        백작약(Paeonia japonica)의 항산화, 트롬빈 저해, 암전이억제 및 암세포사멸 평가

        백작약 물 추출물과 유기용매 분획물들의 항산화 활성, 트롬빈 억제 및 구강암세포주에서의 암전이 억제 및 암세포 사멸능을 확인하였다. 에틸아세테이트 분획물이 85.13%의 높은 항산화 활성과 87.54%의 높은 트롬빈저해 효과를 나타냈으며, 또한구강암세포주에서도 트롬빈 처리에 의해 활성화된 MMP-2/ pro-MMP-2이 높은 암전이 억제 활성을 나타냈다. 그리고 구강암세포주에 대한 세포사멸 효과는 물 추출물이 5배 이상의 높은 능력을 보였다. 이와 같은 연구 결과를 바탕으로 백작약 에틸아세테이트 분획물은 새로운 항산화제, 트롬빈억제제 및 암전이억제제의 개발을 위한 우수한 천연물 소재 후보 물질로서의가능성을 제시하고 있다. Paeonia japonica is a perennial flowering plant used in traditional medicine therapy. The purpose of this study was to investigate the effect of water extract and solvent fractions obtained from P. japonica on anti-oxidative, anti-thrombin, anti-invasive and pro-apoptotic activities in YD-10B cells, human oral squamous carcinoma cell line. Water fraction revealed the highest extraction yield at 11.44% (w/w). Anti-oxidative activity was the highest in ethyl acetate fraction (85.13%). In the thrombin inhibitory activity test, ethyl fraction was the highest, with a value of 87.54%. Release and activation of MMP-2/pro-MMP-2 ratio in thrombin-treated YD-10B cells were significantly inhibited in the ethyl acetate fraction. At a concentration of 120 ㎍/㎖, water extract and solvent fractions of P. japonica inhibited cell proliferation in YD-10B cells except water fraction. Pro-apoptotic effect on human oral squamous carcinoma cell using the Bax/Bcl-2 ratio analysis was higher in water extract than other fractions. These findings suggest that the ethyl acetate fraction of P. japonica potentiates a promising antioxidant, anti-thrombin and anti-invasive agents.         Fisetin의 항산화 활성 및 미용기능식품 소재로서의 가능성 고찰

        The term of nutri-cosmetics refers to nutritional supplements which can support the function and the structure of the skin. The fisetin is a flavonol which belongs to the flavonoid group of polyphenols. Although the anti-carcinogenic property of fisetin have been widely studied, anti-oxidative function of fisetin in HaCaT keratinocytes is unknown. In this study, we examined the in vitro chemical and cellular anti-oxidant activities of fisetin in HaCaT keratinocytes and we further discussed its potential as an anti-oxidant nutricosmetics. We employed scavenging assay for the 1,1-diphenyl-2,5-picrylhydrazyl (DPPH) radicals and cellular anti-oxidant activity of fisetin was investigated in H2O2-treated HaCaT keratinocytes as judged by the 2’7’-dichlorofluorescein-diacetate (DCFH-DA)-based assay. The fisetin effectively scavenged DPPH radicals (IC50 54 μM) when compared to the scavenging activities of L-ascorbic acid (IC50 87 μM) acid and trolox (IC5074 μM), well-known anti-oxidants. In addition, the fisetin significantly ameliorates H2O2-induced oxidative stress in HaCaT keratinocytes in dose-dependent manner. The fisetin showed more strong cellular anti-oxidant activity than those of L-ascorbic acid and trolox in H2O2-treated HaCaT keratinocytes. In agreement with result of the cellular anti-oxidant activity, we observed markedly suppressed H2O2-induced oxidative stress by the pretreatment of 100 μM fisetin under the fluorescence microscopy analysis. These results provide the scientific evidences for the development of skin targeted nutri-cosmetics which has anti-oxidant function.

