Quercetin Enhances Cisplatin Sensitivity of Human Osteosarcoma Cells by Modulating microRNA-217-KRAS Axis

        Zhaohui Chen,Xian Zhang,Qinggong Guo,Jingtao Chen 한국분자세포생물학회 2015 Molecules and cells Vol.38 No.7

Quercetin can suppress osteosarcoma cell growth and metastasis. However, other effects of quercetin on osteosarcoma remain largely unknown. This research aims to evaluate the effects of quercetin in combination with cisplatin as treatment for osteosarcoma and investigate its regulatory mechanism. Cell viability and apoptosis in 143B cell line were determined after treatment with quercetin and/or cisplatin. RT-PCR and Western blot analysis were performed to determine the RNA or protein expression levels. Moreover, transwell assay was used to evaluate metastasis. Furthermore, rescue experiments were performed to investigate the potential regulatory mechanism of the treatment. Results showed that quercetin with concentration that was equal to or greater than 10 μM inhibited 143B proliferation, while 5 μM quercetin enhanced the cisplatin sensitivity of 143B cells. Expression of miR- 217 was upregulated after quercetin and/or cisplatin treatment, while its target KRAS was downregulated both at mRNA and protein levels. MiR-217 knockdown led to the loss of enhanced cisplatin sensitivity while miR-217 overexpression showed the opposite effects, indicating that quercetin regulated cisplatin sensitivity by modulating the miR-217-KRAS axis. In conclusion, 5 μM quercetin enhanced the cisplatin sensitivity by modulating the miR- 217-KRAS axis. This finding suggests that quercetin may be administered with cisplatin to improve the treatment for osteosarcoma.

Exponential L₂-L∞ Filtering for a Class of Stochastic System with Markovian Jump Parameters and Mixed Mode-Dependent Time-Delays

Zhaohui Chen,Qi Huang 제어·로봇·시스템학회 2014 International Journal of Control, Automation, and Vol.12 No.3

This paper discusses the exponential L₂-L∞ filtering problem of a class of nonlinear stochastic systems with Markovian jumping parameters and mixed mode-dependent time-varying delays. By introducing a new multiple mode-dependent Lyapunov-Krasovskii functional, stochastic analysis is conducted. The condition for the existence of mode-dependent L₂-L∞ filter, in which the filtering error is guaranteed to be exponentially stable with prescribed L₂-L∞ performance, is developed. The developed criterion is delay-range-dependent, mode-dependent and decay-rate-dependent. Based on the derived criterion, the L₂-L∞ filtering problems are solved. The mode-dependent filter coefficients can be obtained by solving a set of linear matrix inequalities (LMIs). Numerical simulations are presented to illustrate the effectiveness of the proposed approach.

Thermo-/pH-dual responsive properties of hyperbranched polyethylenimine grafted by phenylalanine

        Chen, Jie,Xia, Jialiang,Tian, Huayu,Tang, Zhaohui,He, Chaoliang,Chen, Xuesi 대한약학회 2014 Archives of Pharmacal Research Vol.37 No.1

Novel thermo- and pH-dual responsive amphiphilic copolymers were synthesized based on hyperbranched polyethylenimine (PEI) by grafting $\small{L}$-phenylalanine. The phenylalanine-modified PEI exhibited lower cytotoxicity than commercial PEI. These copolymers showed the phenomena of phase transitions in response to pH and temperature. The dilute copolymer solution at lower pH displayed the higher LCST. Furthermore, LCST increased with the increasing of phenylalanine grafting density. LCST of these copolymers were tunable from 7.2 to $59.6^{\circ}C$ by the degree of amidation and pH of solution. DLS and TEM experiments certified that the copolymer chains aggregated to form small size particles as increasing the temperature above LCST. For these reasons, the obtained smart copolymers were considered to be potential gene/drug carriers in biomedical field.

Dual-template synthesis of interconnected 3D hollow N-doped carbon network for electrochemical application

        Chen Liang,Yang Lanyun,Xu Chenxi,Chen Kechun,Wang Wei,Yang Liming,Hou Zhaohui 한국탄소학회 2023 Carbon Letters Vol.33 No.2

Recently, hollow carbon spheres (HCS) have aroused great interests in the field of energy storage and conversion owing to their unique morphology, structure and other charming properties. Nevertheless, unsatisfactory electrical conductivity and relatively poor volumetric energy density caused by inevitable gaps between discrete carbon spheres greatly impede the practical application of HCS. In this work, for the first time we propose a novel dual-template strategy and successfully fabricate interconnected 3D hollow N-doped carbon network (HNCN) by a facile and scalable pyrolysis process. By systematical characterization and analysis, it can be found that HNCN is assembled by HCS and lots of mesoporous carbon. Compared to the counterparts, the obtained HNCN exhibits unique 3D interconnected architecture, larger specific surface area, hierarchical meso/macropore structure, higher structure defects, higher N doping amount and more optimized N configurations (especially for pyridinic-N and graphitic-N). As a result, these advantageous features endow HNCN with remarkably promoted electrochemical performance for supercapacitor and oxygen reduction reaction. Clearly, our proposed dual-template strategy provides a good guidance on overcoming the intrinsic shortcomings of HCS, which undoubtedly broadens their application in energy storage and conversion.

