Purpose: This study aimed to investigate the potential role of copine-1 (CPNE1), a calcium-dependent membrane-binding protein encoded by the CPNE1 gene, in colorectal cancer (CRC). Despite previous research on the involvement of copine family members in various solid tumors, the specific role of CPNE1 in CRC remains poorly understood. Methods: We conducted clinicopathological analysis and functional studies to explore the impact of CPNE1 in human CRC. We examined the expression levels of CPNE1 in CRC patients and correlated it with invasive depth, lymph node metastasis, distant metastasis, lymphatic invasion, and TNM stage. Additionally, we performed experiments to assess the functional consequences of CPNE1 knockdown in CRC cells, including proliferation, colony formation, migration, invasion, and the expression of key regulators involved in the cell cycle and epithelial-mesenchymal transition (EMT). Furthermore, we evaluated the effects of CPNE1 knockdown on tumor growth using a xenograft mouse model. Results: High expression of CPNE1 was significantly associated with advanced tumor features in CRC patients. CPNE1 knockdown in CRC cells led to impaired abilities in proliferation, colony formation, migration, and invasion. Furthermore, CPNE1 silencing resulted in the suppression of protein expression related to the cell cycle and EMT. In the xenograft mouse model, CPNE1 knockdown inhibited tumor growth. Conclusion: CPNE1 plays a crucial role in promoting tumorigenesis and metastasis in human CRC. By regulating the cell cycle and EMT, CPNE1 influences critical cellular processes at the membrane-cytoplasm interface. These results provide valuable insights into the potential development of novel therapeutic strategies for CRC targeting CPNE1.

We report a 15-year-old male, who has had dark-brownish, irreguarly bordered patches on the left side of trunk and buttock since birth. The breasts were femine, and limping gait developed at the age of 10 years. On radiologic exarninations, polyostotic fibrous dysplasia was noticed on the both humeri, ulnae, radii, tibiae, fibulae and pelvis. And brain CT scan revealed pituitary tumor. The level of prolactin was elevated.

Using peanut agglutinin(PNA), neurarninidase, and avidin-biotin peroxidase com plex(ABC) technique, normal skin specimens, nevocellular nevi, and malignant melanomas were studied, and different PNA binding patterns between nevocellular nevi and malignant melanomas were observed. The results were as follows : 1. In normal skin, except the basement membrane, epidermis and hair follicle epithelium showed a cell membrane staining of PNA after neuraminidase pretreat ment. Sebaceous glands revealed membranous and cytoplasmic staining of PNA, but sweat ducts were not stained. 2. In nevocellular nevi, none of the nevus cells were stained with PNA aftet neuraminidase preteatment. 3. In malignant melanomas, all of the melanorna cells were stained along the cell mernbrane with PNA after neuraminidase pretreatment. Therefore, the PNA staining after neuraminidase pretreatment on paraffin embedded sections using ABC technique is considered to be a useful probe for the differentiation between malignant malanoma and nevocellular nevus. In tuberous sclerosis angiomyolipomas involving kidney and regional lymph nodes are known to be extremely rare. We report a 21-year-old female, who has had pinhead-sized reddish or brownish papules on the face and skin colored, orange peel like plaques in the lurnbosacral region. She has suffered from tenderness on the left flank since 4 years ago. Abdominal CT scan revealed a large mass in the upper portion of the left kidney and lymphadenopathy of the para-aortic lymph nodes. Biopsy specimens from the mass of the left kidney and regional lymph nodes revealed angiomyolipomas. (Kor J Dermatol 28(3): 382 367, 1990)

Muscle atrophy, a debilitating condition characterized by loss of muscle mass and strength, is a major concern in various clinical settings. Acetyl genistin (AG), a bioactive compound, was evaluated for its role in muscle cell differentiation and its potential protective effects against dexamethasone (dexa)-induced muscle atrophy. Our study demonstrated that AG significantly promoted C2C12 myotube differentiation, as evidenced by enhanced myotube width and increased fusion index. Notably, AG treatment upregulated the expression of myogenic markers, including MHC, MyoD, and MyoG. Moreover, AG displayed protective properties by attenuating dexa-induced muscle atrophy, mainly by suppressing the expression of the atrophy-related genes MAFbx and MuRF1. AG’s protective effects are mechanistically attributed to its regulation of the AMPK/FoxO-dependent signaling pathway. Our results highlighted the dual benefits of AG in fostering muscle differentiation and safeguarding against muscle atrophy, positioning it as a promising agent for muscle health and therapeutic applications.

