Broad-specificity amino acid racemase, a novel non-antibiotic selectable marker for transgenic plants

        Yi‑Chia Kuan,Venkatesan Thiruvengadam,Jia‑Shin Lin,Jia‑Hsin Liu,Tsan‑Jan Chen,Hsin‑Mao Wu,Wen‑Ching Wang,Liang‑Jwu Chen 한국식물생명공학회 2018 Plant biotechnology reports Vol.12 No.1

The broad-specificity amino acid racemase (Bsar) from Pseudomonas putida catalyzes the racemization of various amino acids, offering a flexible and feasible platform to develop a new non-antibiotic selectable marker system for plant transformation. In the present study, we demonstrated that a Bsar variant, Bsar-R174K, that is useful as a selectable marker gene in Arabidopsis and rice that were susceptible to l-lysine and D-alanine. The introduction of wild-type Bsar, Bsar-R174K or Bsar-R174A into E. coli lysine or asparagine auxotrophs was able to rescue the growth of these microorganisms in minimal media supplemented with selectable amino acid enantiomers. The transformation of Arabidopsis with Bsar or Bsar variants based on d-alanine selection revealed that Bsar-R174K had the greatest efficiency (2.40%), superior to kanamycin selectionbased transformation (1.10%). Whereas, l-lysine-based selection exhibited lower efficiency for Bsar-R174K (0.17%). The progenies of selected Bsar-R174K transgenic Arabidopsis revealed normal growth properties. In addition, Bsar-R174K transgenic rice was obtained on l-lysine medium with an efficiency of 0.9%, and the progenies of the transgenic rice revealed morphologically normal phenotypes comparable with their wild-type counterparts. This study presents the first report of broad range amino acid racemase Bsar-R174K as a non-antibiotic selectable marker system applied in transgenic plants.

      • Clinical Application of Endoscopic Inguinal Lymph Node Resection after Lipolysis and Liposuction for Vulvar Cancer

        Wu, Qiang,Zhao, Yi-Bing,Sun, Zhi-Hua,Ni, Jing,Wu, Yu-Zhong,Shao, Heng-Hua,Qu, Jun-Wei,Huang, Xin-En Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.12

Aim: To examine lymph nodes obtained after lipolysis and liposuction of subcutaneous fat of the inguinal region of female vulvar cancer patients to explore the feasibility of clinical application. Methods: The field of operation was on the basis of the range of the conventional resection of inguinal lymph nodes. We injected lipolysis liquid fanwise, started liposuction after 15-20 minutes; then the subcutaneous fatty tissue was sucked out clearly by suction tube. We selected the first puncture holes located on 2-3 cm part below anterior superior spine, the others respectively being located 3cm and 6cm below the first for puncturing into the skin, imbedding a trocar to intorduce $CO_2$ gas and the specular body, and excise the lymph nodes by ultrasonic scalpel. The surgical field chamber was set with negative pressure drainage and was pressured with a soft saline bag after surgery. Results: A lacuna emerged from subcutaneous of the inguinal region after lipolysis and liposuction, with a wide fascia easily exposed at the bottom where lymph nodes could be readily excised. The number of lymph nodes of ten patients excised within the inguinal region on each side was 4-18. The excised average number of lymph nodes was 11 when we had mature technology. Conclusion: Most of adipose tissue was removed after lipolysis and liposuction of subcutaneous tissue of inguinal region, so that the included lymph nodes were exposed and easy to excise by endoscope. This surgery avoided the large incision of regular surgery of inguinal region, the results indicating that this approach is feasible and safe for used as an alternative technology.

      • The compression-shear properties of small-size seismic isolation rubber bearings for bridges

        Wu, Yi-feng,Wang, Hao,Sha, Ben,Zhang, Rui-jun,Li, Ai-qun Techno-Press 2018 Structural monitoring and maintenance Vol.5 No.1

Taking three types of bridge bearings with diameter being 100 mm as examples, the theoretical analysis, the experimental research as well as the numerical simulation of these bearings is conducted. Since the normal compression and shear machines cannot be applied to the small-size bearings, an improved equipment to test the properties of these bearings is proposed and fabricated. Besides, the simulation of the bearings is conducted based on the explicit finite element software ANSYS/LS-DYNA, and some parameters of the bearings are modified in the finite element model to reduce the computation cost effectively. Results show that all the research methods are capable of revealing the fundamental properties of the small-size bearings, and a combined use of these methods can better catch both the integral properties and the inner detailed mechanical behaviors of the bearings.