        人蔘의 配合에 따른 抗酸化作用에 關한 硏究 (1) : 人蔘과 配合하는 數種 韓藥制의 抗酸化 作用에 對한 比較 硏究 Study on comparative anti - oxidative effects of some herbal medicine frequently combined with Ginseng Radix

        This study was carried out in order to investigate the anti-oxidative effects of main tonic herb medicine, Ginseng Radix, Rehmammiae Radix Preparat, Astragali Radix, Poria, Aconiti Iateralis Preparata Radix, Gecko and Bupleuri Radix. These herbs were measured on inhibitory activity against lipid peroxidation of linoleic acid mixture and rat liver and hydrogen-giving-temper effect by DPPH. The study showed the following valuable results: 1. In inhibitory activity of water extract against lipid peroxidation of linoleic acid mixture, Gecko showed the highest anti-oxidant activity among the some herbal medicine and Rehmammiae Radix Preparat showed the lowest anti-oxidant activity when compared with the examined samples. 2. In inhibitory activity of water extract against lipid peroxidation of rat liver, Gecko showed the highest anti-oxidant activity among the some herbal medicine and Rehmammiae Radix Preparat showed the lowest anti-oxidant activity. 3. In hydrogen-giving-temper effect of water extract by DPPH, Gecko showed the highest anti-oxidant activity. 4. In the linoleic acid mixture, Gecko showed the highest anti-oxidant activity in Butanol fraction. 5. In the lipid peroxidation of rat liver, Gecko showed the highest anti-oxidant activity in Butanol fraction. 6. In hydrogen-giving-temper effect of water extract by DPPH, Gecko showed the highest anti-oxidant activity in Butanol fraction. From the above results, it was concluded as follow: All the seven herb medicine including Ginseng Radix showed the anti-oxidant activity. Especially, Gecko showed the highest anti-oxidantactivity among them.

        Endlicheria anomala (Nees) Mez 추출물의 항산화, 항염증 및 미백 활성

        본 연구에서는 Endlicheria anomala (Nees) Mez 메탄올 추출물(EAME)의 항산화, 항염증 및 미백 생리활성을 in vitro assay 및 cell culture model system을 이용하여 분석 하였다. EAME의 항산화능을 분석한 결과 DPPH, H2O2로 유도한 ROS, LPS로 유도한 NO 등 다양한 산화적 스트레스원을 효과적으로 소거하였다. 대표적인 항산화 효소들로 천연물에 의한 항산화능 활성에 의해 주로 발현이 유도되는 세효소인 HO-1, TrxR1, NQO1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 시료 처리 농도의 증가에 따라 세 효소 및 Nrf2의 발현이 유의적으로 증가됨을 보였다. 또한 EAME는 in vitro DOPA oxidation을 강하게 저해하여 tyrosinase inhibitor로서 작용할 가능성을 시사 하였고 이에 B16F10 melanocyte를 이용하여 미백 효능을 분석한 결과 유의적인 melanin 생성억제능 및 tyrosinase 효소 활성 억제능을 보였으며 이는 tyrosinase, TRP-1, TRP-2 등 melanin 생성의 핵심 작용 효소들의 단백질 발현 저해를 통해 일어나는 것으로 나타났다. 이러한 결과를 통해 EAME가 높은 항산화능과 항염증 활성, 그리고 미백 활성을 보유함을 처음으로 밝혔으며 향후 기능성 식품 및 피부 미용 소재로서유용하게 활용될 수 있을 것으로 판단된다. In this study, the anti-oxidative, anti-inflammatory, anti-melanogenic activities of Endlicheria anomala (Nees) Mez methanol extract (EAME) were evaluated by use of in vitro assays and cell culture model systems. The results revealed that EAME scavenges various radicals such as 1,1-diphenyl-2-picryl hydrazyl hydrogen peroxide induced reactive oxygen species, and lipopolysaccharide induced nitric oxide. Furthermore, EAME induced the expression of anti-oxidative enzymes such as heme oxygenase 1, thioredoxin reductase 1, NAD(P)H dehydrogenase 1, and their upstream transcription factor, nuclear factor-E2- related factor 2. Moreover, EAME inhibited in vitro DOPA oxidation and 3-isobutyl-1-methylxanthine induced melanogenesis in B16F10 cells. Its anti-melanogenic activity will have originated from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Taken together, these results provide the important new insight that E. anomala possesses various biological activities such as anti-oxidative, anti-inflammatory, and anti-melanogenic. Therefore, it might be utilized as a promising material in the fields of nutraceuticals and cosmetics.