Purpose : The purpose of this study was to examine the expression of various angiogenic factors during osteoblastic differentiation of periostealderived cells and the effects of osteogenic inductive medium of periosteal-derived cells on the proliferation of endothelial progenitor cells. Materials and methods : Periosteal-derived cells were obtained from mandibular periosteums and introduced into the cell culture. After passage 3, the cells were divided into two groups and cultured for 21 days. In one group, the cells were cultured in the DMEM supplemented with osteogenic inductive agent, including 50g/ml L-ascorbic acid 2-phosphate, 10 nM dexamethasone and 10 mM -glycerophosphate. In the other group, they were cultured in DMEM supplemented without osteogenic inductive agent. VEGF isoforms, VEGFR-1, VEGFR-2, and neuropilin-1 mRNA expression was observed. Human umbilical cord blood-derived endothelial progenitor cell proliferation was also observed. Results : The expression of VEGF isoforms was higher in osteogenic inductive medium than in non-osteogenic inductive medium. The expression of VEGFR-2 was also higher in osteogenic inductive medium than in non-osteogenic inductive medium. However, the expression of VEGFR-1 and neuropilin-1 was similar in both osteogenic inductive medium and non-osteogenic inductive medium. In addition, conditioned medium from differentiated periosteal-derived cells stimulated human umbilical cord blood-derived endothelial progenitor cell numbers compared to conditioned medium from non-differentiated periosteal-derived cells. Conclusion : These results suggest that in vitro osteoblastic differentiation of periosteal-derived cells has angiogenic capacity to support endothelial progenitor cell numbers.

The aim of this study is to understand the characteristics of indoor air pollutants and provide information about the present Indoor Air Quality(IAQ) in 104 public facilities from January 2011 to December 2012. The measured air pollutants are PM10, HCHO(formaldehyde), CO, CO2, and total airborne bacteria(TAB). And the effects of environmental factors such as temperature, humidity and construction characteristics were analyzed in relation to the measured concentrations. The results of this study showed that the mean concentration of PM10, CO2, HCHO, CO, and TAB were 55.3 /, 558.6 ppm, 31.0 /, 1.2 ppm, and 561.3 CFU/, respectively. The correlation analysis showed that CO2 and HCHO (r=0.497, p<0.01), CO2 and TAB (r=0.468, p<0.01), and PM10 and CO (r=0.342, p<0.01) were significant. The result of regression analysis was found that the influence factors associated with the concentration of PM10, HCHO, and TAB were the year of building construction, the temperature and the humidity.

Purpose: The periosteum is a well-known source of osteogenic precursor cells for tissue-engineered bone formation. However, cultured endothelial or endothelial-like cells derived from periosteum have not yet been investigated. This study focused on endothelial-like cell culture from the periosteum. Methods: Periosteal tissues were harvested from the mandible during surgical extraction of lower impacted third molars. The tissues were treated with 0.075% type I collagenase in phosphate-buffered saline (PBS) for 1 hr at $37^{\circ}C$ to release cellular fractions. The collagenase was inactivated with an equal volume of DMEM/10% fetal bovine serum (FBS) and the infranatant was centrifuged for 10 min at 2,400 rpm. The cellular pellet was filtered through a $100{\mu}m$ nylon cell strainer, and the filtered cells were centrifuged for 10 min at 2,400 rpm. The resuspended cells were plated into T25 flasks and cultured in endothelial cell basal medium (EBM)-2. Results: Among the hematopoietic markers, CD146 was more highly expressed than CD31 and CD34. The periosteal-derived cells also expressed CD90 and CD166, mesenchymal stem cell markers. Considering that the expression of CD146 was constant and that the expression of CD90 was lower at passage 5, respectively, the CD146 positive cells in passage 5 were isolated using the magnetic cell sorting (MACS) system. These CD146 sorted, periosteal-derived cells formed tube-like structures on Matrigel. The uptake of acetylated, low-density lipoprotein, labeled with 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI-Ac-LDL) was also examined in these cells. Conclusion: These results suggest that the CD146-sorted positive cells can be referred to as periosteal-derived CD146 positive endothelial-like cells. In particular, when a co-culture system with endothelial and osteoblastic cells in a three-dimensional scaffold is used, the use of periosteum as a single cell source would be strongly beneficial for bone tissue engineering.
