        중년여성의 자각하는 유·수분 유형과 비침습적 방법으로 측정한 안면피부상태

        일부 도시 중년여성을 대상으로 안면부위별 피부상태를 파악하고자 비침습적 측정방법을 이용하여 유 수분, 멜라닌 홍반 정도를 관찰하였고, 주관적으로 자각하는 유 수분 유형을 조사하여 객관적 측정값들과의 관련성을 보고자 하였다. 안면부위별 측정 결과, 수분량은 눈가 > 이마 > 볼, 유분량은 이마 > 눈가 > 볼 부위 순이었으며, 홍반지수는 이마 > 볼 > 눈가, 멜라닌지수는 눈가 > 이마 > 볼 부위 순이었다. 객관적 유분 측정값과 주관적으로 자각하는 유분유형은 일치성을 나타내었다. 유분량 홍반지수, 멜라닌지수의 각 측정값들은 유의한 양의 상관관계를 보였다. The content of lipid or moisture and index of erythema or melanin on each facial part of 170 urban middle-aged women without dermic disease was measured by non-invasive method. This measurement intended to understand the skin condition on each facial part of middle-aged women, and to determine the correlation between objective measured values and subjective symptom of lipid content/moisture content. The results was summarized as follows. At each facial Part, the order of moisture content was eye rims, forehead, and cheek and that of lipid content was forehead, eye rims, and cheek. The erythema index was in the order of forehead, cheek and eye rims and melanin index, in the order of eye rims, forehead and cheek. It turned out that the middle-aged women generally lacked lipid content rather than moisture one. The objective measured values of lipid content coincided with subjective symptom of lipid type. The lipid content, erythema index and melanin index showed correlation significantly.

        느릅나무의 에틸 아세테이트 추출물에 의한 Melanin생성 효과

        Melanogenesis is a physiological process resulted in the synthesis of melanin pigments, which has a role in protecting skin from the damaging effect of ultra-violet (UV)radiation. The main aim of the present study was to examine the effect of Ulmus davidiana var. japonica (UL) on melanogenesis. Cells were cultured in the presence of various concentrations of Ulmus davidiana var. japonica For 48 h, and there were estimated total melanin contents as a final product and activity tyrosinase, a key enzyme, in melanogenesis. Among the four solvent extracts tested, EtOAc extract mostly increased tyrosinase activity And EtOAc extract increased the melanin contents and tyrosinase activity in a dose-dependent manner, Especially it was observed that 100mcg/ml/ EtOAc extract promotes melanin secretion in B16/F10 melanoma cells by 140% at 48 h treatment and activity of increased by 180% in the presence of same concentration. In conclusion, as for EtOAc extract treatment, there was no effect on the viability of B16/F10 cell, only to stimulate melanization.

        해녀콩(Canavalia lineata THUNB. DC.) 추출물의 멜라닌 생성 억제 효과

        피부에서 melanin은 자외선 차단의 주요한 역할을 한다. Tyrosinase는 멜라닌 생합성과정에서 초기 단계에 관여하는 중요한 효소로서 이것의 조절을 통한 피부 멜라닌화 억제에 관해 많은 연구가 되어져왔다. 본 연구에서는 해녀콩 추출물에서 mushroom tyrosinase 활성억제, B16F10 melanoma 세포를 이용한 dopa oxidase 활성억제 및 멜라닌 합성 억제 효과를 확인하였다. Tyrosinase mRNA 발현에서의 억제 효과를 확인하기 위하여 RT-PCR을 이용하였으며, CHCI<sub>3</sub> 층에서 분리해낸 A 분획에서 tyrosinase mRNA 발현을 억제시킴을 확인하였다. Melanin pigmentation in human skin is a major defensive mechanism against ultraviolet light of the sun. Tyrosinase plays a key role in the biosynthesis of melanin. This is why many researches have been focused on regulations in controlling the epidermal melanization. We found that extract of Canavalia lineata inhibits mushroom tyrosinase activity, dopa oxidase activity, and melanin synthesis in B16F10 melanoma cells. To elucidate mRNA level reverse transcription polymerase chain reaction (RT-PCR) technique was used. It was revealed that A subfraction of CHCI<sub>3</sub> extract of Canavalia lineara reduced the tyrosinase mRNA expression of B16F10 melanoma cells by reverse transcription polymerase chain reaction (RT-PCR) technique.

