        눈개승마 분말을 첨가한 쿠키의 품질특성 및 항산화효과

        This study investigated the quality characteristics and antioxidative effects of cookies containing different levels (0, 1, 3, 5, and 7 % Aruncus dioicus var. kamtschaticus powder/400 g of the flour) of Aruncus dioicus var. kamtschaticus powder. The crude protein and crude fiber contents, thickness, hardness, and a color value of cookies increased, and the moisture and carbohydrate contents, spread factor, and L and b values of cookies as the concentrations of Aruncus dioicus var. kamtschaticus powder in the cookies were increased. The results showed similar crude lipid and crude lipid contents in the cookies. The pH and density of the cookie dough decreased significantly as the concentration of Aruncus dioicus var. kamtschaticus powder increased. The sensory score of the cookies containing 3% Aruncus dioicus var. kamtschaticus powder was the most suitable in terms of its taste, flavor, texture, overall acceptability, aroma, and roasted nutty flavor. The total polyphenol content, total flavonoid content, DPPH radical scavenging activity, and ABTS radical scavenging activity of the cookies increased as the amount of Aruncus dioicus var. kamtschaticus powder increased. These results suggest that Aruncus dioicus var. kamtschaticus powder can be an effective ingredient in cookie production, considering its nutritional components, quality properties, consumer preference, and antioxidative activities.

        재배 삼나물 뿌리 추출물의 자유 라디칼소거 및 α-glucosidase 저해활성

        As part of our continuing search for bioactive natural products, the antioxidant and a-glucosidase inhibitory activities of an 80% methanolic extract and organic solvent soluble-portions of Aruncus dioicus var. kamtschaticus roots were investigated by using a bioassay system. The antioxidant activity of A. dioicus var. kamtschaticus roots extract and organic solvent soluble-portions were assessed by examining with 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS+) radical scavenging. In addition, anti-diabetic effects of the A. dioicus var. kamtschaticus root extract and organic solvent soluble-portions were tested via a-glucosidase inhibition assay. The total phenolic contents of the products were determined by applying UV-VIS spectrophotometry. All tested samples showed dose-dependent radical scavenging and a-glucosidase inhibitory properties. In particular, the α-glucosidase inhibitory and radical scavenging effects of the ethyl-acetate (EtOAc)-soluble portion from the roots of A. dioicus var. kamtschaticus were greater than those from other solvent-soluble portions. These results indicate that A. dioicus var. kamtschaticus could be considered a new effective source of natural antioxidants and anti-diabetic materials. More systematic research of the constituents of the roots of this A. dioicus variety will be conducted to further develop its antioxidative and anti-diabetic properties.

        재배 삼나물 뿌리 추출물의 자유 라디칼소거 및 α-glucosidase 저해활성

        삼나물 뿌리를 80% 메탄올로 침지 추출하여 얻어진 추출물에 대해 n-hexane, EtOAc 및 n-BuOH의 유기용매를 활용하여 순차 분획을 실시하였고, 얻어진 결과물에 대하여 DPPH, $ABTS^+$ 라디칼 소거능 및 ${\alpha}$-glucosidase 저해활성을 평가하였다. DPPH 라디칼 소거능은 총 페놀성 화합물의 함량이 상대적으로 높은 EtOAc 층에서 $IC_{50}$값이 $32.3{\pm}2.3{\mu}g/mL$으로 가장 우수한 DPPH 라디칼 소거능을 확인하였고, 삼나물 뿌리 의 EtOAc 가용부에 다량 존재하는 페놀성 화합물과 라디칼 소거능과의 연관성을 시사하였다. 또한 $ABTS^+$ 라디칼 소거능은 EtOAc 층의 $IC_{50}$값이 $18.9{\pm}2.1{\mu}g/mL$의 우수한 라디칼 소거활성이 확인 하였고, 우수한 라디칼 소거 활성물질의 존재가 시사되었다. 또한, ${\alpha}$-glucosidase 저해활성을 평가한 결과, 우수한 라디칼 소거능을 나타낸 EtOAc 층의 $IC_{50}$은 $57.8{\pm}7.8{\mu}g/mL$의 저해활성을 나타내었으며, 이는 positive control인 acarbose의 $IC_{50}$값인 $501.1{\pm}17.5{\mu}g/mL$과 비교하여 우수한 활성이었으며, 추출물 상태의 시료를 단일물질로 정제할 경우 더욱 강한 효능의 화합물이 존재할 가능성을 시사하였다. 향후 이들 활성물질 동정을 통한 활성 기작에 대한 연구가 필요하며 본 연구결과는 보다 우수한 라디칼 소거능 및 ${\alpha}$-glucosidase 저해능을 가지는 새로운 기능성 소재 발굴을 위한 기초자료로 활용 가능할 것으로 사료된다.         포도당 처리로 유도된 뇌신경세포 독성에 대한 눈개승마 추출물의 보호효과

