        감정노동과 감정부조화가 노조몰입에 미치는 영향: 직무소진의 매개효과를 중심으로

        [연구목적] 본 연구는 조직구성원의 감정노동 및 감정부조화에 주목하고 이로 인해 발생하는 직무소진과 노조몰입에 미치는 영향을 실증적으로 검증하기 위한 것이다. [연구방법] 이러한 목적을 달성하기 위해 선행연구의 문헌을 고찰하고 일곱 개의 연구가설을 설정하였다. U시와 B시에 소재하고 있는 대학병원 종사자를 대상으로 총 200부의 설문지를 배포하여 최종적으로 187부의 설문자료를 수집하였으며, 수집된 자료에 대해 신뢰성과 타당성을 검토하였다. 또한, 상관관계분석, 다중회귀분석, 그리고 매개회귀분석을 통해 가설을 검증하였다. [연구결과] 감정노동 및 감정부조화가 직무소진의 하위요소 중 감정고갈과 탈 인격화에 유의한 정(+)적 영향을 미치는 것으로 나타났으며, 감정노동과 감정부조화가 노조몰입에 유의한 정(+)적 영향을 미치는 것으로 나타났다. 또한, 직무소진의 하위요소 중 감정고갈이 노조몰입에 대해 유의한 정(+)적 영향을 미치는 것으로 나타났다. 마지막으로 감정노동이 노조몰입에 영향을 미치는데 있어서 직무소진의 하위요소 중 감정고갈이 부분매개역할을, 감정부조화가 노조몰입에 영향을 미치는데 있어서 감정고갈이 완전매개역할을 하고 있음을 확인하였다. [연구의 시사점] 본 연구를 통해 대학병원 종사자가 직무수행과정에서 겪게 되는 감정노동과 감정부조화의 결과로 직무소진과 노조몰입이 발생함을 확인할 수 있었다. [Purpose] Focusing upon emotional labor and emotional dissonance of organization’s members, this study empirically examined their effects on job burnout and union commitment. [Methodology] For this purpose, we reviewed previous literature and we developed seven hypotheses. To verify these hypotheses, data were collected through questionnaire survey from workers of university hospitals in U city and B city. Data from 187 respondents were tested by reliability, validity, correlation, regression, and mediated regression analysis. [Findings] This study found that emotional labor and emotional dissonance is positively related to job burnout(emotional exhaustion and depersonalization) and union commitment. Also, it is found that job burnout(emotional exhaustion) is positively related to union commitment. The results showed that job burnout(emotional exhaustion) mediated partially the relationship between emotional labor and union commitment and job burnout(emotional exhaustion) mediated fully the relationship between emotional dissonance and union commitment. [Implications] This study empirically assessed job burnout and union commitment as some of main consequences of workers’ emotional labor and dissonance at university hospitals.

        Immobilization of Recombinant Bacterial Biosensors

        본 연구에서는 페놀 화합물들을 현장에서 검출하기 위해 간단하고 간편한 일회용 재조합 박테리아 바이오센서 시스템을 개발하였다. 플라즈미드 pLZCapR을 함유하는 E. coli 세포를 β-galactosidase 기질인 CPRG와 함께 96-well plate. wells. agarose로 고정하였다. 이 바이오센서는 현장에서 발색을 위한 별도의 기질을 추가하거나 불편한 기기 사용 또는 시료의 전 처리를 필요로 하지 않는다. 시료의 측정은 간단히 적은 부피(<100 μl)의 현장 시료를 바이오센서를 포함하는 wells에 넣고 발색을 관찰하여 측정하였다. 또한 6% DMF, 0.1% SDS 그리고 10mM CaCl2를 첨가하여 agarose 고정에 의한 화합물의 세포내 확산 제한을 감소시켜 보다 더 나은 발색을 얻을 수 있었다. 따라서 이 고정된 미생물 유래 재조합 바이오센서 시스템은 현장에서 환경오염물질들을 간단하게 확인하고 정량 하는 유용한 접근 방법이 될 것으로 사료된다. We herein report the development of an agarose-gel-immobilized recombinant bacterial biosensor simple system for the field monitoring of phenolic compounds. Escherichia coli cells harboring the pLZCapR plasmid, which was previously designed to express the β-galactosidase reporter gene in the presence of phenolic compounds, were co-immobilized with a substrate [chlorophenol red β-galactopyranoside (CPRG) in agarose gel, and dispensed to the wells of a 96-well plate. Field samples were added to the wells and color development was monitored. In the presence of 5 μM to 10 mM of phenol, the biosensor developed a red (representing hydrolysis of CPRG) color. Other phenolic compounds were also detected by this immobilized system, with the pattern resembling that previously reported for the corresponding non-immobilized biosensor. The immobilized cells showed optimum activity when the gel was simultaneously supplemented with 6% dimethyl formamide (DMF), 0.1% SDS and 10 mM CaCl2. The immobilized biosensor described herein does not require the addition of a substrate or the use of unwieldy instruments or sample pretreatments that could complicate field studies.