In vitro Evaluation of the Thermosensitive and Magnetic Nanoparticles for the Controlled Drug Delivery of Vitamin D3

        Yi-Hsin Lien,Jhaol-Huei Wu,Jiunn-Wang Liao,Tzong-Ming Wu 한국고분자학회 2013 Macromolecular Research Vol.21 No.5

Thermosensitive and magnetic nanoparticles as “smart” drug carriers have shown great potential in the field of controlled drug delivery owing to their unique properties. Previously, poly(N-isopropylacrylamide)-coated silica/magnetic nanoparticles (PNIPAM/SiO2/Fe3O4 nanoparticles) were synthesized using SiO2-coated Fe3O4 nanoparticles as a template. The properties of these nanoparticles such as the transition of the lower critical solution temperature (LCST), biocompatibility, drug loading efficiency, and drug release kinetics were investigated in vitro for both targeted and controlled drug delivery. The release profile and the in vitro cancer cell inhibition activity of vitamin D3 loading for PNIPAM/SiO2/Fe3O4 nanoparticles were systemically studied. The loading and release behavior of vitamin D3 were found to be dependent on the LCST of PNIPAM/SiO2/Fe3O4 nanoparticles. HepG2 liver cancer cells were incubated with PNIPAM/SiO2/Fe3O4 nanoparticles loaded with vitamin D3 for 5 days, and cell viability significantly decreased, as observed by 3-(4,5-cimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) and lactate dehydrogenase (LDH) assays, which was further confirmed by transmission electron microscopy (TEM)images. In conclusion, the current study demonstrated that PNIPAM/SiO2/Fe3O4 nanoparticles can be used as potential drug delivery systems for controlled release.

      • Object Tracking Benchmark

Wu, Yi,Lim, Jongwoo,Yang, Ming-Hsuan IEEE 2015 IEEE transactions on pattern analysis and machine Vol.37 No.9

<P>Object tracking has been one of the most important and active research areas in the field of computer vision. A large number of tracking algorithms have been proposed in recent years with demonstrated success. However, the set of sequences used for evaluation is often not sufficient or is sometimes biased for certain types of algorithms. Many datasets do not have common ground-truth object positions or extents, and this makes comparisons among the reported quantitative results difficult. In addition, the initial conditions or parameters of the evaluated tracking algorithms are not the same, and thus, the quantitative results reported in literature are incomparable or sometimes contradictory. To address these issues, we carry out an extensive evaluation of the state-of-the-art online object-tracking algorithms with various evaluation criteria to understand how these methods perform within the same framework. In this work, we first construct a large dataset with ground-truth object positions and extents for tracking and introduce the sequence attributes for the performance analysis. Second, we integrate most of the publicly available trackers into one code library with uniform input and output formats to facilitate large-scale performance evaluation. Third, we extensively evaluate the performance of 31 algorithms on 100 sequences with different initialization settings. By analyzing the quantitative results, we identify effective approaches for robust tracking and provide potential future research directions in this field.</P>

      • CNS Efficacy of Osimertinib in Patients With T790M-Positive Advanced Non-Small-Cell Lung Cancer: Data From a Randomized Phase III Trial (AURA3)

Wu, Yi-Long,Ahn, Myung-Ju,Garassino, Marina Chiara American Society of Clinical Oncology 2018 Journal of clinical oncology Vol.36 No.26