        본 연구에서는 눈개승마(Aruncus dioicus var. kamtschaticus)의 in vitro 산화방지활성 및 고당(intensive glucose)과 과산화수소(H₂O₂)로 야기된 산화적 스트레스로부터의 뇌신경세포 손상에 대한 보호효과와 더불어 알파글루코사이드가수분해효소 및 아세틸콜린에스터가수분해효소 억제효과를 확인하였으며 또한 HPLC를 이용하여 주요 물질을 분석하였다. 눈개승마 아세트산에틸 분획물(ethylacetate fraction from Aruncus dioicus var. kamtschaticus: EFAD)은 매우 우수한 총 페놀화합물 함량(430.08 mg GAE/g)과 총 플라보노이드 함량(511.72 mg RE/g)을 나타냈으며, 높은 ABTS 라디칼 소거 활성과 과산화지방질생성물의 억제력을 확인하였다. 또한 고당(intensive glucose)과 과산화수소(H₂O₂)로 야기된 뇌신경세포에서의 산화적 스트레스 및 이로 인한 뇌신경세포 사멸을 측정한 결과, 눈개승마 아세트산에틸 분획물(EFAD)의 유의적인 뇌신경세포 보호효과를 확인할 수 있었다. 추가적으로 다당류 분해효소인 알파글루코사이드가수분해효소 억제효과 및 아세틸콜린 분해 효소인 아세틸콜린에스터가수분해효소 억제효과를 살펴봄으로써 혈당 상승 억제효과와 뇌신경전달물질의 세포 내 유지 효과를 확인하였다. 마지막으로 눈개승마 아세트산에틸 분획물(EFAD)의 주요 페놀성 물질을 확인하기 위해 HPLC분석을 한 결과 카페산으로 추정되었다. 본 연구 결과들을 고려할 때, 눈개승마는 고혈당 지연 또는 개선을 통한 고혈당 예방 소재로서의 가능성 뿐만 아니라 이로 인해 야기되는 산화적 스트레스로부터 뇌신경 세포를 보호함으로써 고혈당에 의한 대사성 뇌신경질환 예방 소재로도 활용 가능성이 있을 것이라 판단된다. To assess the physiological effects of Aruncus dioicus var. kamtschaticus extract on cytoxicity of a neuronal cell line, antioxidant activity, and neuroprotection against intensive glucose-induced oxidative stress were quantitated. Compared to the other fractions, the ethyl acetate fraction of Aruncus dioicus var. kamtschaticus (EFAD) showed the highest total phenolics and flavonoids. The 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assay and malondialdehyde inhibitory effect test confirmed the superior antioxidant activity of EFAD. Moreover, EFAD also decreased the intracellular ROS level and suppressed neuronal cell death against intensive glucose- or H₂O₂-induced oxidative stress. Additionally, assessment of α-glucosidase and acetylcholinesterase inhibitory activities revealed that EFAD was an effective inhibitor of α-glucosidase and acetylcholinesterase. Finally, high-performance liquid chromatography analysis identified caffeic acid as the main ingredient of EFAD. Overall, these results suggest that the EFAD is a good natural source of biological compounds that counteract hyperglycemic neuronal defects.

      • 대기-엽 수증기압차(VPD)에 의한 어수리와 눈개승마의 광합성 반응 비교

        This study was conducted to investigate the effect of atmosphere-leaf vapor pressure deficit (VPD) in Heracleum moellendorffii and Aruncus dioicus var. kamtschaticus. The VPD was rapidly increased with increasing temperature and decreasing relative humidity. Taken as a whole, the stomatal transpiration reaction was slightly late with increasing of VPD. Maximum photosynthetic rate at high-VPD condition was 6.49 µmol CO<sub>2</sub>·m<sup>-2</sup>·s<sup>-1</sup> in Heracleum moellendorffii Hance, which was a little lower than 5.57 µmol CO<sub>2</sub>·m<sup>-2</sup>·s<sup>-1</sup> in Aruncus dioicus var. kamtschaticus, respectively. After 2 p.m, stomatal transpiration of Heracleum moellendorffii at the high VPD condition was rapidly decreased. The results indicated that physiological activities in Heracleum moellendorffii are more limited from high VPD conditions.

