        Effects of Aqueous Extract of Schizandrae Fructus on Lead-Induced Change of Monoamine Neurotransmitters in Hippocampus

        The effects of aqueous extract of Schizandrae Fructus (AESC) on lead (Pb)-induced changes of monoamine neurotransmitters in the hippocampus (HIP) of adult rats were investigated. Male Sprague-Dawley rats were received intraperitoneal (i.p.) administration of Pb acetate (5 mg/kg/d) for 28 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA), serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in HIP were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Pb treated rats (P < 0.05), while pretreatment with AESC (100 mg/kg/d or 300 mg/kg/d, p.o., 2 h before Pb) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC (300 mg/kg/d) significantly increased the reduction of glutathione contents and superoxide dismutase activities in HIP induced by chronic Pb. These results suggest that AESC ameliorates Pb-induced depletion of monoamine neurotransmitters in HIP through its antioxidant activity.

        Structural and photoelectrochemical characterization of TiO2 nanowire/nanotube electrodes by electrochemical etching

        TiO2 nanowire/nanotube electrodes were synthesized by anodization of titanium foils in ethylene glycol solution containing 0.5 wt% NH4F and 1 wt% water at 60 V for 6 h. The microstructure and morphology of the asprepared electrodes were investigated by XRD and SEM. A possible formation mechanism and oxidation parameters of nanocomposite structure were discussed. The relationship between structural characteristics of TiO2 nanowire/nanotube electrodes and its photoelectrochemical characterization were evaluated by electrochemical analyzer and photocatalytic degradation of methylene blue (MB) solution. Furthermore, these TiO2 nanowire/nanotube electrodes promoted the photoelectrochemical characterization due to the larger surface areas, enhanced light harvesting and electron transport rate. The results show that photocurrent density of 1.44mA/cm2 and photocatalytic degradation of 95.51% was achieved for TiO2 nanowire/nanotube electrodes, which were 0.55mA/cm2 and 20.52% higher than the TiO2 nanotube electrodes under a similar condition, respectively.

        Cytotoxic Components in an Extract from the Leaves and Stems of Stauntonia hexaphylla

        An investigation was carried out to identify novel anti-cancer compounds from Korean indigenous plant extracts. Bioassay-guided fractionation and chemical investigation of the EtOAc extract from the leaves and stems of Stauntonia hexaphylla resulted in the isolation of two active compounds, hederagenin 3-O-a-L-arabinoside (1) and quercetin (2). The structures of these compounds were elucidated by spectroscopic methods, including UV, IR, MS, NMR techniques and compared with previous spectroscopic data. The cytotoxic effects of fractions and compounds on HCT116 human colon cancer cells were evaluated using the MTT assay. Quercetin showed a stronger anti-cancer effect when compared to hederagenin 3-O-a-L-arabinoside.

        A One-Step System for Convenient and Flexible Assembly of Transcription Activator-Like Effector Nucleases (TALENs)

        Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the convenient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

        P-hydroxybenzoic acid positively affect the Fusarium oxysporum to stimulate root rot in Panax notoginseng

        Background: Plant health is directly related to the change in native microbial diversity and changes in soil health have been implicated as one of the main cause of root rot. However, scarce information is present regarding allelopathic relationship of Panax notoginseng root exudates and pathogenic fungi Fusarium oxysporum in a continuous cropping system. Methods: We analyzed P. notoginseng root exudate in the planting soil for three successive years to determine phenolic acid concentration using GC-MS and HPLC followed by effect on the microbial community assembly. Antioxidant enzymes were checked in the roots to confirm possible resistance in P. notoginseng. Results: Total 29 allelochemicals in the planting soil extract was found with highest concentration (10.54 %) of p-hydroxybenzoic acid. The HPLC showing a year-by-year decrease in p-hydroxybenzoic acid content in soil of different planting years, and an increase in population of F. oxysporum. Moreover, community analysis displayed negative correlation with 2.22 mmol. L<sup>-1</sup> of p-hydroxybenzoic acid correspond to an 18.1 % population of F. oxysporum. Furthermore, in vitro plate assay indicates that medium dose of p-hydroxybenzoic acid (2.5-5 mmol. L<sup>-1</sup>) can stimulate the growth of F. oxysporum colonies and the production of macroconidia, as well as cell wall-degrading enzymes. We found that 2-3 mmol. L<sup>-1</sup> of p-hydroxybenzoic acid significantly increased the population of F. oxysporum. Conclusion: In conclusion, our study suggested that p-hydroxybenzoic acid have negative effect on the root system and modified the rhizosphere microbiome so that the host plant became more susceptible to root rot disease.

        Transcriptome sequencing reveals non-coding RNAs respond to porcine reproductive and respiratory syndrome virus and Haemophilus parasuis co-infection in Kele piglets

