Are pet foods safe? From perspective of mycotoxins, preservatives and antioxidants

        Kyu-hwan Hwang(Kyu-hwan Hwang),Hye-ra Kim(Hye-ra Kim),Yoon-hwa Choi(Yoon-hwa Choi),Doo-hwan Kim(Doo-hwan Kim),You-Jin Lee(You-Jin Lee),Dong-Hyuk Jeong(Dong-Hyuk Jeong) 한국예방수의학회 2023 예방수의학회지 Vol.47 No.2

The aim of this study was to determine the contamination of mycotoxins and the concentration of preservatives and antioxidants in commercial pet food. 106 pet foods were purchased from online in Korea. Mycotoxin analysis were performed using liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) and preservatives and antioxidants were analyzed by high performance liquid chromatography (HPLC). The contamination of the mycotoxins in all samples was proved to comply with the Korea legislation with regards to animal feed, the analyses revealed presence of aflatoxins, ochratoxin A, zearalenone and fumonisins, with values below 1.6 ㎍/㎏, 12.1 ㎍/㎏, 478.5 ㎍/㎏ and 1873.5 ㎍/㎏, respectively. Furthermore, the simultaneous presences of different mycotoxins were observed in most of positive samples. The levels of dehydroacetic acid (DHA), sorbic acid (SA) and benzoic acid (BA) were, respectively, with values below 0.24 g/㎏, 4.82 g/㎏ and 6.35 g/㎏. The concentrations of butylated hydroxy anisole (BHA) and butylated hydroxy toluene (BHT) were demonstrated to be acceptable, with values below 0.04 g/㎏, and 0.02 g/㎏. This indicated the need for further investigation into the potential risk deriving from chronic exposure to low doses of mycotoxins. Additionally, a co-contamination of mycotoxins which interact in synergic manner are more concern. As DHA that not accepted was detected and very high levels of SA were identified in samples, an upper limit for preservatives in pet foods should be established in pet foods. BHA and BHT were detected within the recommended levels for antioxidant content, so these are considered as safe.

      • Controlled generation of submicron emulsion droplets via highly stable tip-streaming mode in microfluidic devices.

        Jeong, Woong-Chan,Lim, Jong-Min,Choi, Jae-Hoon,Kim, Jong-Hoon,Lee, You-Jin,Kim, Seung-Hyun,Lee, Gaehang,Kim, Jong-Duk,Yi, Gi-Ra,Yang, Seung-Man Royal Society of Chemistry 2012 Lab on a chip Vol.12 No.8

<P>Submicron emulsions could be produced via the tip-streaming process in a flow-focusing microfluidic device. In this article, the stability of the liquid cone and thread for tip-streaming mode could be significantly improved by employing a three-dimensional flow-focusing device, in which the hydraulic resistance was adjusted by modulating the channel heights in the flow focusing area, orifice, downstream and dispersed phase inlet channel. The pressure range for tip-streaming mode was enlarged significantly compared with two-dimensional flow-focusing devices. Therefore, monodisperse emulsions were produced under this tip-streaming mode for as long as 48 hours. Furthermore, we could control the size of emulsion drops by changing the pressure ratio in three-dimensional flow-focusing devices while the liquid cone was easily retracted during the adjustment of pressure ratio in two-dimensional flow-focusing devices. Furthermore, using the uniform submicron emulsion droplets as confining templates, polyethylene glycol (PEG) particles were produced with a narrow size distribution at the sub-micrometre scale. In addition, magnetic nanoparticles were added to the emulsion for magnetic PEG particles, which can respond to magnetic field and would be biocompatible.</P>

      • Anti-inflammatory Effect of Columbianetin on Activated Human Mast Cells

        Jeong, Hyun-Ja,Na, Ho-Jeong,Kim, Su-Jin,Rim, Hong-Kun,Myung, Noh-Yil,Moon, Phil-Dong,Han, Na-Ra,Seo, Jae-Uk,Kang, Tae-Hee,Kim, Jae-Joong,Choi, Youngjin,Kang, In-Cheol,Hong, Seung-Heon,Kim, You-Ah,Seo, Pharmaceutical Society of Japan 2009 BIOLOGICAL & PHARMACEUTICAL BULLETIN Vol.32 No.6