        NaCl 처리가 눈개승마[Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara]의 생육과 생리활성에 미치는 영향

        본 논문은 염(NaCl) 처리 시 눈개승마의 생육과 생리활성에 미치는 영향을 알아보기 위해 0, 100, 200, 300 mM의 농도로 염 처리를 하였다. 처리 30일 후 초장, 엽폭, 엽장 및 무기 이온 함량을 측정하여 생육에 미치는 영향을 조사하였고 생리활성에 미치는 영향을 확인하기 위해 항산화 활성 및 염증 생성 물질인 NO 생산량을 측정하였다. 그 결과, 염 처리 시 대조구에 비하여 생육 저하되었으며 염 농도 100, 200, 300 mM 처리구 간의 차이는 뚜렷하게 나타나지 않았다. 무기이온 함량은 염 처리시 $Na^+$의 함량은 증가하였고 $K^+$의 함량은 감소하였으며 $Ca^{2+}$ 함량은 염 처리 시 대조구에 비해 증가하였고 $Mg^{2+}$ 함량에서는 차이를 보이지 않았으나, 100 mM 이상의 농도에서는 $Na^+$ 함량이 현저하게 증가됨에 따라 식물 생장에 불리한 $K^+/Na^+$, $Ca^{2+}/Na^+$, $Mg^{2+}/Na^+$ 비를 초래하였다. 총 폴리페놀 함량과 플라보노이드 함량은 100 mM 이상의 염 농도에서 모두 감소하였으며 염 농도가 증가함에 따라 감소하는 경향을 보였다. DPPH, ABTS radical 소거활성 및 환원력에서도 대조구에 비해 염 처리농도가 증가함에 따라 감소하였다. 염 처리 시 눈개승마의 마우스 대식세포 Raw 264.7 cell에서의 세포독성 및 항염증 활성에 미치는 영향을 알아보기 위해 CCK assay와 염증 생성 물질인 NO 생산량을 측정한 결과, 염 처리구에서 모두 100% 이상의 생존율을 보여 세포독성이 없음을 확인하였다. 또한, NO 생산량의 경우 항산화 활성의 경향과 마찬가지로 대조구에 비해 100 mM 이상의 염 처리구에서 NO 생산 억제 활성이 감소하는 경향을 보였다. 따라서, 염 처리시 100 mM 이상의 농도에서 눈개승마의 생육과 항산화, 항염증 활성이 저하됨을 확인하였으며, 이는 염 스트레스로 인해 생장과 생리활성 저하를 유발된 것으로 생각된다. 따라서, 눈개승마의 경우 100 mM 미만의 농도에서 생육 및 생리활성 유지에 적합한 조건을 확립하기 위한 추가 연구가 이루어져야 할 것으로 사료된다. This study was conducted to investigate the growth and physiologycal activitis of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara under the NaCl treatment conditions (0, 100, 200, 300 mM). After 30 days treatment, the growth and physiologycal activitis were investigated. In the growth of plants, the plant height, leaf width, leaf length and ion level were reduced at NaCl treatments of more than 100 mM. The total polyphenol content was decreased by NaCl in a concentration-dependent manner compared to the control group. The contents of total flavonoids did not show any difference at the concentration of 200 mM and 300 mM. However, the content of total flavonoid decreased compared with that of control. In antioxidant activity, the DPPH and ABTS radical scavenging activity and reducing power activity were decreased by NaCl concentration compared to the control. When changes in the content of NO production was monitored by ELISA, production inhibitory effect was 94.5%, 70%, 63%, 56.9% in NaCl concentration of 0, 100, 200, 300 mM, respectively. The growth, ion level, antioxidant and anti-inflammation activity of Aruncus dioicus var. kamtschaticus was reduced at NaCl treatments of more than 100 mM.

        차광처리가 눈개승마 유묘의 생장 및 생리적 특성에 미치는 영향

        Background: This study was conducted to investigate the changes in the photosynthetic parameters, chlorophyll content, chlorophyll fluorescence, and growth characteristics of Aruncus dioicus var. kamtschaticus seedlings under different shading treatments. Methods and Results: The shading treatment was regulated with the shading level (non-shaded, 35%, 55%, and 75% shading). Photosynthetic activities, such as net photosynthetic rate, stomatal conductance, stomatal transpiration rate, and performance index on absorption basis (PIABS)were the highest under 35% shading (4.36 µ㏖ CO2·m−2·s−1, 54.2 m㏖ H2O·m2·s−1, 0.66 m㏖ H2O·m−2·s−1, and 1.3, respectively), and the lowest under 75% shading. This implies that the decrease in net photosynthetic rate may be due to an inability to regulate water and CO2 exchanged through the stomata. Thechlorophylla, b, and a + b contents were increased with elevating shading level and the chlorophyll a/b ratio showed non-significant differences. It was found that the dry weight (leaf, shoot, and whole) was the highest (1.14 g, 0.49 g, and 2.31 g, respectively) under 35% shading and the t/R ratio was the highest under 75% shading. Conclusions: It is concluded that 75% shading exhibited a strong reduction of photosynthetic activity, and 35% shading showed the best conditions for the early growth and cultivation of A. dioicus var. kamtschaticus.