Preparation of self‑supporting Co3S4/ S‑rGO film catalyst for efficient oxygen evolution reaction

        Liang Chen,Liying Hu,Chenxi Xu,Lanyun Yang,Wei Wang,Junlin Huang,Minjie Zhou,Zhaohui Hou 한국탄소학회 2023 Carbon Letters Vol.33 No.7

Exploring cheap and efficient oxygen evolution reaction (OER) catalysts is extremely vital for the commercial application of advanced energy storage and conversion systems. Herein, a self-supporting Co3S4/ S-doped reduced graphene oxide ( Co3S4/S-rGO) film catalyst is successfully prepared by a blade coating coupled with high-temperature annealing strategy, and its morphology, structure and composition are measured and analyzed. It is substantiated that the as-synthesized Co3S4/ S-rGO film possesses unique self-supporting structure, and is composed of uniformly dispersed Co3S4 nanoparticles and highly conductive S-rGO, which benefit the exposure of catalytic sites and electron transfer. By reason of the synergistic effect of the two individual components, the self-supporting Co3S4/ S-rGO film catalyst displays outstanding catalytic performance towards OER. As a consequence, the Co3S4/ S-rGO film catalyst delivers an overpotential of 341 mV at 10 mA cm-2, and the current attenuation rate is only 2.6% after continuous operation for 4 h, verifying excellent catalytic activity and durability. Clearly, our results offers a good example for the construction of high-performance self-supporting carbon-based composite film catalysts for critical electrocatalytic reactions.

Hepatic Resection Provides Survival Benefit for Selected Intermediate-Stage (BCLC-B) Hepatocellular Carcinoma Patients

        Zhang Zhaohui,Shen Shunli,Chen Bin,Li Shaoqiang,Hua Yunpeng,Kuang Ming,Liang Lijian,Peng Bao Gang 대한암학회 2019 Cancer Research and Treatment Vol.51 No.1

Purpose The intermediate stage of hepatocellular carcinoma (HCC) (Barcelona Clinic Liver Cancer [BCLC] B) comprises a highly heterogeneous population, and the treatment strategy is still controversial. Because of the heterogeneity, a subclassification of intermediate-stage HCCs was put forward by Bolondi according to the ‘beyond Milan and within up-to-7’ criteria and Child-Pugh score. In this study, we aim to analyze the prognosis of BCLC-B stage HCC patients who received hepatic resection according to the Bolondi’s subclassification. Materials and Methods One thousand and one hundred three patients diagnosed with HCC and treated with hepatic resection were enrolled in our hospital between 2006 and 2012. According to Bolondi’s subclassification, the BCLC-B patients were divided into four groups. Recurrence-free survival (RFS) and overall survival (OS) were analyzed. Results According to Bolondi’s subclassification, the BCLC-B patients were divided into four groups: B1 (n=41, 18.7%), B2 (n=160, 73.1%), B3 (n=11, 5.0%), and B4 (n=7, 3.2%). Significant difference was observed between B1 and other groups (B1 vs. B2, p=0.022; B1 vs. B3, p < 0.001; B1 vs. B4, p < 0.001), but no difference for B2 vs. B4 (p=0.542) and B3 vs. B4 (p=0.542). In addition, no significant differences were observed between BCLC-A and BCLCB1 group for both RFS (p=0.087) and OS (p=0.643). In multivariate analysis, BCLC-B subclassification was not a risk factor for both OS (p=0.263) and RFS (p=0.892). Conclusion In our study, HCC patients at B1 stage were benefited from hepatic resection and had similar survival to BCLC-A stage patients. Our study provided rationality of hepatic resection for selected BCLC-B stage HCC patients instead of routine transarterial chemoembolization.

Grinding Marks on Ultra-Precision Grinding Spherical and Aspheric Surfaces

        Bing Chen,Bing Guo,Shichun Li,Zhaohui Deng,Qingliang Zhao 한국정밀공학회 2017 International Journal of Precision Engineering and Vol.4 No.4

Grinding marks are regard as a great obstacle to manufacture spherical and aspheric surfaces with higher surface quality, lower energy and wastage. The scallop-height was studied for optimizing the grinding parameters firstly to reduce its effect on grinding marks. Secondly, the expression of grinding points distribution was established to characterize the grinding marks caused by the radial run-out of grinding wheel. And then, the aspheric grinding experiments of monocrystalline silicon were carried out to investigate the influence of grinding marks on surface quality. The experiments revealed that the remarkable grinding marks with patterned grinding points distribution would cause more fractures and roughness, deeper grooves, and more inhomogeneous surface quality compared with the weak grinding marks. The discriminating standard of grinding marks was established, and the grinding parameters were optimized for homogenizing the grinding points distribution by this discriminating standard to reduce the grinding marks in actual grinding process. Finally, the large size infrared lens was ground with high surface quality by the optimized grinding parameters, and the results of surface quality demonstrate that the discriminating standard was effective. This research provides references and ideas for grinding aspherical surface with high surface quality and efficiency, low energy and wastage.

      • Joint Deployment and Task Computation of UAVs in UAV-assisted Edge Computing Network

        Yuqing Chen,Zhaohui Zheng 한국통신학회 2020 한국통신학회 APNOMS Vol.2020 No.09

Unmanned Ariel Vehicle(UAV) has been widely used in the edge computing network. Owing to its line-of-sight communication ability and mobility, it has offered task offloading service and some task computation for mobile devices(MDs). Nowadays, as high-intensity calculation application develops rapidly, maximizing the size of offloaded tasks is necessary for meeting the users’ experience in applications. In this context, our paper takes research on how to deploy UAVs at the most proper place to offer task offloading with TDMA protocol. Specifically, we will optimize the task offloading number in the MDs-UAV system by combining the location of UAVs, the task computation capacity of UAV, and the MDs-UAV associations in a certain time. We prove that the joint UAV deployment and task computation problem is NP-hard, and use a greedy algorithm to optimize the result. Compared with randomly selected method, our simulation shows that the greedy algorithm performs greatly and the deployment of UAVs are sensible. We believe that this idea will improve the system tasks processing rate and Quality of Experience(QoE) of mobile users.
