        Brucella abortus 국내 분리주의 36kDa 외막단백을 암호하는 omp2b 유전자의 염기 분석

        B. abortus 국내 분리주의 분자유전학적 성상을 규명하고 분자역학적 연구에 필요한 기초자료를 얻기위해 36kDa OMP를 암호하는 omp2b 유전자를 PCR법으로 확인하고, 그리고 omp2b 유전자의 핵염기 서열과 아미노산 서열을 결정하고 이 결과를 Brucella 표준균주 및 여러 동물에서 분리된 Brucella 분리주와 상호 비교 분석하였던 바 다음과 같은 결과를 얻었다 1. B. abortus 분리주 (7주)에 대해 omp2b primers를 이용하여 PCR을 수행 한 바 공시한 참조균주와 분리균주 모두에서 1,251bp의 특이한 DNA 절편이 각각 증폭되었다 2. omp2b 유전자의 1,089bp에 대한 염기서열과 이에 대한 362개의 아미노산 서열을 결정하였다. 분리균주의 omp2b 유전자의 염기서열을 참조균주와 비교 분석한 결과 B. abortus 2308 (biovar 1) 및 B. abortus B3196 (biovar 5)과는 99.6%~l00% 및99.5%~99.9%의 매우 높은 상품성을 나타내었고, B. abortus 45/20 (biovar 1)과는 97.1%~97.4%의 상품성을 나타내었다. 또한 362개의 아미노산 서열에 대해 분석하였던 바, 분리균주들은 B. abortustus 2308 (biovar 1) 및 B. abortus B3196 (biovar 5)과는 99.2%~l00%의 매우 높은 상품성을 보였으며, B. abortus 45/20 (biovar 1)과는 96.4%~97.2%의 상품성을 보였다. 3. 본 시험에 공시된 브루셀라균주의 omp2b 유전자서열과 여러 동물에서 분리된 브루셀라 분리주의자료를 바탕으로 phylogenetic tree를 작성 한 바, 국내 분리주는 B. abortus 2308 (biovar 1), B. abortus B3196 (blovar 5), B. abortus 45/20 (biovar 1), B. suis 1330 (biovar 1), B. melitensis 16M (biovar 1), B. canis NCTC 01854, B. neotomae 5k33과 97.3%이상의 높은 근연성을 보이며 동일한 cluster에 소속되었으며, omp2b 유전자는 매우 잘 보존됨을 알 수 있었다. omp2b gene of Brucella abortus isolates encoding 36kDa outer membrane protein (OW) was amplified by polymerase chain reaction (PCR) and the sequences of nucleotide and amino acid were determined and analyzed to obtain the basic data for molecular epidemiological features of brucellosis in Korea. In PCR using omp2b primers for the gene of 36kDa OMP, the specific signals at 1,251bp were demonstrated, The nucleotide sequences of omp2b gene of 1,089bp were sequenced by dye terminator cycle sequencing method, and 362 amino acid sequences were predicted. As compared the nucleotide sequences with those of B. abortus reference strains, the isolates showed the very high homology to the reference strains ranging 97.1%-100%. The similar results were also obtained in the analysis of the amino acid sequences. In the phylogenetic tree, the isolates clustered with the reference strains and the majority of strains originated from pig, dog, wood rat and cattle.

      • 태권도 지도자의 자질에 관한 연구

        This study was conducted to identify the most important qualities that Taekwondo masters need for their jobs. It used questionnaire among 175 of the 190 Taekwondo masters and their members from Daejeon city. 71 items suggested by the researcher were divided into seven categories by a panel of professors: (a) characteristics and attitudes, 12 items, (b) abilities and skills, 13 items, (c) knowledge, 14 items, (d) education and experience, 7 items, (e) instructional methods, 12 items, (f) interpersonal relationships, 6 items, and (g) management, 7 items. The respondents were asked to identify the most important qualities that they perceive themselves to be necessary for success on the job. The results of this study showed that the most important qualities regarding characteristics and attitudes were social trait, martial spirit, physical health. With regard to abilities and skills, the item that received the counseling sills was regarded as more important than the other items. In the instructional method category, devise instructional plans and executed them accordingly was regarded as more important than the other items. With regard to knowledge, coaching and instruction was considered to be the most important quality that masters should have. With regard to education and experience category, instructing experience was considered to be the most important quality that masters should have. The results of this study lead to the conclusion that these items should be a major concern for masters who wish to be successful on the job, and they should be aware of the need to develop their qualities accordingly.

