Optimization conditions for cryopreservation of Nelumbo nucifera Gaertn., aquatic plant

This study was conducted to find the optimal cryopreservation conditions for efficiently preserving the genetic resources of the aquatic plant lotus (Nelumbo nucifera Gaertn). The seed viability differed depending on the type of vitrification treatment agent, and the longer the PVS2 treatment time, the lower the viability. Among the treatments, T1 (2M glycerol) and T7 (Loading solution + PVS2) were the treatments that showed viability rates similar to those of the control (92.1 ± 5.95%). When the seeds were stored for 12 weeks, the viability rate of the control dropped to 0%, but the treatments T1 and T7 were maintained unchanged. The change in the germination rate of seeds according to the storage period, the control was 0% after 4 weeks. In the case of the treatments, T1 showed a high germination rate in the 1 day (86.67 ± 6.67%), but after 12 weeks, a lower germination rate (54.67 ± 5.58%). On the other hand, T7 had a slightly lower initial germination rate of 73.33%, but it was maintained even after a prolonged storage period. The shoot growth of the cryopreserved sprouted seedlings was lower than that of the control, and there was no morphological difference, as T7 was the best among the treatments. As a result of RAPD analysis using seedling primers, no genetic variation was found. The above results are expected to be widely used not only for treatment but also for the conservation of genetic resources of other aquatic plants.
Effects of Nelumbo Nucifera Gaertn on Neuropathic Pain in Rats

Neuropathic pam is a disturbance resulted from injury to a nerve or particUlar group of nerves. Neuropathic pain is often reported as having a lancinating or continuous burning character and is often associated with the appearance of abnormal sensory signs, such as allodynia or hyperalgesia. The purpose of the present study was to investigate pain relieving effects of Nelumbo nucifera Gaertn (KHA24) on a rat model of neuropathic pain. Adult male Sprague-Dawley rats were used. Neuropathic pain model was prepared according to the method described by Lee et al. Animals were devided into three groups according to each three dose of Nelumbo nucifera Gaertn and control. The Nelumbo nucifera Gaertn were injected intraperitoneally. A behavioral test for mechanical allodynia were performed using von Frey filament (8mN) for two weeks postoperatively. To quantify cold sensitivity, acetone was applied to the plantar surface of the foot 5 times on each hind paw. After the injectionof Nelumbo nucifera Gaertn, mechanical allodynia and cold allodynia were reduced. These results indicate that Nelumbo nucifera Gaertnmay provide a potent strategy in relieving neuropathic pain.

This study was undertaken to optimize preparation of sweet rice muffins using lotus (Nelumbo nucifera Gaertn) seed powder. The experimental conditions used in the preparation included lotus seed powder (X1) and soybean oil (X2). The muffin formulation was optimized using rheology. Lightness (P<0.01) displayed a quadratic model pattern, whereas yellowness (P<0.01) and redness (P<0.05) was presented a linear model. Texture properties significantly differed in cohesiveness (P<0.05). Evaluating the sensory characteristics of muffins, except for flavor and texture, the remaining properties of color (P<0.01), taste, overall quality, and appearance (P<0.05) were found to be significantly different. The overall quality was also affected by the proportion of lotus seed powder, rather than the amount of soybean oil. The optimal ratio for palatability of muffins was determined to be 47.80 g lotus seed powder and 95.28 g soybean oil. Taken together, our results indicate that sweet rice muffins prepared using lotus seed powder are sufficiently competitive in terms of function and quality.

The nutritional composition of the white lotus(Nelumbo nucifera Gaertn) leaf are moisture(1.87±0.06%), crude ash (9.57±0.07%), crude protein(15.53±0.4%) and crude lipid(2.42±0.1%) per 100g. The highest mineral content was K and Ca. The principal free sugars in the white lotus leaf was sucrose, fructose and glucose. The total phenolics compounds evidenced maximal levels of 8,842.73±20.57 g/100g. DPPH and hydroxyl radical of scavenging ability of each sample tended to increase with increasing degrees of the sample concentration. The IC50 values of the ethanol extract, n-hexane, chloroform, ethylacetate, butanol and water fraction from the white lotus leaf to DPPH radicals were 5.65, 8.5, 8.27, 2.03, 2.3 and 13.7 ㎍/mL respectively. The IC50 values of the hydroxyl radical scavenging ability of the ethanol extract, ethylacetate, butanol and water fraction were 390, 574, 327 and 378 ㎍/mL respectively.
국산 한약재로 이용되는 약용식물의 NCI-H1229 인간 폐암 세포주에 대한 성장 억제효과 분석

Lung cancer, the most common malignant disease worldwide, is the predominant cause of cancer deaths, particularly amongst men. Therefore, various researchers have focused on the growth inhibitory effects of medicinal plants used in traditional Korean medicine. This study aimed to investigate the growth inhibitory effects of ethanol extracts of Rubiae radix, Inulae flos, Nelumbinis receptaculum, Astilbe radix, and Lagerstroemia flos on NCI-H1229 cells. Method and Results: The viability of NCI-H1229 cells was evaluated in vitro using an MTS assay. Treatment with the ethanol extracts of the selected medicinal plants at 500 ㎍/㎖ reduced NCI-H1229 cell viability and increased apoptotic cell death and caspase-3 activation. In addition, treatment with ethanol extracts of Inulae flos and Astilbe radix increases DNA fragmentation, as measured by the TUNEL assay. Conclusions: These results indicated that ethanol extracts of Rubiae radix, Inulae flos, Nelumbinis receptaculum, Astilbe radix, and Lagerstroemia flos exhibited growth inhibitory effects, inducing apoptotic cell death, DNA fragmentation and caspase-3 activation in NCI-H1229 cells. Therefore, these medicinal plant extracts may be used in the development of natural medicines to inhibit the growth of lung cancers. However, further study is needed to determine the active ingredients of the ethanol extracts from medicinal plants that are reposible for the inhibitory effect on lung cancer cell grwoth.