<P>In the present study, we extracted <I>Corydalis heterocarpa</I> with various solvents in order to find the bioactive constituents that demonstrated anti-inflammatory effects. We isolated the active compound, Columbianetin. Anti-inflammatory effect of Columbianetin has been reported but the precise effects of Columbianetin in experimental models have remained unknown. In the present study, we investigate the effect of Columbianetin on the production of histamine, interleukin (IL)-1β, IL-6, IL-8, and tumor necrosis factor (TNF)-α and expression of cyclooxygenase-2 (COX-2) by using the human mast cell line (HMC-1). Various concentrations of Columbianetin were treated before the activation of HMC-1 cells with phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore, A23187. PMA plus A23187 significantly increased IL-1β, IL-6, IL-8, and TNF-α production compared with media control (<I>p</I><0.05). We also show that the increased cytokines IL-1β, IL-6, IL-8, and TNF-α level was significantly inhibited by Columbianetin in a dose-dependent manner (<I>p</I><0.05). Maximal inhibition rates of IL-1β, IL-6, IL-8, and TNF-α production by Columbianetin were about 102.6%, 101.1%, 95.8%, and 103.9%, respectively. Columbianetin inhibited expression of COX-2. In addition, the effect of Columbianetin was investigated on the histamine release from HMC-1 stimulated by substance P, which promotes histamine release. Columbianetin also inhibited the histamine release by substance P. In conclusion, these results indicate that Columbianetin may be helpful in regulating mast cell-mediated allergic inflammatory responses.</P>

Combined treatment of silibinin with either sorafenib or EGFR tyrosine kinase inhibitors enhances growth inhibitory effects in hepatocellular carcinoma cells

        Ha Ra Gu,Su Cheol Park,Su Jin Choi,Jae Cheol Lee,You Cheoul Kim,Chul Ju Han,Ki Young Yang,김연주,Geum Youb Noh,So Hyeon No,Jae-Hoon Jeong 대한간학회 2015 Clinical and Molecular Hepatology(대한간학회지) Vol.21 No.1

Background/Aims: Silibinin, the main component of silymarin, is used as a hepatoprotectant and exhibits anticancer effects against various cancer cells. This study evaluated the effects of a combination of silibinin with either gefitinib or sorafenib on hepatocellular carcinoma (HCC) cells. Methods: Several different human HCC cell lines were used to test the growth-inhibiting effects and cell toxicity of silibinin both alone and in combination with either gefitinib or sorafenib. The cell viability and growth inhibition were assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, trypan blue staining, and a colony-forming assay. Furthermore, changes in epidermal growth factor receptor (EGFR)-related signals were evaluated by Western blot analysis. Results: Gefitinib, sorafenib, and silibinin individually exhibited dose-dependent antiproliferative effects on HCC cells. Combined treatment with silibinin enhanced the gefitinib-induced growth-inhibiting effects in some HCC cell lines. The combination effect of gefitinib and silibinin was synergistic in the SNU761 cell line, but was only additive in the Huh-BAT cell line. The combination effect may be attributable to inhibition of EGFR-dependent Akt signaling. Enhanced growth-inhibiting effects were also observed in HCC cells treated with a combination of sorafenib and silibinin. Conclusions: Combined treatment with silibinin enhanced the growth-inhibiting effects of both gefitinib and sorafenib. Therefore, the combination of silibinin with either sorafenib or gefitinib could be a useful treatment approach for HCC in the future. (Clin Mol Hepatol 2015;21:49-59)

        Na Ra Jeong,Kwang Jin Kim,Hyung Gewon Yun,Seung Won Han,Soojin You 인간식물환경학회 2019 인간식물환경학회지 Vol.22 No.5

This study analyzed the factors affecting urban residents expectations and participation in community gardens to present basic data of guidelines for community garden management. Urban citizens valued the necessity of community gardens overall, especially residents with outdoor gardens. The experience of private gardens was important in perceiving the need for community gardens, which are public spaces. The creation of community gardens had high expectations for social aspects such as improving quality of life, health promotion, and community revitalization. In particular, residents living in individual housing have high expectations for revitalization of the community, and community gardens can be a mediator for forming and maintaining local communities. Through this, the citizens perception on the role and function of the garden as a community space could be examined. Expectations for quality of life, health promotion, and community revitalization are factors affecting the necessity of community garden. Participation was high in the creation and management of the community garden, and the necessity of community gardens, expectation for community revitalization, and expectation for environmental purification are factors that affect participation in the management of community gardens. The amount of donations that the residents are willing to pay for community garden management was KRW 75,222 /year on average. Overall, residents with higher need for community gardens and higher social expectation showed higher participation. Urban citizens have high expectations for community gardens, and they perceive the importance of social aspects more than personal aspects such as community revitalization and quality of life. As a strategy for increasing community involvement, it is necessary to set up the processes, determine participants composition and roles, and establish a system to promote participation. Considering welfare aspects such as community revitalization and improvement of residents’ quality of life, motivation and administrative support for participation in the creation and management of community gardens will serve as important factors.