<P>PurposeIn patients with epidermal growth factor receptor (EGFR) mutation-positive advanced non-small-cell lung cancer (NSCLC), there is an unmet need for EGFR-tyrosine kinase inhibitors with improved CNS penetration and activity against CNS metastases, either at initial diagnosis or time of progression. We report the first comparative evidence of osimertinib CNS efficacy versus platinum-pemetrexed from a phase III study (AURA3; ClinicalTrials.gov identifier: NCT02151981) in patients with EGFR T790M-positive advanced NSCLC who experience disease progression with prior EGFR-tyrosine kinase inhibitor treatment.MethodsPatients with asymptomatic, stable CNS metastases were eligible for enrollment and were randomly assigned 2:1 to osimertinib 80 mg once daily or platinum-pemetrexed. A preplanned subgroup analysis was conducted in patients with measurable and/or nonmeasurable CNS lesions on baseline brain scan by blinded independent central neuroradiological review. The CNS evaluable for response set included only patients with one or more measurable CNS lesions. The primary objective for this analysis was CNS objective response rate (ORR).ResultsOf 419 patients randomly assigned to treatment, 116 had measurable and/or nonmeasurable CNS lesions, including 46 patients with measurable CNS lesions. At data cutoff (April 15, 2016), CNS ORR in patients with one or more measurable CNS lesions was 70% (21 of 30; 95% CI, 51% to 85%) with osimertinib and 31% (5 of 16; 95% CI, 11% to 59%) with platinum-pemetrexed (odds ratio, 5.13; 95% CI, 1.44 to 20.64; P = .015); the ORR was 40% (30 of 75; 95% CI, 29% to 52%) and 17% (7 of 41; 95% CI, 7% to 32%), respectively, in patients with measurable and/or nonmeasurable CNS lesions (odds ratio, 3.24; 95% CI, 1.33 to 8.81; P = .014). Median CNS duration of response in patients with measurable and/or nonmeasurable CNS lesions was 8.9 months (95% CI, 4.3 months to not calculable) for osimertinib and 5.7 months (95% CI, 4.4 to 5.7 months) for platinum-pemetrexed; median CNS progression-free survival was 11.7 months and 5.6 months, respectively (hazard ratio, 0.32; 95% CI, 0.15 to 0.69; P = .004).ConclusionOsimertinib demonstrated superior CNS efficacy versus platinum-pemetrexed in T790M-positive advanced NSCLC.</P>

Synthesis of mesoporous SiO2 xerogel/chitosan mixed-matrix membranes for butanol dehydration

        Yi-Feng Lin,Chang-Yu Wu,Ting-Yu Liu,Kun-Yi Andrew Lin,Kuo-Lun Tung,Tsair-Wang Chung 한국공업화학회 2018 Journal of Industrial and Engineering Chemistry Vol.57 No.-

Novel mesoporous SiO2 xerogel/chitosan (CS) mixed-matrix membranes (MMMs) were successfully prepared by incorporating sol–gel synthesized SiO2 xerogels into glutaraldehyde (GA) cross-linked CS membranes. The effects of the SiO2 doping amount and the feed temperature on the performance of the separation of butanol and water was also investigated in this study. The 0.25 wt% SiO2 xerogel/CS MMMs possessed the best pervaporation performance and the largest PSI value of 1102 kg/m2h with a permeate flux of 736 g/m2h (Permeability of 3.25 × 10−5 g m m−2 h −1 Pa−1) and a separation factor of 1498 at a feed temperature of 50 °C. This PSI value is not only comparable but also exceeds that of other membranes reported in the literature. The separation factor and the flux of the SiO2 xerogel/CS MMMs in this study clearly exceed the upper limit of the previously reported MMMs. As a result, the as-prepared novel SiO2 xerogel/CS MMMs showed an improved performance during the butanol and water separation process, which raises the possibility of future pervaporation applications using this novel SiO2 xerogel/CS MMMs.

Asymptotic normality and mean consistency for the weighted estimator in nonparametric regression models

Yi Wu,Xuejun Wang,성수학 한국통계학회 2019 Journal of the Korean Statistical Society Vol.48 No.3

In this paper, we mainly study the asymptotic properties of weighted estimator for the nonparametric regression model based on linearly negative quadrant dependent (LNQD, for short) errors. We obtain the rate of uniformly asymptotic normality of the weighted estimator which is nearly O(n−1/4) when the moment condition is appropriate. The results generalize the corresponding ones of Yang (2003) from NA samples to LNQD samples and improve or extend the corresponding one of Li et al. (2012) for LNQD samples. Moreover, we obtain some results on mean consistency, uniformly mean consistency, and the rate of mean consistency for the weighted estimator. Finally we carry out some simulations to verify the validity of our results.
