        Analysis of nifH Gene Diversity in Red Soil Amended with Manure in Jiangxi, South China

        In order to understand the community structure of diazotrophs in red soil and effects of organic manure application on the structure, four nifH gene libraries were constructed: the control (CK), low manure (LM), high manure (HM), and high manure adding lime (ML). Totally 150 nifH gene clones were screened and grouped into 21 clusters by RFLP analysis. Existence of dominant patterns was observed in all libraries, which counted for over 96% of clones in library HM and about 56~72% in other three libraries. The nifH sequences of the dominant patterns in all libraries were most similar to sequences of the cyanobacteria. nifH genes showed high diversity in red soil, dispersing throughout the nifH clades (alpha-, beta-, and gamma- Proteobacteria, Firmicutes, cyanobacteria, Verrucomicrobia, and posited group). Bradyrhizobium and Burkholderia were also important diazotrophs in low fertility soil samples. Low manure treatment increased the diversity of nifH genes compared with CK and high manure treatments. Manure and lime treatment led to obvious community succession. Total N to available P ratio, total carbon, and K concentrations were the main factors affecting the diversity of diazotrophs in red soil.

        Continuous Conversion of Rice Starch Hydrolysate to 2-keto-D-gluconic Acid by Arthrobacter globiformis C224

        A continuous conversion process of rice starch hydrolysate to 2-keto-D-gluconic acid (2KGA) by Arthrobacter globiformis C224 was developed. Its feasibility for industrial application was also evaluated. Results showed that the initial cell concentration exceeding 1.25 g/L met the continuous 2KGA production at a stable dilution rate and media composition, while the dilution rate and feeding glucose concentration had a significant effect on 2KGA production performance. The optimal operating parameters were obtained as: 0.090/h of dilution rate and 171.0 g/L of feeding glucose concentration. Under these conditions, the steady state had a produced 2KGA concentration of 124.74 g/L, average volumetric productivity of 11.23 g/L/h,and yield of 0.97 g/g. In conclusion, continuous 2KGA production by the A. globiformis C224 strain would be a superior industrial process for the production of 2KGA in terms of its high 2KGA productivity and yield.

        A study on the impact of homestay sharing platform on guests" online comment willingness

        본 논문의 연구 목적은 공유숙박 비즈니스 바탕으로 숙박 플랫폼이 세입자의 온라인 리뷰 의향에 미치는 영향을 연구하는 것이다. 기존 숙박예약 모델보다 공유숙박 중 숙박 플랫품, 집주인, 세입자 간을 공유하는 독립성이 더 명확하다. 공유숙박 플랫폼은 집주인과 세입자간의 오프라인 숙박서비스를 완료하고 거래를 실현할 수 있도록 다양한 지원 서비스를 제공하는 것은 물론, 공유숙박 플랫폼은 세입자가 집주인에게 객관적이고 적극적으로 평가하도록 장려하는 특정 조치를 파악해야 한다. 공유숙박에 대한 신용 생태를 더 잘 확립할 수 있도록 필요하다. 본 논문에서는 공유숙박 플랫폼을 사용해본적 있는 소비자들을 대상으로, 2주간의 설문 조사를 하고 SPSS24.0 프로그램을 사용하여 데이터가 분석되었다. 이 논문의 연구결과는: (1) 플랫폼 리뷰 기능의 사용 용이성, 세입자의 만족도 및 플랫폼 리뷰 인센티브가 세입자의 온라인 리뷰 의향에 긍정적인 영향을 미친다. (2) 플랫폼의 신용 메커니즘은 세입자의 만족도가 온라인 리뷰 의향에 영향을 미치는 과정에서 긍정적인 영향을 미친다. The purpose of this study is to explore the impact of home stay platform on guests" willingness to comment online under the Shared home stay business model. Shared platform of home stay facility in addition to providing a variety of support services, help the landlord to the tenant do offline accommodation services, implementation, trading, will need to take some measures to actively promote the tenant groups to the landlord, the evaluation is objective, effective and sufficient number in order to better promote the sharing credit ecological establishment of home stay facility. In this study, consumers who have used the Shared home stay platform are taken as the research objects. The survey method adopts network questionnaire survey and Likert seven subscales. The statistical software SPSS24.0 program is used to process the data. Firstly, descriptive statistical analysis was conducted, followed by validity analysis and reliability analysis. After the reliability and validity of the questionnaire were determined, correlation analysis and regression analysis were used to verify the proposed hypothesis. The research results of this study are summarized as follows :(1) the usability of platform comment function, guest satisfaction and platform reward have a positive impact on the guest online comment willingness; (2) The credit mechanism of the platform has a positive regulating effect on the process of tenant satisfaction influencing tenant comment intention.