        산초 추출물 및 분획물의 항산화, 미백 및 항염증 활성

        본 연구에서는 한약재로 사용되고 있는 산초 과피 추출물의 기능성 소재로서의 활용 가능성을 알아보기 위해 산초 추출물 및 분획물이 보유한 항산화, 미백 및 항염증 활성을 in vitro assay system 및 cell culture model system을 이용하여 분석하였다. 그 결과 산초 MeOH, EtOH, 열수 추출물 모두 강한 DPPH radical 소거 활성 및 tyrosinase 효소 활성저해를 통한 DOPA 산화 억제능을 보였다. B16F10 mouse melanoma cell을 이용하여 α-MSH에 의해 유도된 세포 내 melanin 생성에 미치는 산초 추출물의 영향을 알아본 결과 농도의존적인 melanin 생성 억제능을 보였고 세포의 tyrosinase 활성 저해능 또한 이와 일치하는 결과를 보여 산초 추출물에 의한 melanin 생성 억제능이 tyrosinase 효소 활성 억제 및 melanogenesis 관련 단백질 발현 저해에서 기인한 것으로 판단된다. 또한 RAW 264.7 murine macrophage cell을 이용하여 산초 추출물의 항염증 활성을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 단백질 발현 분석을 통해 확인하였다. 산초 추출물에 존재하는 활성물질을 규명하기 위해 용매 분획을 실시한 후 각 분획물의 미백 및 항염증 활성을 비교 분석한 결과 물 층을 제외한 모든 용매분획이 활성을 보유함을 확인하였다. 이러한 결과를 통해 산초 추출물이 높은 항산화능과 미백활성, 그리고 항염증 활성을 보유함을 확인할 수 있었으며, 이러한 결과는 미백활성을 비롯한 생리활성 보유 천연물 소재 탐구의 기초자료로 유용하게 쓰일 것으로 사료된다. This study was designed to explore new nutraceutical and cosmetic resources possessing biological activities from the plant kingdom. To fulfill this purpose, we analyzed the anti-oxidative, anti-melanogenic, and anti-inflammatory activities of Zanthoxylum schinifolium extract (ZSE) and its solvent fractions using in vitro assays and cell culture model systems. Three kinds of ZSE treated with methanol, ethanol, and water exhibited potent anti-oxidative activities through DPPH radical scavenging capacity, and inhibited in vitro DOPA oxidation. Furthermore, Z. schinifolium methanol extract (ZSME) inhibited the α-melanocyte stimulating hormone, which induces melanin contents in B16F10 cells. Its anti-melanogenic activity originates from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Moreover, lipopolysaccharide induced nitric oxide production in the RAW 264.7 cell line was also ameliorated by ZSME treatment in a dose dependent manner. Among the four solvent fractions of ZSME treated with dichloromethane, ethyl acetate, nbutanol, and water, three fractions, except water, showed significant anti-melanogenic and anti-inflammatory activities. Taken together, these results provide important new insights into Z. schinifolium, indicating that it possesses numerous biological activities such as anti-oxidative, anti-melanogenic, and anti-inflammatory activities. Therefore, it may well serve as a promising material in the field of nutraceuticals and cosmetics.

