Thus far, attempts to develop drugs that target corticotropin-releasing hormone receptor 1 (CRF1R), a drug target in stress-related therapy, have been unsuccessful. Studies have focused on using high-resolution G protein-coupled receptor (GPCR) structures to develop drugs. X-ray free-electron lasers (XFELs), which prevent radiation damage and provide access to high-resolution compositions, have helped accelerate GPCR structural studies. We elucidated the crystal structure of CRF1R complexed with a BMK-I-152 antagonist at 2.75 Å using fixed-target serial femtosecond crystallography. The results revealed that two unique hydrogen bonds are present in the hydrogen bond network, the stalk region forms an alpha helix and the hydrophobic network contains an antagonist binding site. We then developed two antagonists—BMK-C203 and BMK-C205—and determined the CRF1R/BMK-C203 and CRF1R/BMK-C205 complex structures at 2.6 and 2.2 Å, respectively. BMK-C205 exerted significant antidepressant effects in mice and, thus, may be utilized to effectively identify structure-based drugs against CRF1R.

Three large scale weirs were constructed 2010 - 2011 in the Namhan river, Korea. The purpose of this study was to investigate the spatial distribution of benthic macroinvertebrates and the influence of environmental factors at the weirs 2014 - 2015. The number of species was higher in the riparian zone than in the transition or the limnetic zone. This seems to be because of the diversification of microhabitats and food sources according to the development of littoral zones. From the riparian zone to the limnetic zone, the individual abundance proportion of gathering collectors among functional feeding groups decreased, and that of filtering collectors increased. In the limnetic zone, sprawlers and climbers among habitat orientation groups decreased, and burrowers increased. This means that coarse particulate organic matter originated from land or riparian zone was transformed to fine particulate organic matter in the limnetic zone. Asian clam (Corbicula fluminea) and chironomids were dominant species based on individual abundance. Asian clam, a major taxon considering biomass, was abundant toward the limnetic zone. This is becasue of the shallow depth, suitable water current, slightly coarse substrate, and good water quality. There was no significant relationship between the water quality and the characteristics of the benthic macroinvertebrate community because the water quality was spatially not heterogenous. The more influential factors for benthic community were physical factors, especially water depth. Water depth showed a markedly significant correlation with Shannon-Weaver’s species diversity (r=-0.90), Margalef’s species richness (r=-0.82), and McNaughton’s dominance (r=0.86). Water depth showed a positive correlation (r=0.68) with the Kong and Kim BMSI (Bentic Macroinverebrates Streambed Index), and this may be related to the coarse substrate of the limnetic zone.
Mesoporous Carbon Additives for Long Cycle Life Sulfur Cathodes of Li-S Batteries

We examine the potential use of disordered mesoporous carbon as a functional additive for confining dissolved Li-polysulfides and improving the cycling performance of Li-S batteries. To promote a better understanding of the correlation between the total pore volume of disordered mesoporous carbon and the cycling performance of Li-S batteries, a series of disordered mesoporous carbons with different total pore volumes are successfully synthesized using a commercial silica template. Based on the electrochemical and structural analyses, we suggest that the total pore volume of disordered mesoporous carbon is a predominant factor in determining its capability for either the absorption or adsorption of Li-polysulfides, which is primarily responsible for enhancing the cycling performance. The addition of disordered mesoporous carbon is also effective in enhancing the homogeneous distribution of active sulfur in the cathode, thereby affecting the cycling performance.
Understanding of Electrochemical Oxidation Route of Electrically Isolated Li<sub>2</sub>S Particles

<P>New insight into the oxidation mechanism of solid Li<SUB>2</SUB>S is presented by investigating a specially designed cell in which the Li<SUB>2</SUB>S particles are electrically isolated from the carbon cathode. Surprisingly, the cell containing the isolated Li<SUB>2</SUB>S particles delivered considerable charge and discharge capacities despite the prevention of any charge transfer process between the Li<SUB>2</SUB>S particles and the carbon cathode. This fact directly indicates that the electrochemical oxidation of Li<SUB>2</SUB>S occurs not through a direct charge (electron) transfer between solid Li<SUB>2</SUB>S and conducting materials but through chemical reactions coupled with the charge transfer process. We believe that these unexpected results will greatly contribute to a deep understanding of the exact working mechanism of Li-S batteries and Li<SUB>2</SUB>S cathodes as well as a paradigm shift toward an innovative and rational design of Li-S batteries.</P>

Physical environmental factors (water depth, current velocity and substrate) and fish community were surveyed in the Gapyeong stream, Korea. The fish group of Gapyeong Stream was divided into three types. Lithophilic fish, Koreocobitis rotundicaudata and Pseudopungtungia tenuicorpa preferred shallow depth, low-velocity current, and coarse bed condition, whereas Coreoleuciscus splendidus and Microphysogobio longidorsalis were adapted to high-velocity current and bed materials. Nektonic fish, Zacco koreanus and Zacco platypus appeared in a wide range of physical conditions. Intermediate fish, Hemibarbus longirostris, Pungtungia herzi and Coreoperca herzi adapted to moderate water depths and current velocities. Among them, H. longirostris and C. herzi were adapt to various bed materials. C. splendidus, M. longidorsalis and P. herzi showed high niche overlap for current velocity, water depth and substrate with Z. koreanus and Z. platypus. The occurrence of M. longidorsalis in a relatively low-velocity current compared to Z. koreanus and Z. platypus suggests that the current velocity act as a isolation factor for these species. The competition, isolation and character displacement among these species investigated detail in the future. Based on canonical correspondence analysis, the relative importance of each environmental factor was determined as substrate > water depth > current velocity.

<P>We designed and synthesized seven α-GalCer analogues with a pyrazole moiety and varying positions of a phenyl group in the sphingosine backbone to polarize cytokine secretion. On the basis of in vitro and in vivo biological evaluations, we found that analogue <B>5</B> induced greater polarization toward Th2 and greater secretion of the immunomodulatory cytokine, IL-4, over secretion of pro-inflammatory cytokines, IFN-γ and IL-17. Treatment of a single dose of analogue <B>5</B> markedly ameliorated disease pathogenesis in an animal model of an inflammatory demyelinating disease of the central nervous system, compared to that of KRN7000 (<B>1</B>). <P>De novo analysis of protein N-terminal sequence is important for identification of N-terminal proteolytic processing such as N-terminal methionine or signal peptide removal, or for the genome annotation of uncharacterized proteins. We introduce a de novo sequencing method of protein N terminus utilizing matrix-assisted laser desorption/ionization (MALDI) signal enhancing picolinamidination with bromine isotopic tag incorporated to the N terminus. The doublet signature of bromine in the tandem mass (MS/MS) spectrum distinguished N-terminal ion series from C-terminal ion series, facilitating de novo N-terminal sequencing of protein. The dual advantage of MALDI signal enhancement by the basic picolinamidine and b-ion selection aided by Br signature is demonstrated using a variety of peptides. The N-terminal sequences of myoglobin and hemoglobin as model proteins were determined by incorporating the Br tag to the N terminus of the proteins and obtaining a series of b-ions with Br signature by MS/MS analysis after chymotryptic digestion of the tagged proteins. The N-terminal peptide was selected for MS/MS analysis from the chymotryptic digest based on the Br signature in the mass spectrum. Identification of phosphorylation site as well as N-terminal sequencing of a phosphopeptide was straightforward.</P>
