        Melatonin protects endothelial progenitor L255cells against AGE-induced apoptosis via autophagy flux stimulation and promotes wound healing in diabetic mice

        Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.

        Defect Detection of Steel Wire Rope in Coal Mine Based on Improved YOLOv5 Deep Learning

        The wire rope is an indispensable production machinery in coal mines. It is the main force-bearing equipmentof the underground traction system. Accurate detection of wire rope defects and positions exerts an exceedinglycrucial role in safe production. The existing defect detection solutions exhibit some deficiencies pertaining tothe flexibility, accuracy and real-time performance of wire rope defect detection. To solve the aforementionedproblems, this study utilizes the camera to sample the wire rope before the well entry, and proposes an objectbased on YOLOv5. The surface small-defect detection model realizes the accurate detection of small defectsoutside the wire rope. The transfer learning method is also introduced to enhance the model accuracy of smallsample training. Herein, the enhanced YOLOv5 algorithm effectively enhances the accuracy of target detectionand solves the defect detection problem of wire rope utilized in mine, and somewhat avoids accidentsoccasioned by wire rope damage. After a large number of experiments, it is revealed that in the task of wirerope defect detection, the average correctness rate and the average accuracy rate of the model are significantlyenhanced with those before the modification, and that the detection speed can be maintained at a real-time level.

        Spinal Angiolipomas : Clinical Characteristics, Surgical Strategies and Prognosis

        Objective : Angiolipomas are usually found in the subcutaneous tissue of the trunk and limbs. Spinal angiolipomas (SALs) are uncommon and have rarely been reported. We report a series of nine SALs patients who received surgical treatment in our department. To summarize the clinical characteristics of SALs, propose our surgical strategies, and evaluate the effects of the operation. Methods : This was a retrospective review of nine SALs patients who received surgical treatment from August 2015 to March 2020. Total or subtotal resection was determined by the axial localization (dorsal or ventral) and the extent of intervertebral foramen involvement. The outcomes were assessed based on the modified Japanese Orthopaedic Association (mJOA) scoring system utilized before surgery and at various follow-up points. Results : Among the nine patients, the mean mJOA score before surgery was 6.6±2.3, compared with 10.1±1.1 at the last follow-up time point (33.4±11.8 months). All patients achieved good outcomes, even in cases of subtotal resection. Conclusion : Early surgical resection of SALs is recommended, and the specific procedures should be determined by the axial localization (dorsal or ventral) and the extent of intervertebral foramen involvement. Most of the patients had a good prognosis, even in cases of subtotal resection.

      • Profit Distribution Method for Innovative Customer Based on Linguistic Consistency

        Rewarding the innovative customers in collaborative innovation can increase the initiative of customer involvement and improve the performance of product innovation. Therefore, a method of customer profit distribution based on contribution degree was proposed. Firstly, customer contribution evaluation index system was established from knowledge perspective. Secondly, customer contribution evaluation method based on group linguistic consistency was put forward to determine the contribution degree, and customer profit was distributed according to the contribution degree. Two-tuple linguistic consistency was used to integrate the multi-attribute evaluation information. Finally, the feasibility and effectiveness of the proposed method was verified.

        Structural and Magnetic Properties of the Gas Atomized Mn-Al Alloy Powders

        Ferromagnetic Mn-Al powders were produced by a gas-atomization method followed by heating treatment. The gas-atomized powders were ε-phase, which is a high temperature phase in the Mn-Al system. The ε-τ phase transformation took place by subsequent heat treatment at temperatures from 500 to 700 °C. The gas-atomized powders with a smaller particle size formed the τ-phase faster and thus exhibited better magnetic properties. On the other hand, the annealing temperature and time similarly played important roles in determining the magnetic properties of the products. The Mn-Al powders of 25-38 μm annealed at 550 °C for 120 min exhibited a high coercivity of 3.2 kOe with a remanence of 38 emu/g.

        Numerical study on the gaseous radioactive pollutant dispersion in urban area from the upstream wind: Impact of the urban morphology

        The radioactive pollutant could migrate to the downstream urban area under the action of atmospheric dispersion due to the turbulent mixing under actual pollution accidents. A scenario in which radioactive contaminants from the upstream (for example, a nearshore nuclear power plant accident) migrates to the downstream urban blocks have been considered in this study. Numerical simulations using computational fluid dynamics (CFD) are then conducted to investigate the effects of the urban morphology (building packing density and layout) on the atmospheric dispersion of radioactive pollutants in this scenario. The building packing density and structure can significantly affect urban areas’ mean flow pattern and the turbulent kinetic energy (TKE). The flow pattern and the TKE distribution influence the radioactive pollution dispersion. It is found that the radioactive pollution at the urban canyons is significantly affected by the vertical transport at the canyon. A comparison of the distributions of radioactive and traditional non-radioactive pollutants is also provided

