      • Application of potentiometric method using a cell without liquid junction to underway pH measurements in surface seawater

        This study evaluated a method to carry out underway pH measurements of surface seawater by means of a cell without liquid junction using glass electrodes for hydrogen and sodium ions as follows:Glass-electrode-Na<SUP>+</SUP>Test solution (reference solution)H<SUP>+</SUP>-glass-electrodeThe Pitzer method was used as the theoretical basis for the calibration of the above cell and calculations of the pH in total hydrogen concentration scale. It was shown that abovementioned cell has long-term stability and can be calibrated only once a day. A buffer solution, TRIS-HCl-TRIS-NaCl-H<SUB>2</SUB>O, is recommended for calibration. This approach has been applied to a cruise in the Sea of Okhotsk on the R/V Akademik M.A.Lavrentyev during September and October 2007.

        On Board Comparison of Total Hydrogen Ion Concentration (pH) and Total Alkalinity Measurements in Seawater

        해수 중의 수소이온농도(pH)와 총알칼리도(TA) 측정 방법에 대한 선상 비교 연구가 동해 표층에서 저층에 이르는 약 550개의 실제 해수 시료에 대해 이루어졌다. 분광광도법과 전위차법에 대한 pH의 비교가 이루어졌으며, 두 방법에 의한 해수의 pH는 전반적으로 잘 일치하였으나, pH가 낮은 심층의 경우 피펫을 사용한 분광법에 의한 값이 전위차법 보다 높은 값을 보였다. 그러나 피펫을 사용하지 않은 분광법과는 두 방법의 정밀도 내에서 동일한 값을 보였다. 이는 피펫에 의해 해수 내의 이산화탄소가 제거되면서 시료 내의 pH를 증가시키는 것으로 판단된다. TA 측정법의 비교를 위해 폐쇄 용기를 이용한 전위차법과 개방 용기를 이용한 전위차법이 사용되었으며, 개방형 용기를 사용했을 때의 값이 폐쇄형 용기를 사용했을 때의 값에 비해 약 5~10 ${\mu}mol\;kg^{-1}$ 작은 값을 보인다. 이는 폐쇄형 용기를 사용했을 때, 적정 도중에 발생하는 이산화탄소가 용액 속에 남아 적정액인 염산 이외의 산을 용액 속에 추가하는 효과를 내기 때문으로 판단된다. 따라서 pH혹은 TA의 분석에 있어서 본 연구에서 비교한 어느 방법을 사용할 수도 있지만 각각의 방법에 대해 특별히 요구되는 주의를 기울일 때 만족할 만한 분석결과를 얻을 수 있다. On board comparison of pH and total alkalinity measurement in seawater was carried out during the JES expedition on R/V Roger Revelle in the East Sea using 550 real seawater samples from the surface to the bottom. Spectrophotometry and potentiometry without liquid junction were compared for pH measurement. The pH values of two methods are generally in a good agreement. Spectrophotometry with a pipette provides higher value compare with the potentiometry in the deep layer, where pH values are lower. However, spectrophotometry without a pipette shows same values with potentiometry within their precision range. The pipetting procedure may remove of $CO_2$ in the sample, which causes increase of pH. Potentiometric titration methods using a closed-cell and an open-cell were compared for the total alkalinity measurement. Values from open cell method are smaller by about 5~10 ${\mu}mol\;kg^{-1}$ than those from closed cell method. This may be caused by the bubble formed in the closed cell during the experiment. Although any analytical method compared in this study for the pH or TA measurement can be applied, special attentions should be paid for satisfactory results.

        Seasonal Variability of Polycyclic Aromatic Hydrocarbons (PAHs) in a Coastal Marine Area in the Northwestern Region of the Sea of Japan/East Sea (Possiet Bay)

        Contamination by persistent organic pollutants including polycyclic aromatic hydrocarbons (PAHs) constitutes a major concern in relation to aquatic ecosystems. To study the seasonal variability of the vertical distribution of PAHs in Possiet Bay (the Sea of Japan/East Sea), surface and near-bottom water samples were collected in February, May, July and November 2013. In the surface layer, the total concentration of 13 PAHs ranges from 11 to 29.6 ng/L over the year with a peak in February. Near-bottom PAHs concentrations varied from 13 to 29.5 ng/L with a peak in May. Seasonal changes in the suspended particulate matter with sorbed PAHs (PPAHs) distribution were detected. Surface–bottom depletion of PPAHs was observed in winter. For spring and summer, the higher near-bottom PPAHs content in comparison with surface PPAHs is attributed to resuspension of particles with PAHs from the sediment in storm events. In autumn, there were similar PPAHs concentrations in the surface and near-bottom layers as a result of the strong mixing typical for the study area at that time. The PAHs isomer ratio indicates both petrogenic and pyrogenic PAHs sources in winter and spring, whereas in summer and autumn, PAHs are derived only from combustion. The partitioning of PAHs is also influenced by seasonality. These findings are useful for understanding the dynamics of PAHs pollution in a coastal marine area of the Sea of Japan (East Sea).

        Identification of a mutant from Arachis veigae with enhanced seed oleic and very long‑chain fatty acid content

        High oleate is an important seed quality trait frequently incorporated in peanut varieties. Crop wild relatives (CWR) are potentially useful genetic resources for cultivar improvement through genetic introgression; but for wild peanut species, many chemical or nutritional traits are not well characterized. A mutant from Arachis veigae S. H. Santana & Valls (2n = 2x = 20), with increased oleic and very long chain (C ≥ 22) fatty acid content was identified from screening 209 accessions of 45 species using gas chromatography (GC). The A. veigae (formerly A. sylvestris) accession, VVeSv 8373 (PI 688970) contained 55.5% oleic acid in seeds, significantly higher than the average (18.3%) of other accessions within the same species and also significantly higher than the average (37.0%) of all wild peanut accessions evaluated. A C37T substitution was identified by sequencing the coding region of FAD2H, resulting in the nonsense mutation of Q13* (a premature stop codon). This functional mutation may significantly reduce the fatty acid desaturase (FAD) activity and result in the enhanced oleate level. Arachis veigae also contained a high percentage of very long-chain (C ≥ 22) fatty acids, and their variation identified in this study is also discussed and compared with other species. The mutant with such an altered fatty acid composition may be useful for potentially improving seed or food nutrition quality.