        Ultrathin Two-Dimensional Atomic Crystals as Stable Interfacial Layer for Improvement of Lithium Metal Anode

        <P>Stable cycling of lithium metal anode is challenging due to the dendritic lithium formation and high chemical reactivity of lithium with electrolyte and nearly all the materials. Here, we demonstrate a promising novel electrode design by growing two-dimensional (2D) atomic crystal layers including hexagonal boron nitride (h-BN) and graphene directly on Cu metal current collectors. Lithium ions were able to penetrate through the point and line defects of the 2D layers during the electrochemical deposition, leading to sandwiched lithium metal between ultrathin 2D layers and Cu. The 2D layers afford an excellent interfacial protection of Li metal due to their remarkable chemical stability as well as mechanical strength and flexibility, resulting from the strong intralayer bonds and ultrathin thickness. Smooth Li metal deposition without dendritic and mossy Li formation was realized. We showed stable cycling over 50 cycles with Coulombic efficiency ∼97% in organic carbonate electrolyte with current density and areal capacity up to the practical value of 2.0 mA/cm<SUP>2</SUP>and 5.0 mAh/cm<SUP>2</SUP>, respectively, which is a significant improvement over the unprotected electrodes in the same electrolyte.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/nalefd/2014/nalefd.2014.14.issue-10/nl503125u/production/images/medium/nl-2014-03125u_0005.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl503125u'>ACS Electronic Supporting Info</A></P>

        Effect of Graphene Oxide Incorporation into Electrospun Cellulose Acetate Scaffolds on Breast Cancer Cell Culture

        Graphene-based nanomaterials have been used as biomaterials to enhance cell adhesion, growth, anddifferentiation. However, the effect of graphene materials on cancer cell behavior has not been thoroughly investigated. Herein, we have incorporated graphene oxide (GO) into cellulose acetate (CA) to develop nanofibrous scaffolds for in vitrocancer cell culture, which is a crucial step for drug screening and cancer research. The GO/CA scaffolds were seeded withbreast cancer cells and cell viability, proliferation, adhesion, infiltration, and morphology were assessed. Mechanicalcharacterization demonstrated that the mechanical properties of GO/CA scaffolds were significantly better than bare CAscaffold and improved with increasing GO content. More importantly, the in vitro cell studies showed that the cancer cells onGO/CA scaffolds had significantly higher viability and better cell adhesion and growth than bare CA. Our results confirm animportant role of GO in improving mechanical properties and cancer cell performance on GO/CA scaffolds. These resultssuggest the potential of the GO/CA scaffolds as a promising candidate for in vitro cancer models.

        MAPK4 silencing in gastric cancer drives liver metastasis by positive feedback between cancer cells and macrophages

        Liver metastasis is a major cause of death in gastric cancer patients, but the underlying mechanisms are poorly understood. Through a combination of in vivo screening and transcriptome profiling followed by quantitative RT-PCR and tissue array analyses, we found that mitogen-activated protein kinase 4 (MAPK4) downregulation in gastric cancer tissues from patients is significantly associated with liver metastasis and poor prognosis. The knockdown of MAPK4 in gastric cancer cells promotes liver metastasis in orthotopic mouse models. MAPK4 depletion in gastric cancer cells induces the secretion of macrophage migration inhibitory factor (MIF) to polarize tumor-associated macrophages (TAMs) in orthotopic xenograft tumors. Moreover, TAMs activate epithelial–mesenchymal transition of gastric cancer cells to suppress MAPK4 expression, which further increases MIF secretion to polarize TAMs. Taken together, our results suggest a previously undescribed positive feedback loop between cancer cells and macrophages mediated by MAPK4 silencing that facilitates gastric cancer liver metastasis.

      • Role of Nucleation-Promoting Factors in Mouse Early Embryo Development

        <B>Abstract</B><P>During mitosis nucleation-promoting factors (NPFs) bind to the Arp2/3 complex and activate actin assembly. JMY and WAVE2 are two critical members of the NPFs. Previous studies have demonstrated that NPFs promote multiple processes such as cell migration and cytokinesis. However, the role of NPFs in development of mammalian embryos is still unknown. Results of the present study show that the NPFs JMY and WAVE2 are critical for cytokinesis during development of mouse embryos. Both JMY and WAVE2 are expressed in mouse embryos. After injection of JMY or WAVE2 siRNA, all embryos failed to develop to the morula or blastocyst stages. Moreover, using fluorescence intensity analysis, we found that the expression of actin decreased, and multiple nuclei were observed within a single cell indicating that NPFs-induced actin reduction caused the failure of cell division. In addition, injection of JMY and WAVE2 siRNA also caused ARP2 degradation, indicating that involvement of NPFs in development of mouse embryos is mainly through regulation of ARP2/3-induced actin assembly. Taken together, these data suggested that WAVE2 and JMY are involved in development of mouse embryos, and their regulation may be through a NPFs-Arp2/3-actin pathway.</P>
