      • Relationship between Metabolic Syndrome and Bone Fracture Risk in Mid-aged Korean Women using FRAX scoring system

        Objective: The aim of this study is to investigate the relationship between metabolic syndrome (MetS) and bone fracture risk assessed by FRAX tool in mid-aged Korean women. Methods: Retrospectively, the study reviewed medical records of 1,975 female patients with or without MetS who had underwent routine medical check-ups from 2010 to 2016 University Hospital. The patients who met diagnostic criteria for metabolic syndrome based on NCEP-ATPIII were included, and each of the patients was assessed through self-report questionnaires and individual interview with a healthcare provider. The FRAX tool was used for bone fracture risk. Results: Univariate logistic regression analysis showed C-reactive protein (CRP), FRAX1, FRAX2 and calculated glomerular filtration rate (GFR) with the higher statistical significance in comparing the MetS group and its control group, while the patient’s high-risk status of bone fracture was not significantly different between the two groups. Through Pearson’s correlation coefficient study, correlation of the patient’s high risk status of bone fracture with age, serum glucose level, triglycerides (TG), HDL-cholesterol and percent body fat were found. Multivariate analysis of such variables confirmed that only the age of patients was statistically significant in relation to high risk of fracture by FRAX tool. Conclusion: MetS was not significantly associated with the patient’s high-risk status of bone fracture analyzed by using FRAX; however, the absolute value of FRAX was increased in MetS patients.

        <P><B>Abstract</B></P> <P>Mesenchymal stem cells (MSCs) are a promising therapeutic option for cell-based therapy due to their immunomodulatory and regenerative properties. They can be isolated from various adult tissues, including bone marrow, fat, dental tissue, and glandular tissue. Although they share common characteristics, little is known about the biological differences between MSC populations derived from different tissues. In this study, we used MS to compare the endogenous metabolite level in the human MSCs originating from the bone marrow, adipose tissue, periodontal ligaments, and salivary glands. Using an optimized metabolomics technique, we verified that human MSCs exhibit differences in the endogenous metabolite level depending on their source material, while the multivariate analysis showed that 5 lysophosphatidylcholines and 3 lysophosphatidylethanolamines can serve as markers for the discrimination between MSC sources and may be related to differences in their differentiation capacity.         Aspergillus oryzae로 생물전환한 감귤박의 항비만 효과

        서구화된 식습관의 변화와 편리한 생활에 따른 운동 부족, 유전적 요인, 심리적 요인 등이 복잡하게 얽히면서 비만이 꾸준히 증가하는 추세이다. 한편, 감귤류에는 carotenoid, coumarin, phenylpropanoid, limonoid 등 60여 종의 생리활성 물질이 함유되어 있으며 감귤박에는 bioflavonoid, carotenoid, hesperidin, terpenes 등이 풍부하게 함유되어 있다. 감귤박은 항산화 효과, 고혈압 예방, 혈중 LDL 콜레스테롤 함량 감소 효과 등이 있음이 보고되어 있고 최근 들어 감귤박의 비만을 억제하는 효능이 밝혀져 항비만 효과가 입증되었다. 본 연구에서는 고지방식이로 유도한 비만모델 ICR 생쥐에서 Aspergillus oryzae로 발효한 감귤박의 증강된 항비만 효과를 확인하고자 하였다. 항비만 효과를 확인하기 위해 생쥐의 체중변화와 조직의 무게변화를 관찰하였고, 그룹별 혈청 지질 농도, 간 지질농도 및 동맥경화지수를 관찰하였다. 그 결과, 고지방식이 투여군(HFD)에 비해 감귤박 추출물 투여군(CP)과 감귤박 발효 추출물 투여군(CPA)은 체중 증가량과 증체율이 감소하였고, 혈청과 간의 total cholesterol (TG), triglyceride (TC) 함량이 현저히 감소하였다. 한편, 유익한 cholesterol로 알려진 high density lipoprotein cholesterol (HDL-C) 함량은 일반식이 투여군(NFD)과 CPA 투여군이 비슷하였다. CP 투여군과 CPA 투여군의 조직 무게에서도 HFD 투여군보다 감소하여, 전체적으로 CPA 투여군에서 특히 유의적으로 감소하였다. There is a rising trend in obesity due to various factors, including changes in eating habits, lack of exercise, and genetic and psychological factors. Citrus peel has been reported to prevent obesity via antioxidative, antihypertensive, and LDL cholesterol-lowering effects. This study investigated the effects of citrus peel extract fermented with or without Aspergillus oryzae in a mouse model of diet-induced obesity. The animals were divided into four groups: a high-fat diet group (HFD), a normal fat diet (NFD) group, a citrus peel extract (CP) group, and a citrus peel extract fermented with A. oryzae (CPA) group. The citrus peel extract improved lipid metabolism and weight loss in the high-fat diet-induced obese mouse model. As expected, the body weight was higher in the HFD group compared with the NFD, CP, and CPA groups. However, the concentrations of total cholesterol (TG) and triglyceride (TC) in the serum and liver of the CP and CPA groups were lower than in the HFD group. There were no significant differences in the HDL cholesterol concentration among the groups. Taken together, our results suggest that extract of citrus peel biotransformed with A. oryzae had more antiobesity activity than citrus peel not transformed by A. oryzae through the fermentation of metabolites.

