      • 골절 환자 혈청내 Alkaline Phosphatase와 Osteocalcin의 변화에 관한 연구

        연구목적: 골절 후 비교적 측정이 용이한 골 형성 지표인 혈청 내 alkaline phosphatase와 osteocalcin의 변화를 분석하여 생화학적 측면에서 골절 치유 과정을 알아보고 이들이 골 형성 및 골 유합을 예측할 수 있는 객관적인 지표로 이용될 수 있는지 알아보고자 하였다. 대상 및 방법: 22예의 골절 환자에서 수상 직후, 수상 후 2주, 수상 후 4주, 수상 후 8주때의 혈청 내 alkaline phosphatase, bone specific alkaline phosphatase, osteocalcin 등의 골형성 지표의 증가 정도를 비교 분석하였다. 결과: 혈청 내 alkaline phosphatase는 수상 직후부터 오르기 시작하여 4주경 가장 높게 나타났으며 osteocalcin은 수상 후 1주 때부터 점차로 증가하여 8주 때 가장 높게 나타났다. 결론: 골절 치유 과정에서 혈청 내 alkaline phosphatase와 osteocalcin이 증가시기에 있어 차이를 보이는 이유는 먼저 골 기질이 성숙되는 시기에 alkaline phosphatase가 증가하고 이 후 무기질화가 진행되며 osteocalcin이 주로 발현되기 때문이며 이러한 골 형성 지표의 변화는 골절 치료에 있어 치료 방법을 결정하거나, 어떤 약물이나 치료 방법의 효과를 평가하는 또 다른 지표가 될 수 있을 것이다. Purpose: This study was to evaluate the changes of osteoblastic marker during fracture healing. Materials and Method: The study included 22 fresh fractured patients. Serum alkaline phosphatase, bone specific alkaline phosphatase and osteocalcin were analyzed at first day, 1st week, 2nd week, 4th week, and 8th week after injury. The bone specific alkaline phosphatase was quantified by electrophoresis. Osteocalcin was quantified by enzyme-linked immunosorbent assay (ELISA). Result: At the time of first week, second week, fourth week and eighth week after injury, mean values of serum alkaline phosphatase increased 1.26 times, 1.45 times, 1.63 times, and 1.2 times than values of first day after injury, respectively. During the same period, bone specific alkaline phosphatase increased 1.38 times, 1.33 times, 1.73 times and 1.25 times. And serum osteocalcins increased 1.32 times, 1.2 times, 1.64 times and 2.09 times than values of first day Conclusion: Serum alkaline phosphatase increased during early soft callus phase. But serum osteocalcins, a protein thought to be expressed only by osteoblastic cells, were not increased during soft callus phase but were increased during late hard callus phase during fracture healing. During the fracture healing, the differentiation of osteochondral progenitor cells into chondrocytes and osteoblasts is the first step. As being hypertrophy of these cells followed by mineralization of the cartilage, concomitant elevation of alkaline phosphatase level was known. As ossification proceeded, the expression of chondrocyte decreased, but expression of osteoblast increased. As woven bone converted to lamellar bone, expression of osteoblasts decreased. With further study, serum alkaline phosphatase and osteocalcin seemed to be useful prognostic indicators of fracture healing.

        호르텐스극구흡충에서 Alkaline Phosphatase 및 Acid Phosphatase의 특성

        호르텐스극구흡충 성충 (adult worm)을 재료로 하여 acid phosphatase (Acp)와 alkaline phosphatase (Alp)의 조직 세포에 대한 분포 및 동위효소의 특성을 규명하기 위하여 Gomori lead nitrate 및 Gomori calcium 방법 그리고 전기영동법 등을 시행하였다. 조직 내의 Acp 및 Alp의 분포는 충체의 소장 (intestine)과 난황선 (vitellaria) 그리고 인두 (pharynx)에서 강한 반응을 나타냈다. 동위효소 유형은 Acp에서 179.5 및 209.4 kDa 두 개의 분획이 분리되었고, Alp는 145.9, 207.5 및 220.8 kDa의 세 개의 분획이 분리되었다. 열에 대한 안정도는 Alp가 90℃에서 12초 경과 후에 완전히 불활성화 되었으며, Alp의 최적 온도 및 pH는 40℃와 pH 9이었고 Acp의 최적 pH는 5였다. 그리고 Acp 및 Alp의 최대 활성도 (unit)는 Acp가 71 범위였고, Alp는 189의 범위에 있었다. 이상에서와 같이 호르텐스극구흡충은 phosphatase가 소화관과 난황선에서 주로 분포하고 있었으며 숙주 체내에서 소화관을 중심으로 대사가 활발하게 이루어지고 있음을 알 수 있었다. This study was aimed to investigate the enzyme-histochemical localization and characteristics of alkaline and acid phosphatase extracted from adult of Echinostoma hortense. Using the Gomori calcium stain and the Gomori lead nitrate satin method, we found that the alkaline and acid phosphatases were localized mostly in the intestine, vitellaria and pharynx of Echinostoma hortense. The three isozymes of alkaline phosphatase and two isozymes of acid phosphatase were separated from Echinostoma hortense by electrophoresis. The isozymes of alkaline phosphatase were 145.9, 207.5, 220.8 kDa and the isozymes of acid phosphatase were 179.5 and 209.4 kDa. The activity of alkaline phosphatase was denatured completely after heating at 90℃ for 12 seconds. The optimum pH and temperature for activity of alkaline phosphatase were about pH 9 and 40℃, while the optimum pH for activity of acid phosphatase was about pH 5. The maximum activity of alkaline phosphatase was at 189 unit, but maximum activity of acid phosphatase was at 71 unit. As the result from above, we observed that alkaline and acid phosphatases funtion mainly in the alimentary tract and vitellaria. Echinostoma hortense performs the parasitism in the intestine of host by using proper isozyme of phosphatase.

