<P>The effects of intrinsic feature of feedstock in air plasma-sprayed (APS) coatings on the microstructure and mechanical properties of vertical-cracked thermal barrier coatings (TBCs) were investigated in thermal exposure. The microstructure after the thermal exposure for 400 h is densified, while after the thermal exposure for 800 h various defects such as interlamellar cracks and the vertical and horizontal cracks are newly developed, even though the total porosity is decreased. The microstructure of the TBC prepared with 204 C-NS is more porous than that of the TBC with 204NS, showing higher mechanical properties and better thermal stability in the TBC with 204 NS.</P>

<P>The tumor-associated antigen GA733 is a cell-surface glycoprotein highly expressed in colorectal carcinomas. In this study, 3 recombinant genes were constructed as follows: GA733 tagged to the ER retention sequence KDEL (GA733K), GA733 fused to the immunoglobulin Fc fragment (GA733-Fc), and GA733-Fc fused to the ER retention sequence (GA733-FcK). <I>Agrobacterium</I>-mediated transformation was used to generate transgenic plants expressing recombinant genes. The presence of transgenes was confirmed by genomic PCR. Western blot, confocal immunofluorescence, and sandwich ELISA showed the expression of recombinant proteins. The stability, flexibility, and bioactivity of recombinant proteins were analyzed and demonstrated through N-glycosylation analysis, animal trials, and sera ELISA. Our results suggest that the KDEL retained proteins in ER with oligomannose glycan structure and enhanced protein accumulation level. The sera of mice immunized with GA733-FcK purified from plants contained immunoglobulins which were at least as efficient as the mammalian-derived GA733-Fc at recognizing human colorectal cancer cell lines. Thus, a plant system can be used to express the KDEL fusion protein with oligomannose glycosylation, and this protein induces an immune response which is comparable to non-KDEL-tagged, mammalian-derived proteins.</P>

Colorectal cancer is the third leading cause of cancer-related death with 655,000 deaths worldwide per year in the Western world. The tumor-associated antigen GA733 is a glycoprotein highly expressed on colorectal carcinoma cell surface. We have fused GA733 to Fc fragment of human IgG as a vaccine candidate in plant expression system. The fusion protein GA733-Fc can improve their expression in plant cells and induce cellular and humoral host immune responses. GA733-Fc is glycosylated, but the N-glycanstructures of plant-derived GA733-Fc differ from their parental counterparts. Plant-derived specific N-glycoforms can potential to induce immunogenicity and allergenic responses in human therapy. Thus, we have tried to generate oligomannose glycan structure by retaining GA733-Fc and GA733 in ER with KDEL sequence. Agrobacterium-mediated transformation was conducted to generate transgenic plant expressing GA733-Fc and GA733. We have compared protein expression, accumulation and biological activity of GA733-Fc with KDEL, GA733-Fc without KDEL, GA733 with KDEL and GA733 without KDEL in transgenic plants by western blot, confocal immunofluorescence analysis, ELISA and mouse immunization, respectively.

The effects of coating thickness on the delamination and fracture behavior of thermal barrier coating (TBC) systems were investigated with cyclic flame thermal fatigue (FTF) and thermal shock (TS) tests. The top and bond coats of the TBCs were prepared by electron beam-physical vapor deposition and low pressure plasma spray methods, respectively, with a thickness ratio of 2:1 in the top and bond coats. The thicknesses of the top coat were 200 and $500{\mu}m$, and those of the bond coat were 100 and $250{\mu}m$. FTF tests were performed until 1140 cycles at a surface temperature of $1100^{\circ}C$ for a dwell time of 5 min. TS tests were also done until more than 50 % delamination or 1140 cycles with a dwell time of 60 min. After the FTF for 1140 cycles, the interface microstructures of each TBC exhibited a sound condition without cracking or delamination. In the TS, the TBCs of 200 and $500{\mu}m$ were fully delaminated (> 50 %) within 171 and 440 cycles, respectively. These results enabled us to control the thickness of TBC systems and to propose an efficient coating in protecting the substrate in cyclic thermal exposure environments.

