 계육의 품질을 개선할 수 있을 것으로 사료된다.

        Aspergillus awamori로 발효한 황기 열수 추출물의 이화학적 특성과 항염증 효과

        Background: Fermentation of medicinal plants increases their absorption rate and bioavailability in the body. Astragalus membranaceus has been used as a raw material, but research in its use as a food ingredient is lacking. Therefore, the purpose of this study was to identify the physicochemical characteristics and anti-inflammatory effect of fermented Astragalus membranaceus. Methods and Results: Astragalus roots were fermented using Aspergillus awamori for 4 days and their extracts prepared using hot water. The pH, total acidity (%), and reducing sugar (%) of the extracts were then investigated. The pH and total acidity decreased during fermentation. After fermentation, the pH and total acidity decreased, whereas the reducing sugar level increased. The active ingredients in fermented Astragalus were calycosin-7-O-ßd-glucoside, ononin, calycosin and formononetin. The calycosin contents was highest in the hot-water extracted samples fermented for 4 days. The other components were similar to those in control. Nitric oxide level was lower in the hot-water extracted samples fermented for 4 days than in lipopolysaccharide control group. The sample fermented for 4 days was confirmed to inhibit the production of tumor necrosis factor-α and interleukin-1β. Conclusions: Our results showed the physicochemical properties and anti-inflammatory effects of A. membranaceus after fermentation using Aspergillus awamori. These results indicated that fermented Astragalus membranaceus can be used as a functional food.

        Radix Astragali, the root of Astragalus (A.) membranaceus, has been applied in a variety of diseases for a long time in Asian countries such as Korea and China. In addition, the aerial parts such as leaves and stems of A. membranaceus have received a great deal of attention. Recently, the polysaccharide fraction showing a potent immunomoduating activity was isolated from the aerial parts of A. membranaceus. Thus, the aerial parts of A. membranaceus would be worthy enough for a food material and a dietary supplement. However, they should be safe even though valuable. In our previous study, it was estimated that NOAEL for female rats are 5000 mg/kg/day of the crude polysaccharide fraction from A. membranaceusaboveground parts. As a series of safety evaluation, genotoxicity test for the crude polysaccharide fraction was carried out in this study. In conclusion, the three genotoxicity assays provided strong overall support that the crude polysaccharide fraction lacks mutagenic and/or clastogenic potential under the GLP-based test conditions. This indicates the aerial parts of A. membranaceus would be safe enough for a food material and a dietary supplement.

        랫드를 이용한 황기의 지상부 추출물에 대한 90일 반복경구투여 독성시험

        Pharmacological studies and clinical practices have indicated that Radix Astragali, a dried root of Astragalusmembranaceus possesses a lot of biological activities, including antioxidant, hepatoprotective, anti-diabetic, tonic, diuretic,antimicrobial, antiviral, and immunological activities. These biological activities approved by the modern pharmacologicalstudies are mainly due to the constituents of Astragalus membranaceus including polysaccharides, saponins, flavonoids,amino acids, and trace elements. In resent, the main constituents in the root part showing a lot of biological activities hasbeen isolated also from the aboveground parts such as leaves and sprouts in our laboratory. However, the safety evaluationfor the aboveground parts of Astragalus membranaceus should be checked before expanding their application as one of food.In the study, a 90-day rat oral gavage study has been conducted with the extracts from Astragalus membranaceus-above-ground parts at doses of 1000, 3000, and 5000㎎/㎏/day. The following endpoints were evaluated: clinical observations, bodyweight, gross and microscopic pathology, clinical chemistry, and hematology. Based on the analysis of these endpoints, it wasestimated that NOEL (no observed effect level) for male rats and NOAEL (no observed adverse effect level) for female ratsare 5000㎎/㎏/day of the water-extracts from Astragalus membranaceus-aboveground parts.

        Effect of Electron Beam Irradiation on Microbial Growth and Qualities in Astragalus membranaceus

        Electron beam irradiation was applied to examine the microbial growth and qualities of vacuum-packaged Astragalus membranaceus, a Korean medicinal herb. Samples were irradiated at dose of 2, 4, 8, 12, and 16 kGy, respectively. Microbiological data on A. membranaceus showed that populations of total bacteria, yeast and mold, total coliforms were significantly reduced with increase of irradiation dose. Populations of microorganisms in A. membranaceus were decreased by 2-3 log cycles at 8 kGy irradiation. Color measurements showed that electron beam treatment caused negligible changes in Hunter color L, a, and b values of A. membranaceus. Sensory evaluations showed that there were no significant changes among the samples. These results suggest that electron beam irradiated A. membranaceus have better microbial safety and qualities, compared with the non-irradiated control.

        Peptide Hydrolysates from Astragalus membranaceus Bunge Inhibit the Expression of Matrix Metalloproteinases in Human Dermal Fibroblasts

        Inhibition effects of peptide hydrolysates from Astragalus membranaceus Bunge. on the expression of the matrix metalloproteinases (MMPs) in human dermal fibroblasts were evaluated in vitro. Crude peptides were obtained by the hydrolysis of proteins extracted from A. membranaceus. Peptides were purified partially by the basis on the molecular weight using 40% polyacrylamide gel electrophoresis before treatment with human dermal fibroblasts. Basis on the doseeffect experiments, expressions of MMPs including MMP-1, MMP-3, MMP-8, MMP-13 in human dermal fibroblasts were evaluated. Expressions of MMP-1, MMP-3, MMP-8 and MMP-13 were reduced in 43%, 5%, 22% and 57% respectively. The mass spectrometric analysis of partially purified peptides from A. membranaceus, which strongly inhibit expressions of MMPs, indicated that the peptides were composed of molecules below 1500 Da.

        과도 운동 후 면역능 개선에 미치는 황기 다당체의 효과

        Background: Polysaccharides are the most important functional constituent in Astragalus membranaceus. The purpose of the present study was to evaluate the effect of polysaccharides isolated from the aboveground parts of A. membranaceus (AMA) and polysaccharides isolated from the roots of A. membranaceus (AMR) immune function by modulated cytotoxic T cell and Th1- and Th2- related cytokines kinetics. Methods and Results: Sprague-Dawley rats were randomly divided into exhaustive exercise case groups and non-exercise case, AMA and AMR samples were administered orally for 30 days (500 ㎎/㎏/day and 10 ㎎/㎏/day, respectively) and were compared to those rats in the groups fed commercial sports drink (SPD) and vehicle. Both exhaustive exercise groups and non-exercise groups had a lower ratio of CD4+ and CD8+ cells in the spleens of the rat fed AMA and AMR compared to those in the rats fed SPD and vehicle group. These results suggested that AMA and AMR promote an increase in the proportion of cytotoxic T cells. The IL-4- producing T lymphocytes decreased significantly in the AMR (10 ㎎/㎏/day) group compared to SPD and vehicle, whereas the AMA group increased the IL-4 concentration more than the SPD and vehicle in exhaustive exercise group. However, the populations of IFN-γ-producing T lymphocytes of AMR and AMA increased. AMA decreased the concentration of IFN-γ to inhibit the Th1 response and thereby increased the concentration of IL-4 to induce a Th2 response that was related to humoral immunity in the non-exercise group. Conclusions: These results showed that, in addition to Th1/Th2 regulation, AMR and AMA played an important immuno-modulatory role after exhaustive exercise-induced Th1/Th2 lymphocyte imbalance, which might be correlated with cytokine producing immunoregulatory cells.