        Co-infection with porcine reproductive and respiratory syndrome virus (PRRSV) and Haemophilus parasuis (HPS) has severely restricted the healthy development of pig breeding. Exploring disease resistance of non-coding RNAs in pigs co-infected with PRRSV and HPS is therefore critical to complement and elucidate the molecular mechanisms of disease resistance in Kele piglets and to innovate the use of local pig germplasm resources in China. RNA-seq of lungs from Kele piglets with single-infection of PRRSV or HPS and co-infection of both pathogens was performed. Two hundred and twenty-five differentially expressed long non-coding RNAs (DElncRNAs) and 30 DEmicroRNAs (DEmiRNAs) were identified and characterized in the PRRSV and HPS co-infection (PRRSV-HPS) group. Compared with the single-infection groups, 146 unique DElncRNAs, 17 unique DEmiRNAs, and 206 target differentially expressed genes (DEGs) were identified in the PRRSV-HPS group. The expression patterns of 20 DEmiRNAs and DElncRNAs confirmed by real-time quantitative polymerase chain reaction (RT-qPCR) were consistent with those determined by high-throughput sequencing. In the PRRSV-HPS group, the target DEGs were enriched in eight immune Gene Ontology terms relating to two unique DEmiRNAs and 16 DElncRNAs, and the unique target DEGs participated the host immune response to pathogens infection by affecting 15 immune-related Kyoto Encyclopedia of Genes and Genomes enrichment pathways. Notably, competitive endogenous RNA (ceRNA) networks of different groups were constructed, and the ssc-miR-671-5p miRNA was validated as a potential regulatory factor to regulate DTX4 and AEBP1 genes to achieve innate antiviral effects and inhibit pulmonary fibrosis by dual-luciferase reporter assays. These results provided insight into further study on the molecular mechanisms of resistance to PRRSV and HPS co-infection in Kele piglets.

        Size control of Co-doped ZnO rods by changing the solvent

        In this work, the Co-doped ZnO rods were prepared by the hydrothermal method. The size of these rods can be changed from micro-size to nano-size by using different solutions during the preparation. The results of transmission electron microscopy (TEM) and selected area electron diffraction (SAED) showed that the as-prepared nano-sized Co-doped rods have single-crystal structure. The polarized Raman experiments were presented on an individual micro-sized Co-doped ZnO rod in the $X(YY)\vec{X}$, $X(ZY)\vec{X}$ and $X(ZZ)\vec{X}$ configurations, the results of polarized Raman indicated that these rods are crystallized and their growth direction is parallel to c-axis.

        Fragrance Composition in Six Tree Peony Cultivars

        Tree peony is a traditional famous flower of China, and plays an important role in Chinese traditional culture. But the floral scent of tree peony in vivo is little known. In this study, in order to explore the floral composition of tree peony, floral volatiles of six cultivars, including Paeonia suffruticosa 'Zhaofen' (ZF), P. suffruticosa 'Luoyanghong' (LYH), P. ostii 'Fengdanbai' (FDB), P. ${\times}$ lemonei 'High noon' (HN), P. ${\times}$ lemonei 'Renown' (R), and P. rockii 'Gaoyuanshenghuo' (GYSH) were collected by dynamic headspace and then identified by Automated Thermal Desorption-Gas Chromatography/Mass Spectometry. The results showed that floral fragrances of the six cultivars were qualitatively and quantitatively distinct. A total of 105 volatiles involving ten categories were detected. But not all volatile categories were emitted from these cultivars. The six peony cultivars emitted some shared compounds and peculiar compounds. The total released amounts of volatiles emitted from six cultivars were found significantly different, which was greatest for 'GYSH'. The most abundant volatile compounds detected from 'ZF', 'LYH', 'FDB', 'HN', 'R', and 'GYSH' were respectively ${\alpha}$-pinene, 2,3-dihydroxy propanal, 3-methyl-1-butanol, 2-ethyl-1-hexanol, acetic acid 1-methylethyl ester, and 5-ethyl-2,2,3-trimethyl heptane. This result may contribute to exploring the biosynthesis and emission mechanism of floral scent in tree peony.

        Estimation in partially linear time-varying coefficients panel data models with fixed effects

        A partially time-varying coefficient time series panel data model with fixed effects is considered to characterize the nonlinearity and trending phenomenon in panel data model. To estimate the linear regression coefficient and the time-varying coefficient function, two methods are applied with the help of profile least squares. The first one is taking cross-sectional average to eliminate the fixed effects. The second one is local linear dummy variable approach. In each method we derive consistent estimates for both the parametric component and non-parametric trend function. The asymptotic distributions of the estimates are established when T and N tend to infinity simultaneously, where N is the cross section size, T is the time series length. The asymptotic results reveal that the parametric component (non-parametric coefficient function) estimate based on crosssectional average has a rate of convergence T)^−1/2 ((Th)^−1/2) that is slower than that based on local linear dummy variable approach, which is (NT))^−1/2((NTh)^−1/2), where h is the bandwidth. Furthermore, block bootstrap method is used to construct confidence interval for parametric and nonparametric components, respectively. At last, some simulation studies are conducted to examine the finite sample performance for the proposed methods and a real data example is analyzed.

      • Guided waves of porous FG nanoplates with four edges clamped

        Based on the nonlocal strain gradient (NSG) theory and considering the influence of moment of inertia, the governing equations of motion of porous functionally graded (FG) nanoplates with four edges clamped are established; The Galerkin method is applied to eliminate the spatial variables of the partial differential equation, and the partial differential governing equation is transformed into an ordinary differential equation with time variables. By satisfying the boundary conditions and solving the characteristic equation, the dispersion relations of the porous FG strain gradient nanoplates with four edges fixed are obtained. It is found that when the wave number is very small, the influences of nonlocal parameters and strain gradient parameters on the dispersion relation is very small. However, when the wave number is large, it has a great influence on the group velocity and phase velocity. The nonlocal parameter represents the effect of stiffness softening, and the strain gradient parameter represents the effect of stiffness strengthening. In addition, we also study the influence of power law index parameter and porosity on guided wave propagation.
