      • 대장균에서 재조합 Rat Guanine deaminase 유전자의 발현, 정제 및 분석

        Guanine deaminase(EC, 3.5.4.3, ;Guanine aminohydrolase, GAH, GDA) catalyzes the deamination reaction of guanine to xanthine irreversibly. The cDNA encoding rat guanine deaminase had been isolated from a λZAPII rat brain expression library using antibody generated against purified rat guanine deaminase. Toe obtain recombinant GDA and analyze the property of catalytically conserved retion, here we expressed the recombinant GDA in E.coli and showed the retention of its catalytic activity similar to native rat GDA. To make the construct carrying coding region to GDA in pGEX4T prokaryotic expression vector, the region encompassing open reading frame of pBlue-GDA was PCR amplified and subcloned into pGEX4T prokaryotic expression vector with correct reading frame of fusion carrier glutathione S-transferase. After transformation to E.coli DH5a. The bacteria carrying pGEX-GDA was grwon in the condition of IPTG induction. The fusion protein GST-GDA was purified with GSH-sepharose affinity chromatography. The purified GST-GDA was subsequently digested with biotin-conjugated thrombin, and thrombin was removed by streptavidin agarose. Then sample were treated with GSH-sepharose affinity chromatography to remove the contaminant GST and GST-GDA. The purified recombinant GDA showed 70% of specific activity relative to that of purified rat GDA. The recombinant GDA showed identical molecular weight with rat GDA in SDS-PAGE.

        폐경 전후 여성의 신체활동 및 생활습관과 골 밀도 수치와의 관련성

        To evaluate the degree of bone mineral density according to physical activity, lifestyle in pre- and postmenopausal women. The subjects were 951 premenopausal and 1,167 postmenopausal women who have examined in health promotion center of Yeungnam University Hospital in Daegu city from January, 1999 to Januaty, 2001. The test for bone mineral density. The physical activity and life style were evaluated by questionaire through interview. The results were summarized as follow. 1) The prevalence rates of reduced bone mineral density and osteoporosis in premenopausal women were 23.8% and 1.6%, respectively. However, these values were increased to 46.1% and 28.1% in postmenopausal women. 2) The regular exercise was revealed to have preventive effect for osteoporosis in pre- and postmenopausal women. Among the types of exercise, the most effective exercise was bowling and moderately effective exercise was swimming on prevention of osteoporosis, however, bare hand physical exercise was not significant effect on bone mineral density. 3) The risk for reducing bone mineral density was decreased in women who have higher educational background and income in pre- and postmenopausal. The bone mineral density was highest in white color worker and lowest in blue color worker. 4) The premenopausal women who favor meat were revealed higher bone mineral density than the women who favor vegetable. The women who have drunk none or small amount of alcohol were revealed increased bone mineral density and smoking was a risk factor for osteoporosis. These results suggested that to prevent osteoporosis, the systemized program for promoting anti-osteoporotic factor have to be developed and apply from the thirties whom have maximal bone mineral density.

      • 참깨의 栽培環境이 收量構成要素, 脂肪酸組成 및 잎마름病 發生에 미치는 影響

        This experiment was carried out to investigate the effect of environments on yield components, fatty acid composition and leaf blight caused by Corynespora cassicola in sesame. Comparing yield components with different experimental locations, weight of 1,000 grains and maturation rate of grains were lower in Boeun area than that in other areas and there were no significantly different among soil textures. The repeated cultivation of sesame, 2-6 years, increased the occurrence rate of Corynespora leaf blight and fungi density of soil more than that with cultivation of first year and crop rotation. Yield and yield components were also decreased more with the repeated cultivation than that with first year cultivation. In fatty acids of sesame seeds, concentration of oleic acid was slightly high with sesame seeds harvested in Jungweon and Boeun areas, while linoleic acid was slightly high in Jecheon area located on the northern part of Chungbuk province. Concentration of palmitic and stearic acids which are saturated fatty acids were high in loam soil, while linoleic acid which is unsaturated fatty acids was no different among soil texture. Concentration of oleic acid was decreased more in the repeated cultivation of first year cultivation. Oil content was high in Jungweon area and with loam soil, and it was decreased through the repeated cultivation of sesame.