        지속적 및 간헐적 가압력이 MC3T3-E1세포의 Alkaline phosphatase 활성도에 미치는 영향

        교정력에 의한 치아이동은 세포반응의 결과로 나타난다. 이에 힘의 양상에 따른 세포의 반응을 알아보고자 MC3T3-E1세포를 24well 배양접시에 배양한 후 밀생상태가 되었을 때, incubator 속의 특수 제작된 사각상자에 배양 접시를 넣고 타이머에 연결된 Diaphram pump를 이용하여 300 gm/㎠의 압력을 10분간 가압후 10분간 가압이 중지되도록 한 간헐적 가압군과 지속적으로 가한 가압군으로 하여 각각 실험 24시간, 28시간, 72시간후의 alkaline phosphatase 활성도를 대조군과 비교하여 다음과 같은 결과를 얻었다. 1. 가압 24시간 후에서는 각 군간에 alkaline phosphatase 활성도에 유의한 차이가 나타나지 않았다. 2. 가압 48시간 후에서는 간헐적 가압군과 지속적 가압군 모두 대조군 보다 alkaline phosphatase 활성도가 증가 되었다. 3. 가압 72시간 후에서는 간헐적 가압군의 alkaline phosphatase 활성도 증가가 대조군에 비하여 더욱 현저하였으나 지속적 가압군에는 대조군에 비하여 유의한 차이가 나타나지 않았다. 4. 가압 72시간 후에서는 간헐적인 가압군이 지속적인 가압군에 비하여 alkaline phosphatase의 활성도가 증가되었다. 5. 가압에 의한 세포의 뚜렷한 형태학적인 변화는 관찰되지 않았다. The propose of this study was to evaluate the difference of cellular activity dependent on intermittent compressive force by determining the alkaline phosphatase activity. Alkaline phosphatase activity was measured on control and experimental groups every 24, 48, 72hours. Experimental groups consisted of continous and intermittent compressive group which were compressed by 300g/㎠ of diaphram pump. The intermittent compressive group was connected by timer which was worked on 10 minutes an off 10minutes. The results were as follows ; 1. The alkaline phosphatase activity between control and experimental groups showed not significant difference at compressed 24 hours. 2. The alkaline phosphatase activity of experimental groups were more increased than control group at compressed 48 hours. 3. The alkaline phosphatase activity of intermittent compressive group showed significant increased to control group. Whereby continuous compressive group showed not significant difference to control at 72 hours. 4. The alkaline phosphatase activity of intermittent compressive group were stringly increased than continuous compressive groups. 5. Between experimental groups and control group no other morphologic changes were detected by microscopic findings.

      • Adriamycin이 랫트 신장조직의 Alkaline Phosphatase, Adenosine Triphosphtase와 Acid Phosphatase 활성에 미치는 영향에 관한 조직화학적 연구

        Anthracycline계 항암항생제인 adriamycin투여에 따른 랫트 신장 독성을 투여 시간 경과에 따라, alkaline phosphatase, adenosine triphosphatase와 acid phosphatase의 활성 변화로 관찰하였다. This experiment was carried out to investigate the effect of adriamycin on renal alkaline phosphatase, adenosin triphosphatase and acid phosphatase in rat. The experimental animals were administered with single dose of adriamycin, Img/kg, by the intravenous injection. The following result were obtained 1.The alkaline phosphatase activity appeared to be moderately decreased in the proximal convoluted tubules at the 8 hours. This decrease maintained by the second day and recovered to the normal state at the fourth day. 2.The adenosine triphosphatase activity appeared to be moderately increased in the proximal convoluted tubules at the 12 hours This increase maintained by the eighth day. 3.The acid phosphatase activity is moderately increased in the proximal convoluted tubules at the 4 hours. This in crease maintained by the 18 hours and recovered to the normal state at the first day.