Since more elements are involved in the construction projects, the decision making process of Resource-constrained Project Scheduling Problems (RCPSP) cannot be determined by a manager, but multiple level decision makers. This study focuses on the RCPSP in a practical grounding grid system project in Jin’ An Qiao hydropower station, which consider the hierarchical organization structure and hybrid uncertainty environment simultaneously. In this practical RCPSP, the construction contractor is the Upper Level Decision Maker (ULDM), while the outsourcing partner is the Lower Level Decision Maker (LLDM). Considering the operations difficulties and practical decision making process in the RCPSP, a bi-level multi-objective RCPSP model with bi-random coefficients is developed. To deal with the bi-random variables in the model, the Dependent-Chance Programming (DCP) method is introduced and the equivalent crisp model is derived. Subsequently, motivated by the particular nature of model, Bi-level Global-local-neighbor Particle Swarm Optimization Algorithm (BGLN-PSO) is designed to obtain the optimal schedule for grounding grid system project. A practical application is presented to verify the efficiency of proposed bi-level multi-objective RCPSP model and algorithm.
A Large Scale System Model-Order Reduction Method Based on SVD-Krylov

A SVD-Krylov method for large scale MIMO(multi-input multi-output) system model-order reduction is proposed in this paper. Its aim is to combined the singular value decomposition(SVD)and Krylov methods by retaining the best feature that can be applied for solving some problems for the large scale system model-order reduction. The method matches the first r Markov parameters and minimizes the error in the remaining ones in the least-squares sense. The reduced model is asymptotically stable, matches a certain number of moments, and minimizes a weighted error in the discrete time case. The effectiveness of the proposed approaches is tested by the Iss (international space station)model that in the SLICOT library, getting the frequency-response, the error and the error bounds of different order reduced model. The result shows that the proposed method is efficiently.
Improved Performances of AlN/Polyimide Hybrid Film and its Application in Redistribution Layer

The AlN/polyimide (PI) hybrid film was studied as thedielectric layer in the redistribution layer (RDL) in this work. The incorporation of the AlN into the PI matrix was achievedby mechanical ball-milling process. The spin-coating processwas used to fabricate the AlN/PI hybrid film, which iscompatible with micro-electro-mechanical system (MEMS)technology for fabricating RDL. The AlN/PI hybrid film wascharacterized by Fourier transform infrared (FTIR) spectrumand thermogravimetric analysis (TGA). The effect of the AlNcontent on the thermal stability, thermal expansion coefficient,hardness and water adsorption of the AlN/PI hybrid film wasstudied. The results indicated that the addition of AlN nanoparticlesimproved the thermal stability and hardness, butdecreased the thermal expansion coefficient and waterabsorption of the pure PI film. As an example of its typicalapplication, the AlN/PI hybrid film with 8 wt.% AlN waspatterned using micromachining technology and used as thedielectric layer in RDL successfully.

Aiming to green machining of single crystal sapphire, polysaccharide-bonded soft abrasive tool using hydrothermal gelatinisation method was proposed. Mechanical properties of prepared polysaccharide films show that when the weight ratio of natural polysaccharide to cross-linked polysaccharide is 1:1, the tensile strength and extension rate of the film are relatively optimal. Composition and making process of polysaccharide bonded abrasive tool was built. Samples of polysaccharide bonded soft abrasive tools were made by which the single crystal sapphire substrates were machined. It is found that by adopting guar gum as dispersion agent and hydrothermal gelatinisation, the abrasive sedimentation during making process of abrasive tool is substantially eliminated, and better abrasive holding ability is attained. XPS and Raman microscope analysis showed that material of single crystal sapphire was removed by mechano-chemical reaction with good surface integrity. Abrasive tools with abrasive to binder ratio of 4:1 presented relatively better self-sharpening and machining performance, and the material removal rate reached 0.427 μm/min. More interesting, XPS and FT-IR analyses both indicated that the polysaccharide binder dehydrated by the friction heat during machining process.