        Effects of an artificial hole on the crystal growth of large grain REBCO superconductor

        This study presents that various grain boundary junctions are prepared by controlling the seed orientation combined with an artificial hole in a melt process REBCO bulk superconductor. Large grain YBCO superconductors have been fabricated with various grain boundary junctions that the angle between the grain boundary and the <001> axis of Y123 crystal is $0^{\circ}$, $30^{\circ}$ and $45^{\circ}$, respectively. The presence of the artificial hole is beneficial for the formation of clean grain boundary junction and single peak trapped magnetic field profiles have been obtained. Artificial hole makes two growth fronts meet at a point on a periphery of the artificial hole. The presence of artificial hole is not likely to affect on the distribution of Y211 particles. The newly formed <110> facet lines are explained by the formation of new Y123/liquid interface with (010) crystallographic plane.

        Influence of phosphoric acid treatment on hydrogen adsorption behaviors of activated carbons

        The scope of this work investigates the relationship between the amount of oxygen-functional groups and hydrogen adsorption capacity with different concentrations of phosphoric acid. The amount of oxygen-functional groups of activated carbons (ACs) is characterized by X-ray photoelectron spectroscopy. The effects of chemical treatments on the pore structures of ACs are investigated by N2/77 K adsorption isotherms. The hydrogen adsorption capacity is measured by H2 isothermal adsorption at 298 K and 100 bar. In the results, the specific surface area and pore volume slightly decreased with the chemical treatments due to the pore collapsing behaviors, but the hydrogen storage capacity was increased by the oxygen-functional group characteristics of AC surfaces, resulting from enhanced electron acceptor-donor interaction at interfaces.

        Genetic analysis of endangered species Crested Ibis (Nipponia nippon) microsatellite markers

        세계적으로 멸종위기종인 따오기 (Nipponia Nippon)는 한국에서도 멸종위기종으로 구분되어 있으며, 이를 복원하기 위해 2008년 10월에 중국에서 따오기 1쌍을 도입하여 한국 최초로 인공번식에 성공하였다. 이 후 우포늪 따오기 종 복원을 통해 2017년까지 개체수가 200마리 이상으로 늘어났다. 본 연구에서는 우포늪 따오기 228 개체를 대상으로 성별을 결정하고, microsatellite 마커를 이용하여 유전적 유연관계를 분석하였다. 그 결과, 115마리의 암컷과 113마리의 수컷으로 판별되었으며, 2016년보다 2017년에 서식하고 있는 개체의 이형접합도와 다형성정보지수가 모두 감소한 것으로 나타났다. 이는 작은 집단으로부터 개체 수를 늘려나가다 보니 2017년에 근친율이 증가한 것으로 사료된다. 향 후 본 연구는 한국 따오기의 번식사업 및 복원사업을 위한 기초자료로 유용하게 활용될 것으로 기대된다. The Korean Crested ibis Nipponia Nippon is an endangered species. A pair of Crested ibis was introduced from China in October 2008, and a successful program of artificial incubation of the species, and over 200 animals have been successfully bred through the restoration project up to 2017 at Upo ibis restoration center. We assessed genetic diversity and sex determination in the Korean Crested ibis. In total, 228 Crested ibis (115 females and 113 males) were identified. And genetic diversity measures, observed heterozygosity, expected heterozygosity, and polymorphic information content values were lower in 2017 than those in 2016. The inbreeding coefficient showed that the degree of ancestry increased in 2017. The decrease in polymorphism and increase in the degree of ancestry is thought to be due to inbreeding in such a small group. In this study provided important insight into protocols for genetic management of the breeding population of Korean Crested ibis in Korea and will help in extending the restoration program.

        Layer-specific cholinergic modulation of synaptic transmission in layer 2/3 pyramidal neurons of rat visual cortex

        It is known that top-down associative inputs terminate on distal apical dendrites in layer 1 while bottom-up sensory inputs terminate on perisomatic dendrites of layer 2/3 pyramidal neurons (L2/3 PyNs) in primary sensory cortex. Since studies on synaptic transmission in layer 1 are sparse, we investigated the basic properties and cholinergic modulation of synaptic transmission in layer 1 and compared them to those in perisomatic dendrites of L2/3 PyNs of rat primary visual cortex. Using extracellular stimulations of layer 1 and layer 4, we evoked excitatory postsynaptic current/potential in synapses in distal apical dendrites (L1-EPSC/L1EPSP) and those in perisomatic dendrites (L4-EPSC/L4-EPSP), respectively. Kinetics of L1-EPSC was slower than that of L4-EPSC. L1-EPSC showed presynaptic depression while L4-EPSC was facilitating. In contrast, inhibitory postsynaptic currents showed similar paired-pulse ratio between layer 1 and layer 4 stimulations with depression only at 100 Hz. Cholinergic stimulation induced presynaptic depression by activating muscarinic receptors in excitatory and inhibitory synapses to similar extents in both inputs. However, nicotinic stimulation enhanced excitatory synaptic transmission by ~20% in L4-EPSC. Rectification index of AMPA receptors and AMPA/NMDA ratio were similar between synapses in distal apical and perisomatic dendrites. These results provide basic properties and cholinergic modulation of synaptic transmission between distal apical and perisomatic dendrites in L2/3 PyNs of the visual cortex, which might be important for controlling information processing balance depending on attentional state.