      • Cisplatin이 흰쥐 십이지장의 alkaline phosphatase 활성에 미치는 영향에 대한 연구

        Cisplatin is a platinum compound, which has been widely used as an anticancer chemotherapeutic agent. During the treatment of cancer patients cisplatin acts on normally proliferating cells, which results in nephrotoxicity, nausea, vomiting, anorexia, bone marrow suppression and inner ear damage. Cisplatin inhibits the DNA synthesis, protein synthesis and cell division by incorporation to DNA strands. The author has investigated the effect of cisplatin on duodenum of rats histochemically observing the changes in the activity of alkaline phosphatase. The rats treated with 6mg per kg of cisplatin were sacrificed at 6 hours, 12 hours, 24 hours and 3 days after the drug administration. The rats of control group were administered only water for injection. The duodenum specimens were fixed in 10%-neutral formalin at 4 。and sectioned 16 。thickness in frozen cryostat. The activity of alkaline phosphatase was evaluated histochemically by Gomori's method and incubation time of sliced specimen was 30 minutes. The results were as follows. 1. In control rats, the activity of alkaline phosphatase were moderate positive in the epithelium of mucosa and weak or trace positive in lamina propria, submucosa and muscle layer. 2. In the epithelium of the mucosa the activity of alkaline phosphatase were weak positive in 6 hours of cisplatin treated rats, moderate or strong positive in 12 hours and 3 days of cisplatin treated rats and strong positive in 24 hours of cisplatin treated rats. 3. There are trace or weak reactivities in the lamina propria, submucosa and muscle layer of duodenum of all groups. Consequently, it is suggested that cisplatin reduces the activity of alkaline phosphatase at 6 hours after administration and the activities of alkaline phosphatase by regeneration and differentiation of epithelium in the duodenum of cisplatin treated rats.

      • Cytarabine이 마우스 간장의 Alkaline Phosphatase와 Adenosine Triphosphatase 활성 세포의 조직화학적 분포에 미치는 영향에 관한 연구

        미정맥으로 투여된 Cytarabine은 마우스 간장의 효소 분포 양식에 영향을 미쳐 투여 4시간군부터 alkaline phosphatase의 활성이 문맥관 주변부와 중간대 간세포에서 감소되기 시작되어 투여 8시간군에서는 현저히 저하되었고 adenosine triphosphatase 활성은 이때부터 중심정맥 주변부와 중간대 간세포에서 증가되었으며, alkaline phosphatase의 활성 감소는 투여 24시간군까지 지속되었으나 adenosine triphosphatase의 활성 증가는 24시간군에서 정강으로 회복되었다. This experiment was carried out to investigate the effect of cytarabine on the distribution pattern of the hepatic alkaline phosphatase and adenosine triphosphatase in mouse. The experimental animals were administered with single dose of cytarabine, 1.6 mg/kg, by the intravenous injection. The following results were obtained. 1.In the normal hepatic lobule of mouse, the alkaline phosphatase activity was concentrated particularly in the portal canal and the midzonal area. And the adenosine triphosphatase appeared to the mild activity in the overall area. 2.The activity of alkaline phosphatase began to be slightly decreased from the 4 hours and appeared to be markedly decreased at the 8 hours in the portal canal and midzonal area. And the activity of adenosine triphosphatase began to be markedly increased from the 8 hours and recovered to the normal state at the 24 hours in the central area and midzonal area.