        Case Report : Severe ischemic bowel necrosis caused by terlipressin during treatment of hepatorenal syndrome

        Terlipressin is a vasopressin analogue that is widely used in the treatment of hepatorenal syndrome or variceal bleeding. Because it acts mainly on splanchnic vessels, terlipressin has a lower incidence of severe ischemic complications than does vasopressin. However, it can still lead to serious complications such as myocardial infarction, skin necrosis, or bowel ischemia. Herein we report a case of severe ischemic bowel necrosis in a 46-year-old cirrhotic patient treated with terlipressin. Although the patient received bowel resection, death occurred due to ongoing hypotension and metabolic acidosis. Attention should be paid to patients complaining of abdominal pain during treatment with terlipressin.

        Minor Coat Protein pIII Domain (N1N2) of Bacteriophage CTXф Confers a Novel Surface Plasmon Resonance Biosensor for Rapid Detection of Vibrio cholerae

        Bacteriophages are considered excellent sensing elements for platforms detecting bacteria. However, their lytic cycle has restricted their efficacy. Here, we used the minor coat protein pIII domain (N1N2) of phage CTXφ to construct a novel surface plasmon resonance (SPR) biosensor that could detect Vibrio cholerae. N1N2 harboring the domains required for phage adsorption and entry was obtained from Escherichia coli using recombinant protein expression and purification. SDS-PAGE revealed an approximate size of 30 kDa for N1N2. Dot blot and transmission electron microscopy analyses revealed that the protein bound to the host V. cholerae but not to non-host E. coli K-12 cells. Next, we used amine-coupling to develop a novel recombinant N1N2 (rN1N2)-functionalized SPR biosensor by immobilizing rN1N2 proteins on gold substrates and using SPR to monitor the binding kinetics of the proteins with target bacteria. We observed rapid detection of V. cholerae in the range of approximately 10<sup>3</sup> to 10<sup>9</sup> CFU/ml but not of E. coli at any tested concentration, thereby confirming that the biosensor exhibited differential recognition and binding. The results indicate that the novel biosensor can rapidly monitor a target pathogenic microorganism in the environment and is very useful for monitoring food safety and facilitating early disease prevention.

      • A Study on the Effects of Mobile-based LMS on Flipped Learning: Focused on the Affective Pathway in Pre-service Teacher Education

        Flipped learning is a type of blended learning that has recently emerged as an innovative teaching and learning method. In this study, a mobile-based learning management system (LMS) is developed in order to support flipped learning for mobile-friendly learners. The purpose of the study is to investigate the effects of flipped learning focused on a problem-solving approach on the affective pathway. In addition, the study examines the ways in which mobile-based LMS facilitates flipped learning compared to the PC-based Web environment. A series of instructional design of flipped learning for a math class was developed and implemented for undergraduate students at C University in South Korea. The participating students were asked to write self-reflection journals for 6 weeks (12 periods). In-depth interviews were conducted with the students and the contents of the discussion board in the mobile-based LMS were analyzed using Nvivo, in order to track a series of changes of the affective pathway. The results obtained from the qualitative data revealed that students gained high self-esteem, motivation, and interest regarding academic activities, which eventually cultivated a positive local affect. Furthermore, there was convincing evidence of changes from the “flipped classroom” to “flipped learning”, where students were able to take advantage of the mobile-based LMS. Compared to the PC-based Web environment, students were able to overcome the limitation of time and space, using the new environment proactively and creatively.