Effects of Medicinal herb Extracts and their Components on Steatogenic Hepatotoxicity in Sk-hep1 Cells

        Choi, You-Jin,Yoon, Yu-Jin,Choi, Ho-Sung,Park, So-Ra,Oh, Se-Hee,Jeong, Se-Mi,Suh, Hyo-Ryung,Lee, Byung-Hoon Korean Society of ToxicologyKorea Environmental Mu 2011 Toxicological Research Vol.27 No.4

Herbal medicines are widely used in many countries for the treatment of many diseases. Although the use of herb extracts as alternative medicine is growing, their toxicological properties have not been thoroughly investigated. In this study, we have investigated the effects of water and ethanol extracts of 18 herbs on the hepatic lipid metabolism and steatogenic hepatotoxicity. Ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa induced lipid accumulation in Sk-hep1 human hepatoma cells as determined by Nile red staining. These extracts increased the luciferase activity of sterol regulatory element (SRE) and decreased that of peroxisome proliferator response element (PPRE), indicating the possibilities of enhanced fatty acid synthesis and decreased fatty acid oxidation. To identify the components responsible for the fat accumulation, we tested 50 chemicals isolated from the nine herbs. Apigenin, luteolin, pectolinarin and lupeol from Cirsium japonicum, 8-methoxypsoralen and umbelliferone from Foeniculum vulgare and pomonic acid and jiocerebroside from Rehmanniae glutinosa significantly increased the accumulation of lipid droplets. These results suggest that ethanol extracts of Cirsium japonicum, Carthamus tinctorius, Rehmanniae glutinosa (preparata), Polygala tenuifolia, Foeniculum vulgare, Polygonum multiflorum, and Acorus gramineus and water extracts of Polygonum multiflorum and Rehmanniae glutinosa can cause fatty liver disease by decreasing ${\beta}$-oxidation of fatty acid and increasing lipogenesis.

A Study on the Application of IPA Method for Exploring the Properties of Urban Residents Choice of Indoor Plants

        Na Ra Jeong,Kwang Jin Kim,Ji Hye Yoon,Seung Won Han,Soojin You 인간식물환경학회 2020 인간식물환경학회지 Vol.23 No.6

Background and objective: This study was conducted to understand urban residents perception of indoor plants in order to assist in the preparation of guidelines for growing plants indoors. Methods: Questionnaires were distributed to urban residents currently growing plants or with experience of growing plants. The data of 247 respondents were subjected to analysis to assess their level of interest and ability with regard to growing plants, and their recognition of the selection and function of plants. Results: Respondents showed high interest in growing plants at a level of 6.77, but their ability was moderate. This suggests that information regarding plant maintenance should be provided to urban residents. They recognized the function of plants for improving the environment as important, and expected that indoor plants would improve their mood and visual experience. Satisfaction with growing plants was high from an emotional perspective. They recognized that the function of air purification function was important even if the actual performance or effect was not great. This indicates that the function of air purification should be emphasized more. As the result of the IPA, plants should be selected in consideration of morphological characteristics such as leaf, flower and fruit, and continuously managed to maintain their characteristics. Since the performance of pots was less satisfactory compared to their importance, they should be selected so that they harmonize better with plants and are of the appropriate size to fit the space. The type of plants is an important factor in plant selection in order to perform an environmental function, and the flower color and pot size are important in terms of aesthetics and healing. Conclusion: When horticulture information on indoor plants is provided to urban residents, first, it should be provided to satisfy the functions of the plants required by urban residents and in order to promote and maintain the proper growth of plants. Second, various contents that utilize components should be developed to achieve plant function so that resident can determine the function of plants and select the type, quantity, and shape of plants to achieve the desired function.
