        Molecular Cloning, Expression and Characterization of Bovine UQCC and Its Association with Body Measurement Traits

        Ubiquinol-cytochrome c reductase complex chaperone (UQCC) involved in the development and maintenance of bone and cartilage is an important candidate gene for body measurement traits selection through marker-assisted selection (MAS). The expression of UQCC is upregulated in many human and animal models of height as well as other stature indexes. We have cloned the cDNA sequence coding UQCC gene in bovine. Genomic structural analysis indicated that bovine UQCC shares a high similarity with human UQCC. Furthermore, Real-Time PCR analysis showed that the expression of bovine UQCC is remarkably different in diverse tissues, including high level expression in the spleen, heart and windpipe, and relatively low expression in other tissues. We also analyzed allele frequencies in different cattle breeds and an association study on the selected SNPs. SNP DraI A2691T in intron 1 and SNP Bsh1236I A3150G in intron 8 are significantly associated with Body Length (BL), Rump Length (RL), Chest Depth (CD) and Pin Bone Width (PBW). For the A2691T SNP marker, there are significant effects on the RL (p = 0.0001), CD (p = 0.0059) and PBW (p < 0.0001) in 679 individuals; with A3150G SNP marker, there are significant effects on the BL (p = 0.0047) and CD (p = 0.0454). Regarding association analysis of combination of the two SNPs, there are significant effects on the BL (p = 0.0215), CD (p = 0.0282) and PBW (p = 0.0329) in the total population. The results suggest that the UQCC gene is a candidate gene of body measurement traits in bovine reproduction and breeding, and provide data for establishing an animal model using cattle to study big animal body type.

        Performance of Database Driven Network Applications from the User Perspective

        An understanding of performance of database driven network applications is critical in combating slow performance of e-commerce web sites, besides efficient web page design, and high quality and well-maintained web server equipment. This paper describes a method of measuring performance from the user viewpoint, which can help enormously in making realistic assessment of true performance of database driven applications. The performance measurements were taken at user locations by using several specially designed JavaScript functions along with ASP scripts. A performance study is presented in this paper, comparing performance of data access using stored procedures with the traditional way of querying a database. It is generally believed that stored procedures have performance benefits as they are pre-compiled. However, our study shows that the data access approach using stored procedures provides significant benefits (by about 30%) over the traditional approach for querying a commercial MySQL database, only when retrieving a substantial amount of data (at least 10,000 rows of data).

        Experimental Study on the Longitudinal Mechanical Behavior of Shield Tunnel in Soft-Hard Uneven Strata and the Reinforcement Effect of Longitudinal Channel Steel

        This paper investigates the longitudinal behavior and failure pattern of shield tunnel, as well as the reinforcement mechanism of longitudinal channel steel, using reduced-scale tunnel models assembled with 24 lining rings. The longitudinal behavior of the shield tunnel and the reinforcement effect are discussed in terms of vertical deformation, joint deformation, and concrete cracking. The results indicate that the shield tunnel without reinforcement longitudinallydeforms in a “bending and dislocation” mode, with the failure pattern dominated by bending deformation. In contrast, the failure pattern of the shield tunnel reinforced by channel steels is characterized by brittle shearing dislocation fracture of the circumferential joint. The load at which damage occurs in the reinforced tunnel model is 1.74 times and 2.04 times that of the non-reinforced staggered and straight jointed tunnels. The reinforcement reduces the horizontal convergence discrepancy of lining rings above the soft foundation spring and the adjacent lining rings, which improves the overall integrity of the tunnel structure.

        Enabling Efficient Verification of Dynamic Data Possession and Batch Updating in Cloud Storage

        Dynamic data possession verification is a common requirement in cloud storage systems. After the client outsources its data to the cloud, it needs to not only check the integrity of its data but also verify whether the update is executed correctly. Previous researches have proposed various schemes based on Merkle Hash Tree (MHT) and implemented some initial improvements to prevent the tree imbalance. This paper tries to take one step further: Is there still any problems remained for optimization? In this paper, we study how to raise the efficiency of data dynamics by improving the parts of query and rebalancing, using a new data structure called Rank-Based Merkle AVL Tree (RB-MAT). Furthermore, we fill the gap of verifying multiple update operations at the same time, which is the novel batch updating scheme. The experimental results show that our efficient scheme has better efficiency than those of existing methods.
