        Up‑regulation of miR‑27 extenuates lipopolysaccharide‑induced injury in H9c2 cells via modulating ICAM1 expression

        Background MiR-27 has been found to present an overt myocardial expression during cardiogenesis. However, whether miR-27 involves in myocarditis development and the possible molecular mechanism remain unknown. The purpose of this study was to investigate the biological characteristic of miR-27 in LPS-damaged H9c2 cells. Methods H9c2 cells were treated with lipopolysaccharide (LPS, 10 μg/ml) for 12 h to form cell injury. MiR-27 mimic and inhibitor were used to up-regulate or down-regulate miR-27 expression. MTT assay and flow cytometry analysis were conducted to test cell viability and apoptosis. The relative RNA expression level of miR-27 and intercellular adhesion molecule 1 (ICAM1) was determined by qRT-PCR. Luciferase reporter gene assay was utilized to confirm the interaction between miR-27 and ICAM1. Western blot was used to determine the protein expression levels. Results We observed that LPS treatment significantly decreased the level of miR-27 in H9c2 cells. Moreover, LPS exposure suppressed cell viability, promoted cell apoptosis and increased the relative expression of p-NF-κB p65/NF-κB p65 and p-IκBα/IκBα. Up-regulation of miR-27 increased cell proliferation and reduced cell apoptosis, while down-regulation of miR-27 suppressed cell growth and promoted cell apoptosis. ICAM1 was predicted and verified as a target of miR-27, and the expression of ICAM1 is negatively regulated by miR-27. The relative expression of p-NF-κB p65/NF-κB p65 and p-IκBα/ IκBα was dramatically decreased by miR-27 mimic and increased by miR-27 inhibitor. Conclusion Our study illustrated that up-regulation of miR-27 exhibits a protective effect on LPS-damaged H9c2 cells, which may be achieved by regulating ICAM1 and NF-κB signaling.

        A method of X-ray source spectrum estimation from transmission measurements based on compressed sensing

        A new method of X-ray source spectrum estimation based on compressed sensing is proposed in this paper. The algorithm K-SVD is applied for sparse representation. Nonnegative constraints are added by modifying the L1 reconstruction algorithm proposed by Rosset and Zhu. The estimation method is demonstrated on simulated spectra typical of mammography and CT. X-ray spectra are simulated with the Monte Carlo code Geant4. The proposed method is successfully applied to highly ill conditioned and under determined estimation problems with a good performance of suppressing noises. Results with acceptable accuracies (MSE < 5%) can be obtained with 10% Gaussian white noises added to the simulated experimental data. The biggest difference between the proposed method and the existing methods is that multiple prior knowledge of X-ray spectra can be included in one dictionary, which is meaningful for obtaining the true X-ray spectrum from the measurements.

        “Going Native”: The New Imperialism in Late Victorian Adventure Novels She and Heart of Darkness

        The late Victorian era witnessed the flowering of New Imperialism in literary circles. Critics like Edward W. Said and Homi K. Bhabha single out English novels, especially English adventure fiction, as a unique cultural form to explore the imperial attitudes and experiences during the period. Among late Victorian works of fiction, H. Rider Haggard’s She and Joseph Conrad’s Heart of Darkness are prominent that they both endeavor to bring forth white men’s bodily and spiritual “encounters” on their imperial journey to Africa. Current scholarship on the two texts has tended to simplify them either as a critique or approval of New Imperialism and thus categorizes the writers as pro- or anti-imperialists. However, the dissemination of the ideology in popular literature at the end of the nineteenth century was not stereotyped and homogeneous. Therefore, this study traces the similarities and differences in depicting New Imperialism through the lens of “going native.” The paper will investigate the embodiment of the idea in individual texts and inspect the two adventure fictions together within the discourse of New Imperialism. Conventional textual interpretations and computerized analysis of texts are to be conducted.

        MiR-188-5p regulates the proliferation and differentiation of goat skeletal muscle satellite cells by targeting calcium/calmodulin dependent protein kinase II beta

        Objective: The aim of this study was to reveal the role and regulatory mechanism of miR-188-5p in the proliferation and differentiation of goat muscle satellite cells. Methods: Goat skeletal muscle satellite cells isolated in the pre-laboratory were used as the test material. First, the expression of miR-188-5p in goat muscle tissues at different developmental stages was detected by quantitative reverse transcription polymerase chain reaction (qRT-PCR). In addition, miR-188-5p was transfected into goat skeletal muscle satellite cells by constructing mimics and inhibitors of miR-188-5p, respectively. The changes of differentiation marker gene expression were detected by qPCR method. Results: It was highly expressed in adult goat latissimus dorsi and leg muscles, goat fetal skeletal muscle, and at the differentiation stage of muscle satellite cells. Overexpression and interference of miR-188-5p showed that miR-188-5p inhibited the proliferation and promoted the differentiation of goat muscle satellite cells. Target gene prediction and dual luciferase assays showed that miR-188-5p could target the 3'untranslated region of the calcium/calmodulin dependent protein kinase II beta (CAMK2B) gene and inhibit luciferase activity. Further functional studies revealed that CAMK2B promoted the proliferation and inhibited the differentiation of goat muscle satellite cells, whereas si-CAMK2B restored the function of miR-188-5p inhibitor. Conclusion: These results suggest that miR-188-5p inhibits the proliferation and promotes the differentiation of goat muscle satellite cells by targeting CAMK2B. This study will provide a theoretical reference for future studies on the molecular mechanisms of skeletal muscle development in goats.