        Nitro-oleic Acid Decreases Transcription of the Angiotensin II Type I Receptor Gene in Aortic Smooth Muscle Cells

        Nitroalkene derivatives of nitro-oleic acid(OA-NO2) regulate pluripotent cell signaling in vivo underphysiological and pathological conditions. Angiotensin IItype 1 receptor (AT1R) plays an important role in thecardiovascular system. In this study, OA-NO2 reduced theAT1R mRNA level, specifically in primary smooth musclecells, showing a 70% reduction in rat smooth muscle cells(RASMCs) and a 50% reduction in pig smooth musclecells (PASMCs). These effects were not observed in CHOcells, which highly express AT1R. The AT1R mRNA decayrate was unchanged after OA-NO2 compared with OAtreatment. Nitric oxide (NO) and peroxisome proliferatoractivatedreceptor gamma (PPARγ) did not alter thereduced effects of OA-NO2 on the AT1R mRNA level inSMCs. However, Sp1-mediated activation of the AT1Rpromoter was reduced in response to OA-NO2 in RASMCsbut not 293T cells. In addition, the nuclear factor-kappa B(NF-κB) pathway was involved in the OA-NO2-mediateddownregulation of AT1R transcription in SMCs. Takentogether, our results demonstrate that OA-NO2 specificallyinhibits AT1R mRNA expression in primary smooth musclecells via the NF-κB pathway.

        Small molecule natural compound agonist of SIRT3 as a therapeutic target for the treatment of intervertebral disc degeneration

        Oxidative stress-induced mitochondrial dysfunction is implicated in the pathogenesis of intervertebral disc degeneration (IVDD). Sirtuin 3 (SIRT3), a sirtuin family protein located in mitochondria, is essential for mitochondrial homeostasis; however, the role of SIRT3 in the process of IVDD has remained elusive. Here, we explored the expression of SIRT3 in IVDD in vivo and in vitro; we also explored the role of SIRT3 in senescence, apoptosis, and mitochondrial homeostasis under oxidative stress. We subsequently activated SIRT3 using honokiol to evaluate its therapeutic potential for IVDD. We assessed SIRT3 expression in degenerative nucleus pulposus (NP) tissues and oxidative stressinduced nucleus pulposus cells (NPCs). SIRT3 was knocked down by lentivirus and activated by honokiol to determine its role in oxidative stress-induced NPCs. The mechanism by which honokiol affected SIRT3 regulation was investigated in vitro, and the therapeutic potential of honokiol was assessed in vitro and in vivo. We found that the expression of SIRT3 decreased with IVDD, and SIRT3 knockdown reduced the tolerance of NPCs to oxidative stress. Honokiol (10 μM) improved the viability of NPCs under oxidative stress and promoted their properties of anti-oxidation, mitochondrial dynamics and mitophagy in a SIRT3-dependent manner. Furthermore, honokiol activated SIRT3 through the AMPKPGC- 1α signaling pathway. Moreover, honokiol treatment ameliorated IVDD in rats. Our study indicated that SIRT3 is involved in IVDD and showed the potential of the SIRT3 agonist honokiol for the treatment of IVDD.

        Pyramid-like CdS nanoparticles grown on porous TiO<sub>2</sub> monolith: An advanced photocatalyst for H<sub>2</sub> production

        <P><B>Abstract</B></P> <P>Efficient production of H<SUB>2</SUB> via solar-light-driven water splitting by a semiconductor-based photocatalyst without noble metals is crucial owing to increasingly severe global energy and environmental issues. However, many challenges, including the low efficiency of H<SUB>2</SUB> evolution, low solar light absorption, excited electron–hole pair recombination, and slow transport of photoexcited carriers, must be resolved to enhance the H<SUB>2</SUB> photoproduction efficiency and photocatalyst stability. Here, a two-step method is used to synthesize advanced H<SUB>2</SUB>-generating photocatalysts consisting of pyramid-like CdS nanoparticles grown on a porous TiO<SUB>2</SUB> monolith, which show promising photocatalytic activity for the hydrogen evolution reaction. Furthermore, the stability of the photocatalysts is examined through long-term tests to verify their good durability. Without noble metals as cocatalysts, the photocatalyst can reach a high H<SUB>2</SUB> production rate of 1048.7μmolh<SUP>−1</SUP> g<SUP>−1</SUP> under UV–vis irradiation when the ratio of the CdS nanoparticles to TiO<SUB>2</SUB> is 5mol%. This unusual photocatalytic activity arises from the wide-region light adsorption due to the narrow band gap of CdS, effective separation of electrons and holes due to conduction band alignment at the CdS–TiO<SUB>2</SUB> interface, and favorable reaction sites resulting from the porous structure.</P>