      • 정상 성인에서 Amitriptyline이 REM 수면에 미치는 영향

        To investigate the effects of amitriptyline on REM sleep in normal adults, author applied polysomnography of sleep structure to the volunteer before and after administration of amitriptyline 25㎎. The subjects were 8 healthy young adults, aged from 20 to 24 years old. The results were summarized as follows. 1. Compaired with the baseline, the decrease of the absolute value of REM sleep, the decrease of total REM sleep period and duration, the increase of REM sleep interval occured in amitriptyline administration night. 2. Compaired with the baseline, the increase of the absolute value and the relative of REM sleep, the increase of total REM sleep duration, the decrease of REM sleep latency occured in the first recovery night. 3. Compaired with the baseline, the increase of the relative value of REM sleep, the increase of REM sleep efficiency occured in the second recovery night. 4. In amitriptyline administration night, REM sleep supression effect of amitriptyline occured and in the first and in the first and the second recovery night, rebound phenomenon of REM sleep occured. 5. REM sleep supression effect of amitriptyline occured in totally each REM period time rather in specific sequential REM period, but rebound phenomenon of REM sleep markedly occured in the first REM period in first recovery night.

        시화호의 계절변화에 따른 지화학적 환경요인 특성 연구

        Seasonal variation of biogeochemical characteristics was determined in Lake Shihwa from October 2002 to August 2003. When the lake was artificially constructed for the freshwater reservoir in 1988, the development of the strong haline density stratification resulted in two-layered system in water column and hypoxic/anoxic environment prevailed in the bottom layer due to oxidation of accumulated organic matters in the lake. Recently, seawater flux to the lake through the sluice has been increased to improve water quality in the lake since 2000, but seasonal stratification and hypoxic bottom layer of the lake still developed in the summer due to the nature of artificially enclosed lake system. As the lake is still receiving tremendous amount of organic matters and other pollutants from neighboring streams during the rainy summer season, limited seawater flux sluicing into the lake may not be enough for the physical and biogeochemical mass balance especially in the summer. The excess of accumulated organic matters in the bottom layer apparently exhausted dissolved oxygen and affected biogeochemical distributions and processes of organic and inorganic compounds in the stratified two-layered environment in the summer. During the summer, ammonia and dissolved organic carbon remarkably increased in the bottom layer due to the hypoxic/anoxic condition in the bottom layer. Phosphate also increased as the result of benthic flux from the bottom sediment. Meanwhile, dissolved organic carbon showed the highest value at the upstream area and decreased along the salinity gradient in the lake. In addition to the sources from the upstream, autochthonous origin of particulate organic carbon from algal bloom in the lake might be more important for sustaining aggravated water quality and development of deteriorated bottom environment in the summer. The removal of trace metals could be attributed to scavenging by strong insoluble metal-sulfide compounds in the hypoxic/anoxic bottom layer in the summer.

      • 잡음환경을 적용한 음성인식기 구현

        조용한 환경에서 훈련된 음성인식시스템은 여러 종류의 잡음이 섞인 실생활에서 적용될 경우 급격히 성능이 저하된다. 본 연구에서는 여러 대역별 소음을 추가한 음성으로 훈련된 모델들을 가지고서, 다양한 잡음에 대한 인식실험을 수행하여 잡음환경에 따른 인식률의 상관관계를 구하였다. 깨끗한 음성에 비해 40-45snr의 잡음을 인식엔진에 추가하였을 경우 17%의 인식률 향상을 보였다. A Speech recognition systems trained in the silent environment may seriously degrade performance at real world wit various. In this paper, we make a recognition experiment for the various noises using the model trained with a multiple range of noises and find the correlation between noise environment and recogniton rate. Comparing with clean speech, there is an impovement by 17% when we insert noise into recognition engine with 40-45snr