        화장품 소재로서 고마리 추출물의 생리활성

        Purpose: The purpose of this study was to investigate the potential of Persicaria thunbergii (P. thunbergii) extracts as physiologically active cosmetic ingredients. Methods: To elucidate the anti-oxidative, anti-inflammatory, and anti-microbial properties of P. thunbergii , its ethanol and hot-water extracts were examined for 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, nitric oxide (NO) production, protective effects against oxidative stress in HaCaT cells, anti-inflammatory activity, anti-microbial activity, and inhibition of β-hexosaminidase expression, an allergy factor. Results: The anti-oxidative activity of the two P.thunbergii extracts was compared, and the anti-oxidative activity of the ethanol extract was found to be superior to that of the hot-water extract. The polyphenol contents of P. thunbergii ethanol and hot-water extracts were 132.00 and 89.46 μg/ mL, respectively. And total flavonoid contents of P. thunbergii ethanol and hot-water extracts were 115.27, 84.94 μg/mL, respectively. The DPPH radical scavenging activity was found to be 79.80% for the ethanol extract and 70.33% for the hot-water extract at a concentration of 500 μg/mL. No significant cytotoxicity was observed in HaCaT, RAW 264.7 and RBL-2H3 cells. The protective effect of the extracts on HaCaT cell against oxidative stress induced by hydrogen peroxide (H 2 O 2 ) was confirmed by 23% for ethanol extract and 18% for hot water extract. The anti-inflammatory activity of the extracts was examined in RAW 264.7 cells, and nitric oxide (NO) production was suppressed in a concentration-dependent manner. Both ethanol and hotwater extracts also inhibited the degranulation of immune cells in a concentration-dependent manner, as assessed by the secretion of β-hexosaminidase. In addition, the concentration dependent anti-microbial activities of the extracts were demonstrated in several bacterial strains, such as those of Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), and Propionibacterium acnes (P. acnes). Conclusion: Based on the findings from this study, P. thunbergii extracts could be used as functional cosmetic ingredients that possess anti-oxidative, anti-inflammatory, and anti-microbial properties. In order to determine their applicability, further research should be systematically conducted to verify their efficacy through clinical studies in animals and humans. 목적: 고마리 추출물에 대한 생리활성 및 그에 따른 화장품 소재로서의 응용 가능성을 알아보고자 하였다. 방법: 고마리 에탄올및 열수 추출물의 항산화, 항염, 항균 효능을 확인하기 위하여 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical 소거능, nitric oxide (NO) 생성량 측정, 산화적 스트레스에 대한 HaCaT 세포 보호 효과, 항염, 항균, 알러지인자인 β-hexosaminidase 발현량 억제를 측정하였다. 결과: 고마리 추출물의 항산화 효능을 비교한 결과, 에탄올 추출물이 열수 추출물보다 우수하였으며, 고마리 에탄올, 열수 추출물의 폴리페놀 함량은 각각 132.00, 89.46 μg/mL이고, 총 플라보노이드 함량은 각각 115.27, 84.94 μg/mL으로 확인되 었다. 그리고 DPPH radical 소거능 측정결과, 고마리 에탄올, 열수 추출물의 농도가 500 μg/mL 일 때 각각 79.80, 70.33%로 확인 되었으며, 세포 독성 실험결과, HaCaT, RAW 264.7, RBL-2H3 세포에서 유의한 세포독성은 나타나지 않았다. Hydrogen peroxide (H 2 O 2 )에 의해 유발되는 산화적 스트레스에 대한 HaCaT 세포 보호 효과 확인결과, 고마리 에탄올, 열수 추출물은 23%, 18%로 세포 보호 효과가 확인되었다. RAW 264.7 세포에 대한 고마리 추출물의 항염 효과를 확인한 결과, 농도 의존적으로 nitric oxide (NO) 생성이 억제되었다. β-Hexosaminidase의 분비를 측정한 결과, 고마리 에탄올과 열수 추출물은 농도 의존적으로 면역세포의 탈과 립을 억제하였다. 고마리 추출물의 항균효능을 측정한 결과, Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), Propionibacterium acnes (P. acnes)에서도 농도 의존적으로 항균 활성이 확인되었다. 결론: 이상의 연구결과를 통해 고마리 추출물은 항산화, 항염증 및 항균 효과를 가지는 기능성 화장품 소재로서의 효능이 확인되었다. 앞으로 기능성 화장품 소재 로서의 활용 가능성을 판단하기 위하여 임상적으로 동물, 인체실험을 통한 고마리 추출물의 효능 검증 연구가 체계적으로 진행되어야 할 것이다.