      • 淫羊藿이 Tyrosinase 酵素活性및 Melanin 生成에 미치는 影響

        With a view to investigating how Epimedium koreanum Nakai affects formation of melanin content in skin, we have measured change in the number of cell, melanin content, and tyrosinase activity by treating its aqueous extracts at various concentrations with B16 melanoma cells, the result of which is as follows: Firstly, cell viability has been shown as over 95% when treating with the aqueous extract of Epimedium koreanum Nakai up to 200 ug/mL, from which the effect of cell toxicology has not nearly appeared. Secondly, the number of cell has been slightly decreased than that of the control group when treating with the aqueous extract of Epimedium koreanum Nakai, which has not shown a statistical noteworthiness. Thirdly, the aqueous extract of Epimedium koreanum Nakai has increased in tyrosinase activity. Fourthly, the aqueous extract of Epimedium koreanum Nakai has promoted the formation of melanin content, its final product. According to the result, it is confirmed that Epimedium koreanum Nakai has the influence on promoting melanization of B16 melanoma cell, and it is considered that Epimedium koreanum Nakai can be applicable for curing vitiligo caused by decrease or loss of pigment through a systematic study.

        蘇木의 에틸아세테이트 추출물이 B16/F10 흑색종세포의 멜라닌 합성에 미치는 효과

        Caesalpinia sappan L. has been commonly used as analgesic, a cure for contusion and sprain as well as a remedy for thrombosis in the oriental medicine. The main constituent of Caesalpinia sappan L. is brazilin having a flavonoid structure. Melanin is a main pigment found in skin, hair and eyes, and melanin synthesis is controlled by a cascade of enzymatic reactions regulated at the level of tyrosinase. The aim of this study was to examine the effect of Caesalpinia sappan L. on proliferation and melanogenesis of B16/F10 melanoma cells. After 48h treatment of cells with various concentrations of EtOAc extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. Therefore, the growth retardation may be due to the cell arrest, not due to cytotoxicity. And there were estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. EtOAc extract increased the melanin contents and tyrosinase activity in low concentration (0.01-10㎍/ml), whereas it decreased those in high concentration. In conclusion, it was observed that EtOAc extract of Caesalpinia sappan L. differently regulates melanization of cells dependent of its concentrations.

        甘草水抽出物이 HM3KO 細胞의 멜라닌 생성에 미치는 영향

        This study was conducted to evaluate the effects of Glycyrrhizae Radix water extract, known as depigmenting agent, on melanin biosynthesis in the HM3KO human melanoma cells. The inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis was identified by mushroom tyrosinase assay in vitro. To determine whether Glycyrrhizae Radix water extract suppress melanin synthesis in cellular level, HM3KO cells were cultured in the presence of different concentrations of Glycyrrhizae Radix water extract and the effects on cell proliferation, melanin contents and tyrosinase activity were examined after 3 days. Treatment with Glycyrrhizae Radix at various concentrations did not exhibit any change of cell viability, and increased the cell proliferation. And the water extract of Glycyrrhizae Radix inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. These results suggest that the inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis is due to the suppression of tyrosinase in HM3KO cells.

        미소안면침의 얼굴 축소 및 피부상태 개선 효과

        Objectives : The purpose of this study is to evaluate the effects of Miso facial acupuncture on facial reduction and improvement of skin condition. Methods : From March to May 2012, 20 women in their twenties to forties with no facial diseases were recruited. Miso facial acupuncture was performed on the both side of their face twice a week, total seven times. The 3D face photographs of each participant were checked 5 times ; before treatment, after 1st treatment, 4th treatment, 7th treatment and 10 days from the last treatment using RS-400FL(Morpheus Co. Korea) and REAL FACE. The surface distances of the 4 lines on the face were measured using Renai MEF program. The improvement of skin condition was evaluated by water content, trans epidermal water loss(TEWL), erythema index and melanin index using MPA 5. They were checked 4 times ; before treatment, 4th treatment, 7th treatment and after 10 days from the last treatment. We analyzed data using Paired t-test(p<0.05) and Wilcoxon signed rank test(p<0.05). Results : One case was dropped out due to pain and edema after 4th treatment. The surface distances of the 4 lines on face were significantly reduced and maintained after 10 days from last treatment. Water content was significantly increased and maintained after 10 days from last treatment. TEWL was significantly decreased. Erythema index was significantly decreased from the 3rd evaluations. Melanin index was slight decreased with no significance at the 2nd evaluations. Conclusions : These results show that Miso facial acupuncture can reduce sizes of the face and improve skin condition such as water content, TEWL and erythema index except melanin index.