      • 홍모기(Culex pipiens pallens)의 난성숙 과정 중 alkaline phosphase의 활성

        Culex pipiens pallens에 존재하는 Alkaline phosphatase의 연구를 위한 적정 분석조건과 우화 후, 시간 경과에 따른 Alkaline phosphatase 활성 경향에 대해서 연구하였다. C. pipiens에 존재하는 Alkaline phosphatase의 활성은 우화 직후부터 지속적으로 감소하다가, 흡혈 자극에 의해서 급격하게 증가한다. 흡혈 후 30시간이 경과했을 때, 최대의 활성도를 보이고 감소하나, 흡혈 48시간 이후에는 다시 증가하여 지속적으로 유지됨을 알 수 있다. 기관별 분석에서 첫번째 활성 증가는 중장에서 일어나고, 두번째 활성 증가는 난소에서 일어남을 알 수 있다. 그리고 흡혈 후 30시간된 성체에서는 5개의 동위효소 밴드가 보이는데, 난소에서 ALP-1와 ALP-2가 나타나고, 가슴에서는 ALP-3, ALP-4와 ALP-5가 보인다. 지방체에서는 ALP-4와 ALP-5가, 중장에서는 ALP-3, ALP-4와 ALP-5가 나타남을 알 수 있다. 그리고 흡혈 후 72시간된 성체에서, ALP-1, ALP-2가 동일하게 존재함을 알 수 있다. Alkaline phosphatase from Culex pipiens pallens was examined to determine the optimal assay condition and to assay the activity during ovarian development. The activity of alkaline phosphatase in a male and a nongravid female continuously were declined after eclosion. But by the stimulus of a blood meal, the enzyme activity was increased dramatically. At 30 hr. after a blood meal, the maximal activity was reached and then declined. And after 48 hr. after a blood meal, the second activity increase was revealed. This second increase was maintained up to oviposition. The first activity increase was revealed in the midgut and the second increase was done in the ovary to assay the organ distribution of alkaline phosphatase. In electrophresis data, it was shown 5 isozyme bands, ALP-1 and ALP-2 in the ovary, ALP-3 in the thorax and the midgut, and ALP-4 and ALP-5 in the thorax, the fatbody and the midgut in crude extract at 30 hr. after a blood meal. One the same ovary pattern were shown at 72 hr. after a blood meal.

      • Prevalance and Factors Associated with SGOT, SGPT and Alkaline Phosphatase Among Suspected Liver Patients Attending Salyan District Hospital, Salyan, Nepal

        Aims: Serum glutamic-oxaloacetic transaminase (SGOT), serum glutamic-Pyruvate transaminase (SGPT) and alkaline phosphatase are reasonably sensitive indicators of liver damage or injury from different types of diseases or conditions. The study aimed to determine the level of SGOT, SGPT and alkaline phosphatase; and SGOT/SGPT ratio among suspected liver patients attending salyan District hospital of Nepal. Methods: Blood samples were collected from suspected liver patients visiting Salyan hospital. The samples were collected from April, 2019 to October 2019. Enzyme kinetics method was used to process the serum samples. Descriptive statistics and chi-square test were computed at 5% level of significance Results: Of total 116 suspected liver patients, 64.7% patients had high SGOT, 53.4% had high SGPT and 7.8% had high alkaline phosphatase. The prevalence of SGOT and SGPT and was higher among males (65.8% Vs 62.2%), (54.4% Vs 51.4%) respectively. Regarding alkaline phosphatase, there was no any higher level among female. Alcoholic behavior was statistically associated with high SGOT in the study (P<0.05); it was highest among alcoholic group. However, there was no statistical association of high SGOT with sex and ethnicity. Similarly, Alcholic behavior and ethnicity was statistically associated with high SGPT in the study (P<0.05). Likewise, Sex and ethnicity was statistically associated with high SGPT in the study (P<0.05); however, there was no statistical significance with alcoholic behavior. Conclusions: The study shows high prevalence of SGOT, SGPT and alkaline phosphatase; among suspected liver patients attending Pyuthan hospital of Nepal.

        지속적 및 간헐적 가압력이 치주인대 배양세포의 Alkaline Phosphatase 활성도에 미치는 영향

        지속적 및 간헐적인 가압력이 배양치주인대 세포의 ALP활성도에 미치는 영향을 알아보기 위하여 치주인대세포를 24 well 배양접시에 배양한 후 밀생상태가 되었을 때, 세포배양기 속의 특수제작된 사각상자에 배양접시를 넣은 후 실험기간동안 300g/㎠의 압력을 10분간 가압시킨 후 10분간 가압이 중지되도록 한 간헐적 가압군, 그리고 압력을 가하지 않은 군을 대조군으로 하여 각각 실험 24시간, 72시간 후의 alkaline phosphatase의 활성도를 측정하여 다음과 같은 결과를 얻었다. 1. 가압 24시간 군에서는 간헐적 가압군이 대조군에 비하여 ALP활성도가 낮게 나타났다(p>0.05). 2. 가압 48시간 군에서는 실험군은 대조군에 비하여 ALP활성도에 유의한 차이는 나타나지 않았다. 3. 가압 72시간 군에서는 지속적 가압군이 대조군에 비하여 ALP활성도가 높게 나타났다(p>0.01). An intermittent and continuous compressive force were applied on PDL cells at the confluent stage. The alkaline phosphatase activity was measured on control and experimental groups every 24, 48, 72hours. The experimental group were consist of continuous and intermittent compressive group which were compressed by 300g/㎠ of diaphram pump. The intermittent compressive group was connected by timer which was worked on 10 minutes and off 10minutes. The results were as follows; 1. The alkaline Phosphatase activity of intermittent compressive group was lower than control at 24 hours(p>0.05). 2. The alkaline Phosphatase activity between each groups showed no significant differences at 48hours. 3. The alkaline Phosphatase activity of continuous compressive group was significantly higher than control group at 72 hours(p>0.01).