        보중익기탕 (補中益氣湯)의 한국, 중국, 일본 처방에 대한 항염증 및 항산화 효과 비교 연구

        Objectives : Bojungikgitang (BJT), the Oriental medical prescription has been traditionally used about improvement of immune response and infective disease at Asian nation. In this study, we has compared about the anti-inflammatory and antioxidative effects on BJT of three countries including Korea (Korean Traditional Medicine, KTM), China (Traditional Chinese Medicine, CTM) and Japan (Japanese Traditional Medicine, JTM). Methods : We has basically using LPS-stimulated RAW 264.7 cells. The expression of these inflammatory mediators has measured using enzyme-linked immunosorbent assay (ELISA) and reverse transcription polymerase chain reaction (RT-PCR). Also, free radical scavenging assay has tested for anti-oxidative activity as well as the contents of total flavonoid and polyphenol. Results : As a result, we were founded the inhibitory effects of BJT (KTM, CTM, JTM) on LPS-induced production of NO, TNF-α and IL-6 as well as the anti-oxidative activities. Especially the KTM was most effective in anti-inflammatory and anti-oxidative activities. Conclusions : These results indicate that BJT (KTM, CTM, JTM) has a good anti- inflammatory and anti-oxidative effects. But, there were degree of effects on between pharmacopoeia of the countries. Thus, further study is required that find appropriate methods for extracting as well as establish of standardized processes in order to improve the quality of BJT (KTM, CTM, JTM) as an anti-inflammatory and anti-oxidative agent for treatment of inflammatory diseases.

        Anti-oxidant and anti-inflammatory effects of Salix Koreensis Andersson in DC. leaf methanol extract in vitro models

        Oxidative rancidity in foods causes undesirable changes in nutritive value, aroma, flavor, and color. Salix Koreensis Andersson in DC. (SK) has anti-oxidative and anti-inflammatory effects and is traditionally used to treat neuralgia, edema, pain, and inflammatory diseases. However, the regulatory effects of SK on oxidative and inflammatory reactions have not been elucidated. In this context, we scientifically validated the anti-oxidative and anti-inflammatory activities of SK leaf (SKL). The methanol extract of SKL was evaluated for in vitro anti-oxidative activities. SKL showed increased superoxide dismutase (SOD)-like activity and 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity. The in vitro anti-oxidant and anti-inflammatory activities of SKL were also investigated in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. LPS resulted in decreased SOD activities compared with the unstimulated cells, but SKL significantly increased SOD activities reduced by LPS. In addition, LPS-induced nitric oxide, tumor necrosis factor-${\alpha}$, and interleukin-6 productions were significantly and dose-dependently reduced by SKL in RAW264.7 macrophages without inducing cytotoxicity. In conclusion, these results indicate that SKL will be able to be effectively used as a food additive with anti-oxidative and anti-inflammatory effects.

        고마리 추출물의 항산화, 항균, 항염, 항알레르기 효과

        Objectives : Reactive oxygen species (ROS) are involved in a wide spectrum of diseases including chronic inflammation and cancer. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the extracts from the herbal teas such as Lonicera japonica Thunberg (L. japonica), Chrysanthemum morifolium Ramat (C. morifolium), Mentha arvensis L. (M. arvensis), and P.rhizoma. Methods : Anti-oxidant activity was evaluated using DPPH radical scavenging assay and $Fe^{2+}$ chelating assay. And DNA cleavage assay was performed to evaluate an anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell line. Results : L. japonica scavenged DPPH radical by 9.8% at 12.5 ${\mu}g/ml$, 24.8% at 25 ${\mu}g/ml$, 34.3% at 50 ${\mu}g/ml$, 61.1% at 100 ${\mu}g/ml$ and 75.8% at 200 ${\mu}g/ml$, respectively. In addition, C. morifolium and M. arvensis removed DPPH radical by 15.6% and 10.4% at 12.5 ${\mu}g/ml$, 34.8% and 22.8% at 25 ${\mu}g/ml$, 66.9% and 43.3% at 50 ${\mu}g/ml$, 87.4% and 69.1% at 100 ${\mu}g/ml$, and 92.1% and 73.2% at 200 ${\mu}g/ml$, respectively. However, P. rhizoma did not affect on DPPH radical scavenging. The $Fe^{2+}$ chelating activity was highest in L. japonica, but lowest in P. rhizoma among the herbal teas. In addition, the extracts from L. japonica, C. morifolium and M. arvensis inhibited oxidative DNA damage via its anti-oxidant activity. In anti-inflammatory effect, the extracts from C. morifolium inhibited NO production. In addition, it suppressed the $NF-{\kappa}B$ signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, this study indicates that L. japonica, M. arvensis and C. morifolium possess the protective effect against the oxidative DNA damage. Furthermore, C. morifolium exerts an anti-inflammatory effect.