        Isolation, Structural Characterization, and Lymphopoiesis Stimulant Activity of a Polysaccharide from the Abalone Gonad

        A novel polysaccharide (AGP-32) from the gonad of Haliotis discus hannai Ino was isolated using a protease-assisted process and successive ion-exchange and gel-filtration chromatography. The backbone of AGP-32 was determined using hydrolysis with trifluoroacetic acid. FTIR, NMR, and methylation analysis, and periodate oxidation and Smith degradation analysis revealed that the AGP-32 backbone mainly consisted of (1→6)-linked mannose, (1→3)-linked galactose, and (1→3)-linked glucose in a proportion of 2:3:1. An in vitro cell assay indicated that AGP-32 promoted mice splenic lymphocyte proliferation by 26% at a concentration of 50 μg/mL. AGP-32 had an effect on immune protection and is a candidate for consideration as a functional food.

        Physiological oxygen level is critical for modeling neuronal metabolism in vitro

        <P><B>Abstract</B></P><P>In vitro models are important tools for studying the mechanisms that govern neuronal responses to injury. Most neuronal culture methods employ nonphysiological conditions with regard to metabolic parameters. Standard neuronal cell culture is performed at ambient (21%) oxygen levels, whereas actual tissue oxygen levels in the mammalian brain range from 1% to 5%. In this study, we examined the consequences of oxygen level on the viability and metabolism of primary cultures of cortical neurons. Our results indicate that physiological oxygen level (5% O<SUB>2</SUB>) has a beneficial effect on cortical neuronal survival and mitochondrial function in vitro. Moreover, oxygen level affects metabolic fluxes: glucose uptake and glycolysis was enhanced at physiological oxygen level, whereas glucose oxidation and fatty acid oxidation were reduced. Adenosine monophosphate‐activated protein kinase (AMPK) was more activated in 5% O<SUB>2</SUB> and appears to play a role in these metabolic effects. Inhibiting AMPK activity with compound C decreased glucose uptake, intracellular ATP level, and viability in neurons cultured in 5% O<SUB>2</SUB>. These data indicate that oxygen level is an important parameter to consider when modeling neuronal responses to stress in vitro. © 2011 Wiley Periodicals, Inc.</P>

        Monstrosity and Contagion: Response to Non-Human Beings in Frankenstein and Alien (1979)

        This paper deals with the dreadful non-human characters in literary and cinematic works Frankenstein and Alien (1979) respectively. By observing the Frankenstein monster, the cyborg and the xenomorph in the two literary outputs from the perspective of post-humanism, the current study aims to rethink the relations between human beings and their non-human counterparts. How does humankind respond to non-human beings? What research has been done on the sapient creature, the man-made robot, and the alien species historically? What are the meanings of monstrosity and contagion? And how do the two concepts function in the novel and the film? All these questions are to be addressed in the paper. The article expects to take a post-humanistic view on the two notions and to provide references for the future post-human studies. It concludes that in the two works, the monstrous and infective nature of the inhuman others that prompts human beings not to embrace their non-human counterparts reflects the same feature of humans themselves and proves the vulnerability of human-centered humanity.

        Divine or Devilish: Power behind “Mother” in the Sci-fi Horror Alien

        The current study re-examines the science-fiction horror Alien (1979) by locating and analyzing the power hidden behind the computer mainframe named “Mother”. In the movie, six human crew were dispatched to outer space on an installer cruise Nostromo with the computer “Mother” and the cyborg Ash. It was not until the moment when they were slaughtered one after another by an apparently omnipotent alien creature that the remaining human group realized that the ultimate purpose of the space trip was to search for the unknown species. Traditionally, the creepy-looking xenomorph is viewed as a demon, whereas the chamber of “Mother” is regarded as a place with temporary peace and divine. However, the study claims that the seemingly sacred mission of the company Weyland Yutani is no less devilish in nature than the destructive behavior of the alien species. The first stage will provide some background information of the film and study. In the second stage, I will analyze the design and operational system of the spacecraft. The third stage will discuss the value of the computer “Mother” and the cyborg Ash. In the fourth stage, I attempt to expose the intrinsic quality of the corporation by linking the construction of the spaceship and the arrangement of “Mother” and Ash to the disgraceful action and ideology of the capitalistic firm. And finally, the paper will conclude that the self-flattered divine enterprise capitalizes human staff in feminist and fetishist concepts. This study presents various clues to reinterpret the combination of religious and technological elements in sci-fi horrors. It could also serve as a practice to identify and elucidate the inhuman aspect of capitalism in the contemporary world.

        Isolation and Characterization of a Novel Bacterium Burkholderia gladioli Bsp-1 Producing Alkaline Lipase

        Active lipase-producing bacterium Burkholderia gladioli Bps-1 was rapidly isolated using a modified trypan blue and tetracycline, ampicillin plate. The electro-phoretically pure enzyme was obtained by purification using ethanol precipitation, ion-exchange chromatography, and gel filtration chromatography. The molecular weight was 34.6 kDa and the specific activity was determined to be 443.9 U/mg. The purified lipase showed the highest activity after hydrolysis with p-NPC<sub>16</sub> at a pH of 8.5 and 50°C, and the K<sub>m</sub>, k<sub>cat</sub>, and k<sub>cat</sub>/K<sub>m</sub> values were 1.05 mM, 292.95 s<sup>-1</sup> and 279 s<sup>-1</sup>mM<sup>-1</sup>, respectively. The lipase was highly stable at 7.5 ≤ pH ≤ 10.0. K<sup>+</sup> and Na<sup>+</sup> exerted activation effects on the lipase which had favorable tolerance to short-chain alcohols with its residual enzyme activity being 110% after being maintained in 30% ethanol for 1 h. The results demonstrated that the lipase produced by the strain B. gladioli Bps-1 has high enzyme activity and is an alkaline lipase. The lipase has promising chemical properties for a range of applications in the food-processing and detergent industries, and has particularly high potential for use in the manufacture of biodiesel.
