      • S. mutans의 固定化에 依한 Insoluble glucan의 製造方法 및 齒面 細菌膜 分解酵素인 α-1, 3 glucanase 生産菌珠 Streptomyces SW-522에 關한 硏究

        In other to investigate α-1, 3 glucanase from microorganisms for hydrolysis of dental caries, S. muatns was immobilized by cubes of 28% polycrylamide and coated with same polyacrylamide. The insoluble glucan obtained 1.13g per one liter in TH medium with 5% sucrose by PA immobilization of S. muatns, which was very pure by IR and TLC analysis. α-1, 3 glucanase produced by of the Strptomyces SW-522, was developed by treatment with 5% EMS. The new Sw-522 M1 strain obtained, which improved activity of α-1, 3 glucanase.

        복합 세라믹스(TiC-Al_2O_3)의 방전가공특성에 관한 연구

        TiC-Al_2O_3 composite ceramics has high hardness, high strength, high wear and corrosion resistance. Therefore, composite ceramics have been concerned significantly with some excellent properties and many functions as new industrial materials to the industry at large. In present research, experiments are carried out to obtain the machinability and machining properties by BDM. As a result, the most suitable machining conditions of TiC-Al_2O_3 composite ceramics was that the pulse duration is 10-60㎲, the peak current is 10-16A. The machining speed and the wear of the tool electrode increased with the increase in peak current.

      • 주제학습을 통한 ICT 활용 교육 모형 개발

        In coming 21st century, which is forecasted that it will be a knowledge-information oriented society, abilities of information acquirements are required. For this, the Ministry of Education has been encouraging the business of information oriented school education and promoting application of ICT more than 10% in all classes of the 7th curriculum. Moreover, after extending applied classes into 20% by 2005, the Ministry of Education will promote editing textbooks by subjects reflecting the content of ICT application. But, actually, classes applied ICT aren't fulfilled in education fields. The main reason for this is that teachers don't understand fully ICT application model. On the basis of the preceding studies, this study designed subject-settlement class model to develop students' abilities of information acquirements. The subject-settlement class model is made up of 5 steps--confirmation of studying subjects, searching information and collecting, using information, arrangement of results, communication of information, and evaluation. By experiencing search of information, arrangement of information and creation of new information, the application of this model helps the students develop abilities of information acquirements. Nowadays, development of information communication and of computer makes the world change from information oriented society into knowledge oriented society. Future society will focus on the ability of production to create knowledge information. This study is the result presented as a part of the ability of creating information. Furthermore, the development of various learning and teaching models, which are required in information oriented age, are expected.

        Streptomyces sp. Y9343이 生産하는 齒面細菌膜 分解酵素의 精製와 特性

        치면세균막을 제거하거나 형성을 억제하여 치아우식증 예방제를 개발할 목적으로 α-1,3 glucanase를 분비하는 새로운 균주로 부터 분리 정제하여 그 특성을 조사하였다. 불용성 glucan을 유일탄소원으로 하는 agar plate를 제조하여 토양으로 부터 α-1,3 glucanase 분비균주를 탐색한 결과 Streptomyces sp. Y9343을 얻었다. 액체배양시 최적 효소생산조건은 탄소원으로 1% soluble starch와 indcer로 0.5% insoluble glucan을 첨가하였을 때 가장 효율적이었다. α-1,3 glucanase는 황산 암모늄 염석, DEAE-CEllulose 이온교환 크로마토그래피, Sephadex G-75 겔 여과 등에 의하여 32.1배까지 정제되었고 수율은 0.53%이었으며, 이 때의 활성도는 7840.9 U/mg protein이었다. 정제된 α-1,3 glucanase를 SDS-PAGE로 분석한 결과, 단일체임을 확인하였으며, 이 때 분자량은 22,500이었다. 효소의 최적 pH는 6.5이었다. 효소의 최저온도는 37℃이었고, 열에 대한 안정성은 70℃ 이상에서 40%~60%의 효소활성이 상실함을 보였다. Detergent의 영향은 SDS에 의해 83%, Tween 20에 의해서는 약 27% 정도의 활성저해를 받았다. 효소활성의 금속이온에 의한 영향은 Co^2+, Mn^2_+에 의해 각각 81.8, 69.7%의 활성의 증가를 보였고 이들의 최적농도는 10mM이었으며, 반면에 Hg^2+에 의해서는 93.9%의 효소활성의 저해를 나타내었다. 또한 초기속도(30분 이내)에 금속이온에 의한 영향이 큰 것으로 나타났다. α-1,3 glucanase의 불용성 glucan에 대한 K_m 값은 2.50mM이었고, V_max는 0.0431 mM/min이었다. α-1,3 glucanase의 기질특이성을 조사한 결과, 반응 30분 후 IG와 soluble starch에는 각각73, 100%의 높은 분해력을 보였으며, raw starch, dextran T-10에 대해서는 낮은 분해력을 보였다. 한편, 인조치면세균막을 S. mutans로부터 시험관 벽에 제조한 후, α-1,3 glucanase를 처리한 결과 2시간 이내에 완전히 분해 제거되는 것을 알 수 있어 강력한 치아우식예방제로 개발될 수 있음을 보여 주었다. Streptococcus mutans has been implicated as primary causative agents of dental caries by insoluble glucan (IG) in human and experimental animals. An attempt was made to search for the α-1,3 glucanase that degrades IG produced by S. mutans. α-1,3 glucanase was detected in the culture supernatant of microorganisms, which are isolated from soils on agar medium containing IG as a sole carbon source. This Streptomyces sp. hydrolysed IG produced by immobilized S. mutans and was named as Y9343. This enzyme required α-1,3 glucan (IG) as an inducer. The optimum conditions for enzyme production were studied. The enzyme was purified by 30~70% (NH_4)_2SO_4 precipitation, anion exchange chromatography on DEAE-cellulose and gel filration on Sepadex G-75. The purified enzyme has a specific activity of 7840.0 U/mg protein giving 32.1-fold purification and final yield of 0.53%. The molecular weight was estimated to be about 22.5 kDa by SDS-PAGE. The optimum pH and temperature for enzyme reactiorr were 6.5 and 37℃, respectively and the enzyme was relatively stable at the temperature below 60℃. The activity of purified enzyme was enhanced by adding Co^2+, Mn^2+, and Mg^2+ into the medium, whereas inhiited by adding Hg^2+, Zn^2+ and SDS. The K_m and V_max value of α-1,3 glucanase for IG were estimated to be 2.50 mM and 0.0431 mM/min, respectively. The thin layer chromatographic analysis of hydrolysates from IG with α-1,3 glucanase showed that glucose was the main product of reaction. This enzyme activity was about 14 times higher than marketing dextranase as preventive agent against artificial dental caries by S. mutans in TH medium including 5% sucrose after 30 minutes.