        Bioactive Constituents from the n-Butanolic Fraction of Aruncus dioicus var. kamtschaticus

        Six compounds were isolated from the n-BuOH fraction of the aerial parts of Aruncus dioicus var. kamtschaticus including: sambunigrin (1), prunasin (2), aruncide A (3), aruncide C (4), 1-O-caffeoyl-β-D-glucopyranose (5), and caffeic acid (6). Their structures were confirmed by comparing the spectral data with those reported in the literature. The isolated compounds (1 - 6) were then examined for their cytotoxic effects towards MCF-7, HL-60, and HeLa cancer cell lines, as well as their DPPH radical scavenging activity. The results indicated that compound 4 possessed the strongest inhibitory effect toward HeLa cell line with IC50 value of 5.38 ± 0.92 μM. Compound 3 possessed selective cytotoxic activity on HL-60 cells with IC50 value of 6.27 ± 0.17 μM, compound 5 was found as the best in inhibiting proliferation with IC50 value of 2.25 ± 0.09 μM, and the other compounds showed significant inhibition with IC50 values ranging from 6.10 to 11.27 μM. Compound 5 also displayed the strongest cytotoxic effect toward MCF-7 cell line (IC50 4.32 ± 0.15 μM). Both 5 and 6 demonstrated strong radical scavenging activity (IC50 6.87 ± 0.03 and 4.33 ± 0.22 μM, respectively). Compounds 1 and 5 were isolated for the first time from this plant.

        열풍, 진공 및 동결건조가 눈개승마의 항산화 및 품질특성에 미치는 영향

        The purpose of this study was to evaluate the effect of drying methods including freeze, hot-air, and vacuum drying on the nutritional properties, antioxidant activities, and drying properties of Aruncus dioicus var. kamtschaticus. The optimum temperature of hot-air drying was 100℃ based on the total phenolic and flavonoid content and antioxidant activities, which were altered as a function of drying temperatures of 60, 80, 100, and 120℃. In addition, based on the drying curve, the optimum condition of hot-air and vacuum drying was 100℃ for 10 min, whereas that of freeze drying was 210 min. Vacuum drying was most effective for removal of moisture compared with the other drying methods. The water activity and water solubility index of dried samples obtained by the different drying methods were not significantly different. The water absorption index and rehydration ratio increased in the order of freeze, vacuum, and hot-air drying because of differences in porous structure of A. dioicus. Thus, vacuum drying of A. dioicus leads to the highest total phenolic and flavonoid content and antioxidant activities, followed by hot-air, and freeze drying.

        건조방법에 따른 눈개승마의 영양성분 및 항산화효과 비교

        The purpose of this study was to compare the effects of two drying methods (hot air drying [HAD] or vacuum freeze drying [FAD]) on the physicochemical components and antioxidative activities of Aruncus dioicus var. kamtschaticus powder. The crude fat content in the FAD was higher than that in the HAD. There were no significant differences in the proximate compositions, such as contents of moisture, crude ash, crude protein, and carbohydrates, depending on the drying methods. The total amino acid and essential amino acid contents were significantly higher in FAD than in HAD. The valine, phenylalanine, lysine, histidine, aspartic acid, asparagine, glutamic acid, arginine, ornithine, and γ-amino-n-butyric acid (GABA) contents in FAD were higher than in HAD, while the serine, phosphoserine, proline, glycine, alanine, tyrosine, and taurine contents in HAD were higher than in FAD. There were no significant differences in saturated fatty acid, monounsaturated fatty acid, and polyunsaturated fatty acid contents based on the drying methods. The most abundant fatty acid in the FAD was linolenic acid, and the linolenic acid content was approximately the same, regardless of the drying method used. The contents of malic acid and total organic acids in the FAD were higher than in the HAD, while the content of succinic acid was higher in the HAD than in the FAD. Vitamin C and E contents in HAD were higher than those in FAD. The total polyphenol and total flavonoid contents, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino- bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activities of the FAD sample were higher than those of the HAD sample. These results indicate that vacuum freeze drying is a better drying method for preserving the nutritional components and antioxidative activities. These study results may provide the basic data for follow-up studies seeking a better understanding of the functional properties of the FAD powder.