      • The Impacts of Adding Nutritional Supplement in Permanent Hair Colorants on Hair Dyeing

        In this study, the hair damage mitigation effects when applying the Bombyx mori powder, Bombyx mori oil, which are the natural extracts, commercialized argan oil, LPP, and sodium glutamate to the damaged hair in the alkaline dyeing procedure, which is the chemical procedure, were observed and following results were drawn. In the results of observing the cuticle, it was observed that adding the Bombyx mori powder and the Bombyx mori oil, which contain abundant protein, was effective, and the experiment group, which represented the similar brightness to the control group was the group applied with the Bombyx mori oil. In the tensile strength measurement, which the degree of hair damage can be observed, the argan oil, which is the natural vegetable oil, represented the highest tensile strength, and the sodium glutamate, which is the artificial flavor, represented the lowest tensile strength. In the results of observing the amino acid composition change, the adding the Bombyx mori powder and Bombyx mori oil, which contain rich protein, and the sodium glutamate, which is composed of 95% of amino acid, represented the highest amino acid composition value. As a result, the Bombyx mori powder and the Bombyx mori oil, which are the natural extracts, is deemed to be the effective application to be able to reduce the damage not by the pre-treatment nor post-treatment but by adding in the hair colorants in the damaged hair dyeing procedure.

        A Rat Liver Lysosomal Membrane Flavin - Adenine Dinucleotide Phosphohydrolase

        An enzyme that hydrolyzes flavin-adenine dinucleotide (FAD) was found to be present in rat liver lysosomal membrane prepared from Triton WR-1339 filled lysosomes (tritosomes) purified by flotation on sucrose. This FAD phosphohydrolase (FADasej exhibited optimal activity at pH 8.5 and had an apparent Km of approximately 3.3 mM. The activity was decreased 50^-70% by dialysis against EDTA and this was restored by Zn^(2+). Mg^(+2), Hg^(+2), and Ca^(+2) ions inhibited the enzyme, but F and molybdate had no effect. The enzyme was also inhibited by p-chloromercuribenzoate (pCMB), reduced glutathione and other thiols, cyanide, and ascorbate. The presence of ATP, ADP, AMP. α-β-methylene ATP, AMP-p-nitrophenyl phosphate (PNP), GMP, and coenzyme A (CoA) decreased the activity on FAD, but pyrimidine nucleotides, adenosine, adenine, or NAD^+ were without effect. Phosphate stimulated the activity slightly. FAD phosphohydrolase activity was separated from ATPase and inorganic pyrophosphatase activities by solubilization with detergents and polyacrylamide gel electrophoresis and by linear sucrose density gradient centrifugation suggesting that the enzyme is different from ATPase, inorganic pyrophosphatase, and soluble lysosomal FAD pyrophosphatase. Paper chromatography showed that FAD was hydrolyzed to flavin mononucleotide (FMN) and AMP which were further hydrolyzed to riboflavin and AMP by phosphatases known to be present in lysosomal membranes. Incubation of the intact lysosomes with pronase showed that the active site of FAD phosphohydrolase must be oriented to the cytosol. The FAD hydrolyzing activity was detected in Golgi, microsome, and plasma membrane, but not in mitochondria or soluble lysosomal preparations.

      • Incidental Findings on Brain Magnetic Resonance Imaging in Children with Central Precocious Puberty

        Objectives: To investigate brain magnetic resonance imaging (MRI) findings in patients with central precocious puberty (CPP) by age at onset and sex. Methods: We included 130 CPP patients with brain MRI findings of the pituitary gland treated at Ewha Womans University Mokdong Hospital between February 2007 and October 2013 and divided them by age and sex: boys, girls aged ≤6 years, and girls aged>6 years. The control group comprised 224 patients who underwent brain MRIs, and we compared their incidental brain findings with those of the CPP group. Results: In the CPP subgroups who underwent pituitary MRIs, the frequency of incidental brain lesions was 31.6% in boys, 47.1% in girls ≤6 years and 29.8% in girls >6 years. The incidence of pituitary abnormalities was 42.1% in boys, 64.7% in girls ≤6 years and 47.9% in girls >6 years. Among pituitary abnormalities, pituitary hypoplasia had a significantly higher incidence rate in girls ≤6 years (41.2%) than in boys (15.8%) or girls >6 years (13.8%, P=0.027). Hypothalamic hamartomas were detected in one girl aged ≤6 years and in one boy, but not in girls aged 6 years (P=0.075). The incidence of pineal cysts was higher in the CPP groups and significantly higher in girls ≤6 years (47.1%) than in the control group (11.2%, P=0.001). Conclusion: There was a higher incidence of brain abnormalities on pituitary MRIs and a higher incidence of pineal cysts, possibly associated with CPP pathogenesis, in younger CPP patients than in other patients.