      • 참깨의 栽이 收量構成要素, 脂肪酸組成 및 잎마름病 發生에 미치는 影響

        This experiment was carried out to investigate the effect of planting time on growth character, yield components, fatty acid composition and leaf blight caused by Corynespora cassiicola in sesame. Dry weight of plant was the highest in sowing on April 25 and it was reduced with delaying sowing dates. Growth stage which increased dry weight of plant most rapidly was maturation period and capsule weight was drastically increased on 60 to 70 days after sowing. Corynespora leaf blight was severe with delaying the sowing date. Ansan variety had the highest occurrence rate of Corynespora leaf blight in sowing on June 25, 70 days after sowing and Danbaek variety in sowing on June 10, 80 days after sowing. Period when showed rapid increase of Corynespora leaf blight was from August 10 to August 20. Yield components were decreased with delaying the sowing date and capsule number per plant was most severely decreased with planting after June 10. Weight of 1,000 grains was decreased to 0.4-0.5g in sowing on June 25 compared to 2.8g of 1,000 grain weight in sowing on April 25. Maturation rate was also reduced from 87 to 97% in sowing on April 25 to 29-32% in sowing on June 25. Yield with sowing on April 25 was 81 and 89kg/10a with Ansan and Danbaek varieties, respectively, while it was decreased to 36 and 37kg/10a with sowing on June 25. Concentration of linoleic acid was high in sowing on June 25, while concentration of oleic acid was high in sowing on both May 10 and May 25.

        This paper presents a miniaturized electromagnetic wave detector. The dimension is reduced down to 31×18×13.5 ㎣ by using common OP-AMPs for amplification and oscillation. The strength from wave radiators is indicated by a color LED; green, orange and red colors mean the low, the middle and the high level, respectively. An alarm bell rings as the wave strength becomes above high level. The detector uses 3V battery. The detector could be used for a cellular phone decoration, a necklace pendant and a key holder.

        Many school age children spend time in self-care arrangements while their parents are away from home. In this study, 133 latchkey children, their parents, and 16 teachers completed the questionnaire assessing their knowledge and perception of self-care skills. Data suggest that most children are poorly prepared to remain home alone. The methods of self-care preparation for latchkey children were discussed.

        Chlorpromazine이 백서 해마에서 전기경련 충격에 의한 MAPK 타이로신 인산화에 미치는 영향

        Object : In order to examine the interaction mechanisms of electroconvulsive shock(ECS) and antipsychotic drug at the level of molecular biology, we observed the effect of chlorpromazine pre-treatment on the activation of mitogen activated protein kinase(MAPK) induced by electroconvulsive shock(ECS) in rat hippocampus Method : Male Sprague-Dawley rats were divided into 2 groups. To the experimental group chlorpromazine(20㎎/㎏) was given intraperitoneally, and to the control distilled water was given instead. Thirty minutes later, ECS was given and the hippocampus was dissected out 2 minutes thereafter. Immunoblotting with antiphosphotyrosine antibody was carried out, and the signal intensity at 42kDa band was quantitized using densitometer. The obtained result was compared by student t-test between the experimental and the control group. The absolute amount of MAPK was measured by immunoblotting with anti-MAPK antibody. Result : The tyrosine phosphorylation of MAPK reached peak at 2 minutes after ECS. However, in the chlorpromazine pre-treated group, the peak level of MAPK tyrosine phosphorylation was significantly attenuated(t=-3.12, df=14, p=0.008) compared to the control. In contrast to this, the absolute amount of MAPK did not differ between the pretreated and the control group. Conclusion : Chlorpromazine attenuated the tyrosine phosphorylation of MAPK by ECS. This finding seems to be related to the fact that chlorpromazine pre-treatment changed the cfos expression by ECS in rat brain. Antipsychotic drug and ECS might interact at the level of MAPK signal transduction system, and this might explained the observed synergistic effect of two treatment modality.