        얌빈 추출물이 함유된 마스크 팩의 피부개선효과

        Purpose: We had previously demonstrated that yam bean (Pachyrhizus erosus) extracts have anti-oxidant and anti-melanogenic effects. Accordingly, the present study explored skin improvements resulting from the use of a mask pack containing yam bean extracts. Methods: Subjects included 15 women in their 20s who met the selection and exclusion criteria. Subjects applied the mask pack three times a week for 5 weeks. To determine the effects of the mask pack on the skin, we measured oil, moisture, and melanin content as well as erythema of the facial skin once a week for 5 weeks (i.e., a total of five measurements). Results: After using the mask pack for 5 weeks, the moisture and oil content of the facial skin increased. In addition, the erythema and melanin content significantly decreased. Furthermore, the subjects were highly satisfied with the improvement in skin conditions such as dryness and skin tone. Conclusion: Our results indicate that a mask pack containing yam bean extracts has beneficial effects on facial skin, and it should be considered for use in new functional cosmetics. 목적: 이전 연구에서 우리는 얌빈 추출물이 항산화 효과와 항 멜라닌 생성 효과가 있음을 보고하였다. 따라서 본 논문의 목적은 얌빈 추출물이 함유된 마스크 팩의 피부개선 효과를 알아보기 위한 것이다. 방법: 연구대상자는 선별 기준을 충족한 20대 여성을 대상으로 하였다. 15명의 연구대상자들은 얌빈 추출물 함유 마스크 팩을 5주간 주 3회 10 min 이상 부착하였다. 그 후 피부의 유수분량, 홍반량, 멜라닌량, 주관적인 피부상태개선 여부를 조사하였다. 결과: 얌빈 추출물이 함유된 마스크 팩을 5주간 사용한 결과, 유수분 함량이 대체적으로 증가되었고, 특히 홍반량, 멜라닌량은 유의하게 개선된 것을 확인하였다. 설문조사를 통한 주관적인 피부상태 또한 건조함을 느끼는 증상과 색소침착에 관한 항목에서 높은 만족도를 나타내었다. 결론: 본 연구는 얌빈 추출물이 함유된마스크 팩의 사용이 피부의 유수분량 및 홍반량, 멜라닌량을 개선한다고 판단되며, 얌빈 추출물이 임상적으로도 우수한 미용 소재임을 시사한다.

      • 연교 메탄올추출물의 멜라닌생성 억제효과

        The aim of this study was to investigate the effect of Fructus Forsythiae on the melanogenesis. To determine whether Fructus Forsythiae methanol extract suppress melanin synthesis in cellular level, HM3KO human melanoma cells were cultured in the presence of various concentrations of Fructus Forsythiae methanol extract and the effects on cell proliferation, tyrosinase activity and melanin contents were examined. Treatment with Fructus Forsythiae methanol extract inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis, in a dose-dependent manner. And also suppressed melanin contents as a dose dependent manner without cytotoxicity morphological change. It was observed that the color of cell pellets was totally different from the control. These results suggest that the inhibitory effect of Fructus Forsythiae methanol extract on melanogenesis is due to the suppression of tyrosinase in HM3KO cells and Fructus Forsythiae is a candidate for an efficient whitening agent.

        Inhibition of Melanin Production and Tyrosinase Expression of Crocetin Derivatives from processed Gardenia jasminoides

        The crocetin derivatives, crocin (1), gentiobiosyl glucosyl crocetin (3), and mono-gentiobiosyl crocetin (4) were isolated from the fruit of Gardenia jasminoides (Gj) and crocetin (2) from the processed fruit of Gj (PGj) by column chromatography. Their structures were determined on the basis of spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were evaluated for their inhibition activity on melanin production in a-MSH (melanocyte stimulating hormone) activated B16F10 cells. Compounds 1 - 4 reduced melanin content in a dose-dependent manner at concentrations of 20 - 60 uM. They also suppressed tyrosinase protein and m-RNA expressions dose dependently, assayed by western blot analysis, and RT-PCR experiment in B16F10 murine melanoma cells.