      • 방향족 화합물의 할로 메칠화 반응

        Biphenyl의 para position에 methylene group을 도입 시키는 방법으로 paraformaldehyde와 dimethylamine을 반응시켜 Mannich Base인 (1,1'-biphenyl-4,4'-dimethyl)-dimethli amine을 합성해 보았으나, 전혀 반응이 진행되지 않았다. 즉 , Biphenyl에 강한 activating group이 없으므로 Mannich Base을 사용한 방법은 효과적이 못된다. 따라서, 다른 방법으로 biphenyl에 bromomethylation을 진행 시켜서, 두 개의 methylene group을 도입시켰다. Bromomethylation은 다음과 같이 사용하는 시약 및 실험 방법 등을 고려하여 3가지 방법으로 시도 하였다. 1)1,3,5-trioxane + HBr in AcOH + paraformaldehyde 사용법, 2)Hbr in AcOH + paraformaldehyde 사용법, 3) NaBr + H₂SO₄+paraformaldehyde 사용법, 이상의 3가지 방법 중에서 NaBr+ H₂SO₄+ paraformaldehyde 사용법이 가장 좋은 결과를 얻었다. 이러한 system에서 반응을 진행시킨 결과 69% 의 높은 수율 (87%의 purity)로 4,4'-dibromomethylbiphenyl 합성에 성공할 수 있었다. The synthetic method for the induction of methylene group on the para position of arenes is of importance. Since the weak activation effect of phenyl group on the electrophilic substitution reaction, the induction of methylene group by way of Mannich base using paraformaldehyde and dimethylamine was unsuccessful. It was found that halomethylation of byphenyl is only the way for the preparation of di-p-methylenebiphenyl. Especially, bromomethylation was turn out to be more reliable synthetic method than chloromethylation. Herein, we developed 3 protocols for the synthesis of 4,4'-dibromomethylbiphenyl. In addition, the best result was obtained using NaBr + H₂SO₄paraformaldehyde protocol in 69% yield.

      • 천연자원의 간기능 보호 효과에 관한 연구 : Benzo(a)pyrene에 의한 간기능 장해에 미치는 차전자의 효과 The Effect of Plantaginis Semen on Benzo(a)pyrene induced Hepatotoxicity

        B(a)P로 인하여 나타난 간손상 정도가 개선되어지는 현상은 예방효과보다 치료효과가 더 좋은 경향을 보였으며, B(a)P로 인한 간장해의 적응수단으로 높아진 효소활성을 차전자추출물의 유효성분이 대사를 촉진시키므로 간장해 예방효과 및 보간작용이 있을 것으로 생각되므로 차전자의 성분연구 및 작용기전에 대하여 더 많은 연구가 계속적으로 수행되어야 할 것이다. This study was conducted in an attempt to investigate the effectsof Plantaginis Semen on the activities of serum and liver enzyme and to evaluatethe utility of Plantaginis Semen as a hepatotropic agents in liver disease. The results were as follows ; 1. Significantly increased enzyme activity was noted in benzo(a)pyfene-intoxicated group. 2. Plantaginis Semen administration group prevented the leakage of AST,ALT, LDH and ALP in serum. 3. Insignificantly hepatic glutathione in Plantaginis Semen administrationgroups was decreased as compared to that of the control group. The contentof lipid peroxide was decreased in B(a)P-intoxicated groups with PlantaginisSemen administration. 4. In the contents of HDL-cholesterol and total cholesterol Plantaginis Semenadnninistration group was increased as compared to the control group significan-tly, while Plantaginis Semen administraton group increased in B(a)P-intoxicatedgroup and preventive group was higher than curative group. 5. Insignificantly the contents of phospholipid and triglyceride in the Planta-ginis Semen administration group was noted. 6. Significantly the B(a)P-induced liver toxicity was decreased in Plantagi-nis Semen administration.