      • mecA 양성 메티실린 내성 포도구균의 mec 조절유전자에 대한 분자역학적 연구 및 다양성 분석

        목적 : 병원 감염과 지역 감염의 원인균으로서의 중요성이 증가되고 있는 MRSA와 MRCNS의 메티실린 내성 관련 유전자에 대해 분자역학적인 조사를 시행하고 mec 조절유전자의 다양성을 분석하여 분자유전학적 연구와 감염 관리에 기본적 자료를 제공하고자 연구를 시행하였다. 방법 : 본 연구에서는 국내 동일 대학병원에서 분리된 mecA 양성 MRSA 78균주와 MRCNS 36균주를 대상으로 항균제 감수성 검사와 중합효소연쇄반응을 시행하여 표현형과 유전형의 분포를 알아보고 내성 정도와 유전형의 상관성을 분석하였다. 결론 : 최소억제농도를 측정하여 oxacillin 감수성 양상을 알아본 결과 총 90개의 MRSA 임상검체 중 3균주에서 감수성을 보였고, MRCNS 36균주는 모두 내성을 보였으며, 고도 내성을 보이는 균주일수록 다제내성을 보이고 있었다. 중합효소연쇄반응으로 mecA 유전자 유무를 분석하여 mecA 양성을 보인 MRSA 78균주와 MSCNS 36균주 만을 대상으로 mec 조절유전자에 대한 중합효소연쇄반응을 시행하여 유전형의 분포를 조사하였다. 그 결과 MRSA와 MRCNS 모두에서 mecA-Pr-mecA-mecR1-mecI형, mecA-Pr-mecA-mecR1형과 mecA-Pr-mecA-mecR-1의 5′말단형의 3가지 유전형이 공통적으로 관찰되었는데, 이외에 MRSA에서는 mecA-mecR1형, mecA-mecR1의 5′말단형이, MRCNS에서는 mecA-Pr-mecA형과 mecA형이 추가로 분리되었다. 유전형의 분포를 연도별로 분석한 결과 연도별로 우세한 유전형이 서로 다른 것을 알 수 있었다. 유전형과 내성 정도의 상관성을 분석한 결과, mecA-Pr-mecA-mecR1을 보유한 MRSA의 경우 mecA 억제유전자로 생각되는 mecI 유전자 유무에 관계없이 모두 고도 내성을 나타내어 mecI의 유무는 내성 정도와는 일정한 관련이 없었으나, MRSA와 MRCNS 모두에서 mecI와 동시에 mecR1의 3′말단이 결혼된 유전형이 저도 및 중등도 내성을 보이는 균주에서 유의하게 많이 관찰되어 mecI가 결손된 균주에서는 mecR1의 3′말단부위의 유무가 내성 정도의 결정에 중요한 역할을 할 것으로 생각하였다. 결론 : 국내에서 분리되는 메티실린 내성 포도구균의 유전형의 분포가 국외 분리주와 차이를 보이는 것으로 관찰되었다. 또한 기존의 보고와는 달리 메티실린 내성 정도와 유전형 사이에 유의한 연관성이 있는 것으로 관찰되어 mec조절유전자가 내성 발현 뿐만 아니라 내성 정도의 결정에 중요한 역할을 하는 것으로 관찰되었다. MRSA가 중요한 감염균임에도 불구하고 아직까지 MRSA의 내성기전에 대해 확실하게 규명되지 않아 치료가 더욱 어려워지고 있는 실정이므로 이에 대한 더욱 종합적인 연구와 동시에 발생과 확산을 막기 위한 노력이 필요할 것으로 생각된다. Background : High prevalence of methicillin resistance has been noticed in staphylococci which also have been recognized as important nosocomial and multi-drug resistant pathogens. In this study, we investigated the distribution of mec regulator genes and the presence of the mutation in mecIgene to reveal the resistance mechanism at molecular level. Methods : The isolates included 90 clinical isolates of MRSA, 36 MRCNS of which methicillin resistance were determined by disk diffusion test and isolated in a single hospital during 1996-1999. We performed microdilution MIC test for oxacillin resistance and Kirby-Bauer test for other antibiotics. Genotypes of mecA positive (determined by PCR) isolates (78 MRSA, 36 MRCNS) were investigated by PCR amplification of mec regulator genes such as 3' end and 5' end of mecR1. mecI, and Pr-mecA. Results : In MIC assay, all MRSA strains except 3 strains and all MRCNS strains showed oxacillin resistance. The level of resistance correlated to the multi-drug resistance. Seven genotypes were observed in mecA positive MRSA and MRCNS. Differences of predominant genotypes among years isolated and among species were observed. Deletions of the 3' end of mecR1 and the mecI were more frequently observed in the low- and borderline-level resistant MRSA and MRCNS (P(0.05). So the important role of 3' end of mecRI in determining resistance level was suggested. Conclusion : This study demonstrated that genomic distribution of methicillin-resistant staphylococci isolated in Korea was different from that of strains isolated in other countries. And important role of mec regulator genes for expression of methicillin resistance was suggested.

      • 水分과 脂肪 添加量에 따른 소시지의 乳化安定性과 品質에 미치는 影響

        In order to find out and show the maximum amount of water and fat which can be mixed and emulsified with the given meat ingredients, the chemical components and specific properties of the followings are investigated. The chemical content of pork and lard were water 74.3%, 15.2%, protein 21.3%, 6.7% , fat 4.7%, 81.3% and w/p of pork was 3.48, F/P was 0.22. Among the protein, the content of salt soluble proteins of pork was 43.2. The moisture was decreased by the increasing fat level and the fat contents were increased by the increasing of fat level so, by adding fat of 50% level, the fat content of the finished sausages will be less than 30%. But the minimum protein level was 10.5% which is above the 10% that is considered as the minimum level for the acceptable meat emulsion. The lower stability was found in the 20~40% of fat levels in the formulations that consists of higher levels of fat and added water.
